Morphological Characteristics of Chinese Holly (Ilex cornuta) Leaves in Korea (우리나라 호랑가시나무 잎의 형태적 특성)

  • The present study was carried out to analyze the leaf variation of hex corn uta which are one of very important native landscape woody materials, but rapidly destroyed by habitat alteration in the past decades. The leaf characters were ranged as 0.9-11.8cm for the leaf length, 0.4-7.7cm for the leaf width, 0.2-13.0mm for the petiole length, $0.7-46.7cm^2$ for the leaf area and 1-13 for the number of leaf serrations. Due to the leaf shape, number of spines, and colours of leaf and fruits, which are the main characters of the cultivars of the Chinese holly, the hollies in the wild and planted as identified as 9 cultivars and one hybrid including 'Cholbuam', 'Inchon' and 'Sun Chun'.

Phylogenetic Analysis using mtDNA D-loop Sequences in Korean Native Ducks (mtDNA D-loop 염기서열 변이를 이용한 토종오리의 계통 분류)

  • Recently, the consumption of duck meat has been gradually increased in Korea. However, most of the duck breeds in Korea were imported from overseas. Based on the large demands for the breeding stocks of native ducks, a new project for the commercial use of the Korean native ducks has been launched. For the initial investigation of the relationships between Korean native duck (KND) with other duck breeds, the sequences from D-loop control region in mitochondrial DNA (mtDNA) was used. The results from phylogenetic analysis indicated that both KND and White Commercial Duck (WCD) breeds were classified well with wild duck breeds. However, mallard duck was not discriminated well with KND. The haplotype analysis indicated that KND and WCD have eight different haplotypes with eleven SNPs. Three haplotypes (haplotype 1, 3, 4) were shared both in KND and WCD. On the other hand, haplotype 1 was appeared only KND and haplotype 5, 6, 7, 8 were identified only in WCD population. With further verifications, the results presented here can be used for the conservation and commercialization of the Korean native ducks.

Morphological diagnostic characters of Isodon (Lamiaceae) in Korea (한국산 산박하속(꿀풀과)의 외부형태학적 식별 형질)

  • Although a few recent taxonomic studies focusing on infraspecific taxa in each species of Isodon have been reported, a detailed taxonomic revision of the Korean Isodon taxa has yet to be performed. We investigated the morphological characters of Korean Isodon based on approximately 600 herbarium sheets from major herbaria in Korea and Japan. We identified characters which distinguish seven Korean Isodon taxa and created a key to Korean Isodon based on the results from this study and on recent studies of the Korean Isodon taxa. The following unique characters for several taxa were recognized: 1) dense non-glandular hairs in stems for I. inflexus var. canescens (over 70 ea/mm on one side of the stem), 2) dense glandular hairs on the abaxial surface of the leaf for I. serra (over $40ea/mm^2$), 3) protruding pistils and stamens from the corolla with the length of the protruding part longer than that of the lower lobe of corolla for I. japonicus, and 4) the presence of non-glandular hairs on the entire surface of the fruit of I. inflexus var. microphyllus. Based on this study, we recognized that there are four species, two varieties, and one forma of Isodon taxa in Korea.

Difference of Growth and Yield among Rice Cultivars and Direct Seeding Methods as Affected by Yearly Variation Weather (연차간 기상조건에 따른 벼 품종의 담수직파재배 양식간 생육 및 수량)

  • To identify the differences on plant growth and yield of two rice cultivars among direct water-seeding methods broadcasting on flooded paddy surface(BF), drilling on flooded paddy surface(DF), and puddled-soil drill seeding(PD) under markedly different weather condition between 1995 and 1996. The mean air temperature for duration from May to June, early growth stage of rice in 1995 was lower $1{\sim}3^{\circ}C$ than that in normal or 1996. In 1995 the respiratory consumption index during panicle formation stage and early ripening stage was higher than those of in 1996 or normal year. Number of seedling stand among the methods of direct seeding rice appeared slightly higher in order of BF>DF>PD. Properly in Nonganbyeo, the number of seedling stand was much low in 1995 compared with in 1996. The leaf area index and shoot dry weight at early growth stage of rice plant and culm length at mature in 1995 were larger in direct water seeding rice than those of machanical transplanting rice, but in 1996. Faster ripening speed and shorter ripening period of rice crop appeared in 1996 compared to in 1995. It was due to higher growing degree-days, sunshine hours and solar radiation during rice growing season in 1996. Dongjinbyeo rice showed higher yield than Nonganbyeo which had lower ripened grains especially in 1995.

Studies on DNA Fingerprint for the Korean Native Chicken (한국 재래계의 유전자 지문에 관한 연구)

  • This study was conducted to classify Korean native chicken(KNC) and imported chicken by phenotypic performances and DNA fingerprinting. Two lines, KNC and White Leghorn(WL) , of chicken were maintained in the laboratory of Yeungnam University. Economic traits (body weight, sexual maturity, hen-day egg production, egg weight) and phenotypic characteristics (body-type, head, feather, shank) were checked. The DNA fingerprinting was analyzed for both breeds. The growth rate of the KNC was similar to WLS and sexual maturity of the KNC came later than WL. Hen-day egg production of the KNC was also slightly lower than the WL. The egg weight was about 10g lighter than WL. There was no difference in body weight of female KNC compared to the WL after 28 weeks. The study confirms difference between KNC and WL in DNA fingerprinting as well as its outlook. Thus, we suggest that these should be tested in nationwide districts about chickens known as the KNC using DNA fingerprinting. Then, the confirmed KNC populations should be maintained and used for the genetic improvement. Finally, only confirmed KNC should be in market which induce consumer to seek the KNC by its favorite.

