Toxicity, Uptake, and Transformations of Atrazine by Hybrid Poplar Tree

  • 국내에서 자생하는 포플러중 현사시, 양황철, 이태리포플러를 대상으로 제초제인 atrazine의 제거특성을 조사하였다 Atrazine이 포플러에 미치는 독성을 평가하고, 식물체에 의한 atrazine 제거효율 및 제거속도를 조사하였다. 또한 식물체의 체내로의 흡수에 의해 atrazine이 제거되므로 식물체로 흡수된 후의 생물학적 변환 과정에 대해서도 조사하였다. 현사시, 양황철, 이태리포플러 모두 실험이 진행된 전범위의 농도에서 황백화현상, 낙엽증가, 생체량 감소와 같은 식물독성을 나타냈으며, 현사시인 경우 양황청 및 이태리 포플러에 비해 상대적으로 민감한 반응을 나타내었다. Atrazine 독성여부 평가의 하나로 초기 atrazine 농도가 증가할수록 현사시, 양황철, 이태리포플러의 증산량은 상대적으로 감소함을 관찰하였다. 포플러에 의한 atrazine 생물학적 변환실험에서는 atrazine이 중간생성물질인 HA, DEA, I)IA를 거쳐 ammeline을 포함한 HDAP로 전환되는 것을 확인하였다. 이 결과의 중요성은 atrazine 부산물은 상대적으로 독성이 적고, 미생물에 의해 어렵지 않게 완전 무기화할 수 있기 때문이다. 즉, 식물체내에서도 HBAP 이후 과정을 통한 분해가 이루어질 수 있는 가능성이 있다고 추측할 수 있다.

Selection of Poplar Clones for Short Rotation Coppice in a Riparian Area (수변지 단벌기 목재에너지림에 적합한 포플러 클론 선발)

  • This study aims to select the most appropriate poplar clones for planting on short rotation coppice poplar plantations in a riparian area. The research investigated biomass production, nitrogen and carbon absorption with 2-year-old poplar (Populus spp.) clones including P. euramericana clone Eco28, P. deltoides hybrid clone 97-18, and P. alba ${\times}$ P. glandulosa hybrid clone 72-30. The average number of stems per stump was five and P. euramericana clone Eco28 had the greatest average number of live stems per stump with 5.9. The average stem diameter was 23.2 mm, and P. deltoides hybrid clone 97-18 achieved the largest average diameter with 25.4 mm. The average annual above-ground biomass production of Populus deltoides hybrid clone 97-18 was 16.1 ton/ha/year, followed by P. alba ${\times}$ P. glandulosa hybrid clone 72-30 and P. euramericana clone Eco28, 12.3 and 5.4 ton/ha/year, respectively. The average annual nitrogen uptake of poplar clones was 46.5 kg/ha/year. P. alba ${\times}$ P. glandulosa hybrid clone 72-30 had the highest average, 63.1 kg/ha/year. The average of annual carbon absorption was estimated 5.3 ton/ha/year and Populus deltoides hybrid clone 97-18 showed the best results with 7.7 ton/ha/year. Based on the results given above, P. deltoides hybrid aspen clone 97-18 is considered as the most suitable poplar clones for wood biomass production on riparian areas.

Variation in Resistance of Poplar Clones to Melampsora Leaf Rust (Melampsora 잎녹병에 대한 포플러 클론의 저항성 변이)

  • This study was conducted to determine the susceptibility of 15 clones of native and foreign poplar species and their hybrids to Melampsora leaf rust and to select resistant clones at a nursery stool bed. Rust severities of individual trees were determined by estimating the percentage of infected leaves on the trees in addition to comparing the infected leaves with the infection diagrams. Three hybrids out of 15 clones were selected as resistant clones to the Melampsora leaf rust in Korea. Bong-wha 1 and Hyunsasi 3 belong to section Leuce, and Dorskamp belongs to section Aigeiros. Based on our results, we recommend Dorskamp as the best resistant clone to poplar leaf rust.

