가시오가피 물추출물이 고콜레스테롤식이를 공급한 흰쥐의 지질대사에 미치는 영향 (Effect of Acanthopanax Senticosus Water Extract on Lipid Metabolism in Rats Fed a Hypercholesterol Diet)

  • In order to find the effect of improving hyperlipidemia by Acanthopanax senticosus water extract, a hypercholesterol diet and Acanthopanax senticosus water extract were supplemented to week- old male Spargue Dawley rats for four weeks in different amount. Results showed that serum total cholesterol significantly decreased in the group supplemented with Acanthopanax senticosus water extract by 50mg/kg(ASW-50) and 75mg/kg(ASW-75) compared with the control group.(p<0.05). Serum triglyceride also showed a significant decrease in the group supplemented with 50mg/kg(ASW-50) and 75mg/kg(ASW-75) compared with the control group. Liver total cholesterol showed a significant decrease in the group supplemented with Acanthopanax senticosus water extract by 50mg/kg(ASW-50) and 75mg/kg(ASW-75) compared with the control group(p<0.05), but liver triglyceride did not show a significant decrease in all of the experiment groups. Total cholesterol and triglyceride in feces significantly increased in all of the groups supplemented with Acanthopanax senticosus water extract(p<0.05). Acanthopanax senticosus water extract decreased the level of serum total cholesterol and triglyceride, reduced total cholesterol in the liver, and increased the excretion of total cholesterol and triglyceride in the feces.

Acanthopanax senticosus Extract Prepared from Cultured Cells Improves Lipid Parameters in Rats Fed with a High Fat Diet

  • Acanthopanax senticosus was grown by a novel, proprietary method, of culturing isolated cells in a bioreactor. An extract from the cells was evaluated for its effect on lipid metabolism in rats fed a high fat diet. Male Sprague-Dawley rats (n=6) were fed either an AIN-76 diet (control, NDCon), control diet plus Acanthopanax senticosus extract (ND+Ex), a modified AIN-76 diet supplemented with 20% beef tallow (high fat, HFCon), or a high fat diet plus Acanthopanax senticosus extract (HF+Ex), for 5weeks. Body weight gain was significantly higher in the HFCon group than the NDCon group. Feed consumption was significantly lower, but energy intake higher, in the groups fed high fat diets compared with the groups fed control diets. Serum HDL-cholesterol concentrations were significantly increased but serum LDL-cholesterol concentrations were decreased in the groups fed the Acanthopanax senticosus extract. Abdominal fat accumulation and serum leptin levels were significantly higher in the HFCon group than the other groups. Carnitine palmitoyltransferase-I (CPT-I) mRNA levels were increased in the groups fed Acanthopanx senticosus extract. These results suggest that supplementation of cell cultured Acanthopanax senticosus extract regulates CPT-I mRNA levels in liver and has an effect on the normalization of lipids in rats fed a high fat diet.

가시오가피 추출물이 마크로파지 활성과 우유의 발효 중 젖산균 성장에 미치는 영향 (Effects of Acanthopanax senticosus Extract on Macrophage Activity and the Growth of Lactic Starter Culture during Fermentation)

  • 가시오가피의 마크로파지 활성 효과를 평가하고, 발효유에 첨가할 경우 젖산균 성장에 미치는 영향에 대하여 실시하였다. 가시오가피 추출물 농도가 증가할수록 마크로파지 활성은 증가하였고, 특히 $1,000\;{\mu}g/mL$ 첨가하였을 때 유의성 있게 증가하였다. 열수추출물은 70% 알콜추출물보다 NO 분비능이 더 높은 반면, $IL-1{\alpha}$$TNF-{\alpha}$는 알콜추출물이 더 높은 활성을 보였다. 가시오가피 열수추출물이 젖산균에 미치는 영향을 조사한 결과, 가시오가피 첨가량이 증가할수록 젖산균 성장이 억제되었고, pH는 완만히 감소됨에 따라 발효유에 가시오가피열수추출물을 첨가할 경우 Set type보다는 Stirred type 이나 Drink type으로 해야 발효유 제조에 적합할 것으로 보인다.

