영양분이 결핍된 H460 세포주에서 자가포식이 세포사멸에 미치는 영향 (The Effect of Autophagy to Cell Death in Nutrient-Deprived H460 Cells)

  • Background: Autophagy is an important adaptive mechanism in normal development and in response to changing environmental stimuli in cancer. Previous papers have reported that different types of cancer underwent autophagy to obtain amino acids as energy source of dying cells in nutrient-deprived conditions. However, whether or not autophagy in the process of lung cancer causes death or survival is controversial. Therefore in this study, we investigated whether nutrient deprivation induces autophagy in human H460 lung cancer cells. Methods: H460, lung cancer cells were incubated in RPMI 1640 medium, and the starved media, which are BME and RPMI media without serum, including 2-deoxyl-D-glucose according to time dependence. To evaluate the viability and find out the mechanism of cell death under nutrient-deprived conditions, the MTT assay and flow cytometry were done and analyzed the apoptotic and autophagic related proteins. It is also measured the development of acidic vascular organelles by acridine orange. Results: The nutrient-deprived cancer cell is relatively sensitive to cell death rather than normal nutrition. Massive cytoplasmic vacuolization was seen under nutrient-deprived conditions. Autophagic vacuoles were visible at approximately 12 h and as time ran out, vacuoles became larger and denser with the increasing number of vacuoles. In addition, the proportion of acridine orange stain-positive cells increased according to time dependence. Localization of GFP-LC3 in cytoplasm and expression of LC-3II and Beclin 1 were increased according to time dependence on nutrient-deprived cells. Conclusion: Nutrient deprivation induces cell death through autophagy in H460 lung cancer cells.

산화 스트레스에 대한 폴리아민의 세포보호 효과 (Cytoprotective Effects of Polyamines Against Oxidative Stress)

  • 폴리아민은 모든 진핵세포에서 발견되는 다가 양이온성의 저분자 물질이며 세포성장에 필수적인 것으로 알려져 있다. 본 논문에서는 폴리아민의 역할 중에서 산화적인 스트레스에 대한 세포보호 효과를 연구하였다. 쥐의 간세포주인 $Ac_2F$에 산화 스트레스를 유발하기 위하여 2,2'-azobis(2-amidinopropane)dehydrochloride (AAPH)를 처리하였을 때, 세포증식은 농도 의존적으로 감소하였다. 배지에 폴리아민을 첨가하였을 때 세포성장은 농도 의존적으로 증가하였으며 ROS 발생은 현저히 감소하였다. 폴리아민 가운데 특히 spermidine과 spermine이 뚜렷한 세포증식효과를 보였다. Spermine의 경우, $20{\mu}M$농도에서 AAPH에 의해 유도된 ROS발생을 $45\%$나 감소시켰다. 산화 스트레스에 관여하는 효소들 가운데 주된 효소인 superoxide dismutate (SOD)와 catalase (CAT)의 세포 내 단백질을 Western blotting으로 조사한 결과, AAPH는 이 두 가지 단백질의 생성을 억제한 것으로 나타났다. 그러나 spermine을 처리하였을 때 두 단백질의 생산은 모두 정상적으로 회복이 되었다. 또한 세포주기의 중요한 조절 단백질인 cyclin E 역시 AAPH에 의하여 생성이 억제되었다. 이는 AAPH에 의하여 생성된 ROS가 세포주기의 S phase의 진행을 억제한 것으로 생각된다. AAPH에 의한 cyclin E의 억제는 spermine에 의하여 정상적으로 회복되었다. 위와 같은 Spermine의 항산화 효과는 ethidium bromide와 acridine orange를 이용하여 형태학적으로도 증명되었다.

  • 이 논문은 우리나라에서 지금까지 응용한 바있는 Toxoplasma병의 진단에 관한 시험연구의 성적을 기술하였다. 가. 우리나라에서 1957년 돈으로부터 Toxoplasma 원충을 분류한 것이 최초의 보고였다. 나. Toxoplasma병의 사후진단을 하기 위하여 동물접종시험 (1957), 조직배양시험 (1961), Acridine orange 염색법(1974) 그리고 직접형광항체법(1974)을 이용하였다. 다. Toxoplasma병의 생전진단을 하기 위하여 보체결합조지반응시험(1965), 색소시험(1965), 적혈구응집반응시험(1972) 그리고 피내반응시험(1972)을 응용하였다. 라. 야외돈에 대한 항체보유율 조사성적은 보체결합조지반응시험(1965)에서 $13.4\%$ 적혈구응집반응시험(1972)에서 $25.3\%$ 그리고 피내반응시험(1972)에서 $5.3\%$였으며 점차적 증가하는 경향을 보이고 있다. 마. Toxoplasma병에 대한 방역대책의 일단으로서 본병의 신속 정확한 진단을 하기 위하여 가축위생연구소에서는 매년 생산하는 피내반응진단액은 동물검역소에 그리고 직접형광항체진단액은 각도 가축위생시험소에 공급하고 있다.

