Antioxidant Activity of Dopamine-Modified Hydrogels Containing Cross-linked Hyaluronic Acid (도파민이 적용된 히알루론산 가교 하이드로겔의 항산화 활성)

  • Purpose : : In this study, we made dopamine-functionalized hydrogels containing a cross-linked hyaluronic acid (HA) network and investigated their antioxidant activities. Methods : In the first step, we made poly hydroxyethyl methacrylate(p(HEMA))-based hydrogels post-modified with an interpenetrating polymer network(IPN) structure composed of HA polymers and a p(HEMA) network. The subsequent functionalization with dopamine via an amide coupling reaction resulted in the antioxidant hydrogels. Their antioxidant activities were evaluated using 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) and 2,2-diphenyl-1-picrylhydrazyl radical scavenging assays. Results : The dopamine-modified hydrogels exhibited significant antioxidant activities, when compared to unmodified control. The presence of the HA-IPN structure improved the surface wettability of the hydrogel while dopamine-conjugated IPN hydrogel did not demonstrate the significant difference compared to hydrogel control. Dopamine-modified hydrogels exhibited high transmittance (>88%). Conclusion : The results demonstrate that the development of antioxidant hydrogels based on dopamine-conjugated HA-IPN structures may help develop ophthalmic and biomedical materials.

The physical properties of the cosmetic hydrogels affected by adding various celluloses (셀룰로오즈 첨가에 따른 미용용 하이드로겔의 물성 연구)

  • Hydrogels are natural polymer networks that can contain huge quantities of water and many cosmetical ingredients. Their hydrophilic functional groups creates a matrix, which allows high efficacy in delivering active ingredients into the skin. In industry, hydrating properties and strength of the hydrogels are of great interest in manufacturing hydrogel mask packs. We have used the cellulose in various forms such as powder, cotton fiber and cellulase treated cotton fiber to investigate the property changes of cellulose/hydrogel sheets. When 0.1% and 0.3% of cellulose powder were added to hydrogels, tensile strength of hydrogel sheets were decreased by 10% and 14% respectively. Vise versa, when 0.5 ~ 2 cm of cotton fibers were added, tensile strength of hydrogel sheets were significantly increased by about 20%. The hydrogels which contain cotton fibers also gave an excellent moisturizing effect. Especially cellolose/hydrogels containing cellulase-treated cotton fibers showed the best effect on retaining moisture content increasing upto 380% in comparison with the one containing untreated cotton as well as excellent dispersibility.

조절방출성 신제제를 위한 소프트하이드로겔의 개발

  • 소프트하이드로겔은 소재의 친수-소수성, 조성, 가교도, 약물의 물성에 의해 약물방출을 조절할 수 있었으며 또한 특이적인 전연성, 탄성에 의해 약물의 0차방출이 가능하였다. 메타크릴산-IPN-소프트하이드로겔은 현저하게 높은 PH의존성 팽윤도와 이에 따른 약물조절방출능을 보였으며 아크릴산-IPN-소프트하이드로겔은 우수한 점착성을 나타내었다. 이상 연구된 소프트하이드로겔들은 새로운 소재로서 개발대상으로 설정된 조절방출성 제제로의 활용성을 기대되었다.

Synthesis of PVA/PVP Hydrogel by Irradiation Crosslinking (방사선 가교에 의한 PVA/PVP 하이드로겔 제조 및 드레싱에의 응용)

