Anti-allergic Effect of Zizania latifolia Turcz Extracts (고장초의 항알레르기 효과)

  • The anti-allergic effect of the methanol extract of Zizania latifolia Turcz was investigated. Methanol extract of Z. latifolia Trucz did not display any cytotoxicity activities in the IgE-sensitized RBL-2H3 cells. The extract was found to inhibit compound 48/80-induced degranulation and antigen-induced $\beta$-hexosaminidase release as well as PMA plus A23187-induced TNF-$\alpha$ production in RBL-2H3 mast cells in a dose-dependent manner. Release of $\beta$-hexosaminidase, a marker for the release of histamine in mast cells, was inhibited with an $IC_{50}$ value of $73.8\;{\mu}g/mL$. These results suggest that Z. latifolia extracts may be useful for the prevention of type I allergic reaction.

Inhibitors of Antigen-induced Degranulation of RBL-2H3 Cells Isolated from Amomum tsao-ko (초과(草果)의 RBL-2H3 세포 항원 유도 탈과립 억제성분)

  • Bioactivity-guided fractionation of EtOH extract of the dried fruits of Amomum tsao-ko led to isolation of three compounds (1-3). Their structures were elucidated by spectroscopic methods (MS, 1D and 2D-NMR) and comparison with literature values, as naringenin-5-O-methyl ether (1), helichrysetin (2), and cardamomin (3). Compound 2 was obtained from the genus Amomum for the first time. Among them, compounds 2 and 3 inhibited on the release of β-hexosaminidase from RBL-2H3 cells, with 99.1 and 21.3% at the concentration of 50 μM, respectively.

Ethyl Acetate Extract of Korean Rice Wine Lees Inhibits IgE-Mediated Degranulation in Rat Basophilic Leukemia RBL-2H3 Cells and Passive Cutaneous Anaphylaxis in Mice (주박 에틸아세테이트 추출물의 항알러지 효과)

  • Mast cells, the central effector cells involved in the allergic response, release histamine, arachidonic acid, and proinflammatory cytokines. We investigated the effect of the ethyl acetate fraction (EA), derived from Korean rice wine lees, on RBL-2H3 cell degranulation and passive cutaneous anaphylaxis in an animal model. The EA fraction suppressed the release of beta-hexosaminidase, a marker of degranulation, and the mRNA expression of interleukin-3 (IL-3) and IL-13. EA also successfully suppressed the passive cutaneous anaphylaxis (PCA) reaction in mice in a dose-dependent manner. These results suggest that EA can inhibit mast cell degranulation through the inhibition of IL-3 and IL-13 mRNA expression, and that EA may potentially serve as an anti-allergic agent.

Inhibitory effect of the aqueous extract of a tetraploid 'etteum' variety in Platycodon grandiflorum on degranulation and inflammatory mediator release in RBL-2H3 cells (으뜸도라지추출물이 RBL-2H3 세포에서 탈과립과 염증매개물질의 분비 억제에 미치는 영향)

  • Purpose: Platycodon grandiflorum (a domestic diploid variety, DV-PG) has been used as a food and component of various traditional oriental medicines. Although DV-PG is known to have an anti-allergic effect, little is known about the beneficial health effects of the tetraploid 'Etteum' variety in the Platycodon grandiflorum (TV-PG), which is a recently developed variety. In this study, we investigated the effect of TV-PG on the rat basophilic leukemia mast cell (RBL-2H3)-mediated allergic response. Methods: To examine the effects of TV-PG on the allergic response, RBL-2H3 cells were sensitized with dinitropheny (DNP)-immunoglobin E, treated with various concentrations of TV-PG, and challenged with DNP-human serum albumin. We estimated cell granulation by measuring the release of ${\beta}$-hexosaminidase and production of inflammatory mediators by ELISA. Results: TV-PG had no effect on the proliferation or cytotoxicity of RBL-2H3 cells within the concentration range of 0 to $200{\mu}g/mL$. TV-PG inhibited degranulation of RBL-2H3 cells by antigen stimulation in a dose-dependent manner. TV-PG also suppressed the production of inflammatory cytokines and mediators such as interleukin-4, tumor necrosis $factor-{\alpha}$, prostagladin E2, and leukotriene B4 in RBL-2H3 cells by antigen stimulation. Conclusion: These results indicate that TV-PG exhibits anti-allergic activity via inhibition of degranulation as well as suppression of inflammatory mediators and cytokine release. These findings suggest that TV-PG may have potential as a preventive and therapeutic agent for the treatment of various allergic diseases.