Revegetation of a Lakeside Barren Area by the Application of Plant Growth-promoting Rhizobacteria

  • The growth stimulation of wild plants by several bacterial species showing plant growth-promoting capabilities was examined in a barren lakeside area at Lake Paro, Korea. Microbial numbers and activities in the field soil were monitored for 73 days after inoculation of the bacteria. The acridine orange direct counts for the total soil bacterial populations ranged between $2.0-2.3{\times}10^{9}\;cells/g$ soil and $1.4-1.8{\times}10^{9}\;cells/g$ soil in the inoculated and uninoculated soils, respectively. The numbers of Pseudomonas spp., which is known as a typical plant growth-promoting rhizobacteria, and the total microbial activity were higher in the inoculated soil compared to those in the uninoculated soil. The average shoot and root lengths of the wild plants grown in the inoculated soil were 17.3 cm and 12.4 cm, respectively, and longer than those of 11.4 cm and 8.5 cm in the uninoculated soil. The total dry weight of the harvested wild plants was also higher in the inoculated soil (42.0 g) compared to the uninoculated soil (35.1 g). The plant growth-promoting capabilities of the inoculated bacteria may be used for the rapid revegetation of barren or disturbed land, and as biofertilizer in agriculture.

Inhibition of Cell Growth and Induction of Apoptosis by Euonymus Alatus (Thunb.) Sieb in Human Leiomyomal Smooth Muscle Cells

  • Objective : Euonymus alatus (Thunb.) Sieb (EA) is a traditional Korean herbal medicine, commonly used to treat tumors in Korea and China for centuries. Several earlier studies have indicated that EA exhibits anti-tumor properties, but its mechanism remains to be elucidated. In this study, we evaluated the molecular mechanism of EA in a human uterine leiomyomal smooth muscle cell (ULSMC) line. Methods : This study was evaluated by: (a), morphological changes by using acridine orange/ethidium bromide staining; (b), DNA fragmentation by TdT-mediated dUTP nick end labeling (TUNEL); and (c), sub-G1 cell analysis. Results : This study observed that EA treatment caused apoptotic cell death and depletion of intracellular glutathione (GSH) and that reduction of mitochondrial membrane potential was found to be involved in the initiation of apoptosis by EA. Conclusion : This results show that EA exerted clear cytotoxic effects and strongly inhibited the proliferation of ULSMC.

Canna edulis Leaf Extract-Mediated Preparation of Stabilized Silver Nanoparticles: Characterization, Antimicrobial Activity, and Toxicity Studies

  • A novel approach to synthesize silver nanoparticles (AgNPs) using leaf extract of Canna edulis Ker-Gawl. (CELE) under ambient conditions is reported here. The as-prepared AgNPs were analyzed by UV-visible spectroscopy, transmission emission microscopy, X-ray diffraction, Fourier transform-infrared spectroscopy, energy-dispersive analysis of X-ray spectroscopy, zeta potential, and dynamic light scattering. The AgNPs showed excellent antimicrobial activity against various pathogens, including bacteria and various fungi. The biocompatibility of the AgNPs was analyzed in the L929 cell line using NRU and MTT assays. Acridine orange/ethidium bromide staining was used to determine whether the AgNPs had necrotic or apoptotic effects on L929 cells. The concentration of AgNPs required for 50% inhibition of growth of mammalian cells is far more than that required for inhibition of pathogenic microorganisms. Thus, CELE is a candidate for the eco-friendly, clean, cost-effective, and nontoxic synthesis of AgNPs.

