Instrumental Assessments of Sub-clinical Skin Reactions induced by Cosmetic Ingredients (화장품 원료에 의해 유도되는 미세 피부반응에 대한 기기적 평가 연구)

  • The safety of cosmetics or cosmetic ingredients on human skin is generally evaluated by visual assessment but some early subtle skin changes may not be noticed by the naked eyes. Thus, the present study was conducted to detect skin reactions induced by mildly irritating cosmetic ingredients by using a laser Doppler perfusion imager (LDPI) method that measures blood flow, a $Vapometer^{(R)}$ that measure strans epidermal water loss (TEWL), and a spectrophotometer that measures the skin color as the erythema values ($a^*$). Visual assessment showed that all tested oils and humectants except propylene glycol belong to the low skin irritation ranges (grades 0+ to 2.9+) while all tested surfactants and propylene glycol belong to the moderate-to strong-skin irritation ranges (grades 3+ to 5+). Among three instrumental methods, TEWL assessment appeared to be more sensitive than spectrophotometric or LDPI method and suitable for the detection of subtle skin response invisible to the naked eye (grades 0+ to 2.9+). Skin reactions of grade 3+ to 5+ could be detected by all three instrumental methods. In conclusion, the current study suggested that the sub-clinical skin reactions due to mild irritants contained in cosmetics can be best assessed by TEWL measurements.

A Study on the Evaluation of Cosmetic Stability of Broccoli Extracts (브로콜리 추출물의 화장품 안정성 평가 연구)

  • This study compared antioxidant activity through DPPH radical scavenging activity of broccoli extract, low temperature (5℃ or less), and high temperature (50℃ or higher) storage conditions. As a result of the experiment, high DPPH radical scavenging activity of 95.5% was confirmed at 1% concentration of broccoli extract, and antioxidant power was maintained at a high level even under storage conditions due to temperature changes. Even cream containing 5% broccoli extract has been confirmed to maintain safety and stability in both preparation stability, discoloration and odor change, pH, and primary adhesion tests. Through these studies, it has been confirmed that broccoli extract is a safe and effective cosmetic material for the skin through high antioxidant activity and antioxidant power maintenance due to temperature changes.

Functional Cosmetic Effect of Sargassum Extracts (모자반추출물의 기능성 화장품소재 특성)

  • There is growing interest in natural cosmetic ingredients as natural cosmetics become popular. As a part of effort to look for natureal cosmetic agent, seaweed Sargassums were tested for functional cosmetic agents. Effective materials were extracted from Sargassum coreanum. Sargassum hemiphyllum, and Sargassum patens by simple hot water extraction. Antioxidation, whitening, anti-wrinkle, UV absorption and anti-inflammation effects were studied for functional cosmetic agents. Sargassum extracts indicated excellent cell viability, strong anti-oxidation effect by DPPH radical scavenging activity and showed significant whitening effect from tyrosinase inhibition. However, effects of antiwrinkle, UV absorption and anti-inflammation were negligible. In conclusion, Sargassum coreanum extracts showed good possibility for anti-oxidation and whitening cosmetic agent.

The Sympathetic Skin Responses after Thoracic Sympathicotomy for Patients with Palmar Hyperhidrosis (수장부 다한증환자의 흉부 교감신경절단술후 교감신경 피부반응)

