Photoluminescence Characteristics Y2O3:Eu3+ Thin Film Grown on Al2O3(0001) Substrate by PLD (PLD 방법으로 Al2O3(0001) 기판 위에 증착한 Y2O3:Eu3+ 박막의 형광 특성)

  • $Y_{2}O_{3}:Eu^{3+}$ thin films have been grown on $Al_{2}O_{3}$(0001) substrates by a pulsed laser deposition (PLD) method. The phosphor thin films were deposited at a substrate temperature of 500, 600, and $700^{\circ}C$ under the oxygen pressure of 100, 200, and 300 mTorr. The crystallinity, surface roughness and photoluminescence of the films are highly dependent on the substrate temperature and oxygen pressure. The films grown on $Al_{2}O_{3}$(0001) substrate even under the different substrate temperatures and oxygen pressures exhibited (222) preferred orientation. The luminescent spectra exhibited strong luminescence of ${^{5}D_{0}}-{^{7}F_{2}}$ transition within $Eu^{+3}$ peaking at 612 nm. The crystallinity and luminescence intensity of the films have been improved as the substrate temperature increasing. With increase of oxygen pressure from 50 to 300 mTorr, the crystallinity of the films has been uniformly decreased. The photoluminescence intensity and surface roughness have similar behaviors as a function of oxygen pressure. At 200 mTorr, both photoluminescence intensity and surface roughness show a maximum.

A Case of the Treatment of Surgical Scar with 595nm Pulsed Dye Laser (595nm Pulsed Dye Laser를 이용한 수술 흉터 치료 1예)

  • Pulsed dye laser(PDL) is originally designed for the treatment of vascular lesions but also effective in improving the quality and appearance of surgical scars. Recently, 595nm Pulsed dye laser(V-beam laser), which has the advantage of deeper tissue penetration and lesser amount of purpura, is spotlighted as a new option for the treatment of surgical scar. The authors treated a surgical scar of a female patient with V-beam laser for 3 times between 4 and 12 weeks after surgery. Subjective satisfaction was improved with visual analogue scale (VAS) and objective improvements were found in parameters of vascularity, pliability and height with Vancouver Scar Scale(VSS). We report the effectiveness of V-beam laser in surgical scar of Asian patient and plan the prospective study with larger scale.

  • Background : Axillary osmidrosis is characterized by an unpleasant odor, profuse sweating, and in some instances, staining of clothes that may socially and psychologically impair affected individuals. Various types of surgical procedures have been developed for the treatment of axillary osmidrosis. This study was undertaken to evaluate the effectiveness of subcutaneous pulsed neodymium: yttrium-aluminum-garnet (Nd-YAG) laser treatment for the treatment of axillary osmidrosis. Methods : Twenty-nine patients with axillary osmidrosis were included in this study. Patients were categorized according to the results of an axillary malodor grading system, and a subcutaneous pulsed Nd-YAG laser was applied to all patients. The treatment area for the appropriate distribution of laser energy was determined using the iodine starch test (Minor's test) against a grid pattern composed of $2{\times}2cm$ squares. The endpoint of exposure was 300 to 500 J for each grid, depending on the preoperative evaluation results. The results were evaluated by measurement of axillary malodor both pre- and postoperatively using the grading system and iodine starch test. Results : The average follow-up period was 12.8 months. Nineteen patients had a fair-to-good result and ten patients had poor results. The postoperative Minor's test demonstrated that there were remarkable improvements for patients with mild to moderate symptoms. Complications including superficial second degree burns (n=3) were treated in a conservative manner. A deep second degree burn (n=1) was treated by a surgical procedure. Conclusions : Subcutaneous pulsed Nd-YAG laser has many advantages and is an effective noninvasive treatment for mild to moderate axillary osmidrosis.

Viable Bacterial Cell Patterning Using a Pulsed Jet Electrospray System

  • In the present study, drop-on-demand two-dimensional patterning of unstained and stained bacterial cells on untreated clean wafers was newly conducted using an electrospray pulsed jet. We produced various spotted patterns of the cells on a silicon wafer by varying the experimental conditions, such as the frequency, flow rate, and translational speed of the electrospray system in a two-dimensional manner. Specifically, the electrospray's pulsed jet of cell solutions produced alphabetical patterns consisting of spots with a diameter of approximately $10{\mu}m$, each of which contained a single or a small number of viable bacteria. We tested the viability of the patterned cells using two visualization methods. This pattering technique is newly tested here and it has the potential to be applied in a variety of cell biology experiments.

