Evaluation of Soil Microbial Population of Paddy Fields in Gyeongnam Province Area (경남지역의 논토양에서 미생물의 다양성 평가)

  • To use as a fundamental data for the sustainable agriculture, which is nowadays a major trend to keep the productivity and conserve the environment, 487 paddy soil samples were collected from 21 regions of the Gyeongnam Province and analyzed the chemical characteristics and microbial population of the soil. The microbial population densities were bacteria $298{\times}10^5$($4{\sim}3000{\times}10^5$ range), fungi $63{\times}10^3$($2{\sim}441{\times}10^3$ range), actinomycetes $19{\times}10^5$($0.2{\sim}1250{\times}10^5$ range), Bacillus sp. $53{\times}10^4$($4{\sim}890{\times}10^4$ range) and Pseudomonas sp. $198{\times}10^4CFU\;g^{-1}$($4{\sim}1724{\times}10^4CFU\;g^{-1}$ range), respectively. The microbial populations of the soil were in general higher in southern area than in the northern area of the Gyeongnam Province. The average ratio of bacteria/fungi population was 473. As soil clay content increased, the populations of aerobic bacteria, actinomycetes and Pseudomonas sp. were remarkably decreased. The ratio of aerobic bacteria and fungi was 1554 in sandy loam and clay loam 1144, while Bacillus sp./fungi ratio was 11 in clay loam and 10 in loam. On the topographical differences, aerobic bacteria and Bacillus sp./fungi ratio were the higher in coastal plains than any other areas. The microbial population densities from different soil types were generally lower in ill-drained paddy field than those of other paddy field. The content of $P_2O_5$, K, Ca, $NO_3-N$ and EC in soil were positively correlated to the population densities of aerobic bacteria, actinomycetes, fungi, Bacillus sp. and Pseudomonas sp.. The soil organic matter and Mg content were also positively correlated to the population densities of aerobic bacteria, actinomycetes, fungi and Bacillus sp.

Biological Hazard Analysis of Paprikas, Strawberries and Tomatoes in the Markets (유통중인 파프리카, 딸기 및 토마토의 생물학적 위해요소 분포 조사)

  • The consumption of "ready-to-eat" agricultural products is recently increasing and the safety of these agricultural products is forefront of public concerns. The 120 samples of paprikas, strawberries and tomatoes, which are the representative exported agricultural products, were purchased at the department stores and discount stores in Daejeon. And we determined the microbiological and parasitological contamination level of these agricultural products using culture media, multiplex PCR, commercial bacterial detection kit and microscopy, and also evaluated the decontamination method. Mean counts of total aerobic bacteria from these agricultural products ranged from $1.3{\times}10^4$ CFU/g to $1.8{\times}10^5$ CFU/g, and mean counts of coliforms ranged from $1.4{\times}10^3$ CFU/g to $9.6{\times}10^3$ CFU/g. There was no significant difference in the level of bacterial contamination between the agricultural products from department stores and the ones from discount stores. Strawberry showed the highest contamination level for the bacteria and we also found the unidentified parasite eggs. Enterobacter cloacae was the most frequently isolated bacteria strain, but no food poisoning pathogenic bacteria except Staphylococcus aureus was isolated from the products by multiplex PCR. Compared to unwashed products, tab water-washed ones showed 80% decrease of the counts of total aerobic bacteria on the agricultural products, and the rates decreased more by incorporating detergent or ultrasonic wave treatment. We concluded that the biological contamination levels among paprikas, strawberries and tomatoes were the highest in strawberries, but there were not significant difference according to distribution systems.

  • The purpose of this study was to evaluate microbiological contamination of fresh-cut produce salads and raw cabbage toward climate change. Total aerobic bacteria, coliform and Escherichia coli were monitored to get the contamination levels and E. coli O157:H7, Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes and Salmonella spp. to detect pathogens with risk of foodborne disease from samples. Collection of 360 samples (180 fresh-cut produce salads and 180 raw cabbage), including 60 samples from each area after setting 3 areas depending on annual temperature and annual rainfall. As a result, total aerobic bacteria and coliform group were different was performed areas in raw cabbage but there was no difference between areas in fresh-cut produce salads. In addition foodborne pathogens including E. coli were not isolated from fresh-cut produce salads.