계절 변화에 따른 한국산 가시오가피 줄기의 페놀성 성분 함량 분석 (Seasonal Variation of Phenolic Component Contents in the Stems of Korean Acanthopanax senticosus)

  • Objectives: The aim of this study was to compare the total phenolic content of Korean Acanthopanax senticosus stems harvested in different seasons and provide basic data for developing functional reinforcement products based on the optimal harvesting time. Methods: Each sample harvested in different seasons was extracted and concentrated twice for 2 hours using 70% ethyl alcohol. Phenolic compounds were analyzed using high-performance liquid chromatography for simultaneous multi-component analysis of 14 compounds, including syringaresinol and so on. Results: The results showed that the stem of Korean Acanthopanax senticosus harvested in winter (November 29th) (EAS-5) had the highest phenolic content of 1.038%. The stem of Korean Acanthopanax senticosus harvested in autumn (October 1st) (EAS-4) showed the second-highest phenolic content of 0.764%, followed by the stem of Korean Acanthopanax senticosus harvested in spring (February 2nd) (EAS-1) with a content of 0.390%. On the other hand, the stem of Korean Acanthopanax senticosus harvested in the summer (June 2nd) (EAS-3) showed the lowest content at 0.342%. In conclusion, the stem of Korean Acanthopanax senticosus harvested in winter (EAS-5) showed the highest phenolic compound content. Conclusions: Considering the extraction yield and the total phenolic content, as well as the concentrations of key functional components such as eleutheroside B, chlorogenic acid, and syringaresinol in the 70% ethyl alcohol extract of Korean Acanthopanax senticosus, it is suggested that the stems of Korean Acanthopanax senticosus harvested during the winter season are suitable for the development of novel materials with enhanced anti-obesity functionality.

Acanthopanax and Platycodi Independently Prevents the Onset of High Fat Diet Induced Hyperglyceridemia and Obesity in C57BL/6 Mice

  • Using high-fat-fed C57BL/6 mice, therapeutic effects of oral administration of aqueous extracts of Platycodon grandiflorum and Acanthopanax senticosus on obesity were evaluated by comparing body and liver weights, feed intake, adipose tissue mass, blood lipid profile, and triglyceride and total cholesterol levels in liver and feces. Obesity was successfully induced in high-fat diet group. P. grandiflorum and A. senticosus were effective in reducing food intake, serum lipid levels, adipose tissue accumulation, and weight. P. grandiflorum extract reduced triglyceride and cholesterol levels in liver by excluding them in feces. A. senticosus extract increased triglyceride level in liver but decreased in feces, and decreased total cholesterol in liver and feces, indicating active ingredient of A. senticosus exert antiobesity effect through mechanism different from that of P. grandiflorum extract. These results suggest aqueous extracts of P. grandiflorum and A. senticosus have synergistic effect for prevention of hyperglyceridemia and obesity.

고지방식이 유도 비만 마우스에서 가시오가피(Acanthopanax senticosus)의 지방축적 억제 및 인슐린 저항성 개선 작용 (Suppression of Fat Accumulation and Improvement of Glucose Tolerance in High-fat Diet-induced Obese Mice Treated with an Acanthopanax senticosus Extract)

  • Although it has recently been reported that Acanthopanax senticosus has a protective effect against the development of metabolic disorders such as diabetes and hyperlipidemia, its underlying molecular mechanisms have remained to be elucidated. In this study, we observed that treatment of an extract of A. senticosus (ASE) reduced body weight and adiposity and improved glucose tolerance in high-fat diet (HFD)-induced obese mice. HFD-induced weight gain in white adipose tissues and liver was also significantly reduced in ASE-treated HFD-fed mice, which was found to be contributed by suppression of mRNA expression of genes involved in fatty-acid uptake and fat synthesis in white adipose tissue. ASE treatment also attenuated M1 macrophage activation, increased regulatory T ($T_{reg}$) cell population, and modulated leptin/adiponectin profile that might alleviate chronic inflammation to protect against HFD-induced insulin resistance. Our findings suggest a therapeutic potential of ASE for prevention and treatment of type 2 diabetes without adverse effects such as weight gain and dyslipidemia.

오가피 추출액의 저장조건에 따른 이화학적 특성변화 (Changes of Physicochemical Characteristics of Acanthopanax senticosus Extract during Storage)

  • 오가피 열수추출액의 유통 품질의 변화특성을 규명하기 위하여, 오가피($8{\sim}20$ mesh)를 열수 추출한 액을 nylon/PE(0.08 mm) pouch로 밀봉포장하거나 포장하지 않고, 가온조건(40, $60^{\circ}C$)에서 10일간 보관하면서 1일 간격으로 이화학적 품질특성을 조사하였다. 보관 중 추출액의 탁도, pH, 총폴리페놀 함량 및 DPPH 유리기 소거능 등은 감소하는 경향을 보였으나 저온과 포장에 의해 감소가 억제됨을 보였다. 적정산도는 고온에서 증가함을 보였으나 포장에 의해 증가가 억제되는 경향이었다. 가용성 고형분 함량은 포장과 온도의 영향을 받지 않고 거의 일정하게 유지되었다. 품질변화 kinetics를 model화 하기 위하여 반응속도론에 상사시켜 본 결과 오가피 추출액의 품질변화는 1차반응 속도식이 적합하였으며 품질변화 속도상수에 대한 온도의 영향은 Arrhenius 식에 잘 따른다고 할 수 있었다. 이상의 결과로 볼 때, 오가피 열수추출액의 부패억제와 항산화능 유지를 위해서는 포장과 저온유통이 필수적인 것으로 판단된다.

