Inhibitory Effects of Acanthopanax chiisanensis Ethanolic Extracts on FcεRI α Chain Expression (지리 오갈피의 FcεRI α chain 발현 저해 효과)

  • Basophils and mast cells play an important role in $Fc{\varepsilon}RI-mediated$ allergic reaction as effector cells. We studied the effects of Acanthopanax chiisanensis on $Fc{\varepsilon}RI\;{\alpha}$ chain expression in human basophilic KU812F cells. Ethanol extracts from root and stem of A. chiisanensis were tested for inhibitory effects of $Fc{\varepsilon}RI\;{\alpha}$ chain expression. The cell surface $Fc{\varepsilon}RI\;{\alpha}$ chain expression was examined by flow cytometric analysis. All of the extracts of A. chiisanensis reduced the cell surface $Fc{\varepsilon}RI\;{\alpha}$ chain expression. Furthermore, A. chiisanensis extracts caused a decrease in the level of $Fc{\varepsilon}RI\;{\alpha}$ chain mRNA level and $Fc{\varepsilon}RI-mediated$ histamine release. These results suggest that root and stem extracts of A. chiisanensis play an important role in anti-allergic activity via down-regulation of $Fc{\varepsilon}RI\;{\alpha}$ chain expression and decrease in release of inflammatory mediator such as histamine.

Effect of BNs-3 and BNs-7, Extracts of Citrous Orange Peel, on the Obesity Induced by ad libitum Feeding a Cholesterol-Containing Diet in Rats (콜레스테롤 함유 식이 랫드에서 감귤껍질추출물 BNs-3 및 BNs-7의 항비만 효과)

  • The protective effects of BNs-3 and BNs-7, extracts of citrous orange peel, on the obesity induced by ad libitum feeding a cholesterol-containing diet to rats were investigated. The animals were fed on the diet including cholesterol (0.5%) with or without the citrous orange peel extracts BNs-3 (5%) and/or BNs-7 (0.1%) for 8 weeks. The ad libitum feeding a diet containing cholesterol to rats from 6 weeks of age increased the body weight gain compared with that of rats fed on a normal diet. Such an increase in body weights was markedly attenuated by the addition of BNs-3 or BNs-7 to the diet. Especially, a combinational feeding on BNs-3 and BNs-7 significantly reduced the body weight gain below that of normal diet-fed animals. Interestingly, the weights of abdominal adipose tis-sues surrounding epididymides were greatly reduced by the citrous orange peel extracts, in parallel with the decrease in body weights. In addition, blood concentrations of lipids including cholesterol were also lowered by the combinational treatment with BNs-3 and BNs-7. Taken together, it is suggested that the obesity and overweight produced by unrestricted overfeeding on diet with cholesterol may be partially due to the accumulation of abdominal adipose tissues, around the epididymides in rats, and that citrous orange peel extracts might exert antiobese activities by reducing the adipose tis-sues as well as blood lipid concentrations.

Glasswort Powder Diet Activates Lipid Metabolism in Rat (함초분 첨가 고지방식이 흰쥐의 지방대사 연구)

  • The purpose of this study was to investigate the effects of glasswort powder intake on lipid metabolism. Rats were divided into three groups: a group fed a normal diet (CON), one fed a high fat diet (HFC), and one fed a high fat diet with glasswort powder (HFG). They were fed their respective diet for four weeks. Body weight was significantly lower (9%) in the HFG group than in the HFC group at the fourth week. According to the feces analysis, the HFG group showed the highest fat level (120% vs. CON; 138% vs. HFC) and fecal calories (110%). The concentration level of TG and LDL-C was 71.8% lower in the HFG group compared to the HFC group, while the concentration level of HDL-C was 152% higher in the HFG group. Expression of FABP in the liver was 197% greater in the HFG compared to the HFC group, with the expression of CPT-1 showing a similar tendency. These results suggest that glasswort powder intake suppresses weight gain and improves fat metabolism at the level of liver cells. From these results, we suggest that glasswort powder is effective against obesity by inhibiting the absorption of fat in the digestive tract.

