부자(附子) 및 천오(川烏)를 함유한 처방(處方)이 간기능(肝機能)에 미치는 영향(影響)(II) (The Consequences of Long-period Dosages of Herbal Medicine Including Boo-ja and Cheon-o on Liver Injury.(II))

  • Objectives: Boo-ja and Cheon-o are not commonly prescribed, but are necessary for some clinical conditions, dispite the fact that some negative effects have been known to occur with these medicines. Hence, consequences for aspartate aminotransferase (AST), alanine aminotransferase(ALT) due to herbal medicines, including boo-ja and cheon-o are here reported. Methods: From January 2005 to July 2005, iresults were analyzed for 40 inpatients belonging to the sixth internal medicine department of Dongeui Oriental Medical Hospital who took herbal medicine, including boo-ja and cheon-o, over 20 days. The study is about the comparison and the investigation of LFT, and all results were taken upon their hospitalization and upon their departure. The standard of liver injury was used as a standard for their examinations. Results : 1. No Change in normal limit . 34 of the 40 2. Increased AST. ALT from when they were hospitalized : 2 of 40 Normalized AST, ALT from when they were hospitalized with results higher than normal : 6 of 40 3. These results show no side-effects, therefore suggest that these herbal medicines, including boo-ja and cheon-o. do not cause adverse side-effectsfor patients of this kind if administered as was done in this study.

흰쥐의 일차배양 간세포에서 Daidzin, Daidzein, Genistein 및 Puerarin의 간 보호 활성 평가 (Hepatoprotective Activities of Daidzin, Daidzein, Genistein and Puerarin in Primary Cultured Rat Hepatocytes)

  • The aim of this study was to investigate the protective activities of daidzin, daidzein, genistein or puerarin, active isoflavonoids of Puerariae Radix, on the hepatocyte injury induced by carbon tetrachloride (CCl$_4$, 10 mM), tert-butyl hydroperoxide (TBH, 0.5 mM) and D-galactosamine (GalN, 30 mM). Primary cultures of rat hepatocytes (18 hr cultured) were treated with CCl$_4$, TBH or GalN and various concentrations (0.1, 1, 10 and 100 ${\mu}$M) of daidzin, daidzein, genistein or puerarin. CCl$_4$ significantly increased the levels of lactate dehydrogenase (LDH), alanine aminotransferase (ALT) and aspartate aminotransferase (AST). The increase in LDH level was attenuated by daidzein, genistein and puerarin. Puerarin also inhibited the increase in AST level induced by CCl$_4$. The increases in LDH and ALT levels induced by TBH were significantly attenuated by daidzin and genistein treatments. GalN markedly increased the levels of LDH, ALT and AST These increases were significantly attenuated by daidzein. Daidzin also inhibited the increases in LDH and AST levels induced by GalN. The increases in LDH and ALT levels were attenuated by genistein and puerarin, respectively. These results suggest that daidzin and daidzein possess hepatoprotective activities.

벤조피렌으로 유발된 흰쥐 간독성에 대한 번데기동충하초 배양액의 회복효과 (Effect of Culture Broth of Cordyceps militaris on Recovery of Mice Hepatic Damage Caused by Benzo($\alpha$)pyrene-Treatment)

  • 번데기동충하초의 간독성 회복 효과를 HaM/ICR 계열의 흰쥐를 이용하여 검정하였다. 벤조피렌 복강주사시, 혈중의 aspartate aminotransferase (AST) 및 alanine aminotransferase (ALT)의 효소활성과 간조직의 lipid peroxide (LPO) 농도는 대조군과 비교시 급격히 증가하여 간독성을 유발하였다. 그러나, 벤조피렌 대조군과 비교시, 번데기 동충하초 배양액의 경구 투여시 AST와 ALT의 효소활성 및 LPO 농도는 감소하였으며, 벤조피렌에 유도된 간 조직의 glutathione 농도 및 glutathione S-transferase 효소활성은 증가하여 간독성을 회복하였다. 실험결과는 번데기 동충하초 균사체 배양액이 벤조피렌 유발 간독성에 대해 회복 효과가 있음을 보여준다.

