The Effect of Carbon Monoxide on Contraction, Cytosolic $Ca^{2+}$ Level and Ionic Currents in Guinea Pig Ileal Smooth Muscle

  • The aim of this study was to clarify the mechanism of the inhibitory action of carbon monoxide (CO) on contraction, by measuring cytosolic $Ca^{2+}$ level $([Ca^{2+}]_i)$ and ionic currents in guinea-pig ileum. CO (10%) inhibited 40 mM KCl-induced contraction and this effect was blocked by ODQ $(1\;{\mu}M),$ a soluble guanylyl cyclase (sGC) inhibitor. CO inhibited the 40 mM KCl-induced contraction without changing $[Ca^{2+}]_i.$ Cumulative addition of KCl induced a graded increase in $[Ca^{2+}]_i$ and muscle tension. In the presence of CO, cumulative addition of KCl induced smaller contraction than in the absence of CO. On the other hand, the increase in $[Ca^{2+}]_i$ induced by cumulative addition of KCl was only slightly decreased in the presence of CO, and the $[Ca^{2+}]_i-tension$ relationship shifted downwards. Using the patch clamp technique with a holding potential of -60 mV, we found that CO had little effect on the peak Ba currents $(I_{Ba})$ when voltage was stepped from -60 mV to 0 mV. In addition, CO showed no effect on the depolarization-activated outward $K^+$ currents in the all potential ranges. We conclude that CO inhibits smooth muscle contraction mainly by decreasing the $Ca^{2+}$ sensitivity of contractile elements via a cGMP-dependent pathway, not by involving L-type $Ca^{2+}$ and outward-potassium currents in guinea-pig ileum.

흰쥐 적출 배뇨근에서 콜린성 및 퓨린성 수용체의 존재 (Existence of Cholinergic and Purinergic Receptor on the Detrusor Muscle of Rat Urinary Bladder)

  • 흰쥐(Sprague-Dawley)의 방광에서 적출한 배뇨근절편을 적출 근편 실험조에 현수하고, 등척성 장력 측정기를 사용하여 그 수축력을 묘기하였다. 배뇨근 절편은 전기장자극에 의해 수축하였으며, 이 수축반응은 콜린에스테라제 억제약물인 physostigmine에 의해 증가하였고 신경말단에서의 choline 재흡수를 방해하는 hemicholinium에 의해 억제되었으며, 신경절봉쇄약물인 hexamethonium에 의해서는 영향을 받지 않았으나 신경축색전도 억제제인 tetrodotoxin에 의해서 소실되었다. 이러한 전기장자극유발 수축은 콜린성 무스카린성 수용체봉쇄약물인 atropine에 의해 부분적으로 길항되었으며, atropine에 의해 길항되지 않는 부분은 ATP 탈감작에 의해 완전히 소실되었다. 배뇨근 절편은 콜린성 무스카린성 수용체 흥분제인 bethanechol과 퓨린성 수용체 흥분제인 ATP에 의하여 농도의존적 수축력 증가를 나타내었으며, 이중 bethanechol 유발수축은 ATP 탈감작에 의해 영향을 받지 않았고, ATP 유발수축은 tetrodotoxin에 의하여 영향을 받지 않았다. 이상의 결과로 보아 흰쥐의 적출배뇨근에는 흥분성 신경전달체계로서 퓨린성 수용체와 콜린성 수용체가 존재하며, 이들은 서로 영향을 미침이 없이 독자적으로 배뇨근 수축에 기여하고 있다고 사료된다.

