A Study on Whitening and Anti-inflammatory Effects of Eriobotrya Japonica Leaf Extracts with Different Extraction Methods (추출 방법에 따른 비파엽 추출물의 미백 및 항염활성에 관한 연구)

  • In this study, we prepared Eriobotrya japonica leaf extracts by several extraction processes and then evaluated their biological activities for their potential application as a new raw material of functional cosmetic. Their whitening effects were measured by tyrosinase inhibitory activities, and anti-inflammatory effects were determined by inhibitory activities of nitric oxide (NO) production. Among the several extracts obtained from E. japonica leaf, supercritical fluid extract showed tyrosinase inhibitory activities at the concentration of 10%. Inhibitory activity on NO production effect related to anti-inflammatory efficacy was in the order: supercritical fluid extract > ethanol extract > hot water extract. According to the results of MTT assay, cell cytotoxicity was not observed at all concentrations except for a 5% concentration of the 70% ethanol extract. For whitening effects, 30% ethanol and 70% ethanol extract showed mushroom tyrosinase inhibitory activity at the concentration of 5%. These results indicated that E. japonica leaf extracts could have the functional effects when they are added as ingredients in cosmetics.

Anti-oxidative and Anti-inflammatory Constituents from the Extracts of Hydrangea macrophylla Flowers (수국 꽃 추출물 유래 항산화 및 항염 활성 성분)

  • In this study, the extracts of Hydrangea macrophylla (H. macrophylla) flowers were investigated for the anti-oxidative and anti-inflammatory activities, and their active constituents were identified. The anti-oxidative effects were tested by DPPH and ABTS+ assays. To evaluate anti-inflammatory activities, LPS-induced RAW264.7 cells were examined. Among the extracts, the ethyl acetate fraction showed potent radical scavenging activities and inhibition of nitric oxide (NO) production. Chromatographic purification of the extract led to isolation of the compounds; hydrangenol (1), prunin (2) and astragalin (3). The chemical structures of the constituents were elucidated based on spectroscopic data including NMR spectra, as well as comparison of the data in the literature values. Quantitative analysis by high pressure liquid chromatography (HPLC) determined hydrangenol (1) as the major constituent. Isolated compounds 1-3 decreased the NO level without causing cell toxicities. Based on these results, it was suggested that the extract from H. macrophylla flowers could be potentially applicable as an anti-oxidative and/or anti-inflammatory ingredients.

A Study on the Antioxidant, Whitening and Anti-Inflammatory Activities of Trichosanthis Cucumeroidis Radix Extract (쥐참외뿌리 추출물의 항산화, 미백, 항염증 활성 연구)

  • In this study, the physiological activity effect of trichosanthis cucumeroidis radix extract on antioxidant activity, whitening, and anti-inflammatory activity was checked, and the possibility of its use as a functional material was checked. The purpose of this study was to confirm the antioxidant activity through DPPH radical scavenging activity of trichosanthis cucumeroidis radix extract, whitening activity effect through melanin production inhibition ability for melanin cell B16F10 melanoma cell, and anti-inflammatory activity effect through NO production inhibition ability for macrophage RAW 264.7 cell. As a result of the study, the concentration-dependent DPPH radical scavenging activity of the trichosanthis cucumeroidis radix extract was confirmed, and DPPH radical scavenging activity similar to that of the positive control Ascorbic acid was confirmed. It was confirmed that the melanin production inhibitory activity induced by 100 nM 𝛼-MSH and the NO production inhibitory ability induced by LPS 1 ㎍/mL were significantly suppressed. Accordingly, it is considered that the trichosanthis cucumeroidis radix extract may be used as a functional material having antioxidant, whitening, and anti-inflammatory effects.

A Study on the Physiological Activity of Cosmetics in Bupleurum falcatum L. Extract (시호 추출물의 화장품 생리활성에 관한 연구)

  • Bupleurum falcatum L. is one of the medicinal crops and has excellent pharmacological activities such as reducing fever, sedation, inhibition of hepatopathy, anti-inflammatory, and anticancer. Currently, it is used for food or medicine, but the use as a cosmetic is insufficient, so the suitability and use value as a cosmetic material were reviewed. Using the Bupleurum falcatum L. extract, polyphenol, flavonoid, antioxidant, and anti-inflammatory tests were conducted. The total polyphenol content was 14.71±0.16 mg·TAE/g, and the total flavonoid content was 1.42±0.05 mg·QE/g. The antioxidant activity showed excellent activity in a concentration-dependent activative, and the NO expression inhibition ability was 61.21% at 1,000 ㎍/mL, confirming that the anti-inflammatory activity was excellent. Through this study, the antioxidant and anti-inflammatory effects of Bupleurum falcatum L. extract are excellent, so it is considered to be of sufficient value as a natural material for cosmetics.