  • Hydrogels for wound dressing were manufactured using poly(vinylalcohol)(PVA) and poly(N-vinylpyrrolidone)(PVP). The hydrogels were obtained by exposing to $^{60}$ Co${\gamma}$-rays after freezing and thawing of aqueous solutions of PVA and PVP to improve mechanical strength. Mechanical properties such as gelation, water absorptivity and gel strength were examined after repeating the \"freezing and thawing\" of PVA/PVP hydrogels, and then irradiating them at 40 kGy. The PVA/PVP ratio was in the range of 30:70 ~ 100:0, and the solid concentration of PVA/PVP was 20 wt%. The gelation and strength of hydrogels were much higher when \"freezing and thawing\" and the irradiation process were used than when only the irradiation process was utilized.   • 소프트 로봇의 수요와 관심이 증가함에 따라 생체 모방형 액추에이터 연구가 큰 관심을 받고 있다. 액추에이터란 외부 에너지를 기계적인 동작으로 변환하는 장치이며, 재료 자체가 유연하여 부드러운 움직임을 재현할 수 있는 소프트 액추에이터의 연구가 활발히 진행되고 있다. 고분자 연성 재료 중에 하나인 하이드로겔은 90% 이상이 물로 구성되어 있기 때문에 생체 친화적이면서 동시에 환경 친화적인 재료이며 이를 기반으로 한 액추에이터 연구가 새로이 각광받고 있다. 최근에는 하이드로겔 액추에이터의 성능 향상을 위해 나노재료를 하이드로겔에 첨가하는 연구가 진행되고 있으며, 나노재료가 갖는 고유의 특성을 활용함으로써 하이드로겔 액추에이터의 자극 감응성 향상, 변형 방향의 제어, 높은 변형 효율 그리고 기계적 물성 증가가 보고되고 있다. 이는 헬스케어를 위한 웨어러블 장치, 재활을 목적으로 한 인공 근육 등에 적용이 가능하다. 본 기고문에서는 자극 감응성 고분자와 나노재료를 이용한 하이드로겔 액추에이터 연구에 대해 자극(전기장, 빛, 열, 자기장)의 종류에 따라 분류하여 소개하고, 합성 전략 및 구동 원리에 대해 간략하게 설명하고자 한다.

Synthesis of Electroactive PAAc/PVA/PEG Hydrogel Soft Actuator by Radiation Processing and Their Dynamic Characteristics (방사선을 이용한 전기 활성 PAAc/PVA/PEG 하이드로겔 소프트 액추에이터의 제조 및 구동 특성 분석)

  • Over the last few decades, there have been a lot of efforts to develop soft actuators, which can be external stimuli-responsive and applied to the human body. In order to fabricate medical soft actuators with a dynamic precision control, the 3D crosslinked poly(acrylic acid) (PAAc)/poly(vinyl alcohol) (PVA)/poly(ethylene glycol) (PEG) hydrogels were synthesized in this study by using a radiation technique without noxious chemical additives or initiators. After irradiation, all hydrogels showed high gel fraction over 75% and the ATR-FTIR spectra indicated that PAAc/PVA/PEG hydrogels were successfully synthesized. In addition, the gel fraction, equilibrium water content, and compressive strength were measured to determine the change in physical properties of PAAc/PVA/PEG hydrogels according to the irradiation dose and content ratio of constituents. As the irradiation dose and amount of poly(ethylene glycol) diacrylate (PEGDA) increased, the PAAc/PVA/PEG hydrogels showed a high crosslinking density and mechanical strength. It was also confirmed that PAAc/PVA/PEG hydrogels responded to electrical stimulation even at a low voltage of 3 V. The bending behavior of hydrogels under an electric field can be controlled by changing the crosslinking density, ionic group content, applied voltage, and ionic strength of swelling solution.

소프트하이드로겔 소재에 의한 조절방출성 약제개발

  • 소프트하이드로겔의 합성은 IR, DSC등으로 확인되었으며, 전연성의 물성을 나타내었다. PPO 하이드로겔은 대부분 수팽윤도가 10%미만 정도로서 야굴방출시 팽윤에 의한 변형도가 낮아 부착형, 또는 이식형 조절방출성 제제에의 활용도가 기대되었으며, 약물방출은 주로 가교도/주쇄연장도에 의한 고분자 네트?의 세공크기(mesh size)에 의해 조절되었다. IPN하이드로겔의 경우는 pH의존성이 일반적이 겔보다 현저하게 나타났으며, 하유된 약물이 물성에 따른 약물방출양상을 보였다.

