삼릉추출물이 항산화와 멜라노제네시스에 미치는 영향 (Effects of the Sciripi rhizoma on Antioxidation and Melanogenesis)

  • Whitening effect, which decreases the skin pigmentation, is the one of important targets in cosmetics. This study was investigated the effects of Scirpi rhizoma on ant ioxidation and melanogenesis. S.rhizoma is a rhizome of Scirpus fluviatilis G. a perennial Cyperaceae species of wide occurrence in Asia, Europe, Africa and North America. S.rhizoma shown scavenging activities of free radicals and reactive oxygen species (ROS) with the IC50 of 638${\mu}g/ml$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and 21.7${\mu}g/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. S.rhizoma treatment (48 h) suppressed the biosynthesis of melanin up to 27% and reduced tyrosinase activity up to 31% at 100${\mu}g/ml$ in B16 melanoma cells. S.rhizoma was also able to significantly inhibit tyrosinase and TRP-1 expres- sion in protein level. These results suggest that S.rhizoma inhibited melanin biosynthesis by regulating tyrosinase activity and expression in B16 melanoma cells. Therefore S.rhizoma may be useful as new whitening agent due to the antioxidant effect and the inhibitory effect against melanogenesis.

Lactobacillus plantarum ZLP001: In vitro Assessment of Antioxidant Capacity and Effect on Growth Performance and Antioxidant Status in Weaning Piglets

  • The objective of this study was to evaluate the antioxidant capacity of Lactobacillus plantarum ZLP001 and its effects on growth performance and antioxidant status in weaning piglets. The survival in hydrogen peroxide and free radical-scavenging activity of Lactobacillus plantarum ZLP001 were analysed in vitro. The Lactobacillus plantarum ZLP001 showed high viability in 1.0 mmol/L hydrogen peroxide and high scavenging ability against hydroxyl, superoxide anion, and DPPH (1,1-diphenyl-2-picrylhydrazyl) radicals which was dose dependent. Ninety-six weaning piglets were selected ($7.45{\pm}0.79kg$) and divided into three groups comprising of negative control without any supplementation, treatment group with supplemented $6.8{\times}10^7$ Lactobacillus plantarum ZLP001 CFU/g of diet, and positive control with antibiotic treatment (chlorotetracycline, 80 mg/kg diet). The results showed that Lactobacillus plantarum ZLP001 supplementation enhanced feed conversion rates in piglets compared with control (p<0.05). Supplementation of Lactobacillus plantarum ZLP001 increased the concentration of superoxide dismutase (p<0.05), glutathione peroxidase (p<0.01) and catalase in serum (p<0.10), while decreased the concentration of malondialdehyde (p<0.05). The present study implies that the strain Lactobacillus plantarum ZLP001 had high antioxidant ability and its supplementation improved the growth performance and antioxidant status of weaning piglets, so it can be considered useful to alleviate oxidative stress and increase productive performance of pigs.

In vitro Screening of Jeju Medicinal Plants for Cosmeceutical Materials

  • One of the important functions of skin is protection from harmful environments. Many studies have explored how to prevent skin from wrinkling and the occurrence of pigmentation changes. Skin wrinkling and pigmentation changes could be caused by unusual disruption of connective tissue, the formation of free radicals and ultraviolet radiation. In this study, extracts obtained from 254 different kinds of Jeju medicinal plants were screened for inhibitory effects on tyrosinase and elastase, and for free radical scavenging effects. Four herbs, Phormium tenax, Morus bombycis, Morus alba, and Cudrania tricuspidata, were potent inhibitors of tyrosinase ($IC_{50}$ values 4.62, 5.46, 8.17, and 64.17 ${\mu}g$/mL, respectively). Aleurites fordii [$IC_{50}$: 5.29 ${\mu}g$/mL, 1,1-diphenyl-2-picrylhydrazyl (DPPH)], Distylium racemosum ($IC_{50}$: 6.14 ${\mu}g$/mL), Acer palmatum ($IC_{50}$: 5.44 ${\mu}g$/mL), and Spiraea salicifolia ($IC_{50}$: 5.25 ${\mu}g$/mL) showed good antioxidative effects. Furthermore, Distylium racemosum ($IC_{50}$: 7.51 ${\mu}g$/mL), Diospyros kaki ($IC_{50}$: 15.1 ${\mu}g$/mL), Cornus macrophylla ($IC_{50}:$ 16.59 ${\mu}g$/mL), and Psidium guajava ($IC_{50}$: 40.25 ${\mu}g$/mL) exhibited potent inhibitory effects on elastase. These results suggest that medicinal plants possessing several biological activities may be potent inhibitors of the processes involved in pigmentation increases and aging. Further investigations will focus on in vivo assays and on the chemical identification of the major active components responsible for whitening and anti-aging activity in the screened efficacious extracts.