Enhanced CEA-specific Immune Responses by Tat-LLO Fusion Protein (Tat-LLO 융합 단백질에 의한 CEA 특이 항종양 면역 반응의 증가)

  • Background: Carcinoembryonic antigen (CEA) is well-known soluble tumor marker frequently detectable in peripheral blood of carcinoma patients and considered as good target for antigen-specific immunotherapy. However, it is known that the induction of immune response to CEA is very difficult because CEA is a self-antigen expressed in fetal cells and weakly expressed in normal colorectal epithelial cells. To enhance anti-tumor immunity specific for CEA, recombinant CEA protein was modified using listeriolysin O (LLO) for endosomal lysis and trans activator of transcription (Tat) domain for transducing extracellular proteins into cytoplasm. Methods: After immunization using dendritic cells pulsed with Tat-CEA, both Tat-CEA and LLO, and both Tat-CEA and Tat-LLO, antibody titer to CEA and LLO, cytotoxic T lymphocyte activity and the frequency of IFN-${\gamma}$ producing T lymphocytes were measured. Results: Immunization using DC pulsed with both Tat-CEA and Tat-LLO protein showed the increasement of production of CEA-specific antibody in serum, cytotoxic T lymphocyte activity, the frequency of IFN-${\gamma}$ secreting T cells, compared with DC pulsed with both Tat-CEA and LLO. Furthermore the ratio of CD8+T cell to $CD4^+$ cell among CEA-specific T cells was increased in group pulsed with both Tat-CEA and Tat-LLO. Conclusion: These results suggested that DC vaccine using Tat-LLO could be used for the development of effective immunotherapy for the treatment of tumor.

Induction of CEA-specific Cytotoxic T Lymphocytes by Murine Dendritic Cells Expressing CEA (CEA 발현 수지상 세포를 이용한 CEA 특이 살해 T 세포의 유도)

  • Background: Carcinoembryonic antigen (CEA) is well-known soluble tumor marker frequently detectable in peripheral blood of carcinoma patients and considered as good target for antigen-specific immunotherapy. In this study, we used a replication-deficient adenovirus containing CEA to study CTL induction in vitro after adenovirus-mediated gene transfer into DC. Methods: DC were obtained from mouse bone marrow and cultured with IL-4 and GM-CSF. For measuring CTL activity, splenocytes were harvested from the mice, which were immunized with DC that had been infected AdV-CEA or pulsed with CEA peptide. Untreated DC was used as a control. Splenocytes were re-stimulated in vitro with DC pulsed with CEA peptide for 7 days and CTL activity with CEA peptide-pulsed EL-4 cells were assessed in a standard $^{51}Cr$-release assay. The frequencies of antigen-specific cytokine-secreting T cell were determined with $mIFN-{\gamma}$ELISPOT. Results: DC infected with recombinant adenovirus expressing CEA induced CEA-specific CTL responses in vivo. Splenocyte induced from mice immunized with AdV-CEA-infected DC increase in the number of $IFN-{\gamma}$ secreting T cells compared with those from mice immunized with CEA peptide-pulsed DC. Conclusion: These results suggested that DC infected with recombinant adenovirus has advantages over other forms of vaccination and could provide an alternative approach vaccination therapies.

A STUDY ON THE ORTHODONTIC TOOTH MOVEMENT UNDER HALF SINE-WAVE PULSED ELECTROMAGNETIC FIELDS (Half sine-wave pulsed electromagnetic fields 내에서 교정력에 의한 치아 이동에 대한 연구)

  • The biologic potential, which is different from the piezoelectric signals, relates tooth movement at least in part to changes in bone metaboliosm in bioelectric theory. The purpose of this experiment was to determine wheather the application of half sine-wave pulsed electromagnetic fields (HSPEMF) could increase both the rate and amount of orthodontic tooth movement. Forty-three male Hartley guinea pigs, weighting approximately 255g, were utilized in this study. The animals were 35 days old at the start of the study. Laterally directed orthodontic force was applied to the maxillary central incisors of 40 Hartley guinea pigs (20 experimental, 20 control). According to the amount of orthodontic force (6g, 12g), they were divided into two sub-groups (10 experimental I, 10 experimental II, 10 control I, 10 control II). During the experimental period, experimental animals were placed in plastic animal holders with their heads positioned in an area of uniform electromagnetic field. Control animals were placed in similar plastic holders that did not carry the electric apparatus. The results were as follows : 1. The application of a HSPEMF to the experimental groups significantly increase the final amount of orthodontic tooth movement observed over a 10-day experimental period. 2. The application of a HSPEMF to the experimental groups significantly increase the velocity of orthodontic tooth movement observed over a 10-day experimental period. 3. There was no significant difference in the final amount of orthodontic tooth movement at the fourth day to the eighth day, but there was significant difference in the final amount of orthodontic tooth movement at the nineth, tenth day during a 10-day experimental period. 4. After 10 days of HSPEMF exposure & orthodontic force, the experimental groups demonstrated more osteodasts in the pressure side than control groups.

