Simultaneous Evaluation of Cellular Vitality and Drug Penetration in Multicellular Layers of Human Cancer Cells

  • The multicellular layers(MCL) of human cancer cells is a three dimensional(3D) in vitro model for human solid tumors which has been used primarily for the assessment of avascular penetration of anti-cancer drugs. For anti-cancer drugs with penetration problem, MCL represents a good experimental model that can provide clinically relevant data. Calcein-AM is a fluorescent dye that demonstrates the cellular vitality in a graded manner in cancer cell culture system. In the present study, we evaluated the use of calcein-AM for determination of anti-proliferative activity of anti-cancer agents in MCL model of DLD-1 human colorectal cancer cells. Optical sectioning of confocal imaging was compromised with photonic attenuation and penetration barrier in the deep layers of MCL. By contrast, fluorescent measurement on the cryo-sections provided a feasible alternative. Cold pre-incubation did not enhance the calcein-AM distribution to a significant degree in MCL of DLD-1 cells. However, the simultaneous determination of drug penetration and cellular vitality appeared to be possible in drug treated MCL. In conclusion, these data suggest that calcein-AM can be used for the simultaneous determination of drug-induced anti-proliferative effect and drug penetration in MCL model.

  • Features are well recognized to play an important role for the integration of ACD and CAPP. Majority of pervious works for the feature recognition for prismatic part is based on 3D solid model. But in real factories, 2D drawing are used more than 3D drawings. In this paper, we develope an algorithm of the feature recognition on prismatic parts in 2D drawings, using by the graph method and the heuristic algorithm. Previous algorithms have some conflicts at feature interaction. In this paper, elements are grouped into connection by the graph method. Then features are recognized by using these grouped elements and their relationships of front and side-view. For resolving the problem of feature interaction, the element graphs are modified by an deloped algorithm. This algorithm is applied to a CAPP system for milling process planning.

Moving load response in a rotating generalized thermoelastic medium

  • The steady state response of a rotating generalized thermoelastic solid to a moving point load has been investigated. The transformed components of displacement, force stress and temperature distribution are obtained by using Fourier transformation. These components are then inverted and the results are obtained in the physical domain by applying a numerical inversion method. The numerical results are presented graphically for a particular model. A particular result is also deduced from the present investigation.

  • A method for generating 3D solid models and drawings for a rotor in the steam turbine is proposed. One of the most important design steps is generating the drawing for manufacturing it. This step is a very routine and time-consuming job because each drawing is composed of several kinds of views and many dimensions. To achieve automation for this activity, rotor profiles are composed of 2D entity groups with attributes. Based on this, the improved design process is developed as follows. First, the rotor profiles can be selected by searching for 2D entity groups using the related attributes. Second, the profiles are connected sequentially so that an entire rotor profile is determined. The completed profile is used to generate 2D drawings automatically, especially views, dimensions, and 3D models. The proposed method is implemented using a commercial CAD/CAM system, Unigraphics, and API functions written in C-language and applied to the rotor of steam turbines. Some illustrative examples are provided to show the effectiveness of the proposed method.

  • In this paper, a novel tissue engineering scaffold design method based on triply periodic minimal surface (TPMS) is proposed. After generating the hexahedral elements for a 3D anatomical shape using the distance field algorithm, the unit cell libraries composed of triply periodic minimal surfaces are mapped into the subdivided hexahedral elements using the shape function widely used in the finite element method. In addition, a heterogeneous implicit solid representation method is introduced to design a 3D (Three-dimensional) bio-mimetic scaffold for tissue engineering from a sequence of computed tomography (CT) medical image data. CT image of a human spine bone is used as the case study for designing a 3D bio-mimetic scaffold model from CT image data.

  • We propose a hardware accelerated voxelization method for various 3D object model such as surface models, solid models, and volumetric CSG models. The algorithm utilizes the stencil buffer that is one of modern Open히 graphics hardware features. The stencil buffer is originally used to restrict drawing to certain portions of the screen. The volumetric representations of given 3D objects are constructed slice-by-slice. For each slice, the algorithm restricts the drawing areas constructed inner region of 3D objects using the stencil buffer, and generates slices of the volumetric representation for target objects. As a result, we can provide volume graphics support for various engineering applications such as multi-axis machining simulation, collision detection and finite element analysis.

  • The design methodology of injection molding die has been changed from two-dimensional drafting to three-dimensional solid modeling, which is due to many advantages over the conventional methodology in terms of design modification and data associativity. In addition to the solid modeling capability, it is required for a mold designer to utilize a database management system that facilitates efficient mold design. In the paper presented is the implementation of a software program which automatically generates three-dimensional mold-bases including standard parts and slider parts, conforming to given geometric constraints. It is based on a commercial CAD system (Unigraphics NX) along with related API (application program interface) libraries. The research is expected to reduce design efforts and simplify construction of a complex three-dimensional mold-base model that is comprised of standard parts and slider parts, by use of the three-dimensional database and automatized geometric dimensioning.

Development of the Program Checking the Constructible Possibility of Prestressed Concrete Box Girder Bridges (PSC 박스 거더교의 시공성 검사 프로그램 개발)

  • The objective of this study is to develop the practical program which can check the constructible possibility of prestressed concrete box girder bridges for design. Checking constructible possibility is defined as checking the interference of each elements in a PSC box girder bridge and computing the distances of each elements. To check the constructible possibility of a PSC box girder bride, bridge must be modelled using solid in three dimension. By using a 3 dimensional solid modeling system, engineers can get the photo realistic 3D viewing images of the bridge and produce FEM analytic model of it. Users can manipulate their drawings easier and take off quantity of the whole structure and its elements as well as check the constructible possibility of their PSC box girder bridges.

  • This paper is concerned with the investigation of the structural behavior of waffle structure. Parametric variation on waffle floor thickness, main beam depth and column sizes are imposed to study the effects on mode shapes and natural frequencies of waffle structures. Comparisons between FEM models using shell and 3D-solid elements have also been made. The analysis result has shown that the mode frequencies increase with i) increase in main beam depth at level 2 and ii) decrease in waffle slab thickness at level 3. Both 3D and 2D model shown similar mode shapes. Besides, there is a consistent difference in mode frequencies between 3D and 2D model ranging from 25% to 36%.

Prediction of the Mechanical Erosion Rate Decrement for Carbon-Composite Nozzle by using the Nano-Size Additive Aluminum Particle (나노 알루미늄 입자 첨가 추진제에 의한 탄소복합재 노즐의 기계적 삭마 감소 특성 예측)

  • In this study, the influence of Al particle size, as an additive for solid propellant, on the mechanical erosion of the carbon-composite nozzle was evaluated. A new model which can predict the size and distribution of the agglomerated reaction product($Al(l)/Al_2O_3(l)$) was established, and the size of agglomerate were calculated according to the various initial size of Al in the solid propellant. With predicted results of the model, subsequently, the characteristics of mechanical erosion on the carbon-composite nozzle was estimated using a commercial CFD software, STAR CCM+. The result shows that the smaller the initial Al particles are, in the solid propellant, the lower is the mechanical erosion rate of the composite nozzle wall, especially for the nano-size Al particle.