Artemisia scoparia Inhibits Adipogenesis in 3T3-L1 Pre-adipocytes by Downregulating the MAPK Pathway (비쑥 추출물이 3T3-L1 지방세포 분화 및 MAPK 신호 전달 경로에 미치는 영향)

  • Obesity is epidemic worldwide and has reportedly been linked to the progression of several metabolic and cardiovascular diseases. The natural products are decreasing the side effects of medicines used for obesity and also have health benefits dut to their numerous bioactive compounds. In this context, Artemisia scoparia is a widespread plant that has been suggested as possessing various types of bioactivity. In this study, the crude extract from A. scoparia (ASE) was tested for its ability to suppress adipogenesis in mouse 3T3-L1 pre-adipocytes. The molecular pathway by which ASE affects differentiation of 3T3-L1 cells was also investigated. The introduction of ASE to differentiating 3T3-L1 pre-adipocytes resulted in suppressed adipogenesis, as confirmed by decreased intracellular lipid accumulation. The differentiating cells treated with 10 and $100{\mu}g/ml$ of ASE showed 21.9 and 29.0% less lipid accumulation, respectively, than untreated adipocytes. In addition, the results indicated that ASE treatment lowered the expression of the adipogenesis-related factors $PPAR{\gamma}$, $C/EBP{\alpha}$, and SREBP-1c. Furthermore, treating with ASE notably decreased levels of phosphorylated p38, ERK, and JNK in 3T3-L1 adipocytes. These results indicate that ASE exhibits significant anti-adipogenesis activity by downregulating the MAPK and $PPAR{\gamma}$ pathways during the differentiation of 3T3-L1 pre-adipocytes. Therefore, A. scoparia may be a potential source of natural products against obesity.

Anti-Obesity Effect of Ethyl Acetate Fraction from 50% Ethanol Extract of Fermented Curcuma longa L. in 3T3-L1 Cells (발효울금 주정추출물부터 분리된 에틸아세테이트 분획물에 대한 3T3-L1 세포에서의 지방 형성 억제 효과)

  • In the present study, we investigated the effect of ethyl acetate fraction from 50% ethanol extract of fermented Curcuma longa L. (FCEE) on lipid metabolism in 3T3-L1 cells. The safety range of FCEE was up to $300{\mu}g/mL$. Effects of FCEE on lipid accumulation and intracellular triglyceride (TG) content in 3T3-L1 cells were examined by Oil Red O staining and AdipoRed assay. Compared to adipocytes, lipid accumulation and intracellular TG content were significantly reduced by 10.2% and 13.7%, respectively, upon FCEE treatment at a concentration of $200{\mu}g/mL$. Glucose uptake by 3T3-L1 cells was significantly reduced by 36.6% compared to adipocytes at a concentration of $200{\mu}g/mL$. On day 8, free glycerol release into the culture medium was significantly reduced compared to adipocytes at concentrations of 50, 100, and $200{\mu}g/mL$ of FCEE. FCEE significantly stimulated RNA expression of AMP-activated protein kinase (AMPK) and suppressed mRNA expressions of sterol regulatory element-binding protein-1c (SREBP-1c), CCAAT/enhancer binding proteins ${\alpha}$ ($C/EBP{\alpha}$), and peroxisome proliferator- activated receptor ${\gamma}$ ($PPAR{\gamma}$) in 3T3-L1 cells. These results suggest that FCEE inhibits adipogenesis through activation of AMPK mRNA expressions and inhibition of SREBP-1c, $C/EBP{\alpha}$, and $PPAR{\gamma}$ mRNA expressions.