Evaluation of Antioxidant Activity of Cudrania tricuspidata (CT) Leaves, Fruit Powder and CT Fruit in Pork Patties during Storage

  • The objective of this study was to assess antioxidant activities of leaves and fruit powder of Cudrania tricuspidata (CT) with different particle sizes (crude, 500 ㎛, 150 ㎛), and determine the physicochemical properties and microbial counts of pork patties with various levels of CT fruit powder (CTFP) during refrigerated storage. Total phenolic content of crude leaves had the highest value of 3.54 g/100 g (p<0.05). Overall, CT leaves (CTLP) had higher total phenolic content than CTFP (p<0.05). 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of CTFP was higher than that of CTLP (p<0.05), which showed higher iron chelating ability and reducing power than CTFP (p<0.05). After pork patties were manufactured with 0.5% and 1.0% of CTFP at 500 ㎛, pH, color values, thiobarbituric acid reactive substance (TBARS), and peroxide values (POV) were then measured. The addition of CTFP into pork patties significantly (p<0.05) increased redness and yellowness values of patties. TBARS values of pork patties containing CTFP were lower (p<0.05) than those of CTFP-0 patties after 10 days of storage. Pork patties added with CTFP showed no significant (p>0.05) difference TBARS values among different storage periods. POV values of pork patties containing CTFP were lower than those of the control from 3 days up to the end of refrigerated storage (p<0.05). These results suggest that CTFP could be used as a natural antioxidant to retard lipid oxidation in meat products during refrigerated storage.

잔가시 모자반 추출물의 주름 개선 및 미백 효과 (Anti-Melanogenesis and Anti-Wrinkle Effects of Sargassum micracanthum Extracts)

  • 잔가시 모자반 추출물의 피부 노화 억제 효과를 확인하기 위하여 항산화 활성, tyrosinase 저해 활성, collagenase 저해활성을 측정하였다. 잔가시 모자반 에탄올 및 물 추출물의 폴리페놀 함량은 11.20 mg/g 및 11.70 mg/g으로 차이가 나지 않았다. DPPH radical 소거능에서는 잔가시 모자반 에탄올 및 물 추출물 모두 0.5 mg/ml 농도에서 83.68% 및 79%로 활성이 높게 나타났으며, 금속봉쇄력에서는 에탄올 추출물이 물 추출물보다 높게 나왔으나 대조구인 EDTA보다 활성이 낮게 나타났다. Tyrosinase 및 collagenase 저해 활성은 에탄올 추출물이 물 추출물보다 더 높은 것으로 나타났고, collagenase 저해 활성은 에탄올 추출물이 1 mg/ml 농도에서 80% 이상으로 높은 효소 저해능을 보였다. 따라서, 잔가시 모자반은 에탄올 추출물에서 항산화능, tyrosinase 및 collagenase 저해 활성이 더 높게 나타나 피부 노화 억제를 위한 소재로서 더 효과적으로 이용될 수 있을 것으로 사료된다.