  • Background: Thoracic sympathicotomy has been used safely and successfully to manage palmar hyperhidrosis. The preoperative and postoperative recording of Sympathetic Skin Responses(SSR) was performed for objective evaluation and follow-up of thoracic sympathicotomy in hyperhidrosis patients, and also for ascertaining the clinical usefullness of SSR. Material and Method: The recording of SSR was performed on 15 patients suffering from palmar hyperhidrosis with Medelec Sapphire Plus electromyogragh before and after thoracic sympathicotomy. Eletrical stimuli on the right median nerve was made in patients in supine position and results were recorded on right and left palms with soles at the same time by 4 channels. Skin temperatures were also monitored simultaneously. T2,3 sympathicotomy was performed with VATS in every patients. SSR was done in 2 patients one month later. Result: Clinically, all patients had symptomatic improvement with satisfaction. Postoperative complication was small amount of residual pneumothorax in 5 patients but it was absorbed sponteneously. There was no recurrence during follow-up period and ten patients(66%) complained compensatory hyperhidrosis. After operation, SSR change was shown in every 15 patients. Abolition of SSR on both palms was achieved in 12 patients(80%) and on both soles in 6 patients. In the other 3 patients, the latencies were significantly delayed and the amplitudes were significantly reduced at both palms and soles. In two patients who were examined at one month later after operation, similar results with postoperative SSRs were shown. The skin temperature on preoperative both palm and sole were lower than normal temperature, and those on postoperative both palm and sole were increased. Those had statistical significance(p<0.05), and the temperature on the palm was increased higher that than on the sole. Conclusion: After thoracic sympathicotomy was performed on palmar hyperhidrosis patients, an increment of skin temperatures and SSR changes were achieved at both palms and soles of all patients. Palmar SSRs were completely abolished in 12 patients(80%), and similar results of postoperative SSRs were achieved. The recording of SSR may be useful to easily and objectively assess the completeness of sympathicotomy and the follow-up of recurrence in hyperhidrosis patients.

Development of Novel Sulforaphane Contained-composition to Increase Antioxidant and Whitening Effects (항산화와 미백효과를 증진시킨 새로운 Sulforaphane 함유 혼합 조성의 개발)

  • In this study, we analyzed two components, sulforaphane and PF-3758309 to prove that the mixed composition of them has more effective antioxidant and whitening functions compared to each component. We analyzed the cellular toxicity of each component and also the mixed composition to find the safe concentration level for cell viability. From the single component treatment, we discovered that sulforaphane was safe up to $10{\mu}M$, and PF-3758309 up to 100 nM. Combination treatment of $10{\mu}M$ sulforaphane and 1 nM PF-3758309 did not affect the cell viability. The LPS-stimulated NO generation was significantly reduced by the mixed composition of sulforaphane and PF-3758309. Melanogenesis by ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH) was also inhibited by the mixed composition. In order to confirm the possibility as the cosmetic material, we carried out clinical studies for the mixed composition samples. Skin safety evaluation using patch test was judged to be unstimulated, skin whitening effect was increased, and melanin deposition was suppressed by treatment of mixed composition samples. These results provide us with the opportunity for applying it into the development of new functional cosmetics.

Antimicrobial Activity and Safety Test of Mixed Plant Extracts Including Phellodendron Amurense and Eucommia Ulmides Oliv (황벽나무, 두충나무 등을 포함하는 복합수목추출물의 항균활성 및 안전성 시험)

  • In this study, mixed plant extracts including Phellodendro namurense and Eucommia ulmides Oliv were tested for antimicrobial activity and safety. Antimicrobial activity was measured by disc diffusion method using normal skin flora and opportunistic microbes such as Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Candida albicans. As safety test, single dose oral toxicity test, single dose inhalation toxicity test and repeat dose inhalation toxicity test were done. From antimicrobial test, plant extracts showed significant antimicrobial activity Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans, however, did not indicate any antibacterial effect on Escherichia coli. From 3 kinds of safety tests, toxicity of mixed plant extracts was not observed. From experimental results, mixed plant extracts including Phellodendro namurense and Eucommia ulmides Oliv showed good potential for natural antimicrobial agent.

Antimicrobial, antifungal effect and safety verification using BCOP assay of extracts from Coptis chinensis (황련(Coptis chinensis) 추출물의 항균, 항진균 효과와 BCOP 분석을 이용한 안전성 검증)

  • Coptis chinensis is used in oriental medicine for soothing, anti-inflammation, antimicrobial and antipyretic properties, and its main ingredient berberine is known to have strong antibacterial activity. In this study, we investigated the anti-microbial effect of hot water extract of Coptis chinensis (CW) on skin related microorganism and the airborne microbe, the antifungal effects of fungi, which are frequently detected in residential environments. CW showed antibacterial effect against Propionibacterium acnes, Staphylococcus aureus and Staphylococcus epidermidis, against the airborne microbe, which was collected in four different places. At the concentration of 100 mg/mL, the antimicrobial activity continued for 42 days, showed heat stability without change in the antimicrobial activity even after heat treatment. The MIC and MBC of CW against S. aureus was 0.03, 0.05 mg/mL, against S. epidermidis was 0.50, 0.75 mg/mL and against P. acne was 0.10, 0.15 mg/mL. As a result of measuring the MIC of four kinds of fungi with high detection frequency in the surrounding environment, Gliocladium virens was 65 mg/mL by determined as MIC which can inhibit one hundred percent of mycelial growth. The concentration 90 mg/mL was determined as MIC against Aureobasidium pullulans and 100 mg/mL against Penicilium pinophilum and Chaetomium globosum. CW was considered a safe extract that showed no irritation even in the ocular mucous membrane irritation evaluation test, a patch test. Therefore, these results suggest that Coptis chinensis has antimicrobial, antifungal and safety on human body and can be applied to the development of materials for cosmetic and residential environment industries.

