Comparative Radical Scavenging and Nitric Oxide Production Capacity of Water Extracts of Acanthopanax divaricatus var, Albeofructus and Eleutherococcus divaricatus var. Chiisanensis (지리산오가피와 흰털오가피 열수추출물의 라디칼 제거 활성 및 대식세포에서의 산화질소 생성능 비교)

  • In the present study, water extracts of Acanthopanax divaricatus var, albeofructus and Eleutherococcus divaricatus var. chiisanensis were prepared and their antioxidant and anti-inflammatory effects were compared. The water extract of Acanthopanax divaricatus var, albeofructus showed significantly higher activity in total polyphenol and flavonoid contents, while there were no significant differences in ABTS/DPPH radical scavenging activities and ferric reducing antioxidant power. However, the water extract of Eleutherococcus divaricatus var. chiisanensis at the highest dose showed significantly lower capacity in nitric oxide production than that of Acanthopanax divaricatus var, albeofructus. Taken together, there were differences between Acanthopanax divaricatus var, albeofructus and Eleutherococcus divaricatus var. chiisanensis in total polyphenol/flavonoid contents and anti-inflammatory effect, however, the antioxidant effect was almost similar.

Radical Scavenging and Anti-obesity Effects of Various Extracts from Turmeric (Curcuma longa L.) (추출용매에 따른 울금 추출물의 라디칼 소거능 및 항비만 효과)

  • In the present study, the levels of phenolic compounds and flavonoids, as well as activities of radical scavenging (DPPH and ABTS radical scavenging activity) and anti-obesity were assessed with cold water (CLC), hot water (CLH), and methanolic (CLM) extracts of Curcuma longa L. (turmeric). Our results showed that the phenolic compounds of CLC, CLH and CLM were $3.68{\pm}0.80%$, $3.94{\pm}0.74%$ and $9.01{\pm}0.73%$, respectively. The DPPH and ABTS radical scavenging activities of the CLM were significantly higher than that of the water extracts (CLC and CLH). During the adipocyte differentiation, the treatment of CLM more significantly inhibited lipid accumulation in 3T3-L1 cells than that of the water extracts. These results indicate that the stimulation of radical scavenging potential and the inhibition of adipogenesis were brought on by the lipophilic compounds of turmeric.

  • The present study was conducted to evaluate of the 1,1-Diphenyl-2-Picrylhydrazyl (DPPH) radical scavenging effect, cytotoxicity and tyrosinase inhibition activities using methanol extracts from different parts of four herb plants. The results showed that whole and root extracts of yarrow the highest total polyphenol and flavonoid contents as well as whole of yarrow revealed the highest DPPH radical scavenging effect. In cytotoxicity test against three cancer cell lines, HeLa (uterus), SK-Hep-1 (liver), and YD-15 (oral), the whole extract of feverfew showed the highest toxicity with $IC_{50}$ values of $102.58-138.68{\mu}g/mL$. Also, mallow root extract ($71.24{\mu}g/mL$) exhibited potent tyrosinase inhibitory activity comparable to arbutin ($69.56{\mu}g/mL$), which was used as the control.

Antioxidant and Anti-inflammation Effects of Water Extract from Buckwheat (메밀 추출물의 항산화 및 항염증 효능)

  • This study was conducted to investigate the effects of the hot water extract from buckwheat (WEB) in RAW-264.7 macrophage cells against lipopolysaccharide (LPS). In these experiments, we evaluated the anti-inflammatory effects of WEB by measuring MTT assay, nitric oxide (NO), inducible NOS (iNOS) production, and cyclooxygenase-2 (COX-2) expression by Western blotting. The extracts showed a protective effect by increasing cell viability on LPS in RAW264.7 cells. WEB (0.25, 0.5, and 1.0 mg/mL) significantly suppressed LPS-stimulated production of NO. Also, WEB reduced the expression of iNOS and COX-2 proteins. The present results show that WEB has potent anti-inflammatory effects on RAW264.7 cells. In addition, WEB has various antioxidant effects as a result of 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-Diphenyl-1-picrylhydrazyl (DPPH) which possess a radical scavenging activity. The total polyphenol and flavonoid contents of the WEB were $13.22{\pm}3.69mg\;GAE/g$ extract and $38.53{\pm}5.20mg\;CE/g$ extract respectively. The present results give the understanding of biological activities of buckwheat and encourage their application for supplements.

Screening of Peroxynitrite and DPPH Raoical Scavenging Activities from Salt Marsh Plants (염생식물로부터 Peroxynitrite와 DPPH 라디칼 소거 활성 검색)

  • A peroxynitrite is formed when superoxide and nitric oxide exist at near eqimolar ratio in biological systems. Although not a free radical by chemical nature, peroxynitrite is a powerful oxidant having a wide array of tissue damaging effects ranging from lipid oxidation and inactivation of enzymes and ion channels through protein oxidation and nitration to inhibition of mitochondrial respiration. During our search for new antioxidizing components from natural resources, twenty salt marsh plants were screened for their ONOO and DPPH radical scavenging activities. Among them, methanol extract of Rosa rugosa, lxeris tamagawaensis, Erigeron annus, Tetragonia tetragonoides, Imperata cylindrica, and Suaeda japonica inhibited more than 85% of peroxynitrite produced by 3-morpholinsydnonimine (SIN-1) at a concentration of 5 $\mu\textrm{g}$/$m\ell$. In addition, Rosa rugosa, Artemisia capillaris, Erigeron annus and Ixeris tamagawaensis showed significant scavenging effect against DPPH (1,1-diphenyl-2-picrylhydrazyl radical).