Effects of Meju Manufacturing Periods on the Fermentation Characteristics of Kanjang, Korean Traditional Soy Sauce (메주의 제조기간에 따른 재래간장의 발효특성)

  • This study was carried out to clarify the microorganisms which participated in the fermentation of kanjang. The changes in the viable cell counts of total aerobic bacteria, lactic acid bacteria and yeasts for raw soybean, soybean during cooking, meju during cultivation, and kanjang mash during maturing were investigated along with the changes in components during those periods. Lactic acid bacteria that were found to be $6{\times}10^2\;CFU/g$ in raw soybean were disappeared after cooking process, but total aerobic bacteria were diminished from $1.9{\times}10^6\;CFU/g$ to $10^2\;CFU/g$. Aerobic bacteria of inner and outer parts of meju increased to more than $10^9\;CFU/g$. The higher viable cell counts of lactic acid bacteria in the inner parts of meju were observed than those in outer ones. On the contrary, significantly higher viable cell counts of yeasts in the outer parts of meju were found. Total nitrogen content and color density of kanjang increased by using meju with extended cultivation periods. No significant differences were observed in microbial counts between kanjang mash with aeration and non-aeration during kanajng mash maturing.

Improved Preservation and Shelf-life Extension of Fish Cake by Modified Atmosphere Packaging (변형기체포장에 의한 어묵의 저장성 향상 및 유통기한 연장)

  • Modified atmosphere packaging (MAP) of vacuum, 60% $N_2/40%$ $CO_2$, 100% $N_2$ and 100% $CO_2$ were examined to preserve fried fish cakes and extend their shelf life. When product packages in air were stored at 5, 10, and $15^{\circ}C$ with measurement in quality attributes, aerobic bacterial growth was found to be primary quality factor to determine the shelf life of the product. Huang's model was adopted to obtain the parameters for use in shelf life determination for a variety of packaging and storage conditions. Temperature dependence expressed in Arrhenius equation showed activation energies of 139.4 and 41.8 kJ/mol for inverse of lag time and growth rate of the microbial spoilage, respectively. MAP of 100% $CO_2$ could double the product shelf life at $10^{\circ}C$ compared to air package. High $CO_2$ concentration MAP at low temperature was found to be effective to prolong the shelf life.

Reduction of Pathogenic Bacteria in Organic Fertilizer using Gamma-Irradiation (감마선 조사를 이용한 유기질 퇴비의 병원성 미생물 저감화)

  • Organic compost is a useful fertilizer for organic farming. However, it poses a microbiological hazard to the farm produce because most of the compose are originated from excrementitious matters of domestic animals. Irradiation was performed to improve microbiological safety of organic compost and the effectiveness of gamma-irradiation for inactivating Salmonella typhimurium and E. coli was investigated. Total aerobic bacteria and coliform bacteria in 16 produce or commercial compose were ranged from $10^5\;to\;10^7\;CFU/mL\;and\;0\;to\;10^3\;CFU/mL$, respectively. All coliform bacteria in the composts were eliminated by irradiation at 5 kGy, while about $10^2\;CFU/mL$ of total aerobic bacteria survived up to 10 kGy of irradiation. In the inoculation test the test organisms (inoculated at $10^7\;CFU/mL$) were eliminated by irradiation at 3 kGy. $D_{10}$ values of inoculated Salmonella typhimurium and E. coli in the compost were $0.4{\pm}0.05\;and\;0.39{\pm}0.03kGy$. It was considered that $3{\sim}5kGy$ of gamma irradiation was effective for radicidation (radiation sterilization of pathogenic microbes) of organic fertilizer.

