Oxidative Stability of Grape Seed Oil by Addition of Grape Seed Extract (포도씨추출물을 첨가한 포도씨유의 산화 안정성)

  • Grape seed extract (GSE) was added to grape seed oil to improve the oxidative stability of the grape seed oil during storage. To measure the oxidative stability of grape seed oil, peroxide value, acid value, and conjugated diene value were measured, and changes in browning, vitamin E, fatty acid composition, and polyphenol content of oil were examined. In the case of grape seed oil with GSE, peroxide value, acid value, and conjugated diene value were lower than those of grape seed oil. The magnitude of increase in absorbance of grape seed oil with GSE was less than that of additive-free grape seed oil, whereas the magnitude of decrease in vitamin E isomers in grape seed oil with GSE was less than that of grape seed oil with no additive. Changes in fatty acid composition were also similar. However, polyphenol contents showed the greatest reduction in grape seed oil containing GSE. GSE contributes to the oxidation stability of grape seed oil, but the antioxidant capacity of GSE was lower than that of butylated hydroxytoluene.

Effects of Growth Conditions on Properties of ZnO Nanostructures Grown by Hydrothermal Method (수열합성법으로 성장된 ZnO 나노구조의 성장조건에 따른 특성)

  • ZnO nanostructures were grown on an Au seed layer by a hydrothermal method. The Au seed layer was deposited by ion sputter on a Si (100) substrate, and then the ZnO nanostructures were grown with different precursor concentrations ranging from 0.01 M to 0.3M at $150^{\circ}C$ and different growth temperatures ranging from $100^{\circ}C$ to $250^{\circ}C$ with 0.3 M of precursor concentration. FE-SEM (field-emission scanning electron microscopy), XRD (X-ray diffraction), and PL (photoluminescence) were carried out to investigate the structural and optical properties of the ZnO nanostructures. The different morphologies are shown with different growth conditions by FE-SEM images. The density of the ZnO nanostructures changed significantly as the growth conditions changed. The density increased as the precursor concentration increased. The ZnO nanostructures are barely grown at $100^{\circ}C$ and the ZnO nanostructure grown at $150^{\circ}C$ has the highest density. The XRD pattern shows the ZnO (100), ZnO (002), ZnO (101) peaks, which indicated the ZnO structure has a wurtzite structure. The higher intensity and lower FWHM (full width at half maximum) of the ZnO peaks were observed at a growth temperature of $150^{\circ}C$, which indicated higher crystal quality. A near band edge emission (NBE) and a deep level emission (DLE) were observed at the PL spectra and the intensity of the DLE increased as the density of the ZnO nanostructures increased.

  • Tissue engineering techniques require the use of a porous biodegradable/bioresorbable scaffold, which server as a three-dimensional template for initial cell attachment and subsequent tissue formation in both in vitro and in vivo. Small intestinal submucosa (SIS) has been investigated as a source of collagenous tissue with the potential to be used as biomaterials because of its inherent strength and biocompatibility. SIS-loaded poly(L-lactide-co-glicolide)(PLGA) scaffolds were prepared by solvent casting/particle leaching. Characterizations of SIS/PLGA scaffold were carried out by SEM, mercury porosimeter, and so on. Muscle-derived stem cells can be differentiated in culture into osteoblasts, chondrocytes, and even myoblasts by the controlling the culture environment. Cellular viability and proliferation were assayed by 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium-bromide(MTT) test. Osteogenic differential cells were analyzed by alkaline phosphatase(ALP) activity. SIS/PLGA scaffolds were implanted into the back of athymic nude mouse to observe the effect of SIS on the osteoinduction compared with controlled PLGA scaffolds. Thin sections were cut from paraffin embedded tissues and histological sections were conducted hematoxylin and eosin (H&E), Trichrome, and von Kossa. We observed that bone formatioin of SIS/PLGA hybrid scaffold as natural/synthetic scaffold was better thean that of only PLGA scaffold. It canb be explained that SIS contains various kinds of bioactive molecules for osteoinduction.

