  • Chlorophyll contents in Korean pine (Pinus koraiensis) needles of an advance growth stand and a plantation were compared using 80% acetone and DMSO (dimethylsulfoxide). Total chlorophyll contents in DMSO were 4-6 times higher than those in 80% acetone. There were no significant differences in chlorophyll contents between two stand types in both 80% acetone and DMSO. It appeared that 6 hours in DMSO would be appropriate to successfully extract chlorophyll from Korean pine needles of advance growth and planted trees.

  • The pH titration curves of ampicillin, amoxicillin and cephalexin in aqueous acetone, ethanol and N, N'-dimethylformamide by use of 0.02N-KOH solution showed that 80% acetone was the most effective solvent for alkali titration of them. The pH jumps of samples in 80% acetone were sharp enough to determine the contents of them by use in 80% acetone (9, 4) was coincided with each equivalent point of ampicillin, amoxicillin and cephalexin, but those of phenolphthalein, thymolphthalein, thymol blue and bgrom cresol purple were not. The color change of brom thymol blue at equivalent points was very clear except the case of amoxicillin that the determination of smaples in 80% acetone with 0.02 N-KOH solution by the aid of brom thymol blue could be available.

Studies on the Physiological Functionality of Pine Needle and Mugwort Extracts (솔잎과 쑥 추출물의 기능성 점토)

  • This study was conducted to investigate the physiological functionality of the hot water and 70% acetone extracts obtained from the pine needle and mugwort. Flavanol tannin content was above 60% of the total polyphenol in pine needle extracts. 70% Acetone extract from pine needle exhibited inhibition percentage of about 82.2% on the mutagenicity of Trp-P-1. Angiotensin I converting enzyme(ACE) inhibition activity was 61% and 50% in the hot water extract and in the 70% acetone extract respectively. Electron donating ability(EDA) of the hot water and 70% acetone extracts obtained from the pine needle was significantly good above 80%. 70% Acetone extract from pine needle showed inhibitary effect against the polyphenol oxidase(PPO) of water dropwort. The nitrite scavenging ability was appeared in all the extracts examined and it showed $80{\sim}90%$ at pH 3.0. The oxidative stability was determined by POV. Results showed that ethyl acetate fraction is better antioxidants than chloroform and butanol fractions in the soybean oil. Then, in the mugwort extracts, antimutagenicity of 70% acetone extract was similar to that of pine needle. Ratio of ACE inhibition activity was higher than that of pine needle extracts. EDA of hot water and 70% acetone extracts was 45%. Extracts from the mugwort were lower in nitrite scavenging ability about $20{\sim}30%$ than extracts from pine needle.

Chemical Composition and Antioxidant Activities of Prunus salicina Formosa Produced in Gimcheon (김천산 자두 후무사의 화학적 성분과 항산화 활성)

  • The present study was designed to investigate the chemical composition and antioxidant activities of Prunus salicina Formosa produced in Gimcheon. The free sugar amounts in Prunus salicina Formosa were fructose>glucose>sucrose>maltose, and malic acid contents in Prunus salicina Formosa were the highest among all the organic acids found in the present study. Each Prunus salicina Formosa extract was obtained by treating Prunus salicina Formosa with distilled water, 80% ethanol, 60% acetone, and 80% methanol at 25 and $50^{\circ}C$. The highest contents of total polyphenol and flavonoid were observed in the 60% acetone extract and 80% ethanol extract, respectively. The 60% acetone extract exhibited the greatest DPPH radical scavenging ability, reducing power, and nitrite scavenging ability. However, SOD-like activity was not considerably different for all the extracts studied. The results from the present study could be helpful for developing antioxidant products using Prunus salicina Formosa produced in Gimcheon.

  • A gas chromatographic analytical method for IAA was developed. IAA was extracted with 80% acetone & methanol. Sample was macerated with 80% acetone & methanol to extract IAA. After filtration, the solvent was removed and residual IAA was esterified with 14% $BF_3$/MeOH. The IAA esters can be easily separated with intermediate polar column.

  • Chemical heat pump is a system to upgrade the low level energy such as industrial waste heat and solar energy by using coupled endothermic and exothermic chemical reactions. Dehydrogenation of 2-propanol can absorb heat near 80.deg. C and is transformed into acetone and hydrogen. Hydrogenation of acetone can liberate heat near 200.deg. C. Dehydrogenation of 2-propanol is difficult around 80.deg. C because .DELTA.G has positive value, but dehydrogenation reaction in liquid phase can overcome this problem because vaporized acetone and hydrogen can be rapidly eliminated. In this work, dehydrogenation of 2-propanol was investigated in liquid phase with Raney nickel catalyst. The energy efficiency of the chemical heat pump was estimated by computer simulation.

