Production of Reuterin by Batch and Continuous Reactor and Antimicrobial Characteristics of Reuterin (회분식과 연속식에 의한 루테린 생산 및 루테린의 항균 특성)

  • Reuterin production efficiency of Lactobacillus reuteri, which converts glycerol into reuterin (antimicrobial substance) under anaerobic condition, was examined. When compared at $32,\;37,\;and\;42^{\circ}C$, production rate and total amount produced increased with increasing incubation temperature. Reuterin production terminated earlier at $42^{\circ}C$ than at $32\;and\;37^{\circ}C$. Presence of various amino acids in the reaction mixture generally decreased reuterin production, whereas proline did not inhibit reuterin production. A continuous-type reactor in which glycerol was passed through the chamber containing L. reuteri cells produced greater amount of reuterin than when batch-type process was used.

  • Diacylglycerol(DAG) was produced by enzymatic esterification of glyceryl mono-oleate(GMO) and conjugated linoleic acid(CLA) in a stirred-batch type reactor. The reaction was catalyzed by lipozyme RMIM(an immobilized lipase from Rizomucor miehei). DAG was isolated by a short-path distillation process and decolorized. DAG oil was composed of 87.3% DAG, 11.4% triacylglycerol(TAG), and 1.5% monoacylglycerol(MAG)(all w/w). Major fatty acids in DAG oil were oleic acid(54%), CLA(31.1%), and linoleic acid(7%). DAG oil iodine,and acid values were 108.8, 2.57, and 1, respectively. The DAG oil solid fat index(SFI) and thermograms were obtained using differential scanning calorimetry.

  • The heat of reaction has been estimated from heat balance relationships around the reactor. The heat balance equations were formulated with the assumptions that the reactor temperature is uniformly distributed and the jacket temperatures are axially distributed. We have obtained the temperature distribution of jacket contents by FDM. And then, we have rearranged the heat balance equations so that the heat of reaction can be estimated from the finite number of temperature measurements, i.e., temperatures of the reactor contents, at the jacket inlet and outlet, respectively. The proposed method for reaction heat estimation on were applied to industrial batch reactors ; one is ABS polymerization reactor and the other is SAN polymerization reactor. We have also examined the variation of overall heat transfer coefficients for the reactors during reaction.

  • In operational pont of view, the batch reactor is quite different from the continuous reactor in that it is completely in unsteady states. This makes process variables swing over wide ranges and the process disclose its nonlinerarities. For the most part these nonlinearities are due to reaction heat. Accordingly it is very important to know the informations of reaction heat. This paper presents the method of reaction heat measurement in industrial batch reator which furnishes the limited measurable points. The informations of reaction heat are utilized in modeling of the batch reactor as well as deciding the stability and control variables.

  • The effect of agitation speed on mass transfer coefficient in the extraction of citric acid from mixed aqueous solutions of citric and acetic acid with n-butylacetate solutions of di-isotridecylamine(DITDA) and 50% mixture of tri-n-octyl and try-n-hexyl phosphine oxide(MOHPO), were studied in batch extractor. Experimental results showed that the degree of extraction was higher with increasing agitation speed, and was best at 200rpm and 30 minutes in batch extractor. The higher degree of extraction was obtained in mixed solution of citric-acetic acid by using DITDA than MOHPO as an carrier. Mass transfer coefficient was proportional to the degree of extraction, and $K_r=1.254{\times}10^{-3}Re^{0.536}$ was found for she extraction of citric acid by DITDA.

Production of Glutathione by yeast and Process Monitoring (효모에 의한 글루타치온의 생산과 공정 모니터링)

  • In this work the production of glutathione (GSH) by yeast Saccharomyces cerevisiae and the monitoring of the process were studied. In shaking culture the production of GSH was high at initial pH value of 4 and at temperature of 30$^{\circ}C$. But when L-cysteine was added to the culture medium at the beginning of the cultivation, the productivity of GSH was low. In case 0,5% (v/v) of L-cysteine, glycine and glutamic acid were introduced to the culture medium in the exponential cell growth phase, high concentration of GSH (about 90 mg/L) was produced in the bioreactor. A fed-batch operation with stepwise glucose feeding strategy allowed to produce 102 mg/L of GSH. The cultivation processes were on-line monitored by a 2-dimensional fluorescence sensor. A few off-line data such as cell growth, cystein concentration, phosphate concentration and GSH productivity could be well correlated to the fluorescence intensity of some combinations of excitation and emission wavelengths.