Culture Method of Spore for Entomopathogenic Fungus Using Natural Zeolite Ceramic Ball (천연제오라이트 세라믹볼을 이용한 곤충병원성 곰팡이 포자 생산 방법)

  • BACKGROUND: Entomopathogenic fungi have been studied to develop for biological control agents as an alternative to chemical control agents in insect pest management. This investigated to determine the optimal culture conditions in ceramic balls for maximal sporulation of entomopathogenic fungi Beauveria bassiana M130 by use rice bran extract.METHODS AND RESULTS: METHODS AND RESULTS: A culture of entomopathogenic fungi for 12day on rice bran extract(1:8, w/v) incubated in ceramic matrix at 28℃. Natural zeolite ceramic ball was high production of 4.2×108 conidial/mL. The culture condition optimized initial pH, temperature, rice bran extract concentration, adhesives substance and concentration of NaCl, respectively. The high production of spore optimal conditions were temperature 28℃, initial pH 3, rice bran extract 3 mL, starch 33 g, 5 % NaCl and sopre suspension 7 mL, respectively.CONCLUSION: This study was carried out for the mass production of entomopathogenic fungi conidia recover rate 65% in matrix of natural zeolite ceramic ball, and to develop ingredient-used formulation of Beauveria bassiana M130 conidia for biological control agents.

Effects of Isoamyl Acetate Production in Makgeolli According to Fermentation Conditions (막걸리 발효조건에 따른 isoamyl acetate 생성 비교)

  • Because of a continual reduction in its domestic market share, the quality of the Makgeolli, a Korean traditional liquor, needs to be upgraded. Among the several options for quality improvement, sufficient organoleptic expression of flavor is very important. We analyzed production changes of isoamyl acetate, which has a banana smell, based on fermentation temperature and sugar content through the cultivation of S. cerevisiae 98-5 KCCM 11396P using generally polished rice. The banana flavor of that fermentation mash was organoleptically high at 20℃, but a larger amount of isoamyl acetate was obtained with a higher sugar content at 10℃, based on analysis by GC-MS. Consequently, sufficient production of banana flavor from isoamyl acetate was based on the concentration of isoamyl alcohol as a substrate compound of isoamyl acetate, and the production depended highly on the maintenance of heat stability, since it is unstable in temperature and the minimized inhibition of alcohol acetyl transferase by unsaturated fatty acids. We also found that production of the flavor component required the addition of sugar and a slightly higher temperature of 20~25℃ at the beginning stage of fermentation, with additional mash fermentation and a gradual decrease in temperature to 10~15℃.

Simultaneous Determination of Quinolones in Flatfish and Egg Using liquid Chromatography with Fluorescence Detection (액체크로마토그래피를 이용한 광어 및 계란 중 퀴놀론계의 동시분석법 개발)

  • An analytical method for the simultaneous determination of nine quinolones (QNs) namely, marbofloxacin, norfloxacin(IS), ciprofloxacin, danofloxacin, enrofloxacin, sarafloxacin, difloxacin, oxolinic acid, and flumequine in flatfish and egg was developed and validated using liquid chromatography with fluorescence detection (LC-FD). The samples were extracted using a traditional liquid-liquid extraction process; deproteinization was accomplished by the addition of trichloroacetic acid and acetonitrile (ACN), and defatting was performed with hexane. Chromatographic separation was achieved on a reverse phase C8 column with gradient elution using a mobile phase of 200 mM ammonium acetate buffer (pH 4.5) and ACN. The proposed method was validated according to the CODEX guideline. Mean recoveries of QNs from flatfish and egg were 89.6-106.5% with relative standard deviations (RSDs) below 15% at three different concentrations of 50, 100 and $500{\mu}g/kg$. Linearity was obtained with a correlation coefficient ($r^2$) of 0.9989-1.0000. The LOD for the investigated QNs was $1-16{\mu}g/kg$ depending on flatfish and egg. The present method can be applied simultaneously to determine QNs in muscle of flatfish and egg.

