Studies on Various Egg Traits of Korean Native Chicken (한국 재래계의 난 형질에 관한 연구)

  • The present study was conducted to estimate average weights, and corrlation and regression coefficients among various traits of eggs obtained from Korean native and imported chicken. Data were obtained from 360 layers (180 of Korean native origin, and 180 of imported Decalb Brown) between November 8, 1994 to February 8, 1995. The results obtained are summerized as follows : 1. The mean weights of whole egg, yolk, dense white, thin white, and egg shell of native chicken were lighter (P<0.01) than those of Decalb Brown. 2. The coefficients of variability in each egg trait were lower in native chicken than in Decalb Brown, indicating the higher ability of native chicken to adapt to the local environment. 3. The phenotypic correlation coeffcients between whole egg and weights of yolk, dense white, thin white, and eggshell were 0.842∼0.992(P<0.01) in native chicken, and 0.865∼0.985(P<0.01) in Decalb Brown. 4. The average eggshell thicknesses of native and imported Decalb Brown were 362.5 $\mu\textrm{m}$ and 407.9 $\mu\textrm{m}$, respectively. 5. The regression coefficients of most of the traits of eggs from native chicken were lower than those from Decalb Brown layers, suggesting that Korean native chicken should be improved further in various egg traits measured in this study.

Studies on DNA Fingerprint for the Korean Native Chicken (한국 재래계의 유전자 지문에 관한 연구)

  • This study was conducted to classify Korean native chicken(KNC) and imported chicken by phenotypic performances and DNA fingerprinting. Two lines, KNC and White Leghorn(WL) , of chicken were maintained in the laboratory of Yeungnam University. Economic traits (body weight, sexual maturity, hen-day egg production, egg weight) and phenotypic characteristics (body-type, head, feather, shank) were checked. The DNA fingerprinting was analyzed for both breeds. The growth rate of the KNC was similar to WLS and sexual maturity of the KNC came later than WL. Hen-day egg production of the KNC was also slightly lower than the WL. The egg weight was about 10g lighter than WL. There was no difference in body weight of female KNC compared to the WL after 28 weeks. The study confirms difference between KNC and WL in DNA fingerprinting as well as its outlook. Thus, we suggest that these should be tested in nationwide districts about chickens known as the KNC using DNA fingerprinting. Then, the confirmed KNC populations should be maintained and used for the genetic improvement. Finally, only confirmed KNC should be in market which induce consumer to seek the KNC by its favorite.

Single Nucleotide Polymorphisms (SNPs) Discovery in GHSR Gene and Their Association Analysis with Economic Traits in Korean Native Chickens (GHSR 유전자 내 유전변이의 탐색과 한국재래계의 성장 및 산란 특성에 미치는 연관성 분석)

  • Recently, it was reported that certain polymorphisms in the growth hormone secretagogue receptor gene (GHSR) are associated with the growth of chickens. However, the correlation between GHSR polymorphisms and economic traits has not been investigated in Korean native chickens (KNCs). Therefore, the objective of this study was to confirm the suitability of the GHSR gene as a candidate for genomic selection and identify a genetic marker for KNCs. A total of 220 KNCs from six breeds raised at the National Institute of Animal Science were genotyped for the c.739+726 SNP in the GHSR gene using polymerase chain reaction- restriction fragment length polymorphism (PCR-RFLP), and the sequence for a subset of 30 birds was analyzed using direct sequencing. The association between the SNP genotypes and the economic traits of the KNCs was analyzed using the statistical package for the social science (SPSS) software program. The association analysis between the c.739+726T>C SNP and economic traits revealed that the SNP was significantly associated with body weight at 150 and 270 days (BW150 and BW270, respectively) in all KNCs (p<0.01), BW150 in KNC (Gary) (p<0.05), and egg production number in KNC (White, p<0.05). In addition, the SNPs discovered using direct sequencing (513A>G, 517A>T) had a significant effect on the body weight and egg production traits (p<0.05). In conclusion, these results might be useful as a basis for studies on the improvement of KNC breeds. Furthermore, these results suggest that the SNPs (c.739+726T>C, 513A>G, and 517A>T) located in the GHSR gene could be useful molecular genetic markers for KNCs.

Genetic Composition of Korean Native Chicken Populations - National Scale Molecular Genetic Evaluation Based on Microsatellite Markers (초위성체 표지로 본 한국 재래닭 집단의 분자유전학적 구성)

  • The study was conducted to select and optimize microsatellite (MS) markers for evaluate Korean Native Chicken (KNC) breeds in order to provide standard for the classification and breed definition of the indigenous breeds. The study also aimed to characterize and classify each KNC populations for inventory and management of avian genetic resources. A total of 462 chickens from 11 populations of chicken breeds including eight KNC breeds and three commercial chicken breeds were analyzed with 19 MS markers. KNC breeds, especially Long-Tail Chicken breeds, formed separate cluster from those commercial chicken breeds. Genetic distances between KNC populations (0.11~0.18) were relatively shorter. Genetic uniformity of KNC (except KNCR breed) (0.86~0.88) were higher than that of commercial breeds (except Cornish) (0.95~0.97). On the other hand, genetic uniformity of KNC Long Tail (KNCLT) were relatively higher (0.91~0.97). The result can be used to evaluate and manage animal genetic resources at national scale.