The Status and Prospect of Poplar Research in Korea (포플러 연구현황과 전망)

  • Populus species have been as a model species in tree breeding and we have enormous varieties resulting from the poplar breeding because of their fast growth performance and short rotation age. New varieties developed in Korea are common italian poplar(P euramericana, I-214, I-476), P euramericana“Eco 28”(Italian poplar No.1) and p. deltoides“Lux”(Italian poplar No.2), which were introduced from foreign countries. As hybrid polars, Hyun-Sasi(p. alba ${\times}$ P. glandulosa No.1, No.2, No.3, No4.), P. nigra x P. maximowiczii and P. koreana x P. nigra val. italica, were developed, and P. davidiana was selected as the result of selection breeding The total plantation areas covered with the new varieties are 935,162ha that include 745,773ha of P. euramericana, 184,636ha of P. alba x P. glandulosa, and other new varieties are 4,735ha. The new poplars are contributed to increase farmer's income as well as bare land tree-planting in Korea. The technologies associated with the poplar species were developed, such as the determination of optimum site for new the poplar species, the crossing method between incompatible poplar species, and the vegetative mass propagation. In the future, poplar species will be considered for phytoremediation species at contaminated areas such as landfill sites or with lives stock's waste water as well as wood production, a shade tree like road-side tree and public park tree.

Growth Response and Adaptability of Poplar Species Treated with Liquid Pig Manure (양돈분뇨 처리에 대한 포플러류의 생장반응 및 적응능력)

  • This study was conducted to analyze growth characteristics and adaptability of poplar clones under the treatment of liquid pig manure. The average of survival rate was 33% better at control than the treatment under age of 5 years. But, tree height and DBH growth were higher at the treatment than control. Populus euramericana 'Eco28' showed the highest survival rate (97.9%) under the treatment. P. euramericana 'Eco28' and P. deltoides hybrid 'Dorskamp' could be selected as superior clones for height and DBH growth under the liquid pig manure treatment. The above-ground biomass production was also investigated when the poplar clones were 5 years old. The aboveground biomass under the liquid pig manure treatment was, on average. 52.6 ton/ha, which was 80% higher than control (29.1 ton/ha). P. euramericana 'Eco28' (73.6 ton/ha) and P. deltoides hybrid 'Dorskamp' (71.1 ton/ha) showed superior biomass production than other clones at the treatment of liquid pig manure. Based on survival, growth and demage traits, the adaptability of poplar clones to liquid pig manure treatment was estimated. P. deltoides hybrid 'Dorskamp' and P. euramericana 'Eco28' showed better adaptability to the treatment. P. nigra ${\times}$ P. maximowiczii '62-10' and P. koreana ${\times}$ P. nigra var. italic 'Suwon' were identified as poor adaptability clones.

The Cd and Pb Accumulation in Various Tissues of Rooted Cuttings of Four Populus Species Inoculated with Ectomycorrhizal Fungi, Pisolithus tinctorius (모래밭버섯 균근균(菌根菌)으로 접종(接種)한 포플러 4개(個)개 수종(樹種) 삽목묘(揷木苗)의 체내(體內) 부위별(部位別) Cd과 Pb 축적(蓄積) 특성(特性))

  • The objectives of this study were to understand the characteristics of Cd and Pb accumulation in various tissues of poplar species and the effects of ectomycorrhizal inoculation on the accumulation of above two heavy metals in the tissues. The mycelial inoculum of ectomycorrizal fungus, Pisolithus tinctorius was produced in peatmoss and vermiculite mixtures, and inoculated into potted soil with fresh cuttings of four species of Populus, P. alba ${\times}$ glandulosa (Pag). P. koreana ${\times}$ nigra var. italica (Pkn), P. nigra ${\times}$ maximowiczii(Pnm), and P. euramericana(Pe). The potted soils were added with 0, 30, and 80 ppm Cd, and 0, 50, and 300 ppm Pb. The cuttings were grown outdoors for about five months until the plants were harvested for measurement of growth, mycorrhizal infection, and metal contents in leaves, stems, and roots. The total dry weight of Pe treated with Cd and Pb was increased by mycorrhizal inoculation, while that of three other species was not affected by the inoculation. Cd was accumulated in the highest concentration in Pag and its concentration was increased by four times by mycorrhizal inoculation. The Pag accumulated Cd in the highest concentration in the leaves, while three other species accumulated Cd most in the roots. Pb was accumulated in the highest concentration in the roots of all the four species, while Pkn accumulated Pb in the leaves as much as in the roots. Without mycorrhizal inoculation Pe accumulated Pb most among the four species, while Pkn with mycorrhizal inoculation accumulated Pb two times more than in the same species without mycorrhizal inoculation. It was concluded that Pag was the most effective species among the four poplar species in Cd absorption from contaminated soil, and that Pe instead effectively absorbed Pb. Mycorrhizal inoculation increased the Cd accumulation in the tissues by four times in Pag and also increased Pb accumulation by two times in Pkn, with leaves being the major sites of metal accumulation. It may be possible to use these two poplar species in remedying the metal in the soil through the raking and removing the litter out of the contaminate site.