Microbiological changes and quality characteristics of dried persimmon by chlorine dioxide gas fumigation treatment (이산화염소 가스 훈증 처리에 따른 곶감의 미생물학적 변화 및 품질특성)

  • This study was conducted to investigate the effect of chlorine dioxide fumigation as a substitute for sulfur fumigation which has been used as a method to prevent the quality change of persimmon during storage and distribution process. Dried persimmons were treated with chlorine dioxide gas concentration (0, 15 30, and 45 ppm) and time (0, 15, 30, and 45 min) and microbiological changes, texture properties and color of the treated samples were investigated during storage at room temperature. Total aerobic bacteria, yeast and mold numbers after chlorine dioxide gas fumigation were decreased when compared with the control group. The inhibitory effects of total aerobic bacteria, yeast, and mold were observed during storage. The texture properties and color value of dried persimmons were not affected by chlorine dioxide gas fumigation concentration and time. There was no difference in quality between chlorine dioxide gas fumigation treatment group and control group. These results suggested that chlorine dioxide gas fumigation treatment can be utilized as a processing technique to secure microbiological storage stability of dried persimmons.

Occurrence of acid producing bacteria in Meju leaves (재래식 메주중의 산생성균의 분포)

  • The distribution of acid producing bacteria and general bacteria in 23 samples of Korean traditional Meju loaves was investigated and the strains isolated from the samples identified. The acid producing bacteria occurred more in outer part than inner part and anaerobic acid producing bacteria showed higher tendency of occurrence compared with the aerobes in each part. The average number of nonhalophilic and halotolerant bacteria belonging to aerobes were counted as $24{\times}10^6$ and $33{\times}10^5$ and the average number of those belonging to anaerobes $10{\times}10^7$ and $58{\times}10^5$ cells/g, respectively. The general bacteria isolated more in outer part than inner part and its average number was $62{\times}10^7$ cells/g. In the isolates, 2 aerobic acid producing strains were identified as Micrococcus spp., 3 anaerobic acid producing strains as Streptococcus sp., Pediococcus sp. and Lactobacillus sp., and 2 strains of aerobic general bacteria as Bacillus spp.

  • C-1 compounds are observed in anaerobic sediment of high salt environments. Thus, surface sediments and waters from these environments are therefore potential habitats for aerobic methylotrophic microorganisms. The soil samples collected from saltern and tidal flat as inoculums and methanol as carbon and energy source was supplied. After subculture depending on the salt concentration, methanol oxidizing bacteria growth condition investigated, the results of methanol oxidizing bacteria can grow in salt conditions, and the maximum concentration was 20%. Analysis based on denaturing gradient gel electrophoresis of 16S rRNA genes indicates that Methelyophaga-like bacteria were dominants of methylotrophs in the enrichment culture. Quantitative PCR showed that archaeal cells were about 1-10% of bacterial cells. Additionally archaea were assumed not to be involved in methanol oxidation since bacterial antibiotics completely blocked the methanol oxidation. Our results suggest that Methelyophaga-like bacteria could be involved in C-1 compounds oxidation in hypersaline environments although those activities are sensitive to salinity above 20%.

Effects of Cultivation Environment and Fruit Ripeness on Microbial Load in Mulberry (재배환경 및 과실 숙도가 오디의 미생물학적 부하량에 미치는 영향)

  • This study was conducted to investigate the microbial loads in mulberry fruits depending on cultivation environment and fruit ripeness. The population levels of total aerobic bacteria in mulberry fruits collected from open field orchards were higher than those from three plots protected within plastic green houses. In regards to fruit ripeness, the levels of total aerobic bacteria in ripe black fruits were higher than those in unripe green and red mulberry. From the farms into where livestock animals were allowed to enter, Escherichia coli was detected in soil at a level of 4.26~4.94 log CFU/g and in mulberry fruits at 5.03~6.07 log CFU/g, while no coliform and E. coli were detected from where the intrusion of livestock was prevented. We also examined the density change of inoculated E. coli in mulberry fruits as they were becoming mature. While E. coli did not increase in green fruits, two and four log CFU/g increases at $20^{\circ}C$ and $37^{\circ}C$, respectively, were observed with red and fully mature black mulberries during 48 hours incubation. To ensure the food safety of mulberry, it is suggested that the introduction of E. coli into a farm through livestock should be prevented and more hygienic caution should be taken especially when the fruits are ripe.
