Antioxidative and Antiproliferative Effects of Lindera glauca Blume on Human Colorectal Cancer Cells (감태나무(Lindera glauca Blume) 에탄올 추출물의 항산화 및 인체 대장암세포 증식 억제 효과에 대한 연구)

  • Various medicinal plants were collected, air-dried, and subjected to extraction with ethanol. Ethanol extracts were screened for their efficacies as antioxidative and antiproliferative agents against cancer cells. Among the 15 species, extract of Lindera glauca Blume stem with a total polyphenolic content of $70.99{\pm}1.88{\mu}g/TAE\;{\mu}g$, was found to possess high DPPH radical scavenging ($IC_{50}=30.54{\pm}0.62{\mu}g/mL$), nitrite scavenging ($IC_{50}=787.94{\pm}89.28{\mu}g/mL$), and reducing power activities ($595.76{\pm}1.90{\mu}g/mL$). The antiproliferative activities of plant extracts were determined using MTT assay in human colorectal cancer cells. Extracts of stems and roots from L. glauca Blume were found to possess high anti-proliferative activities in HT-29 and HCT116 cells ($IC_{50}=711.52{\pm}40.27{\mu}g/mL$ and $IC_{50}=85.07{\pm}4.06{\mu}g/mL$, respectively). These results suggest that L. glauca Blume extract could be a useful natural antioxidant and anticancer resource.

Comparative Radical Scavenging and Nitric Oxide Production Capacity of Water Extracts of Acanthopanax divaricatus var, Albeofructus and Eleutherococcus divaricatus var. Chiisanensis (지리산오가피와 흰털오가피 열수추출물의 라디칼 제거 활성 및 대식세포에서의 산화질소 생성능 비교)

  • In the present study, water extracts of Acanthopanax divaricatus var, albeofructus and Eleutherococcus divaricatus var. chiisanensis were prepared and their antioxidant and anti-inflammatory effects were compared. The water extract of Acanthopanax divaricatus var, albeofructus showed significantly higher activity in total polyphenol and flavonoid contents, while there were no significant differences in ABTS/DPPH radical scavenging activities and ferric reducing antioxidant power. However, the water extract of Eleutherococcus divaricatus var. chiisanensis at the highest dose showed significantly lower capacity in nitric oxide production than that of Acanthopanax divaricatus var, albeofructus. Taken together, there were differences between Acanthopanax divaricatus var, albeofructus and Eleutherococcus divaricatus var. chiisanensis in total polyphenol/flavonoid contents and anti-inflammatory effect, however, the antioxidant effect was almost similar.

Biological Activities of Various Solvent Extracts from Propolis (프로폴리스 용매별 추출물의 생리활성)

  • The scavenging effects of four extracts on the DPPH radical were increased with increasing amount of extract. However, these effects were not statistically significant. The reducing power of the extracts is increased as the amount of extract is increased. To compare reducing powers of various solvent extracts, 50% ethanol extract showed the highest reducing power. The various solvent extracts were also capable of scavenging nitrite in a manner dependent on concentration. They exhibited scavenging effects 90% on nitrite at the dose 500 p.g of water extract from propolis. The propolis extract significantly inhibited all the microorganisms tested, showing the largest inhibitory zone for Bacillus subtilis and Bacillus cereus. The 70% ethanol extract from propolis exhibited the strongest inhibitory effect on A549. The inhibitory effect of four extracts showed in that order 70% ethanol extract>95% ethanol extract>water extract>50% ethanol extract.

Chemical Composition and Antioxidative Activities of Platycodon grandiflorum Leaves and Stems (도라지 잎과 줄기의 화학성분 및 항산화활성)

  • The chemical composition and antioxidative activities of Platycodon grandiflorum leaves and stems was investigated in order to evaluate the potential as functional food material. The moisture contents of leaves and stems were 84.31% and 75.91%, respectively. The Ca content was the highest in leaves (351.49 mg%) and stems (217.56 mg%). The major free sugar of leaves was glucose (1,729.87 mg%) However major free sugar of stems was fructose (734.91 mg%). Glutamic acid (242.91 mg%) and arginine (228.60 mg%) in leaves were major amino acids, lysine (110.08 mg%) and glutamic acid (80.40 mg%) in stems were major amino acids. Oleic acid and linoleic acid were major fatty acids in crude fat of both leaves and stems. DPPH free radical scavenging activities of fractions from leaves and stems were rising with increasing amount of fractions. Like antioxidant activity, the reducing power of fractions from leaves and stems was also dependent on concentration while butanol fraction of stems showed the highest reducing power.

Isolation of Antioxidant and Antibrowning Substance from Chionanthus retusa Leaves (이팝나무 잎으로부터 항산화 및 항갈변물질의 분리)

  • This study was performed to examine in vitro antioxidative activities such as DPPH radical scavenging activity, reducing power and tyrosinase inhibitory effect of various solvent fractions from Chionanthus retusa leaves. Ethyl acetate fraction showed potent antioxidative activity and tyrosinase inhibitory effect. The active compound was isolated from the butanol fraction by silica gel column chromatography and MPLC. The isolated compound was luteolin-4'-O-glucoside determined by $^1H$, $^{13}C$-NMR and 2D NMR. Compared with several antioxidant compounds, luteolin-4'-O-glucoside exhibited effective DPPH radical scavenging activity and reducing power in a concentration dependent manner. Bioassay with pure luteolin-4'-O-glucoside showed a dose-independent inhibitory effect on L-DOPA oxidation by mushroom tyrosinase and its $IC_{50}$ values were established as 23.2 ${\mu}g/mL$. Therefore, we may suggest that luteolin-4'-O-glucoside can be used as a food additive possessing the potent antioxidative activity and skin-whitening cosmetic material.