Thickness Dependent Properties of Al-doped ZnO Film Prepared by Using the Pulsed DC Magnetron Sputtering with Cylindrical Target (원통형 타겟 타입 Pulsed DC Magnetron Sputtering에서 두께 변화에 따른 Al-doped ZnO 박막의 특성 변화)

  • Various thicknesses of Al-doped ZnO (AZO) films were deposited on glass substrate using pulsed dc magnetron sputtering with a cylindrical target designed for large-area high-speed deposition. The structural, electrical, and optical properties of the films of various thicknesses were characterized. All deposited AZO films have (0002) preferred orientation with the c-axis perpendicular to the substrate. Crystal quality and surface morphology of the films changed according to the film thickness. The samples with higher surface roughness exhibited lower Hall mobility. Analysis of the measured data of the optical band gap and the carrier concentration revealed that there were no changes for all the film thicknesses. The optical transmittances were more than 85% regardless of film thickness within the visible wavelength region. The lowest resistivity, $4.13\times10^{-4}\Omega{\cdot}cm^{-1}$ was found in 750 nm films with an electron mobility $(\mu)$ of $10.6 cm^2V^{-1} s^{-1}$ and a carrier concentration (n) of $1.42\times10^{21} cm^{-3}$.

The Effect of Continuous Ultrasound Therapy and Pulsed Ultrasound Therapy on Adjuvant Induced Rheumatoid Arthritis in Rat (지속적인 초음파치료와 맥동 초음파치료가 Adjuvant로 유도된 흰쥐의 관절염에 미치는 효과)

  • The drug therapy for rheumatoid arthritis must be maintained constantly or for the whole life but is apt to induce the adverse effects in gastrointestinal or renal system. Therefor, newer methods are paid attention to reduce adverse effects. The thirty-two rat female separated into seven groups depending on the therapy or Freund's adjuvant applied: Normal group(n=8) not received anything, Positive control group(n=8) only received adjuvant, USC group(n=8) received continuous US, USP group(n=8) received pulsed US. The physical and radiological findings by thermal and non-thermal effects of ultrasound were evaluated in groups with continuous mode at 1 MHz, $0.5\;W/cm^2$ for 6 minutes and with pulsed mode (duty cycle 1:9) at 1 MHz, $0.5\;W/cm^2$ for 6 minutes. The result summarized followings. 1. Swelling of forepaw and hindpaw was significantly reduced in USP. 2. Arthritis indices in USP group were significantly reduced than those in PCG. 3. In naked eye and radiologic findings, swelling was significantly prevented in USP group but not in PCG. The change of swelling, arthritis index, gross feature in naked eye, radiologic finding were significantly improved in all groups except for USC group, taking care for using its continuous mode.

Optical Diagnostics for Pulse-discharged Plasma by Marx Generator and Its Application for Modifications of Hemoglobin and Myoglobin Proteins

  • Property of optical diagnostics for pulse-discharged plasma in liquid and its biological applications to proteins are investigated by making use of high voltage Marx generator. The Marx generator has been consisted of 5 stages, where each charging capacitor is 0.5 ${\mu}F$, to generate a high voltage pulse with rising time of $1{\mu}s$. We have applied an input voltage of 6 kV to the each capacitor of 0.5 ${\mu}F$. High voltage pulsed plasma has been generated inside a polycarbonate tube by a single-shot operation, where the breakdown voltage is measured to be 7 kV, current of 1.2 kA, and pulse width of ~ 1 ${\mu}s$ between the two electrodes of anode-cathode whose material is made of tungsten pin, which are immersed into the liquids. We have investigated the emitted hydrogen lines for optical diagnostics of high voltage pulsed plasma. The emission line of 656.3 nm from $H-{\alpha}$ and 486.1 nm from $H-{\beta}$ have been measured by a monochromator. If we assumed that the focused plasma regions satisfy the local thermodynamic equilibrium conditions, the electron temperature and density of the high voltage pulsed plasma in liquid could be obtained by the Stark broadening of optical emission spectroscopy. For the investigation of the influence of pulsed plasma on biological proteins, we have exposed it onto the proteins such as hemoglobin and myoglobin. The structural changes in these proteins and their analysis have also been obtained by circular dichroism (CD) and ultraviolet (UV) visible spectroscopy.