The Study on Anti-obesity Effects of Gamiygin-tang Extract and Ferment (가미이진탕(加味二陳湯) 전탕액과 발효액이 항비만(抗肥滿)효과에 미치는 연구)

  • Objective This study was designed to investigate the effects of Gamiygin-tang (GY) extract (GYE) and fermented solution (GYF) on body weight, serum lipid level and adipocyte differentiation in high fat diet-fed obese mice. Materials and Methods High fat diet-fed obese mice and 3T3-L1 adipocytes mice were treated with GYE and GYF and obesity related markers were assessed. A cytotoxicity assay was carried out by MTS assay. Inhibitory effects of GYE and GYF on adipocyte differentiation were carried out by Oil Red O staining. The effects of GYE and GYF on the expression of adipocyte differentiation regulatory factors, peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) and CCAAT/enhancer binding protein alpha (CEBP-${\alpha}$) were measured by real-time reverse transcriptase-polymerase chain reaction. The effects of GYE and GYF on the expression of adipocyte differentiation regulatory factors were also determined in relation to protein production/protein levels by western blotting. The anti obesity effects of GYE and GYF were measured in high fat-diet induced obese mice. Various factors were measured from the serum of the high fat-diet mice. Results Though GYE did not show toxicity at the concentration of 1mg/ml, GYF showed toxicity at the concentration of 1mg/ml. The GYE at 0.1 and 1mg/ml inhibited the differentiation of 3T3-L1 cells, and the GYF also inhibited the differentiation of 3T3-L1 cells. The effect of GYE on adipocyte differentiation factors including PPAR-${\gamma}$ and CEBP-${\alpha}$ was investigated and compared to the corresponding concentration levels of GYF. GYE and GYF both suppressed the RNA and protein levels of adipocyte differentiation factors. In the animal test both GYE and GYF reduced weight gain. GYE and GYF reduced blood cholesterol, TG and LDL levels. GYF better reduced blood cholesterol levels. Conclusions These results demonstrate that GYE and GYF exerts anti-obesity effect in 3T3-L1 cells and obese mice induced by high-fat diet.

Anti-obesity Effects of Galgeun-tang in High Fat Diet Induced Obese Mice Model (갈근탕이 고지방 식이 유발 백서 모델에 미치는 항비만 효과)

  • Objectives To investigate anti-obesity effects of Galgeun-tang, an herbal formula, in high fat diet induced obese mice model. Methods 24 Male C57Bl/6J mice were randomly assigned to normal group fed with normal research diet (NOR, n=6), high fat diet control group treated with water (HFD, n=6), high fat diet group treated with Orlistat (ORL, n=6, Orlistat 10 mg/kg), and high fat diet group treated with Galgeun-tang (GGT, n=6, Galgeun-tang 700 mg/kg). 12 weeks later, body weight, fat weight, liver weight, blood glucose, total cholesterol, triglyceride, HDL, ALT, AST, obesity related neuropeptides and adipokines, ratio of gut microbiota, and histopathology of liver were evaluated. Results In the GGT group, 1. body weight gain, liver weight gain, and total fat weight gain were significantly less than those in the HFD group. 2. blood glucose level was significantly lower and insulin level was significantly higher than in the HFD group. 3. total cholesterol level and triglyceride (TG) level were significantly lower and high density lipoprotein (HDL) level was significantly higher than in the HFD group. 4. appetite-promoting ARC neuropeptides such as Agrp and Npy were significantly less and appetite-inhibiting ARC neuropeptide, Cart was significantly more than in the HFD group in qRT-PCR analysis. 5. adiponectin level and visfatin level were significantly higher, and resistin level and leptin level was significantly lower than in the HFD group. 6. the relative level of Bacteroidetes was significantly higher, and the relative level of Firmicutes was significantly lower than in the HFD group. 7. the increase of adipose tissue was significantly more inhibited than in the HFD group. Conclusions The present study showed that Glageun-tang exerts anti-obesity effects in that it. 1. inhibited the increase in body weight, liver weight, and total fat weight. 2. decreased the level of TG, and increased the level of HDL. 3. influenced neuropeptides and adipokines that are important in regulating food intake and changes of body weight. 4. modified the beneficial quantitative changes in gut microbiota suppressing the tendency toward obesity.

