Mineral, Nutritional, and Phytochemical Profile, Total Phenolic Content, and Radical Scavenging Activity of Philippine Bamboo "Bolo" Gigantochloa levis (Blanco) Merr. Leaves

  • The study is a pioneering effort to determine the mineral, nutritional, and phytochemical composition and phenolic content and to determine the free radical scavenging activity of Gigantochloa levis (Blanco) Merr, a native bamboo species (locally known as "bolo") in the Philippines. Proximate analysis showed that air-dried G. levis leaves contain 15.8% ash, 22.6% crude protein, 1.2% crude fat, 29.3% crude fiber, and 19.7% total sugar. Phytochemical tests indicated the presence of diterpenes, triterpenes, saponins, phenols, tannins, and flavonoids in both the ethanolic and aqueous leaf extracts, while phytosterols were only detected in the ethanolic extract. Folin-Ciocalteu assay determined the total phenolic content in gallic acid equivalents (GAE) to be $85.86{\pm}3.71$ and $32.32{\pm}1.01mg\;GAE/100g$ dried sample for the ethanolic and aqueous extracts, respectively. The total phenolic content in quercetin equivalents (QE) was $74.44{\pm}3.11$ and $29.43{\pm}0.85mg\;QE/100g$ dried sample for the ethanolic and aqueous extracts, respectively. The radical scavenging activity of the different solvent fractions containing varying concentrations of the extract was determined using the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay. The ethyl acetate and 1-butanol fractions were found to have the highest radical scavenging activity. Mineral analysis via Energy Dispersive X-Ray Spectrometry (EDS) of the ash of G. levis leaves showed that Si is the major component, followed by K and Mg. These results point to the potential of G. levis leaves as a source of minerals and bioactive compounds with medicinal value.

Effects of Extraction and Processing Methods on Antioxidant Compound Contents and Radical Scavenging Activities of Laver (Porphyra tenera)

  • Laver is one of the most consumed edible red algae seaweeds in the genus Porphyra. Laver is primarily prepared in the form of dried, roasted, and seasoned products. We investigated the total polyphenol and flavonoid contents of laver products, and evaluated the in vitro antioxidant properties of solvent extracts from commercially processed laver products. Significant differences in the concentration of phenolic compounds were found among differently processed laver. The total phenolic content for laver extracts ranged from 10.81 mg gallic acid equivalent (GAE)/g extract to 32.14 mg GAE/g extract, depending on extraction solvent and temperature. Laver extracts contained very few flavonoids (0.55 mg catechin equivalent/g extracts to 1.75 mg catechin equivalent/g extracts). 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS), hydroxyl radical, and superoxide anion scavenging assays were used to determine the radical scavenging capacities of laver extracts. These assays revealed that the processing method and extraction condition affected the antioxidant potentials of laver. Antioxidant activity of dried laver, roasted laver, and seasoned laver increased in a concentration-dependent manner ($100{\sim}1,000{\mu}g/mL$). The radical scavenging activities of $37^{\circ}C$ and $100^{\circ}C$ water extracts were lower than that of a $37^{\circ}C$ 70% ethanol extract. The highest radical scavenging capacity was observed in the $37^{\circ}C$ 70% ethanol extracts of dried laver, roasted laver, and seasoned laver. Overall, these results support that notion that laver contains bioactive compounds, such as polyphenols and flavonoids, which may have a positive effect on health.

곤충병원성진균의 총 페놀 함량 및 DPPH 라디칼 소거능 (Total Phenol Contents and DPPH Radical Scavenging Activity of Entomopathogenic Fungi)

  • 곤충병원성진균 20종의 총 페놀 함량, 항산화활성 및 항균활성간의 관계를 조사하기 위해 본 실험을 수행하였다. 총 페놀 함량은 Aspergillus flavus에서 $553.0{\pm}52.15{\mu}g/g$, A. parasiticus에서 $529.9{\pm}60.10{\mu}g/g$으로 가장 높게 나타났으며, 다른 균주는 $26.6{\sim}121.9{\mu}g/g$으로 나타났다. DPPH(1, 1-diphenyl-2-picrylhydrazyl) radical 소거능은 대부분의 균주에서 나타났으며 다른 균주에 비하여 A. flavus와 A. parasiticus에서는 각각 $90.9{\pm}2.90%$$77.9{\pm}4.13%$로 높게 나타나 항산화활성과 총 페놀 함량 간 높은 연관성이 나타났다. 항균활성은 모든 식중독균에 대하여 나타났으며 Listeria monocytogenes와 Escherchia coli에 대하여 특히 강하였다.

