Effect of $\gamma$-Irradiation of Volatile Organic Compounds of Houttuynia cordata Thunb. (방사선 조사에 의한 어성초의 휘발성 유기성분 변화)

  • This study was performed to examine the effect of $\gamma$-irradiation on the volatile organic compounds in Houttuynia cordata Thunb.(H. cordata). 1be volatile compounds of non-irradiated and 10 kGy $\gamma$-irradiated H. cordata were isolated using SDE apparatus and analyzed by GC/MS. For each treatment the number of volatile compounds were detected at 83 and 85, respectively, 1be predominant functional groups of volatile organic compounds from H. cordata were identified as alcohols and ketones. H. cordata was mainly composed of hexahydrofarnesyl acetone (12.81 %), phytol, decanoic acid, dodecanoic acid, octadecanol, caryophyllene oxide, 2-undecanone and menthol. Houttuynum which is characteristic compound of H. cordata was found in all samples and there was no significant difference with irradiation doses. Consequently, $\gamma$-irradiation of H. cordata would be an effective process for sanitation and to increase extraction efficiency.

Antifungal Activity of Phenanthrene Derivatives from Aerial Bulbils of Dioscorea batatas Decne (재배마 (Dioscorea batatas Decne)의 주아로부터 분리된 phenanthrene 유도체의 항진균 활성)

  • Plants of the genus Dioscorea have long been used as oriental folk medicine, and Dioscorea batatas Decne has been cultivated for healthy food in Korea. Although the bulbils were produced 2,000 ton annually, there are few reports for bioactive compounds in bulbils. In this study, three phenanthrenes and two phenanthraquinones were isolated from the aerial bulbils of D. batatas Decne, and their structures were elucidated. Among them, compound 2 (6-hydroxy-2,7-dimethoxy-1,4-phenanthraquinone) has not been reported previously. Evaluation of antimicrobial activities based on disk-diffusion assay, MIC and MFC showed the compound 12 (6,7-dihydroxy-2,4-dimethoxyphenanthrene) has strong antimicrobial activity with $25\;{\mu}g/ml$ of MIC and MFC against Candida albicans. Our results suggested that compound 12 has a potent antifungal activity, and the antimicrobial activity and its spectrum are modulated by hydroxylation and methoxylation of phenanthrene ring moiety of the compound.

  • Recent studies on synthesis of metallic nanomaterials such as silver (Ag), gold (Au), platinum (Pt), cerium (Ce), zinc (Zn), and copper (Cu) nanoparticles (NPs) using plants and microbes are attracted researchers for their wide range of applications in the field of biomedical sciences. The plant contains abundant of bioactive contents such as flavonoids, alkaloids, saponins, steroids tannins and nutritionals components. Similarly, microbes produce bioactive metabolites, proteins and secretes valuable chemicals such as color pigments, antibiotics, and acids. Recently reported, biogenic synthesis of NPs in non-hazardous way and are promising candidates for biomedical applications such as antibacterial, antifungal, anti-cell proliferative and anti-plasmodia activity. All those activities are dose dependent, along with their shape and size also matters on potential of NPs. Microbes and plants are great source of metabolites, those useful in biomedical field, such metabolites or chemicals involved in synthesis of NPs in an ecofriendly way. NPs synthesized using microbes or plant materials are reveals more non-toxic, facile, and cost-effective compare to chemically synthesized NPs. In present review we are focusing on NPs synthesis using biological agents such as microbes (bacteria, fungi and algae) and plant, characterization using different techniques and their antibacterial applications on pathogenic Gram-positive and Gram-negative organisms.

Isolation and Characterization of 𝛽-Glucosidase-Producing Yeast, Rhodotorula sp. GYP-1 (𝛽-Glucosidase 생성 효모 Rhodotorula sp. GYP-1의 분리 및 특성)

  • Nine microbial strains were isolated from the byproduct of ginseng processing and field of ginseng cultivation. Two strains among them were confirmed. Phylogenetic analysis of these 𝛽-Glucosidase strains confirmed that strain GYP-1 belongs to the Rhodotorula and strain GYP-3-3 belong to genus Brachybacterium. Rhodotorula sp. GYP-1 was finally selected due to its high biomass production. The 𝛽-Glucosidase activity of Rhodotorula sp. GPY-1 was assessed at 30 ℃, and Higher than 70% of the enzyme activity was maintained at the temperature range of 20-40℃. Although the optimum pH for the highest enzyme activity was pH 5.0, the enzyme was stable throughout the pH range of 5.0-8.0. In addition, Rhodotorula sp. demonstrated antifungal activity against the ginseng root rot disease caused by Botrytis.

