Biological Activities of Scolopendrid Pharmacopuncture (수순(水醇)추출법으로 조제된 오공 약침액의 생리활성 효과)

  • Reactive Oxygen Species(ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Since tissue damage by free radical increases with age, the reactive oxygen species(ROS) such as hydrogen peroxide($H_2O_2$), nitric oxide(NO). Several lines of evidence provided that ROS appears to cause to develop aging-related various diseases such as cancer, arthritis, cardiovascular disease. Our reserch objective was to examine the in vitro biological activity of Scolopendrid Pharmacopuncture, including the total poly-phenol content, DPPH radical scavenging, ABTS radical scavenging, Superoxide dismutase(SOD)-like activity, Nitrite scavenging ability. The total poly-phenol contents of Scolopendrid Pharmacopuncture was 35.859mg/L. Elctron donation ability on DPPH was 36.82%. The 2,2'-azinobis-3-ehtlbezothiazoline-6-sulfonic acid radical decolorization (ABTS) was 84.7%. The superoxide dismutase (SOD)-like activities of Scolopendrid Pharmacopuncture was 44.33%. The nitrite scavenging effects were pH dependent, and were highest at pH 1.5(45.2%) and lowest at pH 6.0(11.3%). We conclude that Scolopendrid Pharmacopuncture may be useful as potential sources of antioxidant.

Quality and Antioxidant Activity of Granules Containing Herbal Medicine Extracts (약재 추출물을 첨가한 과립의 품질 특성 및 항산화 효과)

  • The aim of this study was to investigate the quality characteristics and antioxidant activities of herbal medicine extract granules for the comparison of commercial powder products such as adlay tea powder and roasted grain powder. The antioxidant activities of samples were estimated using reducing power, 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging activities, and total polyphenolic contents. Herbal extract granules contained $126.44{\pm}7.26mg/100g$ of total polyphenolic contents. Interestingly, the highest antioxidant activity was found in 5.5% herbal extract granules, which showed DPPH and ABTS free radical scavenging activities of 71.82% and 85.28%, respectively, at 100 mg/mL and reducing power of O.D 1.25. These results suggested that addition of herbal medicine extract had beneficial effects on the functionality of granules without any affecting on its sensory characteristics.

Antioxidative Activity of Beverage with Water and Ethanol Extracts of Maegmundong (Liriope platyphylla) (맥문동의 물추출액 및 에탄올추출액 이용 음료의 산화방지 활성)

  • The antioxidative activities of beverages with water or ethanol extracts from Maegmundong (Liriope platyphylla) were investigated with free amino acid and mineral analysis, as well as saponin, total phenolics and flavonoid determination. Antioxidative activity was evaluated by electron donating ability and ABTS radical scavenging activity. There was only a small difference between water extract beverages and ethanol-extract beverages with respect to their electron donating ability and ABTS radical scavenging activity, although the total saponin, total phenolics and flavonoid were found to be greater in the ethanol extract beverages than in the water extract beverages. Three major free amino acids of the Magemundong beverages with water- or ethanol extract were asparagine (58.30, 60.68 mg%), methionine (15.10, 13.95 mg%, respectively) and proline (12.31, 14.00 mg%, respectively). The most abundant mineral in the Maegmundong beverage with water and ethanol extract was potassium (238.68, 244.32 mg%, respectively).

Anticytotoxic and Radical Scavenging Activities of Acer tegmentosum Maxim Stem Extracts (산겨릅나무 줄기 추출물의 세포독성 억제 및 라디칼 소거 활성)

  • The objective of this study was to investigate anticytotoxic and antioxidatative capacities of ethanol extracts from Acer tegmentosum Maxim (A. tegmentosum) stem in vitro. The extract at concentration of 200 ug/mL inhibited 10 and 20 ug/mL arsenic trioxide-induced cytotoxicity of HepG2 cells by 79.3 and 57.5%, respectively. The extract at concentration of 200 ug/mL inhibited 0.2 and 0.5 mM t-BHP-induced cytotoxicity of HepG2 cells by 66.3 and 35.7%, respectively. Antioxidative effects of the extract were examined via measurement of ABTS, superoxide, and peroxyl radical scavenging activities. ABTS radical scavenging activity of the extract was higher than that of ${\alpha}$-tocopherol. Superoxide scavenging activity of the extract was higher than that of catechin. Oxygen radical absorbance capacity of the extract was higher than that of ascorbic acid. Cupric reducing antioxidant capacity of the extract was higher than that of ${\alpha}$-tocopherol. The extract at concentrations of 100 and $500{\mu}g/mL$ inhibited 10 mM t-BHP-induced lipid peroxidation of HepG2 cells by 38.2 and 80.7%, respectively. The extract prevented supercoiled DNA strand breakage induced by hydroxyl or peroxyl radical. Total phenolic and flavonoid contents of the extract at concentration of $100{\mu}g/mL$ were 71.3 nmol/mL gallic acid and 18.8 nmol/mL catechin equivalents, respectively. Thus, strong cytoprotective and antioxidant effects of A. tegmentosum stem extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in polyphenolic contents.

