  • The ultrastructures of the granular glands in the asiatic land salamander (Hynobius leechi) skin were observed by means of electron microsope The results were as follows; 1. The granular gland of the asiatic land salamander skin consisted of a body of gland and a duct. The body of the granular gland consisted of the glandular epithelial cells and the myoepithelial cells. The duct of the granular gland consisted of the keratinocytes. 2. The glandular epithelial cells in the asiatic land salamander skin were divided into the dark cell and the light cells in accordance with the electron density of the cytoplasm. 3. The secretory granules of the granular glands were round or oval in form and were divided into the various granules, showing the secretory granules showing weak electron density had the parts showing strong electron density near the granular membrane. 4. It is supposed that showing the different electron densities of the granules in a glandular epithelial cell is due to different mature stages and to different level of water absorption, and the chemical components of the granules showing different electron densities are identical.

Identification of Antioxidant Activities and Stimulation of Human Keratinocytes Differentiation Effects of Syzygium claviflorum Extract (Syzygium claviflorum 추출물의 항산화 활성 및 각질형성세포 분화유도 효과)

  • We validated the physiological activity of Syzygium claviflorum (Roxb.) Wall. ex A.M. Cowan & Cowan (S. claviflorum) extracts (leaves, stems, fruits, and flowers) as a cosmetic ingredient. Firstly, S. claviflorum extracts removed over 80% of free radicals at various concentrations in antioxidant experiments using the DPPH and ABTS assay. In cytotoxicity experiments using human epidermal keratinocytes, S. claviflorum extracts showed low cytotoxicity. In addition, S. claviflorum extracts significantly increased the expression of keratin (KRT)1, KRT2, KRT9, KRT10, which are differentiation markers of keratinocytes, as well as genes involved in the maintenance of skin barrier function, including involucrin (IVL), loricrin (LOR), filaggrin (FLG), and claudin1 (CLDN1). In particular, the expression of FLG protein, inhibited by interleukin (IL)-4/IL-13 in atopic dermatitis, was restored by S. claviflorum extracts in in vitro experiments. Therefore, S. claviflorum extracts with excellent antioxidant efficacy and skin barrier improvement function will be useful materials for the development of future atopic dermatitis treatments and cosmetics.

Rice Cell Origin Recombinant Human Granulocyte Macrophage Colony-Stimulating Factor (rrhGM-CSF) Could Improve the Wound Healing in Diabetic Hamster (당뇨가 유발된 햄스터 창상치유에 미치는 벼세포 유래 GM-CSF의 효과)

  • GM-CSF is a multipotent growth factor, which also plays an important role during the process of wound healing. rrhGM-CSF was specifically produced from rice cell culture in our laboratory (Hanson Biotech Co., Ltd, Daejeon). The rrhGMCSF contains more oligosaccharide side chains than any other types of GM-CSF. This work was taken to evaluate the influence on wound healing of rrhGM-CSF in male golden hamsters. Full thickness skin defects of 9 mm in diameter were made in the back of hamsters, and 100 ${\mu}L$ ointment containing rrhGM-CSF 50 ${\mu}g/mL$ was applied. Control groups were given ointment without rrhGM-CSF. The wound sizes were relatively reduced and skin was well regenerated in the experimental group compared with the control group. Structurally, reepithelialization and architecture of the skin following injury were well accomplished in the experimental group. And also, positive reaction of PCNA of the skin following injury was more prominent in rrhGM-CSF containing ointment treatment group. Since this type of GM-CSF has highly glycosylated side chains, the effectiveness might be retain longer and stable, regarding acceleration of wound healing in the animal model. The present study has important implications for further development of the therapeutic manipulation of wound healing using rrhGM-CSF.

Forsythiae Fructus Induces VEGF Production via p38 MAPK Activation in Human Keratinocytes (각질형성세포에서 p38 MAPK 활성을 통한 연교의 VEGF 생성 효과)