The Change in the Parameters of Silicone Hydrogel Lens and Objective/Subjective Symptoms induced by Repetitive Dryness of Lens (실리콘 하이드로겔 렌즈의 반복적 건조로 인한 렌즈형태 및 자·타각적 증상의 변화)

  • Purpose: In this study, the changes in the shape of silicone hydrogel lenses, dryness of lens and objective/ subjective symptoms that could be induced by repeating dryness of lens and objective/subjective symptoms were investigated. Methods: After drying and rehydrating of silicone hydrogel lenses with different lens material and thickness for 4 times, their overall diameters and base curves were compared. Subjective symptoms, non-invasive tear break-up time (NIBUTs) and blinking rate were evaluated after wearing dehydrated silicone hydrogel lens. Results: Overall diameter and base curve increased in all tested silicone hydrogel lenses by repeating dryness and rehydration. The degree of change in over all diameter and base curve were variable on the material. There were irregular change of lens parameters in thicker lens. When the subjects wore silicone hydrogel lenses after drying and rehydrating, their NIBUTs were decreased and blink rates were increased regardless of lens material or thickness. In addition, repetitive drying of silicone hydrogel lenses affected the lens fitting. Therefore, subjective symptoms such as itching, pain, irritation, foreign body sensation, dryness tended to be increased. Conclusions: The results obtained from the study may suggest to develop durable silicone hydrogel contact lenses against dry environment since the changes in parameters of silicone hydrogel lens and the subjective discomfort were observed after repetitive drying and rehydration.

  • In this study, hydrogels from mixtures of poly (vinyl alcohol) (PVA)/poly(N-vinylpyrrolidone) (PVP)/glycerin/chitosan were prepared by γ-ray irradiation and the mechanical properties such as gelation, water absorptivity, and gel strength were examined to evaluate the applicability of these for wound dressing. Then PVA:PVP was weight ratio of 6 : 4, the concentration of chitosan was 0.3 wt%, the concentration of glycerin was in the range of 0∼5 wt%t. The solid concentration of PVA/PVP/glycerin/chitosan solution was 15 wt%. Gamma irradiation doses of 25, 35, 50, and 60 kGy were exposed to a mixture of PVA/PVP/glycerin/chitosan to evaluate the effect of irradiation dose. Gel content and gel strength increased as glycerin concentration in PVA/PVP/glycerin/chitosan decreased, and as irradiation dose increased. Swelling degree increased as glycerin concentration in PVA/PVP/glycerin/chitosan increased, and as irradiation dose decreased. The glycerin in PVA/PVP/glycerini/chitosan hydrogel prevented the transformation of shape. These hydrogel dressings had better curing effect than vaseline gauge.

Comparison of Pectin Hydrogel Collection Methods in Microfluidic Device (미세유체 장치에서 수거 방법에 따른 펙틴 하이드로겔 입자의 특성 비교)

  • This study investigated the effect of different collection methods on physical properties of pectin hydrogels in microfluidic synthetic approach. The pectin hydrogels were simply produced by the incorporation of calcium ions dissolved in continuous mineral oil. Then, different collection methods, pipetting, tubing, and settling, for harvesting pectin hydrogels were applied. The settling method showed most uniform and monodispersed hydrogels. In the case of settling, a coefficient of variation was 3.46 which was lower than pipetting method (18.60) and tubing method (14.76). Under the settling method, we could control the size of hydrogels, ranging from $30{\mu}m$ to $180{\mu}m$, by simple manipulation of the viscosity of pectin and volumetric flow rate of dispersed and continuous phase. Finally, according to the characteristics of simple encapsulation of biological materials, we envision that the pectin hydrogels can be applied to drug delivery, food, and biocompatible materials.
