Cymbidium orchids with blight and rot symptoms were collected, and a total of 63 isolates of Fusarium app. was obtained from pseudobulbs, roots, and leaves of the diseased plants. The isolates were identified based on their morphological characteristics. Out of the 63 isolates of Fusatium sup., 51 isolates were identified as F. oxysporum, 10 isolates as F. solani, and the rest as F. proliferatum. F. oxysporum was isolated from all the Cymbidium spp., while F. solani and F. proliferatum were isolated only from Cymbidium ensifolium and C. ginatum, respectively. Isolates of the three Fusarium spp. were tested for pathogenicity to their hosts by artificial inoculation. The strongly pathogenic isolates of Fusarium spp. induced severe dry rot of pseudobulbs and roots of the host plants. The symptoms progressed up to the basal part of the leaves, which later caused blight of the entire plant. The dry root symptoms induced on the plants by artificial inoculation with the isolates of Fusarium app. were similar to those observed in the growers'greenhouses. This is the first report of dry rot of Cymbidium spp. caused by F. oxysporum, F. solani, and F. proliferatum in Korea.
To illustrate the action mechanism of pencycuron on Rhizoctonia solani, two experiments were conducted including the comparison of amino acids of $\beta$-tubulin between R-C (sensitive isolate) and Rh-131 (non-sensitive isolate), and the inhibitory effect of pencycuron on tubulin assembly in vitro. Both $\beta$-tubulin genes of R-C and Rh-131 proved to have 1,582 nucleotides encoding a protein of 445 amino acids, showing 98% homology in amino acid sequences between them. It was found that codons at 103, 236, and 267 for lysine (AGG), valine (GTC) and isoleucine (ATT) in R-C were replaced by codons for methionine (ATG), isoleucine (ATT) and methionine (ATG) in Rh-131, respectively. No inhibitory effect of pencycuron on the tubulin assembly was observed. It suggests that pencycuron may have no direct inhibitory effects on the assembly of tubulin at least in vitro.
In order to find an environment-friendly method to suppress astragal stem rot caused by the isolates of Rhizoctonia solani AG 1 and AG 4, we tested an antagonistic fungus Gliocladium virens G1 was evaluated as a biocontrol agent and estimated inorganic compounds and organic materials were tested for their effect of the disease suppression. G. virens G1 effectively inhibited mycelial growth in a dual culture and caused mycelial lysis in the culture filtrate. No adverse effect was observed when examined for seed germination and seedling growth. Promoted seedling growth was observed with the seed treatment. Seeds of astragal plant were germinated higher in the sterile soil than the natural soil. Of 14 inorganics tested, alum, aluminum sulfate and calcium oxide significantly suppressed the mycelial growth and sclerotial germination. Milled pine bark and oak sawdust also suppressed the mycelial growth. Soil amended with 1% of G. virens G1 composted with pine bark (w/v) significantly controlled astragal stem rot in the glasshouse experiments.
Tolclofos-methyl applied as seed dipping at 1,000 ppm for 3 hrs before sowing and soil drenching at the rate of 300 g ai./10 a in the middle of April protected emerging seedlings of Panax ginseng from damping-off caused by Rhiiutonia solani(AG2-1) in Yangjik Soil artificially infested with the pathogen. Germination rates with tolclofos-methyl, pencycuron, and control were 53.7%, 45.8%, and 7.5%, respectively, while the rate of the seeds at non-infested soil was 62.6%. The effectiveness of Tolclofos-methyl against the pathogen in the soil lasted upto 32 days in vitro. However, the transpiratio of ginseng seedlings increased greatly with chemical treatment, showing 0.02, 0.12, and 0.24 m1/cm2 leaf area/day at 0, 1,2, and 4 ppm a.i. of the fungicide, respectively.
A fungal strain of Trichoderma having strong chitinolytic activity was isolated from field soil enriched with crabshell for several years. Based on 5.8S rRNA, partial 18S, 28S rRNA genes, ITS1, ITS2 sequence analysis and morphological characteristics, the fungus was identified as Trichoderma asperellum and named as Trichoderma asperellum T-5 (TaT-5). The fungus released lytic enzymes such as chitinase and ${\beta}$-1, 3-glucanse, and produced six antifungal substances in chitin broth medium. To demonstrate the protective effect of TaT-5 against damping-off in cucumber plant caused by Rhizoctonia solani, TaT-5 culture broth (TA), chitin medium (CM) and distilled water (DW) were applied to each pot at 10 days after sowing, respectively. Then, the homogenized hyphae of R. solani were infected to each pot at 1 week after TaT-5 inoculation. During experimental period, fresh weight of shoot and root in cucumber plant more increased at TA treatment compared to other treatments. PR-proteins (${\beta}$-1, 3-glucanase and chitinase) activities in cucumber leaves markedly increased at CM and DW treatments, but the activity slightly increased and then decreased at TA treatment at 3 days after infection of R. solani. The activity of PR-proteins activities in cucumber roots at all treatments decreased with time where the degree of decrement was more alleviated at TA treatment than CM and DW. These results suggest that the lytic enzymes (chitinase and ${\beta}$-1, 3-glucanse) and antifungal substances produced by TaT-5 can reduce the pathogenic attack by R. solani in cucumber plants.