파파야씨 추출물 및 분획물의 항산화, QR 활성 (Antioxidant Capacity and Quinone Reductase Activity of Methanol Extracts and Fractions from Papaya Seed)

  • 본 연구에서는 파파야씨의 메탄올 추출몰과 분획물을 이용하여 항산화 활성 및 quinone reductase (QR) 활성을 측정하였다. 파파야씨의 추출물 및 분획물의 총 폴리페놀 함량은 17.74~125.99 ${\mu}g/mg$이며, 총 플라보노이드 함량은 1.60~32.69 ${\mu}g/mg$으로 나타났으며, ethyl acetate (EtOAc) 분획층의 총 폴리페놀 및 플라보노이드 함량이 다른 분획층과 비교했을 때 가장 높은 것으로 나타났다. ${\alpha}$,${\alpha}$-Diphenyl-${\beta}$-picrylhydrazyl (DPPH) radicals, 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) and hydrogen peroxide를 이용하여 파파야씨 메탄올 추출물 및 분획물의 항산화 활성을 조사한 결과, EtOAc 분획층에서 가장 높은 free radical 억제능을 보였다. 또한 EtOAc 분획층 12.5~50 ${\mu}g/ml$의 농도에서 QR의 유도활성을 조사한 결과, 50 ${\mu}g/ml$의 농도에서 3.3배 정도의 QR 유도능을 보였다. 따라서 파파야씨의 EtOAc 분획층에 존재하는 물질들은 QR inducer 로써의 역할이 기대된다.

황련해독탕(黃連解毒湯)·편백(扁柏) 정유 복합추출물의 항산화 및 항균활성에 관한 연구 (Antioxidative and Antimicrobial Activities of Herbal Complex Extract from Hwangryunhaedok-tang and Essential Oil of Chamaecyparis obtusa)

  • Objectives : The present study was designed to investigate effects of mixed extracts (9:1, v/v) of Hwangryunhaedoktang, Chamaecyparis obtusa essential oil. We evaluated the antimicrobial and antioxidant activity by manufacturing mixed extracts as the materials for functional medicinal herb cosmetics. Methods : We performed antimicrobial were tested microbes (Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans, Propionibacterium acnes) by disc diffusion method and measure clear zone. Antioxidant activities were measured by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. Results : Antimicrobial activities of mixed extracts against S. epidermideis, P. acnes, S. aureus, E. coli, and Candida. A were $10.9{\pm}3.0mm$, $16.9{\pm}2.0mm$, $9.2{\pm}2.0mm$, $10.3{\pm}1.0mm$, $11.8{\pm}3.0mm$ respectively. The Chamaecyparis obtusa essential oil had the highest antimicrobial activities against S. epidermideis, P. acnes, S. aureus, E. coli, and Candia. A and clear zone of microbes ware $16.7{\pm}3.0mm$, $28.8{\pm}0.2mm$, $15.9{\pm}2.0mm$, $11.5{\pm}1.0mm$, $16.3{\pm}3.0mm$. Hwangryunhaedoktang extract showed antimicrobial activity but only P. acnes and S. aureus The antioxidant activities of the mixed extracts were tested through the evaluation of DPPH radical scavenging activity. The 100 % mixed extracts were found to have 90 % DPPH radical scavenging activity. The mixed extracts was presented similar antioxidant activities compared with that of ascorbic acid. As a result, A mixture extract is expected to have antimicrobial effects and free radical scavenging activity was found. Conclusions : Accordingly, It can be concluded that mixed extracts has the potential to cosmetic material.

In vitro Antioxidant and Cytoprotective Activities of the Extract of Dangyuja (Citrus grandis Osbeck) Leaves

  • The antioxidant activities of the extracts of dangyuja (Citrus grandis Osbeck) leaves were evaluated. The highest phenolic content was obtained from the ethyl acetate fraction (EF) (202.1$\pm$0.8 mg GAE/g dried extract) and it exhibited the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity. The cytoprotective effects of EF on oxidative damage induced by tert-butyl hydroperoxide (t-BHP) in a human hepatoma cell line, HepG2 cells, were investigated to understand the intracellular antioxidant mechanisms. Treatment of HepG2 cells with EF prior to oxidative stress was found to inhibit reactive oxygen species (ROS) generation, lipid peroxidation, and DNA damage in a dose-dependent manner. Gas chromatography-mass spectrometry (GC-MS) studies on EF resulted in tentative identification of 19 compounds representing 94.3% of the total content. Taken together, these results demonstrated that EF has excellent antioxidant activities and thus dangyuja leaves have great potential as a source for natural antioxidant which can be applied in food products.

