Antifungal Activity of Lactobacillus plantarum Isolated from Kimchi (김치로부터 항진균 활성 Lactobacillus plantarum의 분리 및 특성 규명)

  • A lactic acid bacterium having antifungal activity was isolated from kimchi. It was identified as Lactobacillus plantarum based on its morphological and biochemical properties, and 16S rRNA sequence, and designated as Lb. plantarum AF1. This isolate inhibited the growth of Aspergillus flavus ATCC 22546, A. fumigatus ATCC 96918, A. petrakii PF-1, A. ochraceus PF-2, A. nidulans PF-3, Epicoccum nigrum KF-1, and Cladosporium gossypiicola KF-2 under a dual culture overlay assay. Also, the antimicrobial activity was found to be active against various species of Gram-positive and Gram-negative bacteria. The antifungal activity was found to be stable after heat ($121^{\circ}C$, 15 min) and proteolytic enzyme treatment, but it was unstable over pH 5.0. The antifungal compound(s) was estimated to have a low molecular mass (below 3,000 Da).

Culture Conditions and Antifungal Activity of Bacillus licheniformis KMU-3 against Crop Pathogenic Fungi (작물병원성 곰팡이에 대한 Bacillus lichentformis KMU-3의 항진균활성과 배양조건)

  • Bacillus licheniformis KMU-3 shown a strong antifungal activity was isolated from Swedish forest soils. B. licheniformis KMU-3 produced a maximum level of antifungal substance under incubation aerobically at $24^{\circ}C$ for 24 hours in LB broth containing $1.0\%$ sodium acetate, $1.0\%$ ammonium sulfate at 180 rpm and initial pH adjusted to 8.0. Chloroform extraction of culture broth was confirmed inhibitory zone by plate assay and Rf value 0.49 substance by thin layer chromatography (TLC) represented high antifungal activity against Rhizoctonia solani AG-1. This substance also exhibited against Rhizoctonia solani AG-4, Colletotrichum orbiculare, Colletotrichum gloeosporioides, Cladosporium cucumerinum, Fusarium oxysporum, and Fusarium graminearum.

Bacillus polyfermenticus CJ9, Isolated from Meju, Showing Antifungal and Antibacterial Activities (메주로부터 분리한 항진균 및 항세균 활성의 Bacillus polyfermenticus CJ9)

  • A CJ9 bacterial strain, which showed antifungal and antibacterial activities, was isolated from meju and identified as Bacillus polyfermenticus based on Gram staining, biochemical properties, as well as its 16S rRNA sequence. B. polyfermenticus CJ9 showed the antimicrobial activity against the various pathogenic molds, yeasts, and bacteria. The antibacterial activity was stable in the pH 5.0~9.0, but the activity was lost at $37^{\circ}C$ for 24 hr. The antifungal activity was stable in the pH range of 3.0~9.0 and reduced at $121^{\circ}C$ for 15 min, but antifungal activity was not completely destroyed. The antibacterial activity was completely inactivated by proteinase K, protease, trypsin, and $\alpha$-chymotrypsin. The antifungal activity was also completely inactivated by protease and $\alpha$-chymotrypsin, and reduced its activity by proteinase which indicated that the antifungal and antibacterial compounds have proteineous nature. The apparent molecular mass of the partially purified antifungal compound, as indicated by using the direct detection method in Tricine-SDS-PAGE, was approximately 1.4 kDa. The molecular mass of the antibacterial compound could not be determined because of its heat-liable characteristic.

Comparison of Antifungal Activites of Monoterpenes and Sesquiterpenes in Essential Oil from Chamaecyparis obtusa against Dermatophytes (피부사상균에 대한 편백정유의 Mono- 및 Sesquiterpene 항진균 활성 비교)

  • This study was to compare the antifungal activitiy of monoterpenes with that of sesquiterpenes, which were major constituents of antifungal active essential oil of Chamaecyparis obtusa, to investigate which constituents had more effective against dermatophytes, and to evaluate the synergistic effect of combination of the antifungal active constituents. The antifungal activities of seven fractions (A, B, C, D, E, F, and G) from C obtusa essential oil by column chromatography were tested against Trichophyton mentagrophytes. Fraction D, E, F, and G were more active than fraction A, B, and C, and their major compounds were mono- and sesquiterpenes analyzed by GC/MS. Borneol, linalool, and ${\alpha}$-terpineol were selected as monoterpenes, and ${\alpha}$-cedrol, nerolidol, and ${\beta}$-eudesmol as sesquiterpenes, respectively. The antifungal activities of the constituents were respectively examined against Trichophyton mentagrophytes (KCTC6077), Microsporum canis (KCTC6591), and Microsporum gypseum. The constituents of sesquiterpenes were more active than those of monoterpenes. By comparing single and combined sesquiterpenes with fraction F containing the higher ratio of sesquiterpenes, combined sesquiterpenes were the most active. This result indicated that there was the synergistic antifungal effect against dermatophytes when sesquiterpenes were combined together.