  • Cutaneous microvasculature plays a critical role in age-associated skin changes. A considerable reduction of number and size of vessels has been observed in the upper dermis of elderly skin. Forsythiae fructus (FF), the dried fruit of plant Forsythia suspensa (F. suspensa), has been traditionally used as an herbal medicine to treat inflammatory diseases and bacterial diseases. However, its regulatory effect on angiogenic responses has not been elucidated in skin. Therefore, we analyzed secretory profiles upon treatment of FF extract using array designed to detect angiogenesis-associated mediators in human keratinocytes. Because keratinocyte-derived VEGF (vascular endothelial growth factor) has been regarded as a potent factor for new microvasculature under the epidermis, we further investigated the effect of FF extract on VEGF production. We observed that the VEGF expression of mRNA and protein level was increased by about 2 folds in a dose-dependent manner after FF extract treatment. In signaling experiments, FF extract induced rapid p38 MAPK activation within 5 min, and the activation was totally abrogated by pretreatment with a p38 MAPK specific inhibitor. The FF-induced VEGF upregulation was also significantly attenuated by a p38 MAPK inhibition. Taken together, FF extract induces VEGF production via p38 MAPK activation in human epidermal keratinocytes. These novel findings suggest that FF is useful as a potential agent with pro-angiogenic activity and may help to improve age-dependent reduction of the microvasculature in aged skin or to heal skin wound.

Biology of melanocytes and melanogenesis (멜라닌세포의 특성과 멜라닌 형성)

  • Melanocytes, derived from neural crest, make melanin and protect skin from the hazardous ultraviolet light. Melanocytes with dendritic process has similar morphology with neurogenic cells and share growth factor receptors such as neurotrophin receptors. Melanogenesis can be regulated by ultraviolet light and inflammation of the skin. In addition, several factors such as hormone, cytokines, arachidonic acid can affect the proliferation and melanogenesis of melanocytes. For melanogenesis, melanocytes need expression of various genes including tyrosinase, TRP-1, TRP-2. In addition, melanin need to be transferred from melanocytes to surrounding keratinocytes. The biology of melanocytes is complex and mechanism of melanocytes proliferation and melanogenesis is still under the investigation.

Transcriptome Analysis of Human HaCaT Keratinicytes by Ginsenosides Rb1 and Rg1 (진세노사이드 Rb1과 Rg1에 의한 HaCaT 피부각질세포의 전사체 분석)

  • This study examined the efficacy and the mechanism of action of biological response modifiers, ginsenosides Rb1 and Rg1 isolated from Panax ginseng C.A. Meyer on human keratinocytes HaCaT cell lines. A non-significant cytotoxic response was obtained in the HaCaT cell lines on treatment with various concentrations of ginsenosides Rb1 and Rg1 for different time durations. Furthermore, the global changes in the mRNA profile of HaCaT cells were investigated using DNA microarrays after stimulation with the ginsenosides Rb1 and Rg1. Ginsenosides Rb1 and Rg1 strongly increased FGF2 in HaCaT cells, and were found to be a candidate gene for antioxidant activity and elasticity. Other key candidate genes for antioxidant activity, such as FANCD2, LEPR, and FAS, also show enhanced regulation in HaCaT cells treated with ginsenoside Rb1. This study will be useful for understanding the regulatory genes involved in skin elasticity and signal transduction pathway stimulated by the ginsenoside Rb1. This paper currently focuses on the key factors regulating the interaction of anti-aging principles and skin elasticity.

Effect of Cosmetics Contained Isotonic Water Mimicked Body Fluid on Cell Activities and Skin (생체 모사수 화장품이 세포 활성과 피부에 미치는 효과)

  • Body fluid has been studied for diverse fields like Ringer's solutions, artificial joint fluids, cell growth culture media because it plays a crucial role in controlling body temperature and acts as a solvent for diverse metabolite processes in the body and delivery media of mineral, energy source, hormone, signal and drug from and to cell via blood or lymphatic vessel by osmotic pressure or active uptake. Stratum corneum containing extracellular lipids and NMF (natural moisturizing factor) absorbs atmospheric water residing outside of cells and utilize it to hydrate inside of their own. This process is related to skin barrier function. In this study, we conducted the cell viability test with Cell Bio Fluid $Sync^{TM}$, which mimicks body fluids including amino acids, peptides, and monosaccharides to strengthen skin barrier, and the clinical skin improvement test with cosmetics containing Cell Bio Fluid $Sync^{TM}$. In the cell viability test, HaCaT cell was treated with PBS for 3 hours, followed by the treatment of a cell culture medium (DMEM) and isotonic solution (PBS) and Cell Bio Fluid $Sync^{TM}$ for 3 hours each. Then, MTT assay and image analysis were conducted. In the clinical skin improvement test, twenty-one healthy women participated. Participants applied cosmetics containing Cell Bio Fluid $Sync^{TM}$ on their face for a week and evaluated the skin hydration, skin roughness, brightness and evenness. All measurements were conducted after they washed off their face and took a rest under the constant temperature ($22{\pm}2^{\circ}C$) and constant humidity conditions ($50{\pm}5%$) for 20 minutes. All the data were analyzed by SPSS (version 21) software program. Results showed that Cell Bio Fluid $Sync^{TM}$ improved both the cell viability and in vivo skin conditions such as skin hydration, roughness, brightness and evenness.