Flavonoids의 약리작용(V) - 전기자극 및 과민반응으로 유발된 흰쥐 회장의 평활근수축에서 Flavones 및 Flavonols의 진경효과 - (Pharmacological Activities of Flavonoids (V): Spasmolytic Activities of Flavones and Flavonols on Rat Ileal Smooth Muscle Contraction Induced by Electrical Stimulation and Anaphylactic Reaction)

  • Some flavonoids have spasmolytic activities in various smooth muscles, but structure-activity relationships on their spasmolytic activity and its mechanism are unclear. In this study, effects of flavones (flavone and apigenin) and flavonols (quercetin and rutin) on the rat ileal smooth muscle contraction were studied in vitro and in vitro. In the electric stimulation-induced contraction, all of four flavonoids inhibited concentration-dependently the rat ileal smooth muscle contraction induced by electric stimulation (10 mV, 0.1 cps, 0.1 msec duration), IC$_{50}$ of quercetin, apigenin, flavone and rutin were 0.98${\times}$10$^{-5}$, 1.20${\times}$10$^{-5}$, 1.55${\times}$10$^{-5}$ and 1.85${\times}$10$^{-5}$ M, respectively. Flavonoids at a concentration of 2${\times}$10$^{-5}$ M also significantly inhibited the anaphylactic contraction and decreased concentration-dependently the mast cell degranulation by anaphylactic reaction, IC$_{50}$ of quercetin, apigenin, flavone and rutin were 4.0${\times}$10$^{-5}$, 7.5${\times}$10$^{-5}$, 8.0${\times}$10$^{-5}$ and 9.5${\times}$10$^{-5}$ M, respectively. These results indicated that flavones and flavonols inhibited the rat ileal smooth muscle contraction induced by electric stimulation because of their antagonism against acetylcholine and have spasmolytic activities on anaphylactic contraction which may be due to their mast cell-stabilizing activities. Furthermore, double bond of C$_{2,3}$ in benzene ring of flavonoids may be important in the their antispasmodic activities on the rat ileal smooth muscle contraction induced by electric stimulation and anaphylactic reaction.

COINCIDENCE THEOREMS FOR COMPARABLE GENERALIZED NON LINEAR CONTRACTIONS IN ORDERED PARTIAL METRIC SPACES

  • In this paper, we prove some coincidence point theorems involving ${\varphi}-contraction$ in ordered partial metric spaces. We also extend newly introduced notion of g-comparability of a pair of maps for linear contraction in ordered metric spaces to non-linear contraction in ordered partial metric spaces. Thus, our results extend, modify and generalize some recent well known coincidence point theorems of ordered metric spaces.

  • In this paper, we introduce a fixed point index of weakly inward 1-set-contraction mappings. With the aid of the new index, we obtain some new fixed point theorems, nonzero fixed point theorems and multiple positive fixed points for this class of mappings. As an application of nonzero fixed point theorems, we discuss an eigenvalue problem.

  • We establish a coupled coincidence and common coupled fixed point theorem for hybrid pair of mappings under generalized non-linear contraction. An example supporting to our result has also been cited. We improve, extend and generalize several known results.

  • Vocal fold vibration is essentially the propagation of a mucosal wave, starting from the lower surface of the vocal fold. The mucosal upheaval (MU), where the mucosal wave starts and propagates upward, appears only when the vocal fold vibrates. We investigated the location of the mucosal upheaval in response In variations in vocal fold tension. Vibrations were elicited under three conditions: during bilateral thyroarytenoid (TA) muscle contraction, without TA muscle contraction and during vocal fold lengthening. APPLICATION OF GENERALIZED WEAK CONTRACTION IN INTEGRAL EQUATION

  • This manuscript is divided into three segments. In the first segment, we prove a unique common fixed point theorem satisfying generalized weak contraction on partially ordered metric spaces and also give an example to support our results presented here. In the second segment of the article, some common coupled fixed point results are derived from our main results. In the last segment, we investigate the solution of integral equation as an application. Our results generalize, extend and improve several well-known results of the existing literature.

  • In this paper, we will introduce the notion of 𝜓-type and Jaggi type hybrid contraction in a bipolar metric space and show the existence and uniqueness of fixed point for such type of contractions. In the end, we will provide some corollaries and support our theorems by examples.