사염화탄소를 투여한 MICE 혈청내 효소 활성 및 생화학적 조성의 변화 (Changes of Biochemical Composition and Activity of Enzyme in the Serum Exposed to Carbon Tetrachloride in Mice)

  • Carbon tetrachloride ($CCl_4$) has been used to treat a variety of parasitic infection in both large and small animals, including Fasciola hepatica. Recently, we can easily contest with carbon tetrachloride in air, food, water, rain and Industry area etc. and it is using in order to induce liver injury in laboratory. In this study. we investigated activitis of LDH, ALP, AST and ALT and amount of cholesterol, triacylglycerol, glucose and BUN in mice serum exposed to $CCl_4.$ The mice divided Into a Naive control(A), corn oil control(B) and experimental group(C, D). Naive control group(A) was given feed and water only. Corn oil control group(B) was given corn oil 1ml /100g of body weight(B.W). Experimental group(C) was given carbon tetrachloride 1ml /100g B.W. ($CCl_4$: corn oil=1:20(V /V) ), Experimental group(D) was given carbon teterachloride 1ml /100g B.W. ($CCl_4$: corn oil=1:10(V /V) ). The results obtained were summarized as follows : 1) The body weight was declined after the l0th day In mice exposed to carbon tetra-chloride. 2) The total protein level in serum was significantly in mice exposed to carbon tetra-chloride($P{\leq}0.05$). The albumin and A /G ratio was decresed significantly in mice exposed to carbon tetrachloride($P{\leq}0.01$) 3) All of the activity of LDH, AST, ALT and ALP in mice serum exposed to carbon tetra chloride inclosed significantly activity of LDH ($P{\leq}0.05$), inclosed significantly activity of AST($P{\leq}0.05, \;p {\leq}0.01$), inclosed significantly activityof ALT 3nd ALP($P {\leq}0.05,\;p {\leq}0.01$). 4) The amount of cholesterol and triacylglycerol, lipid metabolite products in serum was inclosed in case of cholesterol but did not change in case of triacylglycerol.

  • The aim of this study was to investigate the protective effects of glycynhizin, active glycosides of Glycyrrhizae Radix, and baicalin, bioactive flavonoid isolated from Scutellariae Radix, on hepatocyte injury induced by carbon tetrachloride(CCl$_4$, 10 mM), tert-butyl hydroperoxide (TBH, 0.5 mM), and D-galactosamine (GaIN, 30 mM). Primary cultures of rat hepatocyte (18 hr cultured) were treated with CCl$_4$, TBH, or GaIN and various concentrations (0.1, 1, 10, and 100 ${\mu}$M) of glycyrrhizin or baicalin. Activity was accessed by determining the release of lactate dehydrogenase (LDH) and aminotransferses. CCl$_4$ significantly increased the levels of LDH, alanine aminotransferase (ALT), and aspartate aminotransferase(AST) and these increases were prevented by baicalin concentrations of 0.1,1, and 100 ${\mu}$M. The increases in ALT and AST levels were reduced by glycyrrhizin concentration of 100 ${\mu}$M. The level of LDH was markedly increased by TBH, and this increase was reduced by both glycyrrhizin and baicalin. ALT and AST levels were increased by TBH, which were prevented by glycynhizin and bacalin, respectively: GaIN markedly increased the levels of LDH, ALT and AST These increases was significantly reduced by both glycyrrhizin and baicalin. These results suggest that glycynhizin and baicalin possess the hepatoprotective activity.

