The purpose of this study was to investigate the effects of black garlic supplementation and exercise on TBARS, HSP 70, and COX-2 expression after high-intensity exercise in rats. Twenty-four male Sprague-Dawley rats (4 weeks) were used in this study. Experimental groups were a control group (A, n=6), black garlic supplementation group (B, n=6), exercise group (C, n=6), and black garlic supplementation with exercise group (D, n=6). High-intensity exercise was performed by treadmill running (every 10 min speed increased gradually; 15~24 m/min, $0^{\circ}$ grade) and 2.86 g/kg black garlic for 4 weeks. The findings of this study were as follows: TBARS levels were significantly lower in the D group compared to the other group and B and C groups were significantly lower in TBARS levels than in the A group. HSP 70 expression was lower in the B group compared to the other groups. COX-2 expression has showed a similar expression in all groups. Therefore, it is considered that black garlic intake and exercise may reduce oxidative stress.
The purpose of this study was to examine how combined exercise and acaiberry intake make effects on insulin and glycated hemoglobin of middle-aged women. The middle-aged women in their 40s and 50s were classified into three groups - group A which only had acaiberry intake, group B which both had acaiberry intake and combined exercise, and group C which only underwent the combined exercise. The combined exercise was held three times a week for 8 weeks, each for 60 minutes including warming up and cooling down. The aerobic exercise was carried out for 20 minutes with a treadmill walk with HRmax 50-60% and the resistance movement was carried out for 20 minutes with a strength of 50-60% based on 1RM. The participants were told to have the acaiberry drink before breakfast and dinner, by melting 5g of acaiberry powder to water. 2-way RGRM ANOVA was carried out to process the data for comparing each groups. In conclusion, the 8-weeks of combined exercise and acaiberry ingestion therapy did not improve the insulin and glycated hemoglobin. It is more likely to show clear changes in both elements with longer treatments and controlling the amount of ingestion and exercise intensity.
Purpose: This study aimed to investigate the effect of shoes sole form on fatigue of lower extremity during treadmill gait. Method: Thirty healthy young adults (15 males and 15 females) were recruited. They performed treadmill gait in two different conditions: double sole(DS) and flat sole(FS). Result: lower extremity fatigue were signficantly decreased in double sole condition(p<.05). Conclusion: These findings suggest that double sole contributes to reduction of lower extremity muscle fatigue.
The present research was conducted to investigate the effect of moderate exercise training on splenocyte inflammatory cytokines production in high fat diet-induced obese mice. To determine the effects of exercise training and low fat diet on splenocyte, C57BL/6 male mice (4 weeks aged, n=20) were fed a high fat diet (45% fat diet) for 5 weeks so that obesity was achieved intentionally. These obese mice were then divided into 2 groups; HLC (low fat diet and control n=10), and HLE (low fat diet and exercise n=10). HLE mice performed 8 weeks of exercise training on a motorized treadmill by running for 30-60 min/day at 10-22 m/min, 0% grade, five times per week. After exercise training, all the splenocyte was collected and Con A (concanvalin A, 10${\mu}g/ml$) was used to stimulate the cells, after which IL-1${\beta}$, IL-6, TNF-${\alpha}$ were measured by bio plex. Independent t-test was used and p value under 0.05 was considered a statistical significance. Splenocyte IL-1${\beta}$, IL-6, TNF-${\alpha}$ production of HLE stimulated by Con A was significantly lower than that of HLC (p<0.01). These findings suggest that moderate exercise has beneficial effects on splenocyte inflammatory cytokines in high fat diet induced obese mice.
The purpose of this study was to find out the Bcl-2 (B-cell leukemia/lymphoma-2), Bax, and caspase-3(cysteine-aspartic proteases-3) protein expression in soleus and EDL muscle according to treadmill exercise intensity in 60 week-old SD rats. The SD rats were randomly divided into four groups (n=10 in each group): control (CON), low intensity exercise (LE), moderate intensity exercise (ME), and high intensity exercise (HE). The exercise was given to the rats for 8 wk, 5 day/wk. The animals underwent treadmill exercise at intensities of 30 min at 8 m/min for the LE group, 15 min at 16 m/min for the ME group, and 9 min at 24 m/min for the HE group. The results were as follows: the expression of Bcl-2 protein was lowest in the HE group and the expression of Bax protein was highest in the HE group. The expression of caspase-3 (cleaved form) protein was observed in the HE group. For the different types of muscle fiber, Bcl-2 protein expression in the soleus muscle was decreased in all groups. Bax protein expression in the soleus muscle was increased in the HE group only. Bcl-2 protein expression in the EDL muscle was decreased in the HE group, and Bax protein expression in the EDL muscle was increased in the ME and HE groups. Consequently, the protein expression related to the aged rats shows a difference according to the intensity of exercise. In addition, caspase-3 protein expression appeared in the HE group; however, in all amounts of intensity, DNA fragmentation was not observed. Therefore, apoptosis on skeletal muscles of aged mice can be intervened with optimal exercise. On the other hand, high intensity exercise can potentially accelerate the apoptosis of muscle fiber in aged rats.
