Rubus coreanus Miquel Bioconversion of Metabolites by Fermenting Bacteria and Their Functional Enhancement (복분자 기능성 증대를 위한 유용미생물 활용 생물전환 공정 도입 방안 연구)

  • 복분자(Rubus coreanus Miquel)는 anthocyanin, phenolic acid, tannin, flavonoid, stilbenoid와 같은 생리활성물질이 풍부하게 존재하는 것으로 보고되었으며 항염증, 항암활성, 항산화효과 등 다양한 생리활성에 대한 효능이 있어 건강식품으로써 애용되고 있을 뿐만 아니라 약용성 건강보조식품 등으로 각광받고 있다. 생물전환(Bioconversion)은 미생물 또는 효소의 생물학적 촉매 기반 천연소재의 생리활성 물질기능성, 생체이용률, 안전성을 증대시키기 위한 방안으로 많은 연구가 진행되고 있으며 아울러 식품, 의약품, 화장품 등 다양한 분야에서 활성화 되고 있다. 본 연구는 불가사리 발효액으로부터 유산균을 분리하였으며, 유전학적 특성을 확인하기 위하여 16S rDNA 염기서열을 분석하였다. 전북 고창에서 수확된 복분자를 분말상태로 유산균과 발효공정을 수행하였으며, 복분자의 최적 추출조건 선정과 발효공정 전 후의 활성을 관찰하였다. 발효공정 후 추출물의 기능성 평가를 진행하기 위하여 DPPH radical scavenging activity, total polyphenol 함량을 확인하여 항산화 효능 및 유효성분 함량을 평가하였다. 또한 대식세포인 Raw 264.7을 사용하여 MTT assay, Nitric oxide (NO) 생성 억제 효능을 확인하여 세포독성 및 항염증 활성을 평가하였다. 실험결과, 발효기간이 다른 불가사리 비료발효액으로부터 16 종의 다양한 균주를 확보하였으며, 생물전환 공정에 유용 균주를 선정하기 위하여 복분자 분말의 발효공정을 실시한 결과 3 종의 유산균 처리군에서 무처리군 대비 DPPH radical 소거능 및 polyphenol 함량이 증가됨을 확인하였다. 그 중 가장 우수한 활성을 나타내는 균주를 16S rDNA 염기서열 분석한 결과 Lactobacillus coryniformis A6-4로 확인되었으며, 발효공정 후 항산화 활성은 무처리군 대비 약 115%, polyphenol의 함량은 무처리군 대비 약 121%로 증가됨을 확인되었다. 또한 발효공정 후 독성활성이 감소되는 경향을 확인되었으며, 항염증 활성이 유의적으로 증가됨을 확인하였다.

The Study on the Physiological Activities of Beta vulgaris such as Antioxidant and Anti-inflammatory in RAW 264.7 cells (RAW 264.7 세포에서 레드비트의 항산화 및 항염증 등의 생리활성 연구)

  • This study was designed to examine the cell cytotoxic, anti-inflammatory, and antioxidant activity for raw material of Beta vulagaris tea. Antioxidative ability was evaluated by bioassays using DPPH and ABTS radical scavenging activity. Cell viability was assessed by MTT assay using RAW 264.7 cells, and investigated production levels of reactive oxide speies, and inflammatory meditors(i.e., nitric oxide, tumor necrosis factor-α, and interleukin-6) in LPS-induced RAW 264.7 cells. As a results, DPPH and ABTS raidcal scavenging activity were increased in a dose-dependent manner, and confirmed no cytotoxicity in all concentration. Also, it was significantly decreased level of ROS, NO, IL-6, and TNF-a in LPS-induced RAW 264.7 cells. Therefore these results suggest Beta vulagaris has considerable potential as a raw material of leached tea with safe anti-oxidative and anti-inflammatory effects.

  • Chrysanthemum coronarium L. contains various antioxidants such as beta-carotene, vitamins A and C, and polyphenols, and is known to have anti-inflammatory effects. Under the assumption that the PDRN contained in the extract can mediate the anti-inflammatory response, the mouse macrophages, RAW264.7 cells, were stimulated with LPS to induce the conversion to inflammatory cells, and then the addition of PDRN extracted from the extract was effective in inhibiting inflammation. It was analyzed whether there was The gene expression of IL-1β and TNF-α was used as an anti-inflammatory index, and the relative expression levels of each gene were confirmed by RT-PCR. As a result, RT-PCR confirmed the effect of PDRN-induced inhibition of inflammation in both IL-1β and TNF-α genes. Therefore, based on this study, it is considered to be a precedent data that can be developed as a treatment for inflammatory diseases, and it will be helpful in research and development of a treatment that can improve the anti-inflammatory mechanism.