복부 초음파 검사로 진단된 지방간과 고지혈증 지표와의 관련성 연구 (The Study for the Relation between Fatty Liver Diagnosed by Abdominal Ultrasonography and Hyperlipidemia)

  • 본 연구는 복부 초음파 검사를 통해 확인된 지방간 환자의 지방간 정도에 따른 혈액학적 고지혈증 지표와의 관련성을 분석하여 관련 연구에 기초자료를 제공하고자 시행되었다. 2017년 1월 1일부터 2017년 12월 31일까지 총 412명의 성인을 대상으로 하였으며, 복부초음파 검사를 진행하여 지방간이 확인된 환자 중 초음파 검사 시행일 기준 3개월 이내의 혈액학적 데이터가 있는 환자를 대상으로 후향적 분석을 실시하였다. 연구결과, 지방간 정도에 따른 혈액학적 수치는 중성지방, AST, ALT에서 통계적으로 유의한 차이가 있었고(p<.05), 지방간은 전체 변수 중 성별, 중성지방, AST, ALT와 관련성이 있었다(p<.05). 결론적으로 지방간 정도는 고지혈증의 혈액학적 수치와 직접적인 영향인자는 아니었지만 그 관련성 확인을 통해 지방간과 고지혈증 동반 관리의 필요성을 확인했다.

  • Objectives : Aconiti Lateralis Preparata Radix (附子) and Aconiti Tuber (川烏) are not commonly prescribed, but are necessary for some clinical conditions, despite of the fact that some negative effects have been known to occur with these medicines. This study shows the consequences for aspartate aminotransferase (AST), alanine aminotransferase (ALT) due to herbal medicines, including Aconiti Lateralis Preparata Radix (附子) and Aconiti Tuber (川烏). Methods : From 1st June 2007 to 10th May 2009, the results were analyzed for 64 patients belonging to the OO Oriental Medical Hospital who took herbal medicine, including Aconiti Lateralis Preparata Radix (附子) and Aconiti Tuber (川烏), more than 20 days. This is the study about the comparison and the investigation of AST, ALT. And the results were taken upon their hospitalization and to their departure. The Criteria of Diagnosis in Liver Injury of CIOMS was used as a standard for their examinations. Results : AST, ALT (total 64). The number of patients who were within normal reference value : 49. The number of patients who exceed the normal reference value after dosage, without satisfying the Criteria of Diagnosis in Liver Injury : 5. The number of patients who were recovered in normal reference value after dosage : 9. The number of patients who satisfying the Criteria of Diagnosis in Liver Injury after dosage : 1. Conclusions : This results suggest that these herbal medicines, including Aconiti Lateralis Preparata Radix (附子) and Aconiti Tuber (川烏), didn't cause adverse side-effects on AST, ALT of the patients who are taking them.

Rotavirus 위장관염에서 혈청 Creatine Kinase의 증가 (Elevated Serum Creatine Kinase Level in Rotavirus Gastroenteritis)

  • 목 적: 로타바이러스에 의한 급성위장관염이 있는 환아들에서 AST/ALT가 증가되어 있는 경우 중 CK가 증가된 경우가 있어 이들 환아들의 임상 양상이나 검사에서 특이 소견이 있는지 확인하고자 하였다. 방 법: 2001년 1월부터 2005년 3월까지 본원에서 로타바이러스 위장관염으로 진단된 환아들 중 AST/ALT가 증가해 있고, CK가 증가해 있는 환아들의 의 무기록을 통하여 임상 양상과 검사결과를 종합하였으며, 통계학적 유의성에 대해서 조사하였다. 결 과: 총 14명의 환아가 AST나 ALT가 증가되어 있으면서 CK가 증가되어있었다. 평균 연령은 1년 5개월이었으며, 연도별 빈도는 차이가 없었다. 동반 증상으로 설사, 구토, 열, 경련, 감기 증상의 유무에 따른 차이는 없었으며, 탈수의 정도가 CK 농도 증가에 영향을 미치지 않았으며, 설사의 기간도 연관성은 없었다. 14명 모두에서 CK가 증가될 수 있는 질환의 증거는 없었으며, 회복기에 정상화되는 것을 확인하였다. AST나 ALT, LDH의 농도가 높을 수록 CK의 농도가 증가하는지 확인하였으나 연관성은 없었다. 3명의 환아에서는 CK가 1,000 IU/L이상 증가되어 있었는데, 이들에서 급성 신부전이나 경련, 근육통 등의 소견은 없었다. 결 론: 로타바이러스 위장관염 환아들에서 AST/ALT가 증가할 수 있음은 이미 알려져 있으나 그 기전은 아직 이해되지 않고 있다. 본 연구에서는 AST 나 ALT의 증가 외에도 CK가 증가될 수 있음을 확인하였으나 어떤 기전에 의하는지, CK가 증가할 수 있는 특정 조건이 있는지는 알 수 없었다.