To achieve the purpose of this study, forty-eight male Sprague-Dawley rats were assigned to control and three endurance exercise group. 36 rats were forced to exercise according to exercise intensity for 8 weeks and 12 rats were untrained for control group. Cardiac cytosol was extracted from cardiac tissue and cardiac mitochondria was purified from the cytosol. Purified mitochondria were separated into four fraction: inner membrane, outer membrane inter membrane space and matrix. The changes of cytosolic and mitochondrial LDH isozymes activity were measure. Relative activity $(\%)$ of cytosol for low and control group showed the following order of prevalence $AB_3>A_2B_2>B_4>A_3B>A_4$ for moderate and high group : $AB_3>B_4>A_2B_2>A_3B>A_4$. Outer membrane for low group showed $AB_3>B_4>A_2B_2$, for moderate group:$ B_4>AB_3>A_2B_2$, for high and control group: $B_4>A_3B$. Inter membrane space for low, moderate and high group showed $B_4>AB_3>A_2B_2>A_3B>A_4$, for control group: $B_4>A_3B>AB_3>A_2B_2>A_4$. Inner membrane for all group showed $B_4>AB_3>A_2B_2>A_3B>A_4$. Matrix for control, low, moderate and high group showed $B_4>AB_3>A_2B_2>A_3B>A_4$. These results suggest that long term exercise intensity effect on cardiac tissue cytosolic and mitochondrial activity and $A_4,\;A_3B,\;A_2B_2,\;AB_3\;and\;B_4$ isozymes were found entirely in mitochondrial fraction.
The purpose of this study was to identify effects of walking speed and a cognitive task during treadmill walking on gait variability. Experiments consisted of 5 different walking speeds(80%, 90%, 100%, 110% and 120% of preferred walking speed) with/without a cognitive task. 3D motion analysis system was used to measure subject's kinematic data. Temporal/spatial variables were selected for this study; stride time, stance time, swing time, step time, double support time, stride length, step length and step width. Two parameters were used to compare stride-to-stride variability with/without cognitive task. One is the coefficient of variance which is used to describe the amount of variability. The other is the detrended fluctuation analysis which is used to infer self-similarity from fluctuation of aspects. Results showed that cognitive task may influence stride-to-stride variability during treadmill walking. Further study is necessary to clarify this result.
The purpose of this study was to estimate the energy expenditure simply and practically during physical activities. The physical activity is quantified by the integration of the accelerometer signals obtained from the triaxial accelerometer attached at the waist level of the human body. To find a relationship between energy expenditure and accelerometer data, 6 male and 5 female subjects walked and ran on the treadmill with speeds of 1.5, 3.0, 4.5, 6.0, 6.5, 7.0, and 8.5 km/hr. Each subject performed walking at the speed lower than 6.0 km/hr and running at the speed higher than 6.5 km/hr. Actual energy expenditure was determined by a continuous direct gas analyzer. Two predictive equations of walking and running mode for energy expenditure which includes gender, body mass index(BMI) and data from accelerometer were developed using multiple regression analysis. The correlation coefficients and coefficients of determination between the estimated and measured energy expenditure were R=0.936, R2=0.876 and R=0.881, R2=0.776 in walking and running mode, respectively. For further study, experiments on a larger scale of test subjects are essential for acquiring more reliable results.
The aim of this study was to investigate the effects of exercise on intrinsic and extrinsic apoptosis signaling pathways in skeletal muscle. ICR-type white male mice were divided into a control group (CON: n=10) and an exercise training group (EX: n=10) after a 1 week adaptation period. EX performed treadmill running at 16.4 m/min with a 4% incline, 40 min/day and 5 days/week for 8 weeks. Cervical dislocation was performed at 48 hours after the last bout of exercise, after which gastrocnemius skeletal muscles were immediately collected. The results of verifying the intrinsic apoptosis pathway showed that there were no significant differences in Bcl-2, Bax, or the ratio of Bax/Bcl-2 proteins between EX and CON. On the other hand, the results of verifying the extrinsic apoptosis pathway showed that caspase-8 proteins were significantly lower in EX than in CON (p<0.05). Apoptosis suppressing protein HSP70 was higher in EX than in CON. In addition, caspase-3, which is the final factor for apoptosis, was not activated. These results indicate that apoptosis did not develop since caspase-3 is non-cleaved by the effects of caspase-8 and HSP70 extrinsic pathways rather than Bcl-2 and Bax intrinsic pathways among signal pathways for apoptosis.
The purpose of this study was investigate the effects of short-term exercise and capsaicin supplementation on serum lipids, antioxidants and Akt contents in diabetic rats whose symptom was induced through the use of 50 mg/kg streptozotocin, 31 white rats were experimented. Rats in one exercise group and another exercise-capsaicin supplementation group exercised on a treadmill maintained by a 15-min run at $0^{\circ}$ inclination five times for a week during two weeks one week after the diabetes was induced. 10 mg of capsaicin per kilogram of weight was orally administered to rats in the capsaicin supplementation group after the diabetes was induced. The serum lipids were analyzed through the use of blood obtained from the incision of the abdomen, and antioxidants and Akt contents were analyzed through the use of livers to reach following conclusions. As for the serum lipids, TG significantly decreased in group DCE and HDL-C was higher than in other groups. ROS significantly decreased in all groups except for group D. CAT significantly increased in group DC, SOD was significantly high in groups DE and GSH was significantly high in all groups except for D. Akt contents significantly increased in group DC and p-Akt in group DE. As a result of this study, it has been found that short-term exercise and the capsaicin supplementation had a significantly effect on the serum lipids, antioxidants and Akt contents.