Effects of Bambusae Caulis in Taeniam Extract on the UVB-induced Cell Death, Oxidative Stress and Matrix Metalloproteinase 1 Expression in Keratinocytes (각질세포에서 자외선B가 유도한 세포 사멸, 산화적 스트레스 및 matrix metalloproteinase 1 발현에 대한 죽여추출물의 영향)

  • Ultraviolet radiation (UV) is a major cause of skin photoaging, and effective UV protecting agents are needed for the skin health and beauty. This study was undertaken to examine the effects of Bambusae caulis in Taeniam extract (BCTE) on UVB-induced cell death, oxidative stress and matrix metalloproteinase 1 (MMP1) expression in cell-based assays. HaCaT human keratinocytes were exposed to UVB in the presence of BCTE at different concentrations and resulting changes in cell viability and biochemical events were determined. The results showed that BCTE enhanced the viabilities of UVB-exposed cells, and attenuated apoptotic events such as cleavage of procaspase 3 to its active form, and the increase of Bax to Bcl-2 ratios. BCTE also attenuated the reactive oxygen generation and lipid peroxidation in cells exposed to UVB. Additionally, it attenuated the expression of matrix metalloproteinase 1 and the phosphorylation of c-Jun N-terminal kinase stimulated by UVB. Conclusively, the present study demonstrated that BCTE pro tected skin cells from the UVB-induced cell death, oxidative stress and MMP1 expression, suggesting its potential use as a cosmetic ingredient mitigating some features of the skin photoaging.

Effect of Zanthoxylum piperitum Extract on Human Skin Protection from UVB by Regulation of COP1 and PPAR-α (초피나무 열매 추출물의 COP1 및 PPAR-α 조절을 통한 자외선에 대한 피부 보호 효과)

  • Ultraviolet (UV) irradiation from the sun is the primary environmental factor that causes skin damages including skin cancer and premature skin aging. Because, even the most powerful sunscreen can't always afford enough protection, it is necessary to enhance the defensive power of skin against UV. Recently, constitutive photomorphogenic protein-1 (COP1) has shown to contribute to the regulation of UVB response of keratinocytes. In this study, we represent that COP1 and its associated protein, de-etiolated 1 (DET1), might participate in photoaging process in human skin as Arabidopsis COP1 does sun-protective function in plants. After UVB irradiation, the decrease of COP1 and DET1 mRNA expression was followed by the increase of c-Jun total protein. Moreover, transfection with DNA vectors expressing COP1 and DET1 down-regulated the c-Jun total protein. We found that Zanthoxylum piperitum extract (ZE) up-regulated the expression of COP1 and DET1 on human keratinocytes, and inhibited the expression of MMP1 which is one of the genes regulated by c-Jun signal. In addition, ZE has been reported to stimulate PPAR-${\alpha}$ and strengthen the skin barrier. We found that ZE decreased the UVB-induced IL-6 and IL-8 in NHEK cells. In human study, ZE protected skin against UV-B induced erythema and erythema-induced pigmentation. These results indicate that ZE could be useful for the protection against the adverse effects of UV irradiation through various mechanisms.

Beneficial Effect of a Collagen Peptide Supplement on the Epidermal Skin Barrier (콜라겐 펩타이드의 피부 장벽 보호 효과)

  • Recent studies have revealed that collagen peptide (CP) plays a protective role in skin by improving the activity of antioxidants and acts as an inducer of skin regeneration by positive feedback. In this study, we focused on the beneficial effect of reinforcing the CP skin barrier. To evaluate the skin barrier, hairless mice were exposed to UVB irradiation and acetone-treatment, with or without oral administration of CP. The effects on skin appearance, trans-epidermal water loss, epidermal thickness, and cytokine content were measured using bioengineering and histochemical methods. In the CP treated group, the skin had better appearance and less damage than that of the control. Furthermore, in HaCaT cells, the amount of serinepalmytoyl transferase (SPT) mRNA increased by about 1.6-fold after treatment (CP, 100 mg/L), reflecting that CP can induce SPT expression and reinforce the recovery of skin barrier function. These results suggest that CP is not only an anti-wrinkling agent but also a potent candidate as an epidermal moisturizer.