청소년기 비만아에서 이상 혈청 간효소치 및 총콜레스테롤치의 유병율 (The Prevalent Rates of Abnormal Serum Aminotransferase Levels and Total Cholesterol Levels among Adolescents with Obesity)

  • 목 적 : 최근 소아 및 청소년의 비만은 증가하고 있으며 이러한 비만은 여러 가지 합병증이 동반될 수 있다. 본 연구에서는 서울지역 청소년에서 비만과 간효소치 및 총콜레스테롤치 이상의 유병율을 알아보고자 하였다. 방 법: 2001년도에 실시된 서울지역 청소년들의 건강 관련 지표 조사에서 실시된 서울지역 고등학교 1학년생 약 3만명의 종합 신체검사 결과 중 유효분석 대상 26,876명(남자 13,287명; 여자 13,589명)의 서울특별시 학교보건원의 검사 결과를 대상으로 하여 BMI의 분포를 남녀별로 알아보고 이들을 정상체중군(BMI 25 백분위수 이상-75 백분위수 이하)과 비만군(BMI 95 백분위수 이상)으로 분류하여 이들에 있어서 간효소인 혈청 AST, ALT 및 총콜레스테롤 이상의 유병율을 분석하였다. 결 과: AST 이상자는 남아에서 비만군 168명(23.7%)과 정상체중군 317명(3.7%)이었으며 여아에서는 79명(11.8%)과 334명(4.9%)으로 남녀 모두 비만군에서 정상 체중군보다 높았다(P<0.01, $x^2-value$; 남아 529.2, 여아 56.3). ALT 이상자는 남아에서 241명(33.9%)과 149명(1.7%), 여아에서는 149명(22.3%)과 264명(3.9%)으로 특히 남아에서 비만군의 이상자 비율이 정상체중군에 비해 20배 가량 높았다(P<0.01, $x^2-value$; 남아 1,693.4, 여아 397.6). 총콜레스테롤 이상자는 남아에서 119명(16.8%)과 300명(3.5%), 여아에서는 69명(9.0%)과 371명(5.4%)으로 비만군이 정상 체중군에 비해 높았다(P<0.01, $x^2-value$; 남아 268.3, 여아 14.2). 비만군의 경우 남자에서 여자보다 AST, ALT 및 총콜레스테롤의 이상자 비율이 높았다(P<0.01). 결 론 : 비만 청소년에서 간기능 효소의 이상과 총 콜레스테롤 이상의 유병율이 높으며 이러한 경우 지방간, 고지혈증 등의 합병증의 가능성이 높으므로 체중 감량 등을 통한 적극적인 예방과 치료가 필요하다.

인삼, 한국 및 중국산 산양삼의 간 보호 효능에 관한 실험적 연구

  • Objectives : This study was aimed at investigating liver protection mechanism of Cultivated Ginseng and Cultivated Wild Ginseng of Korean and Chinese by inducing liver toxicity through $CCl_4$ and t-BHP in mice and evaluated serological findings. Methods : Experiment groups was categorized into untreated normal group, treated control group, and orally administered Cultivated Ginseng and Cultivated Wild Ginseng of Korean and Chinese experimental groups. At the termination of experiment, gross examination of the liver as well as Total bilirubin, AST, and ALT contents in the serum were evaluated. Results : 1. In the $CCl_4$ induced acute hepatotoxicity test, total bilirubin, AST and ALT didn't show significant differences between the control and experimental groups. 2. In the t-BHP induced acute hepatotoxicity test, total bilirubin, AST and ALT didn't show significant differences between the control and experimental groups. Conclusion : Taken together, Cultivated Ginseng and Cultivated Wild Ginseng of Korean and Chinese cannot be effectively used for recovering the liver functions in acute hepatotoxicity tests using $CCl_4$ and t-BHP. Further researches, for example treated long period, must be tried to verify the efficacies.