Application of Breed-specific DNA Markers for the use of Identifying Major Pure Pig Breeds Maintained in Korea (대한민국내 주요 돼지 품종의 순종 식별을 위한 품종특이 DNA marker의 활용)

  • This study was conducted for the identification of pure Landrace, Large White and Duroc breeds which are mainly maintained in Korea using DNA markers. We used known KIT and MC1R mutations, which were related coat color in pigs, and pig mitochondrial DNA variations. The KIT mutation was used to distinguish white and colored animals. Duroc breed could be discriminated from other colored breeds using the MC1R mutation N121D. Discriminating Landrace and Large White was possible using the l l-bp duplication of D-Ioop region and alternative initiation codon of ND2. In conclusion, identification of Landrace, Large White and Duroc breeds was might be possible using the procedure designed in this study.

Current status and prospects to identify mutations responsible for mutant phenotypes by using NGS technology (NGS 기술 활용 돌연변이체 해석 및 연구현황)

  • Next-generation sequencing allows the identification of mutations responsible for mutant phenotypes by whole-genome resequencing and alignment to a reference genome. However, when the resequenced cultivar/line displays significant structural variation from the reference genome, mutations in the genome regions absent in the reference cannot be identified by simple alignment. In this review, we report the current status and prospects in identification of genes in mutant phenotypes, by using the methods MutMap, MutMap-Gap, and MutMap+. These methods delineate a candidate region harboring a mutation of interest, followed by de novo assembly, alignment, and identification of the mutation within genome gaps. These methods are likely to prove useful for cloning genes that exhibit significant structural variations, such as disease resistance genes of the nucleotide-binding site-leucine rich repeat (NBS-LRR) class.

Taxonomic Study on Cirsium Miller (Asteraceae) in Korea Based on External Morphology (외부형태형질에 의한 한국산 엉겅튀속(Cirsium Miller)의 분류학적 연구)

  • Patterns of morphological variation in Korean Cirsium were examined using numerical analysis, and the delimitation of taxa was evaluated. Principal components analysis of individuals of taxa using 61 morphological characters revealed the presence of two major groups. One group included C. lineare, C. vlassovianum, and C. setidens, the other contained C. pendulum, C. japonicum var. japonicum, C. japonicum var. spinosissimum, C. schantarense, C. nipponicum, and C. rhiniceros. Also, the result of cluster analysis recognized three sections, Onotrophe, Pseudo-eriolepis, and spanioptilon. Korean Cirsium was classified into three sections; C. japonicum var. japonicum, C. japonicum var. spinosissimum, C. schantarense, C. rhiniceros and C. nipponicum were belonged to sect. Onotrophe, C. setidens, C. vlassovianum and C. lineare were belonged to sect. Spanioptilon, and C. pendulum was belonged to sect. Pseudo-eriolepis. In conclusion, Korean Cirsium was classified into three sections, eight species, three varieties, and five forms.

Metabolic Discrimination of Papaya (Carica papaya L.) Leaves Depending on Growth Temperature Using Multivariate Analysis of FT-IR Spectroscopy Data (FT-IR 스펙트럼 다변량통계분석을 이용한 파파야(Carica papaya L.)의 생육온도 변화에 따른 대사체 수준 식별)

  • To determine whether FT-IR spectral analysis based on multivariate analysis for whole cell extracts can be used to discriminate papaya at metabolic level. FT-IR spectral data from leaves were analyzed by principal component analysis (PCA), partial least square discriminant analysis (PLS-DA) and hierarchical clustering analysis (HCA). FT-IR spectra confirmed typical spectral differences between the frequency regions of 1,700-1,500, 1,500-1,300 and 1,100-950 cm-1, respectively. These spectral regions were reflecting the quantitative and qualitative variations of amide I, II from amino acids and proteins (1,700-1,500 cm-1), phosphodiester groups from nucleic acid and phospholipid (1,500-1,300 cm-1) and carbohydrate compounds (1,100-950 cm-1). The result of PCA analysis showed that papaya leaves could be separated into clusters depending on different growth temperature. In this case, showed discrimination confirmed according to metabolite content of growth condition from papaya. And PLS-DA analysis also showed more clear discrimination pattern than PCA result. Furthermore, these metabolic discrimination systems could be applied for rapid selection and classification of useful papaya cultivars.

  • In polished barley of 22 varieties protein content was correlated significantly with MgO of $P_2O_5$ content (at p=0. 01), $K_2O$ (at p=0. 05), but it was not related to CaO. Dye binding capacity was correlated significantly (at p=0. 01) with MgO or $P_2O_5$. Thus MgO could be used for mass screening of protein content. Carbohydrate content showed insignificant negative correlation coefficients with above four minerals. There were significant positive correlation (at p=0. 01) among $P_2O_5$, $K_2O$ and MgO. The mean contents of $K_2O$, $P_2O_5$, MgO and CaO were 0. 03, 0. 29, 0. 084 and 0. 043%, respectively.