Anti-inflammatory and antioxidant effects of Barringtonia augusta Kurz extract (Barringtonia augusta Kurz 추출물의 항염증 및 항산화 효능 평가)

  • Barringtonia augusta Kurz is a species of the genus Barringtonia. Although several studies have analyzed the biological activity of B. racemosa Roxb and B. acutangula, the anti-inflammatory and antioxidant effects of B. augusta extract (BKE) remain unclear. Therefore, in this study, we investigated the anti-inflammatory and antioxidant effects of BKE using lipopolysaccharide (LPS) and RAW 264.7. BKE suppressed LPS-induced nitric oxide (NO) and inducible NO synthase expression without affecting RAW 264.7 cell viability. Additionally, BKE showed 2,2-Diphenyl-1-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging capacities and inhibited LPS-induced reactive oxygen species production in RAW 264.7 cells. BKE also suppressed LPS-induced phosphorylation of IκB kinase and nuclear factor kappa-B (NF-κB) and p65 translocation from the cytosol to the nucleus in RAW 264.7 cells. These results suggest that BKE is a possible novel material that exerts beneficial antioxidant and anti-inflammatory effects through the inhibition of NF-κB signaling pathways.

Study on Cosmeceutical Activities and Anti-inflammatory Activities of Magnolia biondii Extracts (신이화(辛夷花, Magnoliae Flos)추출물의 화장품약리활성 및 항염증효과에 관한 연구)

  • Existing pharmaceutical studies show that Magnolia biondii is effective in treating rhinitis and in reducing cholesterol, given its endogenous, volatile ingredients. The study herein seeks to assess the cosmeceutical activities and anti-inflammatory activities of Magnolia biondii extracts for possible application as cosmetic ingredients. The cosmeceutical and anti-inflammatory activities were investigated using hydroxyl radical scavenging, superoxide dismutase (SOD)-like activity, xanthine oxidase (XO) inhibition, cell viability, nitric oxide (NO) inhibition, and inducible nitric oxide synthase (iNOS) expression by Western blotting. Magnolia biondii extracts were identified to have antioxidant activities in hydroxyl free radical scavenging, SOD-like activity, and XO inhibition. In testing the anti-inflammatory activities of the extracts, NO production was inhibited in a dose-dependent manner. Additionally, in a dose-dependent manner, the Magnolia biondii extracts were able to suppress iNOS expression in LPS-stimulated RAW 264.7 macrophage cells. From these results, Magnolia biondii showed adequate potential for application in cosmetic production and related industries as well as a functional material.

Antioxidant and Anti-inflammatory Effects of Dioscorea japonica and Chenopodium album (참마와 명아주의 항산화 및 항염증 효과)

  • In this study, free radical scavenging activity and enzyme-linked immunosorbent assay (ELISA) experiments were carried out using Dioscorea japonica (D. japonica) and Chenopodium album (C. album) extracts to evaluate their anti-oxidative and anti-inflammatory effects. In the free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity, $EC_{50}$ of D. japonica and C. album were measured as 2.386 and 0.524 mg/mL, respectively. The free radical scavenging activity of the mixed sample of D. japonica and C. album was the highest when the D. japonica and C. album ratio was 2 : 1. The IL-6 and $TNF-{\alpha}$ ELISA assay showed that IL-6 in mouse spleen cells treated 1 mg/mL of samples, D. japonica and C. album decreased the production of IL-6 concentration by 27.17%, 72.30%. In the case of $TNF-{\alpha}$, D. japonica and C. album decreased 61.97% and 77.85% of $TNF-{\alpha}$ production, respectively. Through these results, we confirmed that D. japonica and C. album have antioxidant and anti - inflammatory effects and could be applied to natural medicine cosmetic having anti - inflammatory effects.

Anti-inflammatory Activity of Extracts of Hovenia dulcis on Lipopolysaccharides-stimulated RAW264.7 Cells (LPS로 유도된 RAW264.7 대식세포에 대한 헛개나무(Hovenia dulcis) 추출물의 항염증 효과)

  • In this study, the anti-inflammatory activities of the extracts of different parts of Hovenia dulcis such as leaves, stems, and roots were investigated. Among them, the roots extract (RE) showed the most potent suppressive effect against pro-inflammatory mediators in LPS-stimulated mouse macrophage cells. RE induced dose-dependent reduction of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) and concomitantly reduced the production of NO and $PGE_2$. Additionally, pre-treatment with RE significantly suppressed the production of inflammatory cytokines, such as tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), interleukin $(IL)-1{\beta}$, and IL-6, as well as mRNA levels. Moreover, phosphorylation of mitogen-activated protein kinases (MAPKs) and nuclear translocation of nuclear factor-kappa B (NF-kB) were also strongly attenuated by RE in RAW264.7 cell. Furthermore, RE induced HO-1 expression through nuclear translocation of nuclear factor E2-related factor 2 (Nrf2) and increase HO-1 activity in RAW264.7 macrophages. Therefore, these results indicate that RE strongly inhibits LPS-induced inflammatory responses by blocking NF-kB activation, inhibiting MAPKs phosphorylation, and enhancing HO-1 expression in macrophages, suggesting that RE of H. dulicis and a major component, 27-O-protocatechuoylbetulinic acid could be applied as a valuable natural anti-inflammatory material.