From soil samples, 380 antagonistic microorgnisms were isolated. Among the isolates, 42 strains had mycelia growing inhibition ability against Fusariun solani, ginseng root rot causing pathogen. Isolates CHA 1 and S-PFHR 6 were proposed as antagonists for this study and they were identified as Promicromonospora sp. and Pseudomonas pseudoalcaligenes respectively. As an antagonism against hyphae of F. solani in dual culture test, CHA 1 and S-PFHR 6 inhibited linear growing, caused abnormal branching, and the membrane projection which formed by cell wall destruction. The secondary metabolites contained in the culture filtrates which prepared from PD broth and Nutrient broth inhibited the spore germination to 14.3%. The culture filtrate of S-PFHR 6 which prepared by a little amount of soil extract addition to nutrient rich medium had more strongly. inhibited the spore germination and spore germination decreased to less than 4.0% in it. The soil used in this study had fungistasis and the germination rate of macroconidia and chlamydospore of F.solani was 19.4% and 17.7% respectively. The steam sterilized soil lost fungistasis and germination rate of conidia increased to more than 97.9%. The soils amended with the propagule of CHA 1 and S-PFHR 6 increased fungistasis and the germination rate of macroconidia decreased to 14.7% and 11.7% respectively in each treatments. But the soil ammended with glucose and asparagine annulled fungistatic ability and the germination rate of macroconidia increased to more than 48.0%. As an antagonistic activity of the secondary metabolites of two antagonistic isolates in soil, the germination rate of macroconidia of F. solani was 9.3% in the soil amended with the culture filtrate of CHA 1 but the culture filtrate of S-PFHR 6 had no such activity. In the soil which treated with antagonist propagule or culture filtrate, the chlamydospore germination rate was lower than that in natural soil. The addition of glucose and asparagine to antagonist propagule treated soil did not enhanced the chlamydospore germination.
Rhizoctonia solani [Thanatephorus cucumeris (frank) Donk], one of the major soil-borne plant pathogens with world-wide distribution, can cause great damages on various crops. In Korea, sheath blight on rice caused by this pathogen is the major concern, and active studies on this pathogen have been performed. However, most of these studies were concerned with pathogenicity of the isolates instead of molecular analyses of different AGs of R. solani. Therefore, in this study, thirty isolates of Rhizoctonia solani collected from various sources were used for the analyses of genetic relationships among themselves and for the rapid anastomosis grouping with RAPD method. As a result, thirty isolates of known and unknown AGs were grouped into five subgroups and each group included AG-1, AG-2, AG-3, AG-4, and AG-5. RS-1 isolate was found to be closely related to AG-5. Isolates RS-4, RS-14, RS-17, and RS-16 were found to be closely related to AG-2-2(III B). Isolate RS-13 was closely related to AG-4, isolates RS-8 and RS-10 were closely related to AG-1(I B), and isolates RS-7 and RS-21 were closely related to AG-2-2(IV). Isolate RS-19 was closely related to AG-1(I C), and isolates RS-3, RS-5, RS-18, RS-6, and RS-15 were found to be closely related to AG-1.
Among 110 isolates of actinomycetes isolated from ginseng pathogen-suppressive soils, the three actinomycetes showing the effective controls to Fusarium solani or Cylindrocarpon destructans causing ginseng root rots were identified according to their morphological, cultural and physiological characteristics on various culture media. Spore chains of K 6-2, S 2-1 and Y 2-2 were Spira (S), Retinaculum-apertum (RA) and Rectus-flexibilis (RF), respectively. Spore surfaces of K 6-2 were spiny, whereas S 2-1 and Y 2-2 were all smooth. Aerial mass colors of 3 isolates were gray series. As a result of various tests, they were identified as Streptomyces variabilis, Streptomyces virginiae and Streptomyces griseo/us, respectively.

In this study, established soil-borne Bacillus sp. FF-9 with strong antifungal activity was isolated for identification and to determine optimal culture condition. By using 16s rDNA sequencing method, FF-9 of the selected bacteria was identified as genus Bacillus sp., Bacillus sp. FF-9 was cultured at $30^{\circ}C$, for 24 h in the LB medium. Cell growth increased quickly after 6 h and the highest cell growth was indicated at 12 h. The most antifungal activity against Rhizoctoina solani AGII-II appeared at 18 h and the optimal temperature and pH were 30 and pH 8.0, respectively. A testing of carbon and nitrogen sources showed the highest antifungal activity at 1% lactose and 1% yeast extract Furthermore an addition of salt showed the most antibiotic activity in the 0.15% $K_2HPO_4$.
This study was carried out to investigate control efficacy of fungicides mixed with pyroligneous acid on Large patch caused by Rhizoctonia solani AG2-2. R. solani AG2-2 was not inhibited by concentration of 500 times and 1,000 times diluents of pyroligneous acid. It is thought that R. solani AG2-2 was not inhibited by Pyroligneous acid alone. Pencycuron and tebuconazol mixed with pyroligneous acid had a good effect in prevention of R. solani AG2-2 in zoysiagrass. Concurrently, it was observed that those were reduced $50\%$ of usage of fungicides. 500 times and 1,000 times diluents of pyroligneous acid treated group had a significantly effect compared to control turf in the leaf length, leaf width and dry weight. In conclusion, usage of fungicides could be reduced about $50\%$ by a fungicides mixed with pyroligneous acid for protection of R. solani AG2-2. Therefore, It is thought that fungicides mixed with pyroligneous acids can be reduced an economic load and also an environmental pollution such as a tolerance to pathogenic bacteria and a destruction of an ecological system of soil microbes according to use fungicides.