Fungicidal and Hemolytic Activity of Cecropin A-Magainin 2 Analogue Peptides against Tri-chospoon beigelii and Human Red Blood Cells (Cecropin A-Magainin 2 유도체 펩티드의 Trichosporon beigelii에 대한 항진균 활성 및 인간 적혈구 세포에 대한 용혈활성)

  • In order to design a novel synthetic peptide with improved fungicidal activity but low hemolytic activity, a hybrid peptide, cecropin A(l-8)-magainin 2(1-12), and its analogue peptides were synthesized by the solid phase method. Antifungal and hemolytic activities of the synthetic peptides were assessed by the growth inhibition against Trichosporon beigelii and the cell membrane lysis against human red hlood cells, respectively. Analogue 2 in which Lys at position 12 in cecropin A(1-8)-magainin 2(1-12) was substituted with Ala showed most potent antifungal activity (MIC: 2.5.$\mu$g/ml) with minimal hemolytic activity (0.5% hemolysis at the (200.$\mu$g/ml peptide). This peptide (A2), therefore, could be useful as a model for further designing potent antifungal peptides without cytotoxicity.

Detection of Antifungal Activities from Pomegranate (석류에서 항진균성 활성의 탐색)

  • Antifungal activities of pomegranate were investigated. Seventy percent ethanol extracts of seeds, peels, and whole fruits of pomegranate showed similar antifungal activities against Candida albicans in liquid media, whereas extract of whole fruit showed relatively high antifungal activity in solid media. When 70% ethanol extracts were fractionated sequentially with chloroform and ethyl acetate, ethyl acetate fraction exhibited the highest anti-fungal activities against C. albicans. Ethyl acetate fractions of whole fruits and peel portions showed at least 36% and 25% growth against C. albicans, Candida glabrata, Candida tropicalis, and Candida lusitaniae in liquid media, respectively. These results indicate that pomegranate contains antifungal compounds soluble with organic solvents.

  • Bacillus sp. KMU-991 was isolated from Oslo city soils at Norway and shown a strong antifungal activity on red-pepper anthracnose, Colletotrichum gloeosporioides. Bacillus sp. KMU-991 produced a maximum level of antifungal substrate under aerobic incubation at $30^{\circ}C$, 180 rpm for 48 hours in TSB medium(initial pH 7.0) containing 1.0% mannitol and 1.0% ammonium chloride. Precipitate of culture broth by $30{\sim}60%$ ammonium sulfate precipitation exhibited strong antifungal activity against C. gloeosporioides KACC 40804. Butanol extract of cultured broth also shown fungal growth inhibitory activity against Fusarium oxysporum f. sp. radicus-lycopersici KACC 40537, Rhizoctonia solani AG-4 KACC 40142, Botrytis cinerea KACC 40573, Colletotrichum orbiculare KACC 40808, and Phytophthora cambivora KACC 40160 by agar diffusion method.

Characterization of Bacteriocin-Like Substances Produced by Bacillus subtilis MJP1 (Bacillus subtilis MJP1이 생산하는 Bacteriocin-Like Substances)

  • The MJP1 bacterial strain, which possesses antifungal activity, was isolated from meju and identified as Bacillus subtilis based on its morphological and biochemical properties, as well as its 16S rRNA sequence. Antimicrobial activity was found against various species of Gram-positive bacteria, yeasts, and molds, including food-spoilage microorganisms. The antifungal activity was found to be stable after heat and proteolytic enzyme treatment, and in the pH range of $6.0{\sim}10.0$. The antibacterial activity was stable in the pH range of $6.0{\sim}10.0$, but about 50% of the activity was lost after 24 hr at $30^{\circ}C$. The antibacterial compound was also inactivated by proteolytic enzyme treatment, indicating its proteinaceous nature. The apparent molecular masses of the partially purified antifungal and antibacterial compounds, as indicated by using the direct detection method in Tricine-SDS-PAGE, were approximately 2.4 kDa and 4.5 kDa, respectively. These studies suggest that B. subtilis MJP1 produces two bacteriocin-like substances with antifungal and antibacterial activities.
