가시오갈피에 발생하는 다양한 병해충을 분리, 동정하여 가시오갈피의 지속적인 안전재배를 위한 병 ${\cdot}$ 해충방제의 기초자료로 활용하고자 시험을 수행하였다. 가시오갈피에 발병하는 병원균을 분리동정한 결과 Phoma sp., Rhizoctonia solani, Botrytis cinerea 등 3종으로 밝혀졌으며, 병징의 특징에 따라 각각 검은무늬병, 잎마름병, 잿빛곰팡이병으로 명명하였다. 병원균에 의한 가시오갈피의 피해는 Phoma sp.에 의한 검은무늬병이 가장 컸으며, 개체별 병징의 형태나 병발생의 차이는 있었지만 6월 초부터 병반이 관찰되어 9월부터 병반면적이 30%를 넘어 계속 확대되었다. 수집균주의 배양온도별 균사생장량을 검토한 결과 균주 모두 $25^{\circ}C{\sim}30^{\circ}C$에서 생장량이 가장 많아, 여름 고온기에 많이 발생할 것으로 사료되었다. 해충별 발생시기 및 가해부위를 보면 진딧물은 6월 10일부터 관찰되어 어린 줄기를 흡즙하였고, 8월 상순에 다시 발생해 새로 분화되는 어린 화아에 집중적으로 발생하여 흡즙하였으며 노린재와 끝검은말매미충은 8월하, 6-7월에 각각 나타났으나 피해는 관찰되지 않았다.
Rhizoctonia solani AG2-2(IV)에 의한 갈색퍼짐병은 들잔디에서 가장 심각한 병이다. 이 연구의 목적은 파라핀 오일을 이용하여 갈색퍼짐병에 대한 실내에서 살균효과와 포장에서 병 방제 효과를 평가하는 것이다. 포장시험인 골프장에서 파라핀 오일은 200배와 50배액을 1 $m^2$당 1 L을 살포하였다. 실내시험에서는 균사생육억제효과시험을 위해서는 0.25%에서 2%까지 파라핀농도에서 시험하였다. 파라핀 오일의 0.5%의 농도와 2%의 농도에서 각각 43%와 67%의 균사생장 억제효과가 있었다. 두 지역의 포장시험에서 파라핀 오일의 9월에 1회 살포로서 갈색퍼짐병의 발달을 48%까지 억제하였다. 파라핀오일은 친환경적으로 들잔디에 갈색퍼짐병을 방제하는 대체물질로 사용할 수 있을것이다.
Since 2000, severe rots on aerial and underground parts of paprika( Capsicum annum L.) has occurred in most cultivation glasshouses throughout the country. Totally 169 isolates of a fungus were consistently isolated from the diseased plant tissues of fruits, stems, branches, and roots collected from 19 farms in six provinces. Anamorph stage of the fungus was identified as Fusarium solani based on morphological characteristics. However, the fungus readily produced sexual structure of perithecia on infected plant tissues and on agar medium. Since the fungus formed abundant perithecia by single isolate, it was considered as a homothallic strain of Nectria hematococn, the teleomorph of F. solani. Irregularly globose perithecia with orange to red color formed sparsely to gregariously on dead tissues of fruits and basal stems at the 3ate infection stage, which is a diagnostic sign for the disease. Abundancy of perithecium varied among isolates and they sized 125-220$\mu\textrm{m}$ in diam. Asci enveloping eight ascospores were cylindrical and measured 60-80x8-12$\mu\textrm{m}$. Ellipsoid to obovate ascospores are two-celled and measured 11-l8${\times}$4-7$\mu\textrm{m}$. Ascospores are hyaline, slightly constricted at the central septum, and revealed longitudinal striations that is a typical trait of the species. This fungus that has never been reported in Korea previously became a threat to paprika cultivation because of its strong pathogenicity and nationwide distribution.