Antioxidant and Anti-physical fatigue Effects of Polygonati Rhizoma and steamed Polygonati Rhizoma (황정(黃精)과 증숙 황정(蒸熟 黃精)의 항산화, 항피로 효능 비교 연구)

  • Objectives: Polygonati Rhizoma (PR) has containing the bioactive compounds such as poly sccharide A,B,C, oligosaccharide, amino acid, it has reported to anti-diabetes and hypertension, atherosclerosis. In this study, we were evaluates antioxidant and anti-physical fatigue effects of PR and steamed PR.Methods : The sample was divided into 5 groups-PR0 (PR without steaming process), PR1 (PR with once steaming process), PR3 (PR with third steaming process), PR6 (PR with sixth steaming process), PR9 (PR with ninth steaming process). We measured anti-oxidant activity through contents of polyphenol, flavonoid and DPPH, ABTS free radical scavenging capacity. And, anti-physical fatigue effect was evaluated using the swimming test, and the AMPK protein expressions in soleus muscle.Results : As a result, polyphenol, flavonoid, DPPH, ABTS free radical scavenging capacity of PR were increased as steaming times. Anti-physical fatigue effects by swimming test, PR0 have significantly increased, but steamed PR groups were decreased. The AMPK protein expressions of PR0 and PR1 groups were increased comparing with PR3, PR6 and PR9. All groups had effects on decreasing TG, creatine in blood serum, but had no effects on TC in blood serum.Conclusions : In conclusion, PR with 9 steaming process was more excellent than not-processed PR in anti-oxidant effect such as DPPH, ABTS radical scavenging activity and contents of polyphenol, flavonoid, but, not-processed PR increased swimming times than processed PR. These results suggest that processed PR has anti-oxidant effect as steaming times, and not-processed PR may be a novel potential anti-physical fatigue agents than processed PR.

Changes of Bioactive Compounds and Antioxidant Activities in Korean Green Tea (Camellia sinensis) with Different Harvesting Periods (국산 녹차의 채취시기별 활성물질 및 항산화능 변화)

  • Korean green tea has been claimed to have health-promoting effects, which may be related to the antioxidant activity in vitro. Korean green teas (Woojeon, WJ ; Sejak, SJ ; Jungjak, JJ ; Daejak, DJ) were subjected to different harvested times and yet little research has examined their bioactive compounds. To assess the effect of this different harvested times on nutritional and health-related properties such as Korean green tea polyphenols, flavonoids, theanine and free amino acids, antioxidant activities and radical scavenging activities were determined. Total polyphenols in JJ (37.16 mg/g) was higher than in other products (WJ, 19.55 ; SJ 24.65 ; DJ, 23.28 mg/g). Contents of flavonol and flavone glycosides were the highest at DJ (350.83 mg%) as compared to WJ (220.81), SJ (256.88) and JJ (270.36 mg%). Contents of theanine and total free amino acids were the highest at WJ (14.11, 23.62 mg/g, respectively), but decreased thereafter. Antioxidant activities were higher in JJ and DJ, using the linoleic acid peroxidation, DPPH and ABTS free radical-scavenging activities. However, WJ and SJ had less active antioxidant activity and free radical-scavenging activity. Reducing powers were increased depend on the concentration of extracts. Antioxidant activity and free radical-scavenging activity of JJ and DJ seemed to relate with total polyphenol and flavonoid contents.

Study of the Antioxidant Effect of Ojayeonjong-hwan (오자연종환(五子衍宗丸) 추출물의 항산화 효과에 관한 연구)