Identification of Chromosomal Band Markers of the Korean Native Chicken (한국재래계의 염색체 분염 표지 분석)

  • The present study was carried out to establish the standard karyotype of the Korean Native Chicken and to find their chromosomal band markers using high-resolution banding technique. Chromosome analysis was performed on early chick embryos following in vitro culture of fertilized eggs of the yellow-brown and the red-brown lines of the Korean Native Chicken which had been established at National Livestock Research Institute. The high-resolution banding of the chromosome was achieved by treating the embryos with ethidium bromide and colchicine during culture. On GTG-banding, the Korean Native Chicken exhibited a typical chick banding pattern in all the macrochromosomes. Overall chromosomal morphology and positions of typical landmarks of the Korean Native Chicken were virtually identical to those of White Leghorn and International System for Standardized Avian Karyotypes(ISSAK). However, the lengths and G-band numbers of the Korean Native Chicken macrochromosomes were greater than those of White Leghorn and ISSAK. Especially in chromosomes 1 and Z, the Korean Native Chicken exhibited more separated bands in compared with ISSAK. In C-banding patterns, although a lot of observed cells had C-band polymorphic patterns, almost the Korean Native Chicken macrochromosomes had heterochromatic C-band on centromeres and/or near terminal part. However, the heterochromatic C-band was constantly observed at the end of q-arm of Z chromosomes and on the whole W chromosome. In addition, the Korean Native Chicken exhibited distinctive heteromorphic patterns of C-bands on the centromere of chromosome 3 and at the end of q-arm of Z chromosome between homologous chromosomes.

Maternal Origins of the Jeju Native Pig Inferred from PCR-RFLP Haplotypes and Molecular Phylogeny for Mitochondrial DNA CYTB Gene Sequences (미토콘드리아 DNA CYTB 유전자 서열에 대한 분자 계통과 PCR-RFLP 반수체형에 근거한 제주재래돼지의 모계 기원)

  • In an effort to gain greater understanding of the maternal lineages of the Jeju native pig (JNP), we analyzed the mitochondrial DNA (mtDNA) CYTB gene and compared it with those of other pig breeds. PCR-RFLP analysis was conducted with six pig breeds including JNP, and then the RFLP patterns allowed for the separation of the pig breeds into two distinct haplotypes (mtCYTB1 and mtCYTB2). The JNP CYTB sequences were detected in both the European and Asian breed clusters on the phylogenetic tree. The J2 group was sorted with the indigenous cluster of Asian pig lineages and was related closely to Chinese native pig breeds, but a second group, J1, was sorted with the European pig lineages and appeared to be related to Spanish Iberian native pigs, rather than to Asian breeds. These results indicate that the JNP currently raised on Jeju Island have two major maternal origins estimated in Asian and European pigs. We concluded that the JNP that share a common lineage with indigenous Asian pigs were domesticated in the distant past, originating from pigs that were already being raised elsewhere at that time, and that the European pig breeds introduced in the twentieth century have also contributed to the formation of this pig population.

Studies on the Genetic Characters of Korean Native Fowl (한국 재래닭의 유전적 특성에 관한 조사 연구)

  • This study was carried out to obtain the information on the genetic characters of Korean native fowls. The data for this study was collected from fowls located at the Gyunggi, Chunscheong, Yeongnam and Jeju area from March, 1989 to November, 1989. The morphology, genetic characters and economic traits of fowls were investigated. Followings are the major results obtained from this study. 1. The morphology and genetic characters of Korean native fowls are similar to those of brown Leghorn, 2. Colored fowls outnumbered white fowls by 402 to 38 in feather color, and wild type fowls outnumbered black fowls by 317 to 38 in feather pattern. 3. For morphology of chromosomes, chromosome size and shape of Korean native fowls were similar to those of other breeds. 4. Body weights of male was 1.2-1.6kg, and of female 1.5-1.9 kg. Hen-housed egg production was 80-120 eggs.

Fatty Acids Composition and Sensory Characteristics of the Commercial Native Chicken Meat (실용 재래닭의 지방산 조성 및 관능적 특성)

  • Fatty acid composition and sensory characteristics of six commercial chickens(CC-RcDR, RcLR, BcBR, BcDR, BcLR, RcBR), one broiler(Arbor Acre) and three Korean native chickens(KNC-D, L, B) were evaluated. In the fatty acid composition at marketing age, KNC had generally higher unsaturated fatty acid ratio than the others. B, BcBR and BcDR showed the highest in unsaturated fatty acid but the lower than others in saturated fatty acid composition. The results of sensory test showed significant difference(P<0.05) only in the leg. B and RcBR had the highest score and Arbor Acre lowest score in juiciness. In tenderness. B had the highest score and RcDR lowest score. In the case of acceptability, KNC generally had high score. Particularly B had the highest score. Among CC, RcBR had the highest score. There were no significant differences in flavor and aroma.

Genotype Analysis of the Major Histocompatibility Complex Region in Korean Native Chicken (한국 재래닭의 MHC 영역 유전자형 분석)

  • The chicken major histocompatibility complex (MHC) is known to be associated with disease resistance and susceptibility to several pathogens. The microsatellite marker LEI0258 is physically located between the BG and BF of MHC region and variations near this marker have been well documented. In this report, the LEI0258 marker was used to find specific alleles for the Korean native chicken. The MHC haplotype was analyzed by PCR screening and sequencing of LEI0258 region in four different breeds including black Korean native chicken, brown Korean native chicken, Cornish and Rhode island red. The serologically same MHC haplotypes showed the differences in repeat numbers, a few indels or single nucleotide polymorphisms by sequencing analysis. Even though we could not identify specific alleles for Korean native chickens, the genotypes analyzed in these breeds can give valuable information for the relationships with disease resistance and establishment of breeding strategies for the Korean native chicken.