Bacillus megaterium KL39, an antibiotic-producing plant growth promoting rhizobacterium (PGPR), was selected from soil. The antifungal antibiotic, denoted KL39, was purified from culture filtrate by column chromatography using Dion HP-20, Silica gel, Sephadex LH-20, and prep-HPLC. Thin layer chromatography, employing the solvent system of ethanol:ammonia:water=8:1:1, showed the $R_{f}$. value of 0.32. The antibiotic KL39 showed a negative reaction with ninhydrin solution, positive with iodine vapor, and also positive with Ehrlich reagent. It was soluble in methanol, ethanol, butanol, and acetonitrile, but insoluble in chloroform, toluene, hexane, ethyl ether, or acetone. Its UV spectrum had the maximum absorption at 208 nm. Amino acid composition, FAB-mass, $^{1}H-NMR,\;^{13}C-NMR$, and atomic analyses showed that the antibiotic KL39 (MW=1,071) has a structure very similar to iturin E. The antibiotic KL39 has a broad antifungal spectrum against a variety of plant pathogenic fungi including Rhizoctonia solani, Pyricularia oryzae, Monilinia froeticola, Botrytis cinenea, Altenaria kikuchiana, Fusarium oxysporum, and F. solani. An MIC value of $10\;{\mu}g/ml$ was determined for Phytophthora capsici. Macromolecular incorporation studies with P. capsici using radioactive [$^{3}H-adenine$] as the precursor, indicated that the antibiotic KL39 strongly inhibits the DNA biosynthesis of the fungal cell. Microscopic observation of the antifungal action showed abnormal hyphal swelling of P. capsici. The purified antibiotic KL39 was very effective for the biocontrol of in vivo Phytophthora-blight disease of pepper.

Foxglove aphid, Aulacorthum solani (Kaltenbach), is a Hemipteran insect that infected a wide variety of plants worldwide and caused serious yield losses in crops. The objective of this study was to identify the putative genes to foxglove aphid resistance in wild soybean, PI 366121 (Glycine soja Sieb. and Zucc.). One hundred and forty-one F4:8 recombinant inbred lines developed from a cross between susceptible variety, Williams 82 and foxglove aphid resistance wild soybean, PI 366121 were used. The two type of resistance response, antibiosis and antixenosis resistance were evaluated through choice and no-choice test, graded by the degree of total plant damage and primary infestation leaf damage; a genome-wide molecular linkage map was constructed with 29,898 single-nucleotide polymorphism markers utilizing a Axiom(R) 180K soyaSNP array. Using inclusive composite interval mapping analysis for foxglove aphid resistance, one major candidate QTL on chromosome 7 was identified. The major QTL on chromosome 7 showed both antixenosis and antibiosis resistance responses. The newly identified major QTL was consistent with previously reported QTL, Raso2, which showed around 5 times narrow down interval range with 8 candidate genes. Furthermore, total 1,115 soybean varieties including Glycine soja and Glycine max were exposed to germplasm screening, and 31 varieties, which showed significant antibiosis type foxglove aphid resistance were identified. This result could be useful in breeding for new foxglove aphid resistant soybean cultivars and developing novel insecticides.
Antagonistic microorganisms against $Rhizoctonia$$solani$ were isolated and their antifungal activities were investigated. Two hundred sixteen bacterial isolates were isolated from various soil samples and 19 isolates were found to antagonize the selected plant pathogenic fungi with varying degrees. Among them, isolate C9 was selected as an antagonistic microorganism with potential for use in further studies. Treatment with the selected isolate C9 resulted in significantly reduced incidence of stem-segment colonization by $R.$$solani$ AG2-2(IV) in Zoysia grass and enhanced growth of grass. Through its biochemical, physiological, and 16S rDNA characteristics, the selected bacterium was identified as $Bacillus$$subtilis$ subsp. $subtilis$. Mannitol (1%) and soytone (1%) were found to be the best carbon and nitrogen sources, respectively, for use in antibiotic production. An antibiotic compound, designated as DG4, was separated and purified from ethyl acetate extract of the culture broth of isolate C9. On the basis of spectral data, including proton nuclear magneric resonance ($^1H$ NMR), carbon nuclear magneric resonance ($^{13}C$ NMR), and mass analyses, its chemical structure was established as a stereoisomer of acetylbutanediol. Application of the ethyl acetate extract of isolate C9 to several plant pathogens resulted in dose-dependent inhibition. Treatment with the purified compound (an isomer of acetylbuanediol) resulted in significantly inhibited growth of tested pathogens. The cell free culture supernatant of isolate C9 showed a chitinase effect on chitin medium. Results from the present study demonstrated the significant potential of the purified compound from isolate C9 for use as a biocontrol agent as well as a plant growth promoter with the ability to trigger induced systemic resistance of plants.