  • Objective: In this study, the antioxidant activity of Ojayeonjong-hwan extracts was compared, and the following results were obtained. Methods: For hydrothermal and ethanol extracts, DPPH free radical and ABTS cationic radical erasing activity and reducing power using the FRAP method were compared, and the association between the antioxidant power of each extract and total phenol content was investigated. Significant results were obtained through in vitro apoptosis analysis through FFITC staining, mitochondrial membrane potential analysis, and ROS level measurement using C2C12 myoblastoma. Results: 1. In a comparison of DPPH free radical and ABTS cationic radical scavenging activity, water, and 70% ethanol extracts of Ojayeonjong-hwan (WEO and EEO) showed superior radical scavenging ability. 2. In the results of reducing power using the FRAP method, WEO and EEO showed antioxidant activity, which was shown to be dependent on the total phenol content contained in the extracts. 3. In comparison to the protective effect against H2O2-induced oxidative stress in C2C12 myoblasts, water extracts had no significant effect, but 70% ethanol extracts inhibited H2O2-mediated cytotoxicity in a concentration-dependent manner. 4. The cytotoxic protective effect of EEO against oxidative stress in C2C12 myoblasts was correlated with its inhibitory effects on H2O2-induced apoptosis and cell-cycle arrest. 5. In H2O2-treated C2C12 myoblasts, the apoptosis inhibitory effects of EEO were associated with the suppression of mitochondrial dysfunction and DNA damage. 6. The protective effects of EEO against H2O2-induced oxidative stress in C2C12 myoblasts were directly related to the inhibition of ROS generation. Conclusions: Ojayeonjong-hwan extracts all have protective potential against oxidative stress.

Antioxidant and Neuronal cell protective effects of Stachys sieboldii Miq. according to drying methods (건조방법에 따른 초석잠(Stachys sieboldii Miq.)의 항산화 활성 및 신경세포 보호효과)

  • This study investigated the antioxidant and neuronal cell protective effects of activity of water and 70% ethanol extracts from Stachys sieboldii Miq. according to different drying method(hot air drying and freeze-drying). The total flavonoid and total polyphenol content in water extracts was significantly higher after freeze-drying compared to hot air drying(p<0.05). DPPH and ABTS radical scavenging were increased in a dose-dependent manner. The DPPH and ABTS radical scavenging activity of water extract were significantly higher after free-drying compared to hot air drying(p<0.05). In a cell viability using MTT, the water extract according to hot air drying and freeze-drying of Stachys sieboldii Miq. showed protective effect against H2O2-induced nurotoxicity. The results suggest that the water extracts of Stachys sieboldii Miq. after freeze-drying has antioxidant activities and may be useful for neurodegenerative disorders.

Antioxidant activities of ethanol extract of Shinsun-yukza-hwan, a Korean medicinal recipe (신선육자환 에탄올 추출물의 항산화 활성)

  • Objective : The Sinseon-yukza-hwan (SSY), a Korean medicinal formula which includes Radix Rehmanniae Preparata and other medicinal herbs, has long been used for treatment of alopecia and gray hair through oral administration. This study is designed to enhance the utilization of natural materials in hair and scalp-related cosmetics. Possibility of SSY as an antioxidant was examined from its 50% ethanol extract. Methods : The antioxidative capacities were evaluated by determining total phenolic and flavonoid contents, 1,1-diphenyl-2-picryl hydrazyl (DPPH), 2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), reducing power and hydroxyl radical scavenging activity. Results : Total polyphenol and flavonoid contents of SSY were 25.53 mg TE, tannic acid equivalent/g and 18.90 mg RE, rutin equivalent/g, respectively, which correlated strongly its antioxidative activity. The DPPH and ABTS free radical scavenging activities of SSY at $0.1mg/m{\ell}{\sim}5mg/ml$ were ranged from 20% to 85% and 10% to 58%, respectively. Also the hydroxyl radical scavenger activity and reducing power increased in SSY-treated group, which were significantly lower in SSY-compared to BHA-treated group. But the highest reducing power was shown as 79% from SSY-treated group, which was higher value than 65% from BHA-treatment. These results showed that SSY extract effectively inhibited the generation of free radicals in the all assay system with dose-dependent manners. Conclusions : Thus, the present study provide preclinical data to support the expanded application of SSY, which could be potential candidates for natural antioxidants.

Antioxidant Activity and Changes in Major Functional Components of Fermented Gastrodia elata Blume (발효 천마의 기능성 물질 함량 변화 및 항산화 활성)

  • This study was carried out to investigate the antioxidant activity and changes in major functional components of fermented Gastrodia elata Blume. Fermented G. elata Blume powder by Phellinus linteus repeated thrice ($3^{rd}$ FGP) showed more DPPH radical scavenging activity than a non-fermented G. elata Blume powder (NFGP), and once fermented G. elata Blume powder ($1^{st}$ FGP) at a concentration of 500 and 1,000 ppm. Free radical scavenging activity of $3^{rd}$ FGP was similar to that of BHA at a dose of 1,000 ppm. Moreover, the ABTS radical scavenging activity of the $3^{rd}$ FGP increased compared to NFGP and $1^{st}$ FGP at a concentration of 31.25 ppm. Total polyphenols and flavonoid contents were increased as fermentation progressed. Ergothioneine content was increased more than 8 times in the $1^{st}$ FGP, 3 times in the $3^{rd}$ FGP, respectively than NFGP. In conclusion, this study indicated that the antioxidant activity and functional component contents of G. elata Blume were increased depending on the fermentation number.