Prodigiosin Production From Serratia sp. PDGS120915 Isolated From Daeyeon Stream Water in Busan (하천에서 분리한 Serratia sp. PDGS120915의 프로디지오신 생산)

  • Prodigiosin is a red pigment characterized by a common pyrrolylpyrromethane skeleton. It is produced by Serratia marcescens, Vibrio psychroerythrus, Hahella chejuensis, etc. Prodigiosin has been reported to possess anticancer, immunosuppressant, antifungal antimalarial, and algicidal activities. However, despite prodigiosin's diverse range of activities, its production rate is significantly low and biosynthesis conditions are difficult. Consequently, the selling price is high, and its usability is limited. This study aimed to increase the efficiency of prodigiosin production according to the culture conditions of Serratia. In this study, a bacterial strain PDGS120915 producing prodigiosin was isolated from lightly contaminated stream water in Busan and identified as a strain of Serratia sp. based on 16S rDNA gene sequence analysis and physiological characteristics. The reddish pigment from PDGS120915 was directly extracted using acidified ethanol, and a characterization analysis confirmed that it was a prodigiosin compound. The optimal conditions for pigment production were 25℃, pH 7, and 0% NaCl concentration for a duration of 14 hr. Furthermore, by treating carbon and nitrogen sources, such as fructose and beef extract, respectively, prodigiosin production increased approximately six-fold and four-fold. Among the minerals tested, 0.1% KCl was found to be the most effective for prodigiosin production. Moreover, casein was identified as the most suitable source for prodigiosin production.

Isolation and Characterization of Plant Growth Promoting Rhizobacterium Bacillus subtilis YK-5 from Soil (토양에서 식물생육촉진 활성을 가진 균주 Bacillus subtilis YK-5의 분리 및 특성)

  • For the isolation of a plant growth-promoting rhizobacteria, strain YK-5 was selected from approximately 400 thermostable strains isolated from special soil samples. Strain YK-5 produced an antifungal compound, and optimum carbon and nitrogen sources for the production of the antifungal compound were investigated against Aspergillus flavus as a test strain. Modified LB medium containing 1% peptone, 1% yeast extract and 5% black sugar was determined to be the optimal medium for growth and antifungal compound production. Culture broth of strain YK-5 potently inhibited growth of the phytopathogenic fungus Fusarium oxysporum KACC 40052 for 7 days. The plant growth-promotion function of strain YK-5 was tested against radish and rice in pot trials. Leaf number, plant height and root length in YK-5-treated radish markedly exceeded (> 60%) those of untreated radish. Leaf length and white rootlet development were markedly more prominent than in commercially-treated rice plants. Strain YK-5 was determined to be Bacillus subtilis YK-5 by physiological, chemotaxonomical, and phylogenetical analyses.

Thyrotropin-Binding Inhibiting Immunoglobulin(TBII) in Patients with Autoimmune Thyroid Diseases (자가면역성 갑상선질환에서의 혈청 Thyrotropin-Binding Inhibiting Immunoglobulin치)