  • The studies reported herein were conducted to inquire the effect of fused siliceous materials which mixed with sand and quartz added calcium fertilzer on rice grown on low available silica content soil. The applied nitrogen level were examined for the two plots, nitrogen 10kg and 20kg per 10 a., and the siliceous materials(Wollastonite, Fused sand and Fused quartz) were applied as levels to 100kg, 300kg and 500kg per 10a. in the pots. The results are summerized as follows; 1. The available silica content solved in N/2 HCl solution of fused sand was more than of fused quartz but the silica solubility in 2% citric acid of fused quartz were higher than other. 2. The absorbed silica content by plant grown on the fused quartz plot was the highest among other siliceous materials and of fused sand and natural wollastonite were at almost equal. The ratio of absorbed silica of natural wollastonite was highest among them and in fused materials, the quartz was higher than sand. 3. The productivity of rice was increased by percentage of filled grain, weights of 1000 grains, and number of spikelets on account of the large quantity of the silica absorbed by plant Especially fused quartz showed the highest productive effect among siliceous materials and natural wollastonite and fused sand were similar effect. 4. From the above results. the fused quartz was selected as expelled siliceous fertilizer on rice plant among them and the fused sand was equal effect to natural wollastonite.

Physiological and Biochemical Responses to Ozone Toxicity in Five Species of genus Quercus Seedlings (참나무속 5종의 오존 독성에 대한 생리생화학적 반응)

  • Physiological and biochemical changes of five species of genus Quercus exposed to ozone fumigation were investigated to assess their tolerance against ozone toxicity. At the end of 150 ppb ozone fumigation, chlorophyll contents, photosynthetic characteristics, malondialdehyde(MDA) and antioxidative enzyme activities were measured in the leaves of five Quercus species(Quercus acutissima, Q. aliena, Q. palustris, Q. serrata, and Q. variabilis). Chlorophyll and carotenoid contents, net photosynthesis and carboxylation efficiency decreased after ozone treatment, indicating that $O_3$-exposed plants underwent physiological inhibition. The reduction rate of total chlorophyll contents and carboxylation efficiency were respectively 15% and 34% for Q. aliena and 38% and 62% for Q. variabilis. The amount of MDA increased with the highest increase rate of 140% in Q. acutissima which also showed the highest increase rate(60%) of superoxide dismutase(SOD). Ascorbate peroxidase(APX) activity increased in Q. variabilis, Q. serrata and Q. acutissima by ozone treatment. Based on our results, ozone tolerance of the five Quercus species was ranked as Q. aliena>Q. palustris>Q. serrata>Q. variabilis>Q. acutissima. We concluded that chlorophyll contents, photosynthesis, MDA content and antioxidative enzymes were the important physiological markers for tolerance against ozone stress, which were closely related with one another.

Cucumber green mottle mosaic virus Moved into the Non-Host Figleaf Gourd Passing through Cucumber in Grafting System (오이/흑종호박 접목에서 오이녹반모자이크바이러스의 비기주 대목인 흑종호박으로 이동)

  • Cucumber green mottle mosaic virus (CGMMV) was not infected in figleaf gourd by sap inoculation. However CGMMV was detected by RT-PCR from the figleaf gourd collected from a field growing cucumber grafted onto figleaf gourd in Cheonan, Chungcheongnam Province in 2008. Which field showed 100% infection rate of the virus disease. In the experiment grafted with cucumber onto figleaf gourd, transportation of CGMMV through cucumber to figleaf gourd was confirmed by RT-PCR when the virus was mechanically inoculated on the leaves of the cucumber. The amplified DNA concentration of the virus on electrophoresis gel was much higher in the cucumber than in the figleaf gourd. However, the virus particles from the figleaf gourds were not observed under electron microscopy, also sap of the figleaf gourds was not transmittable to Nicotiana benthamiana. To identify the existence of CGMMV particle, the virus was purified from figleaf gourd and cucumber growing together in the graft system. CGMMV solution extracted from the cucumber represented a typical absorption spectrum of the virus but that from the figleaf gourd did not. Only a few CGMMV particles were observed in the purified preparation from the figleaf gourd. These results confirmed that CGMMV only passed through figleaf gourd in the grafting system. This study indicated that figleaf gourd is not a host of CGMMY.