The Effects of Anti-Inflammatory Activities and Active Fractions Analysis of Ethanol Extract from Red Rose Petals (붉은 장미꽃잎 에탄올 추출물의 활성 분획물 분석 및 항염증 활성 효과)

  • Red rose petals are usually disposed but they are an abundant source of phenolics and traditionally used as food supplement and as herbal medicine. Of the Various phenolics, they are known to have anticancer, antioxidant, and anti-inflammatory properties. In this study, we investigated the anti-inflammatory effects of red rose ethanolic extracts(GRP) on lipopolysaccharide (LPS)-activated RAW 264.7 cells. The results demonstrated that pretreatment of GRP(500㎍/mL) significantly reduced NO production by suppressing iNOS protein expression in LPS-stimulated cells. Anti-inflammatory effects byred rose petal were observed in the following. Red rose petal inhibited the translocation of NF-κB from the cytosol to the nucleus via the suppression of IκB-α phosphorylation and also inhibited LPS-stimulated NF-κB transcriptional activity. These findings suggest that red rose petal exert anti-inflammatory actions and help to elucidate the mechanisms underlying the potential therapeutic values of red rose petal. Therefore, red rose petal could be regarded as a potential source of natural anti-inflammatory agents.

Anti-inflammatory and Anti-bacterial Constituents from the Extracts of Daucus carota var. sativa Aerial Parts (당근 지상부 추출물 유래 항염 및 항균 활성 성분)

  • In this study, we investigated anti-inflammatory and anti-bacterial constituents from Daucus carota var. sativa (carrot) areal parts. For the extract and solvent fractions, the anti-inflammatory activities were examined by measuring the nitric oxide (NO) production using LPS-stimulated RAW 264.7 cells. Among them, the ethyl acetate (EtOAc) fraction decreased the NO level in a dose-dependent manner. To elucidate further anti-inflammatory mechanisms, EtOAc fraction was evaluated by estimating their effects on the production of prostaglandin $E_2$ and pro-inflammatory cytokines as well as on the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). As a result, the EtOAc fraction was determined to inhibit the production of $PGE_2$, IL-$1{\beta}$, IL-6 and reduce the iNOS, COX-2 protein expression. Upon the anti-bacterial tests using Staphylococcus epidermidis and Propionibacterium acnes, n-hexane (Hex) and EtOAc fractions showed the most potent activities. Three phytochemicals were isolated form the EtOAc fraction; diosmetin (1), diosmin (2), cynaroside (3). The chemical structures of the isolated compounds were elucidated based on the spectroscopic data including $^1H$ and $^{13}C$ NMR spectra, as well as comparison of the data to the literatures. Anti-inflammatory and anti-bacterial effects were studied for the isolates. All of the compounds (1 - 3) decreased the NO production, effectively. Also, compound 3 showed anti-bacterial activity on P. acnes. Based on these results, D. carota var. sativa extract could be potentially applicable as anti-inflammatory and anti-bacterial ingredients in cosmetic formulations.

The Study on the Physiological Activities of Beta vulgaris such as Antioxidant and Anti-inflammatory in RAW 264.7 cells (RAW 264.7 세포에서 레드비트의 항산화 및 항염증 등의 생리활성 연구)

  • This study was designed to examine the cell cytotoxic, anti-inflammatory, and antioxidant activity for raw material of Beta vulagaris tea. Antioxidative ability was evaluated by bioassays using DPPH and ABTS radical scavenging activity. Cell viability was assessed by MTT assay using RAW 264.7 cells, and investigated production levels of reactive oxide speies, and inflammatory meditors(i.e., nitric oxide, tumor necrosis factor-α, and interleukin-6) in LPS-induced RAW 264.7 cells. As a results, DPPH and ABTS raidcal scavenging activity were increased in a dose-dependent manner, and confirmed no cytotoxicity in all concentration. Also, it was significantly decreased level of ROS, NO, IL-6, and TNF-a in LPS-induced RAW 264.7 cells. Therefore these results suggest Beta vulagaris has considerable potential as a raw material of leached tea with safe anti-oxidative and anti-inflammatory effects.