  • The bubbling fluidized bed (BFB) reactor with a diameter of 0.1 m and a height of 1.2 m was used for experimental study of co-firing and emission characteristics fueled by sewage sludge (SS) and wood pellet (WP). The facility consists of a fluidized bed reactor, feeding system, cyclone, condenser and gas analyzer, The mean particle diameter and minimum fluidization velocity are $460{\mu}m$ and $0.21ms^{-1}$ respectively. SS produced from Korea and WP from Canada were examined. The various mixing ratios of WP were 20, 50, and 80% based on HHV. The equivalence ratio of 1.65, reactor temperature of $800^{\circ}C$, air flow rate of $100Lmin^{-1}$, and fluidization number of 4 were fixed in the BFB experiment. In TGA, the range of combustion temperature of SS was wider than that of WP. It represents that the combustibility of WP is higher than that of SS. The BFB reactor temperature was maintained between 800 and $900^{\circ}C$. CO emission of SS was high because of lower combustibility. $NO_X$ and $SO_X$ formation of SS were higher than that of WP since high nitrogen and sulfur contents of SS. CO, $NO_X$, and $SO_X$ formation were suppressed as the mixing ratio of WP was increased. The slagging and fouling tendencies show high in all test conditions.

  • PBT (polybutylene terephthalate) has excellent mechanical properties such as low absorption, dimensional stability, abrasion resistance. It is used in manufacturing electronic components, the automobile part and the various precise parts. Bis (hydroxybutyl) terephthalate (BHBT) which is a PBT monomer, can be produced by transesterification reaction of DMT (dimethyl terephthalate) with 1,4-butandiol (BD). The kinetics of transesterification reaction of DMT with BD using zinc acetate as a catalyst was studied in a batch reactor. Previous kinetic studies was carried out in a semibatch reactor where generated methanol was removed so that reverse reactions were not considered in the kinetic expressions, resulting in inaccuracy of the kinetic model. Mathematical models of a batch reactor for the transesterification reaction were developed and used to characterize the reaction kinetics and the composition distribution of the reaction products. More accurate models than previous models was obtained and found to have a good agreement between model predictions and experimental data.

  • The kinetics of the transesterification of dimethyl terephthalate (DMT) with diethylene glycol (DEG) was studied in a batch reactor. bis-hydroxyethoxytethyl-terephthalate (BHEET), which is polyester polyol monomer, can be produced by the transesterification reaction. Zinc acetate was used as a catalyst. Previous kinetic studies was carried out in a semi-batch reactor where generated methanol was removed so that reverse reactions were not considered in the kinetic expressions, resulting in inaccuracy of the kinetic model. Mathematical models of a batch reactor for the tranesterification reaction, which took the reverse reaction into account, were developed and used to characterize the reaction kinetics and the composition distribution of the reaction products. More accurate models than previous ones were obtained and found to have a good agreement between model predictions and experimental data. Effect of process variables on the esterification reaction was investigated based on the experimental and simulation results.

  • In this study, we analyzed the desulfurization performance of Zn-based dry sorbents according to the reducing power, water vapor content and $H_2S$ inlet concentration of coal gas in a batch-type fluidized-bed reactor. We used three different coal gas composition with different reducing power such as KRW air-blown coal gas, Shell oxygenblown coal gas and IAE coal gas. The experiments were performed by changing the inlet concentration of water vapor and $H_2S$ in a coal gas. Water vapor content was varied from 5% to 30% and $H_2S$ inlet concentration from 0.5% to 2.0%. As both the water vapor content and $H_2S$ inlet concentration increased, desulfurization performance of Zn-based sorbents decreased regardless of the reducing power of the coal gas. The minimum desuflurization performance was, however, above 99.5% for all experimental conditions, which implied that Zn-based dry sorbents could be used to remove $H_2S$ up to 99%.