A Preliminary Study Protocol to Evaluate Patient Outcomes, Feasibility, Preliminary Effectiveness, Safety and Economic Evaluation of Korean Medicine Treatment for Periodontal Disease (치주질환에 대한 한의치료의 환자성과, 연구 가능성, 예비 효과, 안전성 및 경제성 평가를 위한 예비 연구 프로토콜)

  • Objectives : The purpose of this study is to assess the feasibility of conducting a study on traditional Korean medicine treatments for periodontal diseases, specifically gingivitis and mild periodontitis. Methods : This study will employ a randomized, controlled, parallel-group design. Subjects with gingivitis and mild periodontitis will be recruited in one university hospital. In total, 45 subjects will be randomized into three arms (Acupuncture therapy group, herbal mouthwash group and usual care group), and will be followed up for 4 weeks. We will assess clinical variables such as, pocket depth, bleeding on probing, gingival index, plaque index, visual analog scale, uroqol-5 dimensions-5 levels to analyze changes in microbial flora before and after the intervention. Results : The protocol for this study was approved by the Institutional Review Board (IRB) of Dongguk University Ilsan Oriental Medicine Hospital, and registered with the Korean Clinical Trial Registry on March 29, 2024. Conclusions : This study is the first clinical research on periodontal diseases conducted in a Korean traditional medicine institution. The research aims to broaden the scope of traditional Korean medicine and is expected to serve as crucial data for future large-scale studies.

A study on the Formulation of high-moisturizing Cosmetics containing Ulmus davidiana supercritical fluid extract (초임계 추출 느릅나무 성분을 함유한 고 보습 화장품 제형에 대한 평가 연구)

  • This study was performed to develop the cosmetics containing extract of U.davidiana, which has been confirmed as a functional ingredient of anti-microbial, anti-oxidant, and anti-inflammatory effects, and also to evaluate the improvement effects of dry and sensitive skin such as atopic dermatitis. Toxicity tests of the Ulmus davidiana supercritical fluid residue 60% EtOH extracts(USCFR), Ulmus davidiana supercritical fluid extracts(USCF) and preliminary formulations, DKBP-1903(gel), containing the USCFR and USCF were determined to be 'Non-toxic'. Based on this, the stability test of the lotion, cream, gel formulation was conducted and the results showed that cream and gel formulation were stable. A 48-hour moisturizing sustainability test and the improvement test on the pruritus caused by dryness were selected for the cosmetic efficacy test to evaluate the improvement of atopic dermatitis and as a result both cream and gel have been found to be relatively safe moisturizers that can help dry and sensitive skin.

  • 지구온난화의 폐해 중 하나인 해파리의 대량증식은 환경 생태계에 좋지 않은 영향을 미치고 있고, 또한 과도한 개체수는 원할한 처리가 곤란해 많은 문제점을 초래하고 있는 실정이다. 이러한 해파리의 생산적이고 효율적인 개선방안으로 단순 가공식품 뿐만 아니라 해파리의 추출을 통해 산업적으로 활용가능한 기능성 화장품원료로의 응용가능성도 고려해 볼 수 있을 것이다. 따라서 본 연구는 보름달물 해파리 유래 추출물의 기능성 화장품 원료로서의 안전성 및 유효성을 규명하였다. 인간피부섬유아세포에서의 세포독성, 주름 개선 효과와 관련된 procollagen 발현양의 증가 및 노화로 증가되는 SA-${\beta}$-Gal 효소의 활성을 염색을 통하여 확인하였고, 항노화 화장품 신소재로서의 사업화 가능성을 확인 할 수 있었다.
