The species in genus Saxifiraga distributed in circumpolar arctic are taxonomically difficult to study. RAPD analyses were performed to compare the genetic diversity of the 16 Saxifrages originated from the Norwegian Arctic Svalbard and Korea. The 12 accessions of URP primers were tested and 4 of which showed polymorphism were selected. Total 79 (44.8%) DNA bands were scored and analyzed by UPGMA cluster analysis. The results indicated that all of the 9 Saxifraga species from Svalbard showed high genetic diversity than those from Korea. The Similarity matrix and cluster analyses indicated that the Saxifraga species from Svalbard and Korea can be divided into two different subgroups. RAPDs of the Saxifraga species of Korea showed higher homologous patterns than those of Arctic Saxifrage. Among the Saxifraga species, we found that the morphological similarity reflects the genetic similarity. The geographic distance, clonal reproduction, and environmental condition may contribute the high level of genetic diversity between Saxifraga species from the two isolated regions.

Whereas NIR spectroscopy has been applied in agriculture for more than 20 years, few studies refer to those plant substances occurring only in smaller amounts. Nevertheless there is a growing interest today to support efficiently activities in the production of high-quality medicinal and spice plants by this fast and non-invasive method. Therefore, it was the aim of this study to develop new NIR methods for the reliable prediction of secondary metabolites found as valuable substances in various plant species. First, sophisticated NIR methods were established to perform fast quality analyses of intact fennel, caraway and dill fruits deriving from single-plants [1]. Later on, a characterization of several leaf drugs and the corresponding fresh material has been successfully performed. In this context robust calibrations have been developed for dried peppermint, rosemary and sage leaves for the determination of their individual essential oil content and composition [2]. A specially adopted NIR method has been developed also for the analysis of carnosic acid in the leaves of numerous rosemary and sage gene bank accessions. Carnosic acid is an antioxidative substance for which several health promoting properties including cancer preservation are assumed. Also some other calibrations have been developed for non-volatile substances such as aspalathin (in unfermented rooibos leaves), catechins (in green tea) and echinacoside (in different Echinacea species) [3]. Some NIR analyses have also been successfully performed on fresh material, too. In spite of the fact that these measurements showed less accuracy in comparison to dried samples, the calibration equations are precise enough to register the individual plant ontogenesis and genetic background. Based on the information received, the farmers and breeders are able to determine the right harvest time (when the valuable components have reached their optimum profile) and to select high-quality genotypes during breeding experiments, respectively. First promising attempts have also been made to introduce mobile diode array spectrometers to collect the spectral data directly on the field or in the individual natural habitats. Since the development of reliable NIRS methods in this special field of application is very time-consuming and needs continuous maintenance of the calibration equations over a longer period, it is convenient to supply the corresponding calibration data to interested user via NIRS network. The present status of all activities, preformed in this context during the last three years, will be presented in detail.
The genetic relationships in 5 species of $Ligularia$ were investigated using RAPD (Randomly Amplified Polymorphic DNA) and ITS (Internal Transcribed Spacer) sequences analyses. In RAPD analysis, sixty three of 196 arbitrary primers showed polymorphism. The amplified fragments ranged from 0.2 to 1.6 kb in size. The dendrogram was constructed by the UPGMA clustering algorithm based on genetic similarity of RAPD markers. A total of 16 accessions were classified into 5 major groups corresponding each species at the similarity coefficient value of 0.77. In the ITS sequence analysis, the size of ITS 1 was varied from 248 to 256 bp, while ITS 2 was varied from 220 to 222 bp. The 5.8S coding region was 164 bp in lengths. Forty nine sites (10.2%) of the 478 nucleotides were variable, and the G+C content of ITS region ranged from 49.4 to 53.5%. In the ITS tree, five species of $Ligularia$ were monophyletic, and $L.$$taquetii$ was the first branching within the clade. $Ligularia$$intermedia$ formed a clade with $L.$$fischeri$ var. $spiciformis$ (BS=79), and $L.$$stenocephala$ and $L.$$fischeri$ were also claded. Two data sets were congruent, except of the position of $L.$$fischeri$ var. $spiciformis$.