Biochemical analysis and physiological activity of perilla leaves (들깨잎의 품종에 따른 성분분석 및 생리활성물질 탐색)

  • The biochemical components of Namcheondlggae, Miryangdlkkae 25, Boradlggae and Ipdlkkae 1 were measured. The samples were extracted with hot water, 60% acetone or 80% ethanol for screening physiological activity. The crude protein content (4.36%) was found in the Miryangdlkkae 25 and calcium content (497.5 mg%) was found in the Namcheondlggae among the tested 4 perilla loaves. Fructose was 30.86 mg% in the Namcheondlggae and free amino acids at all perilla leaves was detected seventeen. In Boradlggae, glutamic acid and alanine were 25.37 and 11.91 mg%. Totally nine non-volatile organic acids were also detected and the contents of malic acid and glutaric acid were 28.34 and 14.57 mg% in Boradlggae. The Miryangdlkkae 25 had the highest vitamin C amount which was 113.24 mg%. Angiotensin converting enzyme (ACE) inhibition activity of 60% acetone extract of Miryangdlkkae 25 was 39.20% when added as addition of 200 ppm level and xanthine oxidase inhibition activity of 80% ethanol extract of Boradlggae was 46.71%. Electron denoting activity of 60% acetone extract from Namcheohndlggae was the strongest inhibition activity as 98.19% when 200 ppm level of the sample extracts were added.

Highly Selective and Sensitive Detection of Acetone by ZnWO4-WO3 Hetero-composite Spheres

  • ZnWO4-WO3 hetero-composite microspheres were prepared by ultrasonic spray pyrolysis of a solution containing Zn and W cations, followed by heat treatment at 600℃. The gas-sensing characteristics of 5 at% of Zn-added WO3 (5Zn-WO3; ZnWO4-WO3 hetero-composite) microspheres to 1 ppm acetone, ethanol, 20 ppm hydrogen (H2), 5 ppm carbon monoxide (CO), 25 ppb toluene, and 5 ppm ammonia (NH3) were measured at 325-400℃ under 80% relative humidity (RH). The sensor using 5Zn-WO3 microspheres exhibited highly selective and sensitive gas-sensing properties to acetone at 375℃ even under high humidity conditions. These superior gas-sensing properties were attributed to the increased resistance (electronic sensitization) through n-n heterojunction formation between WO3 and ZnWO4 phases and the acidic property of WO3, which exhibited a low gas response to interfering ethanol gas. The superior acetone gas-sensing characteristics of the 5Zn-WO3 sensor can be utilized in breath acetone analyzers for rapid, real-time ketogenic diet monitoring.

Acetone Enhancement of Cumene Hydroperoxide-supported Microsomal Cytochrome P450-dependent Benzo(a)pyrene Hydroxylation

  • In vitro effects of acetone on cytochrome P450 (P450)-dependent benzo(a)pyrene (B(a)P) hydroxylation supported by cumene hydroperoxide (CuOOH) or NADPH/$O_2 $ systems were studied using 3-methylcholanthrene-pretreated rat liver microsomes. The maximal rate of B(a)P hydroxylation at constant concentration ($80\;{\mu}M)$ of the substrate was observed in the presence of $30\;{\mu}M$ CuOOH. However, at concentrations higher than $30\;{\mu}M$ CuOOH the hydroxylation rates were rapidly decreased. In contrast to CuOOH, at a concentration of $200\;{\mu}M$ NADPH, B(a)P hydroxylation rate reached a plateau. At concentrations higher than $200\;{\mu}M$ NADPH, the rates of substrate hydroxylation were maintained at the maximal rate with no inhibition. Acetone at 1% (v/v) enhanced both CuOOH- and NADPH/$O_2$-supported B(a)P hydroxylation at the optimal concentrations of the cofactors. At concentrations higher than 1% (v/v) acetone, substrate hydroxylation was sterero specific under the support of these two cofactors; it was strongly enhanced with $30\;{\mu}M$ CuOOH, but rather inhibited in the $200\;{\mu}M$> NADPH/$0_2 $ system. The lipid peroxidation rate induced during CuOOH-supported P450-dependent B(a)P hydroxylation was increased as CuOOH concentrations were increased. Acetone in the concentration range of 2.5~7.5%(v/v) inhibited lipid peroxidation during CuOOH supported B(a)P hydroxylation. The finding that CuOOH-supported B(a)P hydroxylation is greatly enhanced by acetone suggests that acetone may contribute more to the activation of oxygen (for the insertion of oxygen into the substrate) in the presence of CuOOH than with NADPH/$O_2$. Acetone may also contribute to the partial inhibition of destruction of microsomal membranes by lipid peroxidation.

Efficiency of Eluting Solvents for Solid-Phase Extraction During Multiresidue Analysis of 16 Pesticides in Cucumber (고상추출카트리지를 이용한 잔류농약 다중분석과정에서 정제용매에 따른 회수율 비교)

  • A study was carried out to optimize the cleanup step using a solid-phase extraction (SPE) cartridge, $SupelClean^{TM}$ Florisil, for multiresidue analysis of 16 pesticides in cucumber matrix. Eluting efficiencies of two solvent systems including acetone/n-hexane and ethyl acetate/n-hexane mixtures were critically evaluated by recoveries of target anaytes from the SPE Florisil column. Based on the recovery as a measure of eluting efficiency, an acetone/n-hexane (20/80,v/v) mixture provided more than 80% recovery for 15 pesticides except bifenthrin. In case of ethyl acetate/n-hexane, 14 pesticides showed recoveries higher than 75% while those far alachlor and bifenthrin were less than 30%.
