3T3-L1 preadipocyte were differentiated to adipocytes, and then treated with 0, 10, 20, and $40{\mu}g/mL$ of peanut sprout ethanol extract (PSEE). The main component of PSEE is resveratrol which contained 5.55 mg/mL of resveratrol. The MTT assay, Oil-Red O staining, glycerol-3-phosphate dehydrogenase (GPDH) activity, and the triglyceride concentration were determined in 3T3-L1 cells. MMP-2 and MMP-9 activities as well as mRNA expressions of C/EBP ${\beta}$ and C/EBP ${\alpha}$ were also investigated. As the concentration of PSEE in adipocytes increased, the cell proliferation was decreased in a dose-dependent manner from 4 days of incubation (P < 0.05). The GDPH activity (P < 0.05) and the triglyceride concentration (P < 0.05) were decreased as the PSEE treatment concentration increased. The mRNA expression of C/EBP${\beta}$ in 3T3-L1 cells was significantly low in groups of PSEE-treated, compared with control group (P < 0.05). The MMP-9 (P < 0.05) and MMP-2 (P < 0.05) activities were decreased in a dose-dependent manner as the PSEE concentration increased from $20{\mu}g/mL$. In conclusion, it was found that PSEE has an effect on restricting proliferation and differentiation of adipocytes.
The present was undertaken to establish a model for the control of adipocytes differentiation by using antibody from egg yolk. The emulsion of membrane protein of 3T3L-1 cell membrane protein with the complete Freund's adjuvant was firstly immunized in layer. Second and third boosting were undertaken with two weeks intervals by injection of the emulsion of the same antigen with the incomplete Freund's adjuvant. After 4 week of the first immunization, eggs were collected and antibody (IgY) was purified from egg yolk. The purity of IgY was 60-98% determined by single radial immunodiffusion (SRID) methods. Titer value of the antibody showed high reactiviy for the preadipocytes membrane protein measured by ELISA. When the IgY was added in the test media containing either 2.5% porcine serum or 10% FBS(control), the differentiation of 3T3L-1 cells and Glycerol-3-phosphate dehydrogenase(GPDH) activities was significantly decreased compared to the control cells(p〈0.05). When mice were subcutaneously injected with IgY raised against membrane protein of 3T3L-1 cells for 3 weeks, adipose tissue mass around ovary was tended to be decreased in female mice compared to those of control mice. It is suggested that a potential for manipulating of lipid accumulation through decrease in 3T3L-1 cell differentiation and fat accumulation in female mice by IgY treatment.
The anti-adipogenic effect of dongchimi nano juice prepared using a nano-filtering process was investigated by measuring leptin and glycerol levels and the expression of a peroxisome proliferator-activated $receptor-\gamma\;(PPAR\gamma)$ gene as indicators of lipid accumulation or lipolysis. Red pepper powder, seeds of red pepper, garlic, and ginger were added in the preparation of dongchimi. Dongchimi was fermented to reach the optimal fermentation period, followed by nano-filtration in the range of $0.0005\sim0.1\;{\mu}m$. The lactic acid bacteria of dongchimi nano juice were removed completely by a nano-filtering process. Treatment of dongchimi nano juice induced glycerol release in the 3T3-L1 adipocytes and decreased the mRNA expression level of $PPAR\gamma$. These results suggested that dongchimi nano juice may enhance lipolysis and modulate adipogenesis in 3T3-L1 cells.
The aim of this study was to examine the cytotoxicity and potential inhibitory effects from four types of edible domestic brown seaweeds, Undaria pinnatifida (UP), Laminaria japonica (LJ), Sargassum fulvellum (SF), and Hizikia fusiforme (HF), on preadipocyte differentiation in 3T3-L1 cell line. Water and ethanol extracts from the four types of seaweeds were prepared and tested for cell viability in the 3T3-L1 cell line by using MTT assay. In addition, various doses of the water extract of seaweeds (WES) and ethanol extract of seaweeds (EES) were treated at the beginning of 3T3-L1 differentiation and continued until the cells were fully differentiated to adipocytes. Oil Red-O staining was performed to determine the potential cell differentiation inhibitory effects of the WES and EES by measuring the levels of lipid droplet accumulation in 3T3-L1 adipocytes. $PPAR{\gamma}$ mRNA expression levels were significantly reduced by WESs of UP, LJ, and HF as well as EESs of LJ and HF. As a result, we observed the superior cell differentiation inhibitory effects of WES compared to that of EES in a dose-dependent manner without any significant cytotoxicity in mouse adipocytes.