Anti-oxidant, Anti-inflammatory and Anti-allergic effects of mixed herb medicines extracts by steaming process (증숙과정에 따른 생약 혼합추출물의 항산화, 항염 및 항알러지 활성)

  • 본 연구는 국내 생약중 도라지를 포함, 인삼, 당귀, 생강, 마 5가지를 선별하여 각 원물별로 항산화 활성 가장 높은 기준으로 증숙시킨 조건의 흑도라지, 흑삼, 흑당귀, 흑생강, 흑마를 확보하였다. 증숙시킨 5가지 생약은 다시 혼합 추출(BMHE; Black Mixed Herbal Extract)하여 증숙하지 않은 생약 혼합 추출물(MHE; Mixed Herbal extract)과 항산화, 항염 및 항알러지 활성을 비교하여 증숙공정이 생약의 생리활성에 미치는 긍정적인 영향을 바탕으로 피부향장 소재로 활용가능성을 확인하고자 연구하였다. 실험은 MHE와 BMHE를 각각 50% Ethyl alcohol로 추출하여 감압농축하여 생리활성을 평가하였으며, MHE와 BMHE 추출물에 대한 세포생존률을 확인한 결과 $1,000{\mu}g/ml$의 농도에서 각각 89.4%와 95.3%의 세포생존률을 보임에 따라 $1,000{\mu}g/ml$ 이하의 농도에서 HME와 BMHE 모두 세포독성이 거의 적은 것을 확인하였다. DPPH radical scavenging effects 항산화 활성에 대하여 MHE와 BMHE을 비교한 결과 농도 의존적으로 항산화 활성을 보였으며 $1,000{\mu}g/ml$의 농도에서는 각각 66.8%와 74.2%의 라디칼 소거능을 확인하였다. NO assay로부터 항염활성을 비교한 결과에서도 MHE와 BMHE 모두 농도 의존적으로 NO의 함량 감소시켰으며 $1,000{\mu}g/ml$로 처리한 농도에서는 59.5%와 64.7%의 억제율을 각각 보임에 따라 항염 효과도 우수한 것을 확인하였다. 항알레르기 효능 실험 중 하나인 5-LO 억제효능을 측정하여 ME와 BME의 항알러지 효능을 측정한 결과 상기의 결과와 마찬가지로 농도 의존적으로 5-Lipoxygenase를 억제하였으며, $1,000{\mu}g/ml$의 농도로 MHE와 BMHE를 처리하였을 시 5-Lipoxygenase 효소활성에 대하여 각각 76.2%와 83.5%의 높은 억제율을 확인 하였다. 이는 양성대조군인 EGCG를 $200{\mu}g/ml$의 농도로 처리한 9.10%에 근접한 억제율을 보임에 따라 항알러지 효과가 우수한을 확인할 수 있었다. 실험 결과를 바탕으로 BMHE가 MHE보다 항산화, 항염, 항알러지 활성이 우수한 것을 확인 할 수 있었으며, 이는 증숙기법이 생약의 생리활성을 높여주는 것은 물론 더 나아가 본 연구를 통해 확보된 5가지 흑도라지, 흑삼, 흑당귀, 흑생강, 흑마는 향후 피부향장 소재로 활용이 용이할 것으로 사료된다.

  • Hyaluronic acid(HA)는 보습성, 생체적합성, 고점도성의 특징이 있는 수용성 다당류로 분자량에 따라 보습, 탄성, 점성 등이 달라진다. 또한 피부의 침투성 또한 분자량에 가장 큰 영향을 받는다. 본 연구에서는 HA를 저분자화하여 10만Da-1천Da의 저분자 히알론론산을 제조하고 이를 아세틸화하여 항주름 및 항염활성을 측정한 결과 분자량이 낮은 분자량일 수록 우수한 항주름 및 항염활성을 나타내었으며 아셀틸화한 것이 하지 않은 것에 비하여 높은 항주름 및 항염활성을 나타 내었다.

  • The anti-inflammatory and analgesic actions of Cedrela sinensis, Oenanthe javanica and ARtemisia princeps var. orientalis were carried out. The methanolic extract of Artemisia princeps var. orientalis showed the inhibitory effect on carrageenin-induced edema as well as vascular permeability in mice. And we also found the alagesic activity in these plants.
