Antioxidant Activity and Acetylcholinesterase Inhibitory Activity of Ark shell (Scapharca broughtonii) (피조개의 항산화 활성과 Acetylcholinesterase 저해 활성)

  • Ark shell (Scapharca broughtonii; Korean name, pijogae) is one of the most widely cultivated and consumed shellfishes in Korea. The purpose of this study was to evaluate the antioxidant activity and acetylcholinesterase inhibitory activity of ark shell. After preparing the methanol extract of ark shell powder, the extract was subsequently fractionated by hexane, diethyl ether, ethyl acetate and water. The antioxidant activity evaluated by DPPH radical scavenging activity, ABTS radical scavenging activity and reducing power was relatively higher in the water fraction; however, the activity was spread out in all fractions. Acetylcholinesterase inhibitory activity was the highest in the diethyl ether fraction. Taken together with the results of both antioxidant and acetylcholinesterase inhibitory activities, it can be suggested that different kinds of physiological compounds were contained in the ark shell.

Chemical Constituents and Their Acetylcholinesterase Inhibitory Activity of Underground Parts of Clematis heracleifolia (조희풀(Clematis heracleifolia) 지하부의 성분과 Acetylcholinesterase억제 활성)

  • To clarify chemical constituents of Clematis heracleifolia, isolation and structure elucidation of the underground parts of C. heracleifolia were performed. Five compounds were isolated from $CHCl_3$ and n-BuOH soluble fraction of this plant. On the basis of spectral and physico-chemical data, the structure of isolated compounds were identified as coniferyl alcohol (1), scoparone (2), (+)-lariciresinol (3), phytosterols (4), and daucosterol (5), respectively. All compounds are isolated from this plant for the first time. To evaluate anti-acetylcholinesterase activity of the isolated compounds, compounds 1, 2, 3, and 5 were tested inhibition activity against acetylcholinesterase. Among tested compounds, daucosterol (5) showed acetylcholinesterase inhibitory activity with $IC_{50}$ value of $6.1{\mu}M$.

Acetylcholinesterase Inhibitors from the Stem of Zea mays

  • Five compounds were isolated from the stem of Zea mays. Based on spectral data, they were identified as 4-hydroxybenzaldehyde (1), N-trans-p-coumaryl tyramine (2), N-trans-ferulyl tryptamine (3), N-(p-coumaryl) serotonine (4), and N-(p-coumaryl)-tryptamine (5). All isolates were evaluated in vitro for their inhibitory activity on acetylcholinesterase. Among tested compounds, compounds 2 - 5 exhibited acetylcholinesterase inhibitory activity, with $IC_{50}$ values of 125, 60.4, 183.5 and 53.3 ${\mu}M$, respectively. Compound 1 did not show acetylcholinesterase inhibitory activity in the present study.

Inhibitory Effect of Corynoline Isolated from the Aerial Parts of Corydalis incisa on the Acetylcholinesterase

  • In the course of screening Korean natural products for acetylcholinesterase (AChE) inhibitory activity, it was found that a methanolic extract of the aerial parts of Corydalis incisa (Papaveraceae) showed significant inhibitory effects on AChE. Corynoline isolated from this plant inhibited AChE activity in a dose-dependent manner, and the $IC_{50}$ value of corynoline was $30.6{\;}{\mu}M$. The AChE inhibitory activity of corynoline was reversible and noncompetitive.

Studies on the biological activity of water extract and solvent fractions of wild Grifola frondosa (야생 잎새버섯 물추출물 및 유기용매 분획물의 생리활성 탐색)

  • Samples (10 mg/mL) of wild Grifola frondosa aqueous extract and solvent fractions were examined for fibrinolytic, thrombin inhibitory, acetylcholinesterase inhibitory, and antioxidative activities to determine the biological activities. The fibrinolytic activity of the aqueous extract and solvent fractions was 0.93 and 0.73 plasmin units/mL, respectively. The thrombin inhibitory activity of the butanol extract was 79.60%. The chloroform fraction had high acetylcholinesterase inhibitory activity (85.88%). The aqueous extract had low antioxidative activity (39.81%). The aqueous fraction hydrolyzed Bβ subunits of human fibrinogen but did not show any reactivity for the γ form of the human fibrinogen. The findings indicate the potential of wild Grifola frondosa for the development of drugs and bio-functional foods to prevent cardiovascular diseases.

Antioxidant and Acetylcholinesterase Inhibitory Activities of Subcritical Water Extracts from Houttuynia cordata Thunb (어성초의 아임계수 추출물의 항산화 활성 및 Acetylcholinesterase 저해 활성)

  • Houttuynia cordata Thunb powder (0.05 g) was placed in a stainless vessel containing 10 mL of water, and subcritical water (SCW) extraction was carried out at various temperatures (50, 100, 200, and $300^{\circ}C$) for 10, 30, and 60 min. SCW treatment significantly affected physiologically important compounds, antioxidant activity, and acetylcholinesterase inhibitory activity of H. cordata Thunb extracts. H. cordata Thunb extracted at $300^{\circ}C$ for 30 min had the highest quercetin content, DPPH radical scavenging activity, total phenolic content, and acetylcholinesterase inhibitory activity, whereas the extract at $100^{\circ}C$ for 10 min possessed the highest flavonoid content. The highest ABTS radical scavenging activity was found in H. cordata Thunb extracted at 200 and $300^{\circ}C$ for 60 min. These results indicate that SCW extraction might be a useful processing method for obtaining valuable materials from H. cordata Thunb.