가시오가피와 양파 혼합 추출물이 Collagen 유발 관절염에 미치는 영향 (Effects of Acanthopanax Senticosus and Onion Mixture Extract on the Collagen-induced Arthritis in Rat Model)

  • This study was conducted to determine the analgesic effect of onion and acanthopanax senticosus mixture extract using the rheumatoid arthritis rat model. Rheumatoid arthritis model was made by the intradermal injection of type II collagen emulsified. Rats were divided into four groups: (1) Sham group(n=5), (2) Control group(administered DW 3 $m{\ell}$/1 day after RA induced, n=7), (3) Experimental group I(administered Onion extractor 600 mg/3 $m{\ell}$/1 day after RA induced, n=7). (4) Experimental group III(administered Onion and Acanthopanax senticosus mixture extractor 600 mg/3 $m{\ell}$/1 day after RA induced, n=7). After that, we examined the arthritic index, paw edema, pain threshold at 1st, 14th, 28th days. And also we examined histopathologic study(safranin-O green), immunohistochamical stain(COX-2) of knee joint at 28th days. Arthritic index, paw edema and pain threshold test were decrease in experimental group I, II than control group. Especially group II was most significantly inhibit effect than the other groups at 28th days. On the histopathologic view, all experimental groups were relieved and reproduced the erosion of arthritic site compared with control group. All experimental groups were COX-2 positive cells in the immunohistological stain of the knee joint were significantly decreased compared with control group. Especially group II was most significantly decreased than the other groups at 28th days. Onion and Acanthopanax senticosus mixture extractor can be used for curing rheumatoid arthritis. Anti-inflammatory effects may be somewhat better in combination of Onion and Acanthopanax senticosus.

민가시오갈피나무 종자(種子)의 성분(成分)에 관한 연구(硏究) (Studies on the Constituents of Seeds of Acanthopanax senticosus for. inermis Harms)

  • Fatty acids, sterols and amino acids were confirmed from seeds of Acanthopanax senticosus for. inermis Harms. Fatty acids were isolated from petroleum ether extract and identified by gas liquid chromatography. Its composition was myristic acid, palmitic acid, stearic acid, oleic acid, linoleic acid, linolenic acid and behenic acid. The composition of sterols that isolated from n-hexane extract were determined by GC/MS. It was confirmed to be a mixture of ${\beta}-sitosterol$ and stigmasterol. And several kinds of amino acids-cysteic acid, threonine, serine, proline, glutamic acid, glycine, alanine, valine, isoleucine, leucine, histidine, lysine and arginine-were confirmed from water fraction.

가시오가피 물 추출물에 의한 마우스 비장세포 및 대식세포 활성의 항진효과 (Enhancing Effect of Acanthopanax senticosus Extracts on Mouse Spleen and Macrophage Cells Activation)

  • In vitro 실험을 통한 가시오가피 물 추출물 첨가가 마우스의 면역세포 증식에 미치는 영향에 대한 연구 결과, 음의 대조군에 비해 가시오가피 물 추출물을 첨가한 모든 농도에서 비장세포 증식능이 증가하였으며, 특히 고농도인 $500{\sim}1,000{\mu}g/mL$ 농도에서 유의적으로 증가하였다. 반면, 복강 대식세포로부터 유도된 사이토카인 생성의 경우, IL-2, IFN-${\gamma}$, TNF-${\alpha}$ 사이토카인 생성량을 측정한 결과, TNF-${\alpha}$ 사이토카인은 가시오가피 물 추출물 $50{\mu}g/mL$ 농도와 $250{\sim}1,000{\mu}g/mL$ 농도에서 대조군보다 유의적으로 높은 분비량을 보인 반면, IFN-${\gamma}$에서는 변화를 보이지 않았다. IL-2의 경우, $100{\sim}500{\mu}g/mL$에서 유의적으로 증가하는 경향을 보여주었다. 이상의 결과에 의하면 가시오가피 물 추출물은 마우스 비장 세포를 증식시키고, 사이토카인 분비량에도 영향을 줄 것으로 보이며, 면역 기관의 주요 기능을 증진시킬 가능성이 있을 것으로 사료된다. 따라서 가시오가피 물 추출물이 면역 증진 기능성식품 개발의 소재로 활용될 가능성이 있을 것으로 기대된다.