Goodyera rosulacea, which is morphologically similar to G. repens, is described recently as a new species based on its distinct morphological characters such as rosette-formed leaves, short rhizome and habitat. To verify the taxonomic identity of G. rosulacea and its taxonomic relationship within Korean Goodyera taxa, sequences of the internal transcribed spacer (ITS) region of nuclear ribosomal DNA and the trnL region of cpDNA from 24 accessions including 1 outgroup accession were analyzed. Aligned sequences were analyzed using maximum parsimony and distance method, and the taxonomic identity and the taxonomic relationships among the related taxa were estimated by the existence of private marker gene and the phylogenetic tree of the aligned sequences. Molecular data indicate that G. rosulacea gas several private marker genes and shows monophyly in phylogenetic trees of both ITS and trnL sequences. the pairwise distance between G. rosulacea and the orher taxa of Korean Goodyera was 3.49-6.68% for ITS region and 5.05-9.53% for trnL region, indicating that G. rosulacea could be treated as an independent species. Therefore, our molecular data support the taxonomic of G. rosulacea as a distinct species of Korea. In phylogenetic trees, G. rosulacea formed same clade with G. repens, which has similar morphological characters with G. rosulacea, and showed the lowest pairwise distance with G. repens among Korean Goodyera taxa. These molecular data sugguested that G. rosulacea and G. repens are closely related taxa.
We investigated changes of weedy rice emergence and seed condition under different flooding depth condition using 100 accessions of weedy rice germplasm collected in South Korea. In 1 cm of soil buried depth condition, 54 and 57% of emergence was reduced under 5 and 10 cm of flooding depth conditions respectively compared to non-flooding condition. In 5 cm of soil buried depth condition, 66 and 84% of emergence was reduced under 5 and 10 cm of flooding depth conditions respectively compared to non-flooding condition. The 94% of weedy rice germplasm showed less than 10% of emergence rate at 10 cm of flooding depth condition. In flooding condition, seed number, which germinated but did not emerged, was increased. As a result of analyzing the correlation between effective accumulated temperature and seed condition, dead seed rate increased and germination rate decreased depending on effective accumulated temperature. However, emergence rate did not show significant correlation with growth temperature condition under flooding condition such as 5 or 10 cm of water depth, it decreased according to flooding period. In order to reduce the emergence rate of weedy rice, longer than 21 days of flooding might be needed.
Investigating endophyte distribution, naturally occurring in native grasses, is important for understanding endophyte-grass associations and using the beneficial effects of endophytes in cultivated plants. The goal of this study was to investigate endophytes from natural Festuca spp. in Yunnan, Guizhou, and Sichuan Provinces of China, and to study the compatibility between the endophytes and turf type tall fescue (Festuca arundinacea) which is widely used for lawn and athletic fields in that region. Of 628 accessions in F. ovina, 421 had endophytes identified in leaf sheath from on-site microscopic examination. From Festuca spp, three isolates were obtained from the seeds and ninety isolates were obtained from seedlings established from the collected seeds. The isolates from F. ovina and F. stapfii were tentatively identified as Neotyphodium typhinum and Neotyphodiumstarii, respectively. We tested compatibility of the two fungal species with seven tall fescue cultivars, Little Hero, Sub Boy, Eldorado, Arid III, Millennium, Crossfire, and Fawn. N. typhinum or N. starii did not infect 'Fawn' with either seed injection or seed soaking method. The highest infection rate byboth N. typhinum and N. starii was in 'Sun Boy' and 'Eldorado'. There were significant interaction effects between tall fescue cultivar and type of endophyte on infection.