Nitric Oxide Production and Acetylcholinesterase Inhibitory of Activity Various Extracts from Codonopsis lanceolata by Steaming Times (증숙 더덕 용매별 추출물의 Nitric Oxide 생성 저해 효과 및 Acetylcholinesterase 저해활성)

  • Steaming is a method that has traditionally been used for medicinal plant extraction. This study investigated nitrite oxide production, ferrous ion chelating activity, α-glucosidase, xanthine oxidase, and acetylcholinesterase inhibitory activities of ethanol, acetone and hot-water extracts of Codonopsis lanceolata prepared by steaming seven times. MTT assay showed that each extract was non-toxic up to a concentration of 700 ㎍/mL confirming that there was no cytotoxicity in all extracts. The α-glucosidase, xanthine oxidase, and acetylcholinesterase inhibitory activities exhibited by the hot-water extract obtained from steaming seven times were higher (83.1%) than the other extracts. Higher production of nitrite oxide and better ferrous chelating activity was recorded with hot-water extract compared to ethanol and acetone extracts. These results indicated that more steaming of Codonopsis lanceolata extracts would be required to validate the possibility of developing antioxidants. Also, further study is needed to determine if the components present in the tested extracts might be useful in the prevention of Alzheimer's disease. These results showed that hot-water extracts may be useful for their antioxidant and the production inhibitory activity of nitrite oxide. It will be helpful in the investigation of the constituent analysis of the steam-processed product of Codonopsis lanceolata.

Production and Purification of Acetylcholinesterase Inhibitor from Pseudomonas sp960903 (Pseudomonase sp. 960903에 의한 acetylcholinesterase 억제제의 생산 및 정제)

  • To screen agent for the treat-ment of Alzhimers Disease several strains of bacteria producing acetylcholinesterase inhibitor ware isolated from soil. Strain 960903 showed strong acetylcholinesteras inhibitory activity and low butyrylcholinesterse inhibitory activity. The strain 960903 was identified as Pseudomonas sp. Acetylcholinesterase inhibitor ws highly achieved in fermentation medium containing soluble starch 3.0%, glycerol 1.0%, pharmamedia 0.5%, KCI 0.3%, $CaCO_3$ 0.2%, MgS $O_4$..$7H_2$O 0.05%, $KH_2$$PO_4$ 0.05%(pH6.5) at $30^{\circ}C$ for 4 days. Acetylcholinesterase inhibitor was purified by Diaion WA-30($OH^{-}$) column charomatography and cellulose column chromatography. Acetylcholinesterase inhibi-tor showd the maximum wavelength at 205 nm and was soluble in water, acetic acid, ethanol, methanol and dime-thyl sulfoxide. The concentration of 50% inhibition($IC_{50}$) of inhibitor against acetylcholinesterase was 25$\mu\textrm{g}$/ml. The inhibitor was inactivated on heating ar $100^{\circ}C$ fro 15 min and more stable in acidic region than alkaline region.n.

Screening of 50 Korean Herbal formulas with Inhibitory Effects on Acetylcholinesterase Activity and Amyloid-β Aggregation (다빈도 한약 처방 50종의 아세틸콜린 분해 효소 활성 및 아밀로이드 베타 단백질 응집 억제 효능 비교 연구)

  • Acetylcholinesterase (AChE) activation and amyloid-${\beta}$ ($A{\beta}$) aggregation are major biological markers of Alzheimer's disease. In the present study, we evaluated the inhibitory effects of 50 kinds of herbal formulas on AChE activity and $A{\beta}$ aggregation. Among them, Hwanglyeonhaedok-tang, Cheonwangbosim-dan, Makmundong-tang, and Gamisoyo-san had a potent effects on the inhbition of AChE activity. Sosiho-tang, Samsoeum, Cheonsimyeunjaeum, and Bunsimgieum exerted to have the inhibitory activity on $A{\beta}$ aggregation. In addition, these 8 herbal formulas showed the 3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity, indicating their antioxidant activities.

Inhibitory Effect of Trans-N-p-Coumaroyl Tryamine from the Twigs of Celtis chinensis on the Acetylcholinesterase

  • The methanolic extract of the twigs of Celtis chinensis was found to show inhibitory activity on acetylcholinesterase (AChE), an enzyme that plays a role in the metabolic hydrolysis of ACh. Bioassay-guided fractionation of the methanolic extract resulted in the isolation of N-p-coumaroyl tyramine. as an inhibitor on AChE. This compound inhibited AChE activity in a dose-dependent manner, and the $IC_50$ value of trans-N-p-coumaroyl tyramine was 34.5 $\mu$g/mL (122 $\mu$M).