Analysis of Total Phenolic Content and Antioxidant Activity from Fruits of Vaccinium oldhamii Miq. (정금나무 열매의 총 페놀 함량 및 항산화활성 분석)

  • Vaccinium oldhamii Miq. is a Korean native tree, which is deciduous and shrub tree with broad leaf. To develop superior cultivars for edible or medicinal purposes, we evaluated total phenolic content and antioxidant activity. Total phenolic content was measured by folin-denis method and antioxidant activity was measured by DPPH free radical scavenging activity (DPPH method) and reducing power (potassium ferricyanide method). Total phenolic content, DPPH free radical scavenging activity (at 400 ppm) and reducing power (at 700 ppm) were 15.8~19.9 mg/g, 69.1~91.4% and 0.56~0.96, respectively. A linear correlation was shown between DPPH-radical scavenging activity and reducing power (r=0969).

Effects of Lime and Phosphate Applications on Growth and Nitrogen Fixation of Alfalfa in Acid Soil (산성토양에서 석회와 인산시용이 Alfalfa의 생장 및 질소고정에 미치는 영향)

  • We investigated the effects of applications of various levels of lime(0, 250, 500 and 1,000 kg/10a) and phosphate (0, 17 and 34 kg/10a) on growth and nitrogen fixation of alfalfa (Medicago sativa L.). Effects of lime and phosphate applications were significantly different on dry matter (DM) weight of each part and on acetylene reduction activity (ARA) of alfalfa at 9 weeks alter sowing (p<.05). The effect of lime on DM of shoot and root was not significantly different at 14 weeks after sowing (early bloom stage), but that of phosphate on DM was significantly improved as increasing of phosphate levels (p<.01). The effects of lime and phosphate on ARA were significantly increased (p<.05). Application of lime and phosphate decreased total nitrogen (TN) content of each part of alfalfa at 9 weeks after sowing (p<.05). The effects of lime application on TN was higher but that of phosphate application on TN was lower than no application of lime or phosphate at 14 weeks after sowing (p<.05).

Rhizobium meliloti 102F51 Mutants Defective in Heme Synthesis (Heme 합성특성이 다른 Rhizobium meliloti 102F51 Mutant의 선별)

  • Rhizobium meliloti 102 F 51, the symbiotic partner of alfalfa, was mutagenized with N-methyl-N'-nitro-N-nitrosoguanidine (NTG) and UV-irradiation. Three group of mutants which form white, white-pink and red nodules were selected. The adetylene reduction activity, nodulation activity, amount of heme synthesis during the nodulation, and ${\delta}-aminolevulinic$ acid synthetase (ALAS) and ${\delta}-aminolevulinic$ acid dehydratase (ALAD) activities in free living rhizobia and bacteroid states of the each group of mutants were compared. The mutants forming white nodules showed lower acetylene reduction activity compared to those of red nodule forming mutants. The two key enzymes for the heme synthetic pathway, ALAS and ALAD activities of the mutants forming red nodules was much higher than those of the mutants forming white nodules in bacteroid state, however no significant difference was observed in free living state. In the nodules the ALAS was detected only in bacteroid fraction, while ALAD was detected both in bacteroid and plant fraction. ALAS was dramatically increased with the heme synthesis during the nodulation, while ALAD was decreased in plant fraction but slight increase was observed in bacteroid fraction.

DPPH Radical Scavenging Effect and Antimicrobial Activities of Plantain (Plantago asiatica L.) Extracts (질경이 추출물의 DPPH 라디칼 소거효과 및 항균활성)

  • To develop the functional food, methanol extracts of dried plantain (Plantago asiatica L.) was fractionated in hexane, chloroform, ethyl acetate, butanol and aqueous fraction. DPPH radical scavenging effect, reducing power, nitrite scavenging effect and antimicrobial activities of these fractions were investigated. Ethyl acetate fraction among these fractions showed the highest effect of all experiments. Ethyl acetate fraction showed about 60% scavenging effect on the DPPH radicals at 400 $\mu$g/5 mL. Nitrite scavenging effect of ethyl acetate and butanol fraction was more than 70~80% at concentration above of 500 $\mu$g. Among various solvent fractions from methanol extracts of plantain, ethyl acetate fraction showed the highest inhibitory effect on the microorganism such as Staphylococcus aureus, Bacillus cereus and Vibrio parahaemolyticus at 5 mg/disc.

Pectin Transeminase produced by Aspergillus niger and two yeast species. (Aspergillus niger와 이종의 Yeast에 의한 Pectin Transeliminase의 생성 및 그 특성)

  • Pectin transeliminase (PTE) was produced by Saccharomyces cerevisiae, Schizosaccharomyces pombe 4683, and Aspergillus niger in the media containing 2% pectin and examined for its characteristics. The Production of the enzyme was higher by Asp. niger than by the two yeast strains, showing that the PTE activity was proportional to reducing power. The enzyme was proved to reduce pectin and produce 4, 5- unsaturated galacturonic acid. The optimum activity of the PTE was found to be at pH 6.0 and $50^{\circ}C$. The activities of these enzyme were stable below $50^{\circ}C$ but decreased at the higher temperature. Substrate inhibition of the PTE activities was appeared at high concentrations of pectin. Those PTE activities were increased under 0.6M of KCI and NaCI, but that maximal activities at the concentration of 0.2M MgC $l_2$.