누룩치(Pleurospermum kamtschaticumin) 추출물의 항산화 효과 및 생리활성 (Antioxidative and Physiological Activities of Fractions from Pleurospermum kamtschaticumin Extracts)

  • 본 연구는 누룩치의 생리활성효과와 미백 및 화장료 및 식품 보존제 등의 소재로서 활용가능성이 있는지 알아보고자 누룩치 에탄올 추출물과 용매별 분획물의 항산화 활성 및 tyrosinase 저해 활성, 항균효과를 측정하였다. 누룩치의 80% ethanol과 분획물의 총 폴리페놀 함량은 116.44~382.73 mg/g GAE로 나타났으며, 분획물 중 ethyl acetate 분획물에서 가장 높은 총 폴리페놀 함량을 나타내어 페놀화합물이 주로 ethyl acetate 분획물에 다량 존재하는 것으로 나타났다. 누룩치 용매분획물의 항산화 활성은 ethyl acetate 분획물에서 DPPH 라디칼 소거활성 $IC_{50}$ 값이 0.04 mg/mL, ABTS 라디칼 소거활성(0.5 mg/mL)이 98.86%로 대조군인 ascorbic acid와 비교 시 전혀 뒤떨어지지 않는 활성을 갖고 있는 것으로 분석되었으며, 다음으로는 n-butanol 분획물이 유의적으로 높은 활성을 나타내었다. 환원력은 ethyl acetate 분획물과 n-butanol 분획물에서 높은 흡광도 수치를 나타내었으며, FRAP value 측정 결과 ethyl acetate 분획물에서 높게 나타났다. SOD 유사활성은 1 mg/mL의 농도에서 ethyl acetate 분획물(86.93%), n-butanol(78.23%) 순으로 높은 활성을 나타내었다. Tyrosinase 저해 활성 측정 결과(5 mg/mL), n-butanol 분획물이 43.52%, 80% ethanol 추출물이 41.64%로 나타났다. Paper disc 방법에 의한 항균활성은 누룩치의 chloroform 분획물이 E. coli에 대해서 16.7 mm(0.5 mg/disc)의 저해환 크기를 나타내었다. 따라서 본 연구 결과 누룩치의 분획물들은 비교적 높은 항산화활성 및 미백 효과, 항균효과를 가지고 있으므로 식품 첨가물 및 화장품 원료 등 기능성 소재로 사용할 수 있을 것으로 여겨진다.

수생관속식물의 라디칼 소거능과 남세균 생장에 대한 억제활성 비교 (A Comparison of Radical Scavenging Activity and Cyanobacteria Growth Inhibition of Aquatic Vascular Plants)

  • 수생식물이 가지고 있는 타감물질과 타감작용을 알아보기 위하여 메탄올 추출물의 DPPH 라디칼 소거능과 대장균($Escherichia$ $coli$)과 남세균($Microcystis$ $aeruginosa$)에 대한 생장억제 활성을 분석하였다. 그 결과 고마리($Persicaria$ $thunbergii$), 물여뀌($P.$ $amphibia$)와 마름($Trapajaponica$), 이삭물수세미($Myriophyllum$ $spicatum$), 순채($Brasenia$ $schreberi$)가 라디칼 소거 활성을 나타냈으며, 순채, 마름, 물여뀌, 이삭물수세미 등은 남세균에 대한 높은 생장억제 활성을 나타냈다. 반면 순채를 제외한 다른 수생식물의 추출물은 대장균의 생장에 대한 억제 활성이 나타나지 않았다. 수생식물 추출물에 대한 타감작용에 대한 상관 분석 결과, DPPH 라디칼 소거 활성으로 측정된 항 산화 활성과 남세균 생장억제 활성간에는 양의 상관이 있는 것으로 나타났다. 반면에 수생식물 추출물의 대장균에 대한 생장억제는 다른 활성 결과와 상관이 낮은 것으로 나타났다. 본 연구의 결과를 이용하면 수생식물체의 남세균에 대한 타감작용은 DPPH에 의한 항산화성 검정을 이용하는 간편하고 신속한 검사로 대체 될 수 있을 것으로 보인다.

복분자, 오디 및 블루베리 첨가 그레이비소스의 품질특성 및 항산화 활성 (Antioxidant Activities and Quality Characteristics of Gravy Sauce Added with Bokbunja (Rub coreanus Miquel), Mulberry and Blueberry)