전통적인 한방약물로 이용되는 인삼(사포닌 Re)과 의이인 추출물을 분화단계별 지방세포에 $100\;{\mu}g/ml$ 씩 12일간 이틀에 한번씩 각각의 물질을 처리하였다. 비만지표 유전자로 알려져 있는 여러 유전자의 발현양을 mRNA와 단백질 수준에서 조사하였다. 완전성숙 지방세포에서 약물의 처리에 따라 $TNF-{\alpha}$ 발현이 비처리 세포에 비해 현저히 억제되었고, lipoprotein lipase는 mRNA와 단백질수준에서 그 발현 양이 증가하였다. 약물처리 세포에서 leptin과 resistin의 유전자 발현은 비처리세포에 비해 유의적인 감소되었다. 이러한 결과는 인삼 사포닌 뿐 아니라 의이인 추출액에서도 유사한 영향이 관찰되어 특히, 곡물인 의이인 추출액이 인삼 사포닌과 거의 비슷한 항비만 활성을 나타내어 장복으로 인한 세포독성을 최소화하면서 지방세포에 긍정적 영향을 미칠 뿐 아니라 인슐린 저항성 개선 효과도 기대되어진다. 향후 대사성 질환과 밀접한 관련이 있는 비만 관리 향상에 이용 할 수 있을 것으로 사료되어진다. 따라서 이 결과들을 바탕으로 동물실험을 통한 더 자세한 조절기능의 확인과 특히, 의이인 추출액에서의 생리활성물질에 대해서도 많은 체계적인 분석과 관련연구가 필요 할 것으로 사료된다.
한의학에서 당뇨병 (소갈) 처방으로 사용되는 옥천산 처방의 성분 중의 하나인 오미자의 포도당 이용에 대한 효과를 조사하기 위해서 오미자를 70% 에탄올로 추출한 후 메탄올과 물을 섞은 용액으로 단계별로 XAD-4 column 으로 분획하였다. 오미자를 복용한 후 혈당을 낮추기 위해서는 인슐린처럼 작용하는 인슐린성 물질이거나, 인슐린 작용을 향상시키는 인슐린 민감성 물질로 작용하거나, 또는 ${\alpha}-glucoamylase$ 활성을 억제하여 탄수화물의 소화를 방해 작용이 있어야 하므로 본 연구에서는 섬유아세포와 지방세포 3T3-L1에서 각 분획층이 이러한 3가지 기전에 관여하는 지를 조사하였다. 오미자 분획물은 인슐린성 물질로 작용하거나 ${\alpha}-glucoamylase$의 활성을 저하시켜 탄수화물의 소화를 방해하지 않았다. 그러나 오미자 분획층은 인슐린의 작용을 향상시켜 포도당의 흡수를 증가시키는 효과가 매우 컸다. 특히 오미자 분획층 중 Fr. 4(메탄올 60%)와 Fr. 5(메탄올 80%)는 지방세포 3T3-L1에서 포도당의 흡수를 현저하게 증가시켜 인슐린을 50 ng/ml를 처리한 것보다도 효과적으로 포도당 흡수를 증가시켰다. Fr. 4와 Fr. 5에서 포도당 흡수가 증가한 것은 Fr. 4와 5가 인슐린 작용을 향상시켜 세포막에 GLUT4양을 증가시진 결과이었다. 결론적으로 오미자 중 특히 Fr. 4와 Fr. 5에 인슐린 민감성 제제가 함유되어 있을 것으로 추정된다.