천연물로부터의 다당류에 의한 항종양효과에 관한 연구 I. 형광기표지와 HPLC에 의한 당의 분석

  • 정상세포의 암세포로의 전환에 수반하는 세포표면상에 당구조 변화가 주목을 받고 있으며 이에 탄수화물이 세포간의 상호작용, 성장 등에 중요한 역할을 하리라고 생각된다. 따라서, 당구조 변화를 야기 시키는 당분해소효소나 전이활성 효소의 활성이 주요한 임상적 지표가 될 수 있다. 또한 천연물에서 유래된 다당류 등이 면역증강 효과를 나타내면서 항종양 효과를 보여준다. 이와같은 일을 원활히 수행하기 위해서는 당을 분석하는 기술이 필요하다. 당을 분석하는 방법으로 많이 이용된 것 중의 하나가 유도체를 만들어 GC에 의해서 확인하는 것이고 또한 당은 chromophore가 없기 때문에 굴절율의 검출기를 이용한 HPLC에 의해서도 분석이 가능하다. HPLC에 의한 또 다른 방법으로 당의 환원당 말단에 형광기를 결합시켜 감도를 매우 높게하여 역상 칼람을 이용해 당을 분리할 수 있다. 환원당에 결합시킬 수 있는 물질로는 대부분 amine기를 띄고 있고 결합하여 형광을 나타내는 특징을 지니고 있다. 본 연구는 형광기를 결합시켜 전기영동과 HPLC에 의해 동시에 추적이 가능한 물질을 결합시켜서 분석을 용이하게 한 방법을 이용하여 단당류를 $C_{18}$ column에 의해서 분리하였고 또한 모델로서 면역증강제로 사용되는 운지버섯 및 영지버섯 다당류와 생약에서 분리한 지유의 당을 분석하였다. 현재까지의 결과로 가장 좋은 방법은 $C_{18}$-->phenyl column을 연결하여 isocratic 방법으로 분리하였을 때이었다. 버섯의 당은 대부분이 glucose로 이루어졌고 지유는 arabinose가 주 구성성분이었다. 위의 분석 기술을 이용하면 피코몰 범위내에서 정확한 구조를 가진 항암효과의 올리고당 및 암세포의 발현에 따른 당단백질의 당의 구조를 규명하는데 도움을 줄 수 있다.aeonol이 aspirin과 같거나 강한 사망 억제 효과가 있었다.cyclopropyl-7-(2-furanyl) -6-fluoro-1,4-dihydro-4-oxo-3-quinoline carboxylic acid (compound 4), 1-cyclopropyl-7-(2-thiophenyl)-6-fluoro-1,4-dihydro-4-oxo-3-quinoline carboxylic acid (compound 6) ,1-cyclopropyl-7-(3-pyridinyl)-6-fluoro-1,4-dihydro-4-oxo-3-quinoline carboxylic acid (compound 8), 1-cyclopropyl-7-(2-fluoro-3-pyridinyl)-6-fluoro-1,4-dihydro-4-oxo-3-quinoline carboxylic acid (compound 10)를 합성하였다.10^{-7}$ M)에 의한 단백인산화에 대하여는 더 미약한 억제-효과를 나타내었다. 이상의 결과는 PDE-1과 항우울약들의 항혈소판작용은 PKC-기질인 41-43 kD와 20 kD의 인산화를 억제함에 기인되는 것으로 사료된다.다. 것으로 사료된다.다.바와 같이 MCl에서 작은 Dv 값을 갖는데, 이것은 CdCl$_{4}$$^{2-}$ 착이온을 형성하거나 ZnCl$_{4}$$^{2-}$ , ZnCl$_{3}$$^{-}$같은 이온과 MgCl$^{+}$, MgCl$_{2}$같은 이온종을 형성하기 때문인것 같다. 한편 어떠한 용리액에서던지 NH$_{4}$$^{+}$의 경우 Dv값이 제일 작았다. 바. 본 연구의 목적중의 하나인