  • Purpose: Bokbunja (Rub coreanus Miquel), mulberry and blueberry have been reported to have powerful anti-antioxidant and anti-aging activities. The purpose of this study was to compare the quality characteristics and antioxidant activities of gravy sauce added with Bokbunja, mulberry and blueberry. Methods: Quality characteristics pH, brix, color and sensory characteristics of samples were evaluated. Antioxidant activities of samples were compared using total phenol, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation radical scavenging and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities. Results: Mulberry juice showed significantly (p<0.05) higher values of pH and $^{\circ}Bx$ compared to Bokbunja and blueberry juice. Bokbunja juice also had significantly (p<0.05) higher value of Color-L value than mulberry and blueberry juice. The total phenol contents and ABTS cation radical scavenging activity of Bokbunja juice were 236.50 mgGAE/100 g and 61.66%, respectively, which were significantly (p<0.05) higher than those of blueberry and mulberry juice. pH value of gravy sauce added with mulberry (MBS) was 4.67, which was significantly higher than that of the control gravy sauce (GS) and gravy sauce added with Bokbunja (BJS) which had the lowest (p<0.05) pH value 4.33. $^{\circ}Bx$ of GS (13.50) was not significantly different from that of MBS and BBS. However, BJS showed the lowest (p<0.05) value $12.33^{\circ}Bx$. Total phenol contents and ABTS cation radical scavenging activity of BJS were 78.09 mgGAE/100 g and 14.47%, respectively, which were significantly (p<0.05) higher than those of MBS or BBS. Values of sensory characteristics color, sour aroma and sour flavor of BJS were significantly (p<0.05) higher value than those of other samples. Conclusion: Bokbunja juice and gravy sauce showed higher antioxidant activities and higher acceptance test compared to blueberry and mulberry. Thus, Bokbunja has the potential to be added to as high value products in the food industry.

다양한 식물배양세포주에서 자유라디칼 포착활성과 Ascorbate 함량 (Free Radical Scavenging Activity and Ascorbate Content in Various Plant Cell Lines)

  • 다양한 유도조건에서 확립한 64종의 식물배양세포주의 MPA추출물을 대상으로 DPPH 라디칼을 이용하여 항산화 활성을 조사하고 활성이 높은 세포주를 대상으로 HPLC로 ascorbate 함량을 분석하였다. 13종의 세포주는 생체중 50mg 이하에서 DPPH 라디칼의 자연산화를 억제하는 높은 항산화활성을 나타내었다. 이중 6종의 세포주의 MPA추출물을 실리카겔 TLC에 전개하여 DPPH용액을 분무한 결과, ascorbate와 같은 Rf 위치에서 DPPH 양성반응을 나타내는 화합물이 존재하였다. DPPH 양성반응을 나타낸 덩굴장미(Rosa multiflora), 황금(Scutellaria baicalensis), 쇠무릎(Achyranthes japonica) 세포주의 ascorbate 함량($\mu\textrm{g}/g$ cell fresh wt)은 각각 48.5, 30.3, 16.8였다. 황금 캘러스 배양에서 ascorbate의 함량은 세포생장에 비례하여 정지기까지 증가한 후 계속적인 생장에 따라 급격히 감소하였다.

항산화 물질을 생산하는 해양 미생물의 분리.동정 및 배양 특성 조사 (Isolation and Identification of Antioxidant-producing Marine Bacteria and Medium Optimization.)

  • 전자 공여능으로 항산화 물질 생산 능력이 우수한 해양 미생물을 분리하여 HJ-14라고 명명하였다. HJ-14는 형태적, 생리적, 생화학적 특성이 Altermonas macleodii ATCC 27126T와 가장 유사하였으며, 주요 세포 지방산이 $C_{14:0}$ , $C_{15:0}$ , $C_{16:1}$$C_{17:1}$ $_{w8c}$로 분석됨으로서 Aiteromonas SP- HJ-14로 동정하였다. Alteromonas sp. HJ-l4의 항산화 물질 생산에 대한 배양 특성을 조사하기 위해, 탄소원, 무기질소원, sodium chloride농도, 초기 pH및 배양온도의 영향을 관찰하였다. 탄소원으로는 2.5% dextrin, 무기 질소원으로는 ammonium sulfate, sodium chloride는 2~6%, pH와 온도는 각각 pH 6~8, 25~37$^{\circ}C$에서 전자공여능이 높게 나타났으며, 최적 조건일 때에 전자공여능은 90~92%로.나타났다 또한, OH소거능도 90.03%로 관찰되어 항산화활성이 높은 것으로 확인되었다.