Effect of Ultrasound on the Decomposition of Sodium Dodecylbenzene Sulfonate in Aqueous Solution (Sodium Dodecylbenzene Sulfonate 수용액의 분해반응에서 초음파 효과)

  • The influence of ultrasound frequency, dissolved gases, and initial concentration on the decomposition of sodium dodecylbenzene sulfonate(DBS) aqueous solution was investigated using ultrasound generator with 200 W ultrasound power. The decomposition rates at three frequencies(50, 200, and 600 kHz) examined under argon atmosphere were highest at 200 kHz. The highest observed decomposition rate at 200 kHz occurred in the presence of oxygen followed by air and argon, helium, and nitrogen. The effect of initial concentration of DBS on the ultrasonic decomposition was decreased with increasing initial concentration and would depend upon the formation of micelle in aqueous solution. It appears that the ultrasound frequency, dissolved gases, and initial concentration play an important role on the sonolysis of DBS. Sonolysis of DBS mainly take place at the interfacial region of cavitation bubbles by both OH radical attack and pyrolysis to alkyl chain, aromatic ring, and headgroup.

Antioxidative Properties of Phenolic Compounds Extracted from Black Rice (흑미 색소물질에 함유된 페놀화합물의 항산화 특성)

  • The composition and antioxidative effects of phenolic compounds in black rice were studied. The contents of free and bound phenolic compounds extracted from black rice were 845.4 and 401.6 mg respectively per 100g sample weight. Free phenolic compounds had higher antioxidation ability than those of bound phenolic compounds. Solvent fractionation of free phenolic compounds revealed that butanol fraction had the highest phenolic compounds contents and antioxidative activity among other solvent fractions. Although butanol fraction showed lower lipid peroxidation inhibition (LPI) ability than that of $\alpha$-tocopherol and BHT, free radical scavenging ability was much higher than that of $\alpha$-tocopherol and BHT, as evidenced by electron donating ability (EDA) and benzoic acid hydroxylation inhibition (BAHI) assays.

Glycoprotein Isolated from Morus indica Linne Has an Antioxidative Activity and Inhibits Signal Factors Induced by Bisphenol A in Raw 264.7 Cells (뽕잎 당단백질의 항산화능과 Raw 264.7 세포에 있어서 bisphenol A에 유도된 신호전달인자의 억제)

  • The present study investigated anti-oxidative and anti-inflammatory activity of glycoprotein isolated from Morus Indica Linne (MIL glycoprotein). We found that MIL glycoprotein has a molecular weight of 32 kD and consists of carbohydrate (40.03%) and protein (59.97%), and that it has a strong scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical $({\cdot}OH)$, and superoxide anion $(O_2{\cdot}\;^-)$ radicals. In addition, MIL glycoprotein had a stable character and an optimal DPPH radical scavenging activity in the alkaline and neutral pH solution, and up to at 105. However, the results indicated that it has a minimal scavenging activity in the metal ionic solution ($Ca^{2+}$, $Mn^{2+}$, and $Mg^{2+}$) in the presence of EDTA. In addition, we further investigated whether MIL glycoprotein scavenges oxygen radicals and blocks inflammation-related signals in the bisphenol A (BPA)-stimulated Raw 264.7 cells. The results in this study showed that it has a character to scavenge the productions of reactive oxygen species (ROS) and nitric oxide (NO) dose-dependently. Also it blocked the activities of inflammation-related signals such as nuclear factor-kappa B ($NF-{\kappa}B$) and inducible nitric oxide synthase (iNOS). For example, it had an inhibitory effect on the activation of $NF-{\kappa}B$ (p50) and iNOS proteins at 200 ${\mu}g/mL$ MIL glycoprotein. Here, we speculate that MIL glycoprotein is one of natural antioxidants and of modulators of the BPA-induced inflammation.

Isolation and Identification of Antioxidant Flavonoids from Salicornia herbacea L. (퉁퉁마디에서 항산화 물질의 분리 및 동정)

  • Two flavonoids were isolated from the aerial parts of Salicornia herbacea L. by column chromatography. The structures of the flavonoid compound 1 and 2 were identified as quercetin $3-O-{\beta}-D-glucopyranoside$ and isorhamnetin $3-O-{\beta}-D-glucopyranoside$, respectively. The antioxidant activity of compound 1 was similar to those of quercetin and rutin, and the activity of compound 2, which contained methoxyl group at flavonoid B-ring, was lower than that of compound 1. The two compounds 1 and 2 were the first to be reported in this plant.