Effects of Genistein on Cell Proliferation and Adipogenesis in Mouse 3T3-L1 Preadipocytes (이소플라본 genistein이 전지방세포 성장 및 지방세포형성과정에 미치는 영향)

  • The effects of genistein on cell proliferation and adipogenesis were examined in mouse 3T3-L1 preadipocyte cells. Genistein decreased viability of 3T3-L1 pre-adipocytes in a dose-dependent manner. Oil Red O staining of these cells also indicated that adipogenesis was inhibited by 50 ${\mu}M$ genistein treatment. We investigated the molecular mechanisms involved in the decrease in cell viability in genistein-treated 3T3-L1 cells by conducting an oligo DNA microarray analysis. We selected the sirtuin-1 gene, one of the upregulated genes, for further experimentation because sirtuin-1 belongs to the sirtuin family, which is associated with anti-obesity and anti-inflammation activities. We found that four phytochemicals (resveratrol, capsaicin, daidzein, and genistein) could increase sirtuin-1 expression. Genistein was the strongest inducer of sirtuin-1 among the tested phytochemicals. The inhibition of adipogenesis by genistein was recovered by surtuin-1 siRNA transfection. Overall, these results may further our understanding of the molecular mechanisms underlying the inhibition of proliferation and adipogenesis by genistein in mouse 3T3-L1 cells.

Anti-obesity and Anti-hyperlipidemic Effects of Taeyeumjowee-tang and its modified prescription (태음조위탕(太陰調胃湯)과 그 가감방(加減方)의 항비만 및 항고지혈증 효과에 관한 연구)

  • Objective : The purpose of this study was to investigate the anti-obesity and anti-hyperlipidemic Effects of Taeyeumjowee-tang and its modified prescription on the animal model of obesity and hyperlipidemia induced high-fat diet. Method : 1) The extracts of Taeyeumjowee-tang (TJT) and its modified prescription, Taeyeumjoweetang gagam-bang (TJGB) were evaluated for its inhibitory effects on obesity. 2) The body weight and feed weight were determined in the pre-treated and post-treated mice and the lipid profiles in the serum were analyzed in order to evaluate the anti-hyperlipidemia action of the extracts. 3) The effect of each extract was investigated for the influences on monoamine oxidase activity and HMG-CoA reductase activity. Results 1. TJT and TJGB extracts dose-dependently reduced the body weight and feed intake in normal mice. The effect of TJGB extract was better than that of TJT extract. 2. TJGB extract diminished the body weight increase and reduced the feed intake in the pre-treatment or post-treatment of the extract 3. TJGB extract decreased the amount of total cholesterol slightly and triglyceride potently after the pre-treatment or post-treatment, but HDL cholesterol exhibited no remarkable change compared with control. 4. TJGB extract weakly potentiated the monoamine oxidase activity, but its effect was better than that of TJT extract. 5. TJGB extract weakly inhibited the HMG-CoA reductase activity, but its effect was better than that of TJT extract. Conclusion : Taeyeumjowee-tang and its modified prescription can clinically be useful as anti-obesity drug and also for the improvement of hyperlipidemia.

Study on Anti-obesity Effect of Chegameuiin-tang (체감의이인탕(體減薏苡仁湯)의 항비만 효과 연구)

  • The aim of this study is to investigate Chegameuiin-tang water extracts (CETE) have potent anti-obesity activities in a high fat diet-induced obesity mouse model. In this study, we designed three group (normal diet group, high fat diet group, high fat diet plus CETE group for 13-week oral administration). Increases in body weight and fat storage were inhibited by 13-week oral administration of CETE at a 500 mg/kg concentration in this animal model, while the amount of food intake was not affected. Results from blood lipid analysis showed that the levels of triglyceride, total cholesterol and LDL-cholesterol were significantly lowered by CETE administration, also HDL-cholesterol was increased more than high fat diet-induced obese mouse. To understand the underlying mechanism at the molecular level, the effects of CETE were examined on the expression of the genes involved in lipogenesis and lipolysis by real-time PCR. In epididymal fat of CETE-treated mice, the mRNA level of lipogenic genes such as sterol regulatory element binding protein 1 and fatty acid synthase were decreased, which was well correlated with the reduction of the epididymal fat weight. Also, CETE administration inhibited decreases of the hormone-sensitve lipase and lipoprotein lipase mRNA expressions, which are genes related with lipolysis. These results suggest that Chegameuiin-tang may have great potential as a novel anti-obesity agent.