곤달비 추출물의 항산화 활성 및 지방세포 분화 억제 효과 (Antioxidant Activity and Anti-Adipogenic Effect of Ligularia stenocephala Extract)

  • The purpose of this study was to evaluate the antioxidant and anti-adipogenic activities of Ligularia stenocephala (L. stenocephala) extract. The contents of the total polyphenol of the extract was 55.950 mg GAE/g residue. Antioxidant activities of L. stenocephala were evaluated by free radical scavenging ability and a reducing power test. 2,2'azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and ${\alpha}$-${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) free radical scavenging activities of the extract were approximately 90% and 70%, respectively. Reducing power of the extract was 258.833 mg TE/g residue. The anti-adipogenic activity of L. stenocephala extract was examined in 3T3-L1 cells. During adipocyte differentiation, the 3T3-L1 cells were treated both with and without the extract. L. stenocephala extract suppressed the lipid accumulation in a concentration-dependent manner in the 3T3-L1 cells. The L. stenocephala extract inhibited the expression of peroxisome proliferator activated receptor ${\gamma}$ ($PPAR{\gamma}$) and adipocyte protein 2 (aP2) proteins, compared with control adipocytes. These results indicate that L. stenocephala could be regarded as a potential source natural antioxidant and an anti-obesity agent.

마카(Lepidium meyenii)의 부위별 함유성분 및 항산화 활성 비교 (Component Analysis and Antioxidant Activity of Maca)

  • This study examined the components and physicochemical properties of Lepidium meyenii as a natural health food source. To accomplish this objective, the general and antioxidative contents of Lepidium meyenii were measured. Total contents of carbohydrates, crude protein, crude lipid, and ash were 56.04%, 33.77%, 1.49%, and 3.7%, respectively(dry wright basis). Total phenol contents of the 70% ethanolic extracts of Lepidium meyenii were 189.9 mg/g, 152.5 mg/g. Total flavonoid contents of the 70% ethanolic extracts were 166.0 mg/g, 78.2 mg/g. The antioxidative activities of Lepidium meyenii were significantly increased in a dose dependent manner on DPPH(1,1-diphenyl-2-picrylhydrazyl) radical scavenging, ABTS (2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging, FRAP(ferric reducing antioxidant power) activity, reducing power. The general nutrients and other antioxidant bioactive materials in Lepidium meyenii were also potential materials for good health food.

Changes in Antioxidant Activity of Rehmannia radix Libosch with Heat Treatment

  • This study evaluated the effects of heat treatment on antioxidant activity of Rehmannia radix Libosch (RRL). RRL was heated at various temperatures ($110-150^{\circ}C$) for various times (1-5 hr), and the total polyphenol, flavonoid content, and antioxidant activity were investigated. With increased heating temperature and exposure time, total content of polyphenol, flavonoid, as well as antioxidant activity increased. The highest total polyphenol and flavonoid contents were 21.65 and 3.56 mg/g, respectively, these values were occurred after heating for 3 hr at $150^{\circ}C$ (RRL was 5.09 and 0.83 mg/g, respectively). The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was highest value of 83.46% after heating for 3hr at $150^{\circ}C$. The 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) cation radical scavenging activity was highest value of 20.43mg ascorbic acid (AA) eq/g after heating for 2 hr at $150^{\circ}C$. There were highly significant differences in the total polyphenol, flavonoid content, and antioxidant activity among heating temperatures and times (p<0.001), with heating temperature having the greater effect.

버섯균사체 발효 뽕잎 추출물의 항산화 활성 (Antioxidant Activity of Mulberry Leaves Extract Fermented by Hericium erinaceum)

  • 본 연구에서는 뽕잎(Morus alba)을 노루궁뎅이 버섯균사체(Hericium erinaceum)로 발효시킨 열수 추출물(MA-HE)의 항산화 활성을 확인하고자 하였다. MA-HE의 항산화 활성을 DPPH radical, ABTS radical 소거활성 측정을 통해 알아보았다. 그 결과 DPPH radical 소거활성은 $500{\mu}g/mL$ 농도에서 61.73%, ABTS radical 소거활성은 $250{\mu}g/mL$ 농도에서 97.39%로 나타났다. MA-HE은 DNA의 산화적 손상을 효과적으로 억제하였다. 또한 생체고분자물질인 사람의 혈청단백질의 산화적 손상을 억제하였다. 세포에 $H_2O_2$를 처리하였을 때 세포생존율에 비하여 발효물을 $100{\mu}g/mL$ 농도로 전 처리한 세포생존율은 8% 증가했으며, 발효물을 $50{\mu}g/mL$ 농도로 처리했을 경우 세포 내 활성산소의 축적이 유의적으로 감소되었다. 본 연구 결과는 MA-HE이 항산화 활성을 가지고 있으며 산화적 스트레스에 의해 야기되는 세포 독성에 대한 보호 작용도 뛰어난 것으로 사료되었다.