Transport of Tetraethylammonium in Renal Cortical Endosomes of Cadmium-Intoxicated Rats

  • Effects of cadmium (Cd) intoxication on renal endosomal accumulation of organic cations $(OC^+)$ were studied in rats using $^{14}C-tetraethylammnium$ (TEA) as a substrate. Cd intoxication was induced by s.c. injections of 2 mg Cd/kg/day for $2{\sim}3$ weeks. Renal cortical endosomes were isolated and the endosomal acidification (acridine orange fluorescence change) and TEA uptake (Millipore filtration technique) were assessed. The TEA uptake was an uphill transport mediated by $H^+/OC^+$ antiporter driven by the pH gradient established by $H^+-ATPase.$ In endosomes of Cd-intoxicated rats, the ATP-dependent TEA uptake was markedly attenuated due to inhibition of endosomal acidification as well as $H^+/TEA$ antiport. In kinetic analysis of $H^+/TEA$ antiport, Vmax was reduced and Km was increased in the Cd group. Inhibition of $H^+/TEA$ antiport was also observed in normal endosomes directly exposed to free Cd (but not Cd-metallothionein complex, CdMt) in vitro. These data suggest that during chronic Cd exposure, free Cd ions liberated by lysosomal degradation of CdMt in proximal tubule cells may impair the endosomal accumulation of $OC^+$ by directly inhibiting the $H^+/OC^+$ antiporter activity and indirectly by reducing the intravesicular acidification, the driving force for $H^+/OC^+$ exchange.

Lactococcus lactis subsp. cremoris ATCC 11602의 Bacteriophage 내성균주 A1의 특성에 관한 연구 (The Characteristics of Bacteriophage-resistant Lactococcus lactis subsp. cremoris ATCC 11602-A1)

  • The ppage resistance mechanism of Lactococcus lactis subsp. cremoris ATCC 11602-A1 was investigated. When parent and A1 were incubated at 30 and 40$^{\circ}C$, A1 grew well and multiplication of phage(MOI=1)on A1 slightly occurred at 40$^{\circ}C$ in contrast with parent. There was a great difference of proteolytic activity between parent and A1, irrespective of the temperature. As a result of ADS treatment oon culture broth, survival rate of A1 was 27% at the lethal concentration of parent and adsorption rate of phage was increased to 95~97%, which was considered to come from the exposure of phage receptor site masked by an unknown component. These results suggest that acridine orange (AO) treatment leads to the modification of cell wall, conferring resistance to high temperature and lytic phage. No change in plasmid profiles of A1 at 30 and 40$^{\circ}C$ were found, which suggests that plasmid is not relative to temperature-resistance of A1.

겨울철에 발생한 Holstein 송아지의 Anaplasma marginale 감염 (Anaplasma marginale infection in Holstein calves during winter)

  • We observed an outbreak of calf anaplasmosis at a farm in Chonbuk area during winter season, which was diagnosed by the hematological and serological tests. The results are as follow. On hematological observation for infected thirteen calves (ages 1 to 25 days) showed anemia with hematocrit $27.7{\pm}7.7%$, erythrocyte $6.9{\pm}1.9{\times}10^6/{\mu}l$, hemoglobin $11.3{\pm}3.2g/dl$, MCV $40.6{\pm}1.5fl$, MCH $16.3{\pm}1.6pg$ and MCHC $40.5{\pm}3.6g/dl$. Anaplasma marginale was observed in all of the calves's erythrocytes by Diff-Quick and acridine orange staining, and were reacted by ELISA.

Molecular Modeling of Small Molecules as BVDV RNA-Dependent RNA Polymerase Allosteric Inhibitors

  • Bovine viral diarrhea virus (BVDV), a major pathogen of cattle, is a well-characterized pestivirus which has been used as a good model virus for HCV. The RNA-dependent RNA polymerase (RdRp) plays a key role in the RNA replication process, thus it has been targeted for antivirus drugs. We employed two-dimensional quantitative structure-activity relationship (2D-QSAR) and molecular field analysis (MFA) to identify the molecular substructure requirements, and the particular characteristics resulted in increased inhibitory activity for the known series of compounds to act as effective BVDV inhibitors. The 2D-QSAR study provided the rationale concept for changes in the structure to have more potent analogs focused on the class of arylazoenamines, benzimidazoles, and acridine derivatives with an optimal subset of descriptors, which have significantly contributed to overall anti-BVDV activity. MFA represented the molecular patterns responsible for the actions of antiviral compound at their receptors. We conclude that the polarity and the polarizability of a molecule play a main role in the inhibitory activity of BVDV inhibitors in the QSAR modeling.