Effects of perilla leaf extracts (PLE) on adipocytes differentiation of 3T3-L1 cells were examined. Ethanol extract of PLE treatment significantly decreased lipid accumulation, a marker of adipogenesis, in a dose-dependent manner. Moreover, gene expression levels of peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), the key adipogenic transcription factor, were markedly decreased by PLE. PLE also suppressed adipocyte fatty acid binding protein (aP2) and glycerol-3-phosphate dehydrogenase (GPDH), which are adipogenic marker proteins. These results suggest that PLE treatment suppressed differentiation of 3T3-L1 adipocytes, in part by down-regulating expression of adipogenic transcription factor and other specific target genes.
Based on the data from our previous studies, four new diabetic remedies were composed with the addition of Coicis Semen into Okchun-san (OCH), Commelinae Herba into Gangsim-tang (GST) , Scrophulariae Radix into Sunki-san (SKS), and Erythrinae Cortex into Yukmijihuang-hwan (YMG). The water extracts of these new remedies were treated in 3T3-L1 fibroblasts and adipocytes in order to investigate insulin-like substances and insulin sensitizers, respectively. With and without differentiation inducers, unmodified SKS (SKS-O) treatment induced 3T3-L1 fibroblasts into adipocytes more than the control. However, without inducers, YMG treatment, but not SKS, induced the differentiation more than the control among modified remedies. Without inducers, SKS, OCH as well as YMG increased the induction of differentiation from 3T3-L1 fibroblasts into adipocytes, compared to the control. The treatment of OCH and YMG with 1 ng/mL insulin increased glucose uptake much more than only insulin 1 ng/mL treatment. Thus, OCH and YMG contained increased insulin actions. In conclusions, the modified remedies, OCH and YMG, contained insulin-like substances and insulin sensitizers, and they can be improved the hypoglycemic effects.
Adipose tissue is an important endocrine organ involved in the control of whole body energy homeostasis and insulin sensitivity. Considering the increased incidence of obesity and obesity-related disorders, including diabetes, it is important to understand thoroughly the process of adipocyte differentiation and its control. Therefore, we performed a differential proteome mapping strategy using two-dimensional gel electrophoresis combined with peptide mass fingerprinting to identify intracellular proteins that are differentially expressed during adipose conversion of 3T3-L1 pre-adipocytes in response to an adipogenic cocktail. In the current study, we identified 46 differentially expressed proteins, 6 of which have not been addressed previously in 3T3-L1 cell differentiation. Notably, we found that phosphoribosyl pyrophosphate synthetase (PRPS), a regulator of cell proliferation, was preferentially expressed in pre-adipocytes than in fully differentiated adipocytes. In conclusion, our results provide valuable information for further understanding of the adipogenic process.

Fucoidan (fucan sulfate) is a fucose-containing sulfated polysaccharide from brown algae such as Fucus vesiculosus, Ecklonia kurome, and Cladosiphon okamuranus. The aim of this study was to investigate the effect of fucoidan on the expression of diabetes-related genes in mouse cell line 3T3-L1. 3T3-L1 adipocytes were cultured for 48 hr with or without fucoidan (10, 100, and 500 ppm) on a 60 mm dish. Reverse transcription polymerase chain reaction (RT-PCR) was used for measurement of peroxisome proliferators activated receptor ${\gamma}\;(PPAR{\gamma})$, CCAAT/enhancer binding protein ${\alpha}\;(C/EBP{\gamma})$, and glucose transporter 4 (GLUT4) RT-PCR analysis revealed that expression level of GLUT4, $PPAR{\gamma}$, and $C/EBP{\alpha}$ mRNAs increased with fucoidan treatment from 10 to 500 ppm in a dose-dependent manner. Fucoidan appears to enhance insulin sensitivity by increasing the expression level of diabetes-related genes in 3T3-L1 adipocytes. Therefore, fucoidan is potentially useful as a natural therapeutic material for hyperglycemia in type II diabetes patients.