Inhibitory Effect of Ethanol Extract of Monascus-fermented Red Yeast Rice on Proinflammatory iNOS and COX-2 Protein Expression in LPS-stimulated RAW 264.7 Macrophage Cells (Monascus sp. BHN-MK로 발효생산한 홍국 에탄올 추출물의 Raw 264.7 대식세포에 있어 친-염증성 iNOS와 COX-2 단백질 발현 억제 효과)

  • Red yeast rice has been extensively used as a food and traditional medicine for thousands of years in Korea. Monascus produces many secondary metabolites during its growth, including pigments, monacolins, and γ-aminobutyric acid. Some metabolites, specifically monacolin K, γ-aminobutyric acid, and dimerumic acid, have been reported to lower cholesterol and blood pressure because of certain antioxidant effects. This study investigated the total phenolic content of ethanol extract from red yeast rice fermented with Monascus sp. BHN-MK and its anti-inflammatory effect on LPS-stimulated RAW 264.7 macrophage cells. To assess its anti-inflammatory effect, the inhibitory activity of the ethanol extract on LPS-induced NO production and expression levels of iNOS and COX-2 proteins in macrophage cells were measured. Its total polyphenol content was higher than that of ordinary non-fermented rice. Its NO production inhibition activity was comparable to that of the negative control group treated with LPS at a concentration of 400 ㎍/ml. Western blot revealed a significant decrease in the inhibition of iNOS and COX-2 protein expression at concentrations of 400 and 800 ㎍/ml, respectively. Red yeast rice ethanol extracts exerted the strongest anti-inflammatory effects. The results indicate that red yeast rice could be used as a functional cosmetic and anti-inflammatory material.

Photoprotective Effect of Grape Pruning Stem Extracts on the UVB Induced HR-1 Mice Skin (포도전정가지 추출물이 UVB에 유도된 HR-1 mice의 피부손상에 대한 광보호 효과)

  • This study intends to analyze the contents of rutin, procyanidin B3, quercetin, kaempferol, known to have antioxidant, anti-inflammatory and anti-carcinogenic effects, among the polyphenol type contained in the grape pruning stem extracts (GPSE), utilizing grape stems being discarded after harvest, measure the effects on the skin moisture, inhibition of skin cell proliferation, anti-inflammatory on the damaged skin of a HR-1 mice induced with UVB, and verify the applicability as a material for functional food and functional cosmetics. The results of verifying the photoprotection effects through the skin proliferation control through of GPSE showed similar result to suncream was achieved at the GPSE concentration of 2,000 mg/kg on the epidermis (p<0.05). The results showed anti-inflammatory effects on all groups applied with GPSE as compared to the control group irradiated with UVB, but at the GPSE concentration of 1,000 mg/kg, a lower COX-2 protein expression at 8%, lower than the 22% of suncream, was observed to achieve an excellent anti-inflammatory effect (p<0.05). The results of this study confirmed the existence of active polyphenol type, such as rutin, kaempferol, querocetin and procyanidin B3, within the GPSE, and GPSE has improvement effects on moisturizing effects, skin proliferation control effect, inflammatory control effect and improvement effects on the skin barrier function through UV ray damage. GPSE is a functional ingredient with a potential for skin protection effects, and has high utilization as an ingredient for functional food and functional cosmetics.

Antibacterial and Anti-inflammatory Effects of Essential Oil from the Magnolia kobus Flower (목련 꽃 에센셜 오일의 항균 및 항염증 활성)

  • Magnolia kobus is known to exert various biological effects, such as antioxidant and hypnotic activity. In this study, we investigated the antimicrobial and anti-inflammatory activity of M. kobus essential oil extracted using steam distillation. Its antimicrobial activity was tested against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa by the paper disk diffusion and minimum inhibitory concentration (MIC) methods. Its anti-inflammatory activity was evaluated by measuring its inhibition ratio on the production of nitric oxide (NO) and PGE2 in lipopolysaccharide (LPS)-induced RAW264.7 cells. Its composition was analyzed by gas chromatography-mass spectrometry (GC-MS). The results showed that M. kobus essential oil exhibited excellent antibacterial activity against S. aureus, with a clear zone of 18 mm and an MIC value of 0.25 mg/ml. Its clear zones against P. aeruginosa and E. coli were 14 mm and 17 mm, respectively, while its MIC values were 1 mg/ml and 0.5 mg/ml, respectively. The essential oil exhibited no cytotoxicity to the RAW264.7 cells at a concentration of 500 ㎍/ml while showing NO (37.7%) and PGE2 inhibition (24.0%). Its three main fragrance ingredients identified were 3-carene (77.07%), β-elemene (6.92%), and caryphyllene (2.86%). The results suggest that M. kobus essential oil has potential as a cosmetic functional material with antimicrobial and anti-inflammatory effects.