  • Protoplasts isolated from leaf mesophyll tissues of Populus koreana ${\times}$ P. nigra var. italica were fused with those of P. euramericana cv. Guardi. Well expended healthy leaves of 5 to 7 week-old-plantlet grown in vitro were used as source materials. Leaves from P. koreana ${\times}$ P. nigra var. italica and P. euramericana cv. Guardi were digested in enzyme solution I (2.0% Cellulase, 1.2% Hemicellulase, 0.4% Macrozyme, 2.0% Driselase, 0.05% Pectolyase ; w/v) and enzyme solution II (1.0% Cellulase, 1.2% Hemicellulase, 0.4% Macrozyme, 2.0% Driselase, 0.05% Pectolyase ; w/v), respectively, The highest frequency of fusion among the protoplasts originated from the two source materials was approximately 21% using 40% PEG or 15% dextran. In addition, fusion frequency was enhanced by incorporating 30mM of $Ca^{2+}$ in eluting solution at pH 10.5. Dividing cells and/or mint-calli were obtained by culturing the fusion products in a liquid 8p-KM medium supplemented with 0.6M sucrose, $0.45{\mu}M$ 2, 4-D, and $0.5{\mu}M$ BA. Shoots were regenerated from the fusion product-derived calli after culture on MS medium containing $5.0{\mu}M$ zeatin. To verify the putative hybrid or cybrid, SDS-PAGE was carried out. From the 24 regenerants, just two plants showed intermediate protein band patterns compared with those of the original source plants.

Studies on the development of mushroom media for bottle culture in new Pleurotus ostreatus 'Miso' (신품종 '미소' 느타리버섯 병재배 배지개발)

  • This study was carried out to develop the best medium of new oyster mushroom for bottle culture. The new oyster mushroom cv. Miso is a Pleurotus ostreatus developed at the Chungnam Agricultural Research & Extension Services. For the bottle culture media, poplar sawdust+beet pulp+cottonseed meal (5:3:2), poplar sawdust+wheat husk meal (8:2), poplar sawdust+beet pulp+cottonseed meal (6:2:2), poplar sawdust+beet pulp+cottonseed meal (4:4:2), and poplar sawdust+beet pulp+wheat husk meal (7:1:2) were used in 850cc PP bottle. The pH was 5~6 and the C/N ratio 19.7~28.3 in bottle culture media. The time of pinhead formation was 5 to 6 days. For the fruiting body formation after inoculation took 29~31 days. The yield of fruiting body of poplar sawdust+beet pulp+wheat husk meal (7:1:2) medium was the highest at 110.4g/bottle compared to other media. Therefore, such cultivation medium would be appropriate for the commercial production of bottle culture in the new oyster mushroom 'Miso'.

Sapflow of Two Poplars under Swine wastewater Irrigation (양돈폐수 처리할 때 포플러의 수액이동)

  • We examined sapflow of two populus tree species (Populus alba ${\times}$ Populus glandulosa and Populus euramericana) grown under Swine wastewater treatment, and measured relationship between sapflow and climate facters. Sapflow of P. euramericana was 1.5 times greater than that of P. alba ${\times}$ P. glandulosa from September to late December 2007. However, sapflow of P. euramericana and P. alba ${\times}$ P. glandulosa was similar after late January. The relationship between the mean temperature and sapflow was relatively high. There was a decrease in sapflow tendency of the two poplar species during the late October when the mean temperature was low. Trends on relationship between the relative humidity and sapflow showed low relationship. However, data showed weak relationship between sapflow and precipitation. During the period of less precipitation, sapflow of P. alba ${\times}$ P. glandulosa has no relationship. Sapflow of two populus species was still active during non-growing season. P. euramericana was considered as a better species than P. alba ${\times}$ P. glandulosa, in terms of phytoremediation.

  • The resistance of a non-transgenic poplar clone, 'Ogy' and three transgenic poplar lines to the cottonwood leaf beetle, Chrysomela scripta F., was evaluated by in vitro feeding. The lines were transformed with neomycin phosphotransferase II(NPT II) as a selectable marker, proteinase inhibitor II(pin2) as a resistance gene, and CaMV 35S as a promoter. An efficient method of sterilizing the beetle eggs and introducing them into plant tissue cultures was developed. The resistance of the transgenic lines was investigated in terms of effects tin leaf area consumed, insect weight, insect developmental stages, and plantlet root dry weight after feeding. Also, leaf area consumed was examined by leaf age as measured through leaf plastochron index(LPI). The leaf area consumed and insect weight were highly significant between transformants and control, and insect development in vitro was significant among the transgenic lines. Larval infestation was the most severe around LPI 4 to 5 which were young leaves. The system provided a quick, highly controlled method to screen developing transgenic plantlets directly.