The antifungal activity of crude methanolic extract and fractions from Yucca smalliana Fern. leaves, roots and flowers were investigated in vitro against a panel of plant pathogenic fungi. The minimal inhibitory concentration(MIC) was determined by an agar dilution method. Preliminary liquid culture and agar plate assays showed that the growth of Fu sarium oxysporum, Phytophthora capsici, Rhizoctonia solani and Botrytis cinerea were inhibited by Y. smalliana extracts. The extracts from flowers and leaves showed antifungal activity of 64.0% and 34.0% against F. oxysporum, 66.0% and 62.0% against P. capsici, and 27.0% and 41.0% against B. cinerea, respectively. The methanolic extract from Y. smallina leaves in distilled water was fractionated using solvents of increasing polarity: hexane, ethyl acetate and butanol. These fractions had a broad spectrum of antifungal activity, found to reside entirely in the butanol and aqueous fraction. The aqueous fraction showed inhibition rate of 60.0, 67.8, 84.6 and 58.3% against F. oxysporum, R. solani, C. gloeosporioides, and B. cinerea, respectively, and the butganol fracgtion showed 36.0, 46.0, 66.1 and 58.3%, respectively. Phenolics(e.g. flavonoids, steroids and terpenoids) were observed in the thin layer profile of the different fractions. Leave extract showed a prominent antioxidant activity totally scavenging the free radical of DPPH at a concentration of 1 mg/ml.
억새즙액을 이용하여 유용한 미생물의 대량배양을 위한 새로운 배지를 개발하고자 본 연구를 수행하였다. 억새즙액의 성분을 조사한 결과 칼륨 (37,952 mg/L), 질소 (14,000 mg/L), 인산 (6,800mg/L), 마그네슘 (5,969 mg/L), 칼슘 (5,910 mg/L) 등이 주성분을 차지하였다. 본 연구를 위하여 먼저 식물병원성균인 뿌리썩음병 원인균인 Rhizoctonia solani, 잿빛곰팡이 병을 일으키는 Botrytis cinerea에 대하여 강력한 저해 활성을 갖는 길항균 G-74를 토양으로부터 분리하였으며, 이 균의 형태관찰과 MIDI 분석을 통하여 Bacillus lentimorbus G-74 균주로 최종 동정하였다. 이 균의 억새즙액 배지에 대한 최적 배양조건을 조사한 결과 30% (V/V) 희석배지, $35^{\circ}C$ 배양온도, 5.0의 초기 배지 pH에서 가장 좋은 성장을 기록하였다. 또한 이 배지에 탄소원으로서 2.0% (W/V)의 corn starch 및 질소 원으로서 1.0% (W/V)의 yeast extract를 추가적으로 첨가하면 최상의 성장을 보여주었으며, 이 경우 대조구인 LB 배지보다도 균체성장이 66% 이상 증가하였다.
본 연구에서 thyme oil의 주성분인 thymol (1)과 유사한 구조를 갖는 4-isopropyl-3-methylphenol (2), 5-isopropyl-3-methylphenol (3), 4-isopropylphenol (4)과 2-isopropylphenol (5)을 출발 물질로 하여 sulfonyl ester 계열 유도체 25종을 합성하였다. IR, $^1H$-NMR과 GC/MS를 이용하여 이들 유도체들이 합성되었음을 확인하였다. 이들 유도체들에 대한 식물병원균의 생장 억제실험은 벼도열병균(Pyrcularia grisea), 벼잎집무늬마름병균(Rhizoctonia solani), 토마토잎마름역병균(Phytophthora infestans), 오이탄저병균(Colletotrichum orbiculare)과 오이흰가루병균(Sphaerotheca fusca)에 대하여 실시하였다. 그 결과, 2-isopropyl-5-methylphenyl o-toluenesulfonate (1a), 2-isopropylphenyl 2,4,5-trich lorobenzenesulfonate (5b)와 2-isopropylphenyl 2-methyl-5-nitrobenzenesulfonate (5e)는 각각 in vivo에서 벼도열병균(Pyrcularia grisea), 토마토잎마름역병균(Phytophthora infestans)과 오이흰가루병균(Sphaerotheca fusca)에 대하여 우수한 항균활성을 나타내었다.