  • In order to evaluate the significance of thyrotropin-binding inhibiting immunoglobulin (TBII) in the patients with autoimmune thyroid diseases, the authors investigated 402 cases of Graves' disease and 230 cases of Hashimoto's thyroiditis comparing 30 cases of normal healthy adult at Kyung Pook University Hospital from February 1993 to August 1994. The TBII was tested by radioimmunoassay and assesed on the dynamic change with the disease course, thyroid functional parameters, and other thyroid autoantibodies : antithyroglobulin antibody (ATAb) and antimicrosomal antibody (AMAb) including thyroglobulin. The serum level of TBII was $40.82{\pm}21.651(mean{\pm}SD)%$ in hyperthyroid Graves' disease and $8.89{\pm}14.522%$ in Hashimoto's thyroiditis and both were significant different from normal control of which was $3.21{\pm}2.571%$. The frequency of abnormally increased TBII level was 92.2% in hyperthyroid Graves' disease, 46.7% in euthyroid Graves' disease or remission state of hyperthyroidism, and 23.9% in Hashimoto's thyroiditis. The serum levels of increased TBII in Graves' disease were positively correlated with RAIU, serum T3, T4, and FT4, but negatively correlated with serum TSH(each P<0.001). The TBII in Graves' disease had significant positive correlation with serum thyroglobulin and AMAb, but no significant correlation with ATAb. In the Hashimoto's thyroiditis, the serum levels of TBII were positively correlated with RAIU, serum T3, TSH and AMAb, but not significantly correlated with serum T4, FT4, thyroglobulin and ATAb. Therefore serum level of TBII seemed to be a useful mean of assessing the degree of hyperthyroidism in Graves' disease and correlated well with thyroidal stimulation. The serum level of TBII in Hashimoto's thyroiditis is meaningful for the degree of both functional abnormality reflecting either hyperfunction or hypofunction and the immune logic abnormality.

  • This study was undertaken to assess the susceptibility of pigmented and nonpigmented strains of Serratia marcescens to antibiotics and human sera, and the effect of culture filtrates from pigmented and nonpigmented of Serratia marcescens on humoral and cellular immune responses in mice to thymus-dependent and indepependent antigens. Humoral immune response was measured by hemagglutinin (HA) and hemolysin (HE) to sheep red blood cell (SRBC), and Arthus or antibody response to polyvinylpyrrolidone (PVP). The cellular immune response was measured by delayed-type hypersensitivity (DTH) determined by footpad swelling reactin to SRBC. The resistance of pigmented strains of Serratia marcescens to the bactericidal action of heat inactivated human serum was insignificantly greater than that of nonpigmented strains. However, the pigmented strains were significantly more resistant to the bactericidal action of heat-untreated human serum than that of nonpigmented strains. The clinical isolates of Serratia marcestens was also tested for their resistance to several antibiotics. There was no difference between the pigmented and non-pigmented strains in the resistance to carbenicillin. However, nonpigmented strains were more resistant to gentamicin, kanamycin and tobramycin than the pigmented strains. The intraperitoneal administration of culture filtrates from the pigmented or nonpigmented strains into mice caused enhancemented of antibody response to SRBC or PVP, and of DTH to SRBC. Besides, their enhancement of immune responses was more prominent when culture filtrate from the pigmented strains was administered.

Fungal frontal sinusitis in a dog (개의 진균성 전두동염)

  • A 6-year-old male Basset hound weighing 9.3 kg was presented with a history of severe mucopurulent discharge from the bilateral fronto-nasal area which had been developed progressively over 1 year. At admission, the physical state and appetite of the patient was poor. There was bilateral thick and mucoprulent discharge on the fronto-orbital area with fistula opening in the skin over the frontal bone. A sample of exudate was cultured and fungi were isolated. Radiographically, there was an increased diffuse opacity of the frontal sinus with decreased definition and a thickening of the mucous membrane lining the sinus. This case was finally diagnosed as chronic fungal frontal sinusitis. After surgical obliteration of the sinus, local and systemic antifungal therapy with chlorhexidine and ketoconazole were applicated. The dog had gradually recovered the physical state and appetite.

  • Chronic granulomatous disease in childhood is a rare inheritable disorder of phagocytic cells in which defective production of the reactive intermediates of oxygen predisposes the patient to severe recuring pyogenic infections. The lung is the most common site of infection and pulmonary disease is the primary cause of death for greater than 50% of children with chronic granulomatous disease. Although the role of surgery in management of this disease remains undefined, rapid diagnosis of the underlying pulmonary problem is crucial to determine the most appropriate antimicrobial therapy and surgical techniques such as lobectomy of involved areas lead to more rapid recovery and thus allow the antibiotics to be more efficacious in these cases. We have treated a one month old male baby who had the chronic granulomatous disease with pulmonary infection. Wide surgical resection of the affected lobe and use of antibiotics and antifungals were carried out with good clinical results. He was well after the operation.