We developed droplet-vitrification protocol, a combination of droplet-freezing and solution-based vitrification, and applied to germplasm collections of garlic, potato, lily as well as cell lines, including hairy roots, somatic embryos. To establish a garlic cryobank, four Korean garlic field collections at Danyang, Suwon, Mokpo and Namhae were cryopreserved last five years. The protocol applied consisted of preculture for 3-4 days at $10^{\circ}C$ on solid MS medium with 0.3M sucrose, loading for 40 min in liquid medium with 35% PVS3, dehydration with PVS3 for 150 min, cooling in $5{\mu}l$ droplets of PVS3 placed on aluminum foil strips by dipping these strips in liquid nitrogen, warming them by plunging the foil strips into pre-heated($40^{\circ}C$) 0.8M sucrose solution for 30s. A total of over 900 accessions of garlic were stored in liquid nitrogen for long-term conservation using unripe inflorescences, cloves or bulbils. Twelve alternative plant vitrification solutions were designed by modifying cryoprotectant concentrations from the original PVS2 and PVS3. The results suggest that PVS2-based vitrification solutions with increased glycerol and sucrose and/or decreased DMSO and EG concentrations can be applied for medium size explants which are tolerant to chemical toxicity and moderately sensitive to osmotic stress. PVS3 and variants can be used widely when samples are heterogeneous, of large size and/or very sensitive to chemical toxicity and tolerant to osmotic stress.
Phylogenetic positions of Sedirea and Neofinetia were addressed using the chloroplast matK and the nuclear ITS sequences. We also evaluate the usefulness of the makers for the identification of species and localities. Sedirea and Neofinetia form an independent monophyletic genus, respectively, in both matK and nuclear ITS trees. The sister genus of the Neofinetia was Vanda in both trees. In addition, our trees support the separate recognition of the Neofinetia from Vanda rather than the inclusion of Neofinetia into Vanda. The sister group of the Sedirea was (Dimorphorchis(Pteroceras(Saccolabiun+Phalaeonopsis))) clade. The Dimorphorchis was one of the most probable sister genus to the Sedirea. The sister group relationship between Sedirea and Aerides was suggested by their similar morphology, but not supported in molecular trees. The identification of species and localities of Neofinetia was possible using our two molecular markers. However, several pseudo-gene sequences are discovered from the public data base. In addition, the horizontal gene transfer of chloroplast genomes is frequent events in orchid hybrids. Therefore, we need a careful evaluation for the data prior to systematic use. Generation of sequence data from multiple accessions of a species may helpful to reduce these types of error.

A total of eleven Chinese cabbage accessions were used for microspore culture and were grown to take basal data. Based on the collected data, breeding materials were chosen to develop new improved Chinese cabbage cultivars. The range of microspore-derived embryoid taken from flower buds was 1.6 to 35.4 embryoids. The embryoids from IT26110 and IT26153 among the Chinese cabbages were more than 34 per flower bud. The viability rate after cold treatment was low from 0.2 to 11.7%. The range of fertility rate was 7.7 to 58.8% in general but the IT26118, IT26122, IT26128, IT26130, and IT26164 were more than 50%. The result of their seed production ability by selfing was 11.9 seeds per siliqua in IT26128 while the others were less than 10 seeds. In the microspore culture using parents of different hereditary, the number of embryoids, the number of plants, the rate of fertility and their pure seed production ability appeared to be very different in doubled haploid lines obtained from fertile plants of Chinese cabbage.
To obtain basic information on forage sorghum F$_1$ hybrid breeding a total of 16 lines were selected from 311 introduced sorghum germplasm accessions, assessed and classified by the taxonomic distance and principal component analysis. The lines of which plant height and morphological characters were diverse and the 50% flowering date was similar to each other, were selected for parental lines in sorghum $\times$ sweet sorghum and sorghum $\times$ sudangrass crossing groups. Three varietal groups were classified by the average linkage cluster analysis based on the D$^2$ computed in eleven characters. Group I, II and III included 6 lines of sudangrass, 4 lines of sweet sorghum and 6 lines of grain sorghum, respectively. In the result of principal component analysis for eleven characters, about 82% of total variation could be appreciated by the first four principal components, the first principal component was highly loaded with head compactness and shape, l00-seed weight, plant color and grain covering, the second principal component with flowering date, plant height and awnness.