Inhibitory Effects of Several Korean Traditional Herbs on Anaphylactic Reaction and Mast Cell Activation (아나필락시스 반응과 비만세포 활성화에 대한 한국전통약제의 억제효과)

  • Korean traditional herbs, especially Anemarrhena asphodeloides (A. asphodeloides), Salvia miltiorrhiza (S. miltiorrhiza), and Terminalia chebula (T. chebula) have been known to have the immunomodulatory, anti-tumor, and anti-inflammatory effects. However, direct cellular mechanism underlying the mast cell-mediated anaphylaxis-like reaction has poorly been understood. In the present study, the effects of the methanol extracts of A. asphodeloides (MEAA), S. miltiorrhiza (MESM), and T. chebula (METC) on anaphylactic reaction were investigated. Using in vitro and in vivo experiments, the influences of MEAA, MESM, and METC on the compound 48/80-induced anaphylaxis-like reaction and mast cell activation, and IgEmediated PCA were examined. Results are below; 1) MEAA, MESM, and METC significantly inhibited systemic anaphylaxis-like reaction, ear swelling response, and IgE-mediated PCA. 2) the compound 48/80-induced mast cell degranulation, histamine release of rat peritoneal mast cells (RPMC) were significantly inhibited by the pretreatment with MEAA, MESM, and METC, and 3) the compound 48/80-induced calcium influx in RPMC was significantly inhibited by the pretreatment with MEAA, MESM, and METC. These results suggest that MEAA, MESM, and METC may be an activity to inhibit the compound 48/80-induced or anti-DNP IgE-mediated anaphylactic reactions by blocking histamine release and calcium uptake into RPMC. MEAA, MESM, and METC potentially may serve as an effective therapeutic tool for allergic diseases.

The Implication of Bandura's Vicarious Reinforcement in Observational Learning for Christian Education (관찰학습에서의 반두라 대리강화에 대한 기독교교육적 함의)

  • This study reviews Bandura's vicarious reinforcement in observational learning process and implies this concept into Christian education in terms of spiritual role modeling. The first part of this study answers three questions: "what is vicarious reinforcement?" "how does vicarious reinforcement take place in observational learning?" and "how does vicarious reinforcement affect observer's behavior change?" Bandura conceptualizes the learning process with observational learning and imitative or non-imitative performance. Based on this concept, Bandura define the roles of vicarious reinforcement in the four steps of observational learning process: attention, retention, motor reproduction, and motivational process. Also, the three effects of vicarious reinforcements are explained in the following categories: the observational learning effect, inhibitory or disinhibitory effects, and eliciting effect. Adapting the structure of observational learning theory in terms of the effect of vicarious reinforcement and the function of role models, the second part of this study examines the biblical concept of imitation of Christ and the modeling strategy of discipleship. Especially Paul's spiritual role model serves as positive vicarious reinforcement for the Christian believers to perform the desired behaviors. Also, Paul's condemnation serves as explicit negative vicarious reinforcement. Then, the last part of this study covers the implication of these findings from observational learning and empirical studies in terms of spiritual role modeling to Christian education.

  • The effects of adenosine and $N^6-cyclopentyladenosine$ (CPA) on superoxide production, myeloperoxidase release and $Ca^{2+}$ mobilization stimulated by fMLP in neutrophils were investigated. The effects were also observed on the stimulatory actions of C5a and PMA and the responses in lipopolysaccharide-primed neutrophils. In addition, the involvement of cAMP in the inhibitory action of adenosine was examined. The fMLP-stimulated neutrophil respiratory burst, degranulation and intracellular $Ca^{2+}$ mobilization may be regulated by activation of adenosine receptors. Adenosine may not affect the stimulated neutrophil responses due to activation of protein kinase C. fMLP-stimulated respiratory burst in lipopolysaccharide-primed neutrophils may be less sensitive to adenosine, compared with nonprimed cells. The inhibitory effect of theophylline in the presence of adenosine on neutrophil responses appears to be ascribed to accumulation of intracellular cAMP.

In vitro Study on the Functionality in Digestive Tract of Chitin and Chitosan from Crab Shell (게껍질 Chitin 및 Chitosan의 소화관내 기능성에 관한 in vitro 연구)

  • Chitin and chitosan samples prepared from crab's shells under different conditions were compared for their physicochemical properties; and functionality in gastrointestinal tract by in vitro test. Their bulk density was in the range of $127{\sim}208\;mg/ml$, and their viscosity was $80{\sim}581\;cP$ in 0.1 chitin and $80{\sim}3,670\;cP$ in 0.5% chitosan solution, showing a wide variation. The degree of deacetylation in chitosan samples as determined by IR spectral analysis was relatively high, showing $81{\sim}93%$. At the same alkali concentration and reaction temperature, a longer reaction period gave an increased degree of deacetylation and lower viscosity. The water holding capacity of chitic substance became greater at higher soaking temperature; chitosan D at $37^{\circ}C$ showed the greatest value. Chitic substance with lower bulk density showed the higher water holding capacity. The retardation effect toward glucose absorption was higher in critic substances of lower density and higher water holding capacity; chitosan D showed the highest value of 38%. The retardation index toward bile acid absorption after 1 hour dialysis was $15{\sim}34%$ in chitic substances, 39% in pectin and 9% in cellulose. The retarding effect showed the highest value of 34% in chitosan D at 3% concentration.

Anti-allergic Effect of Graviola Leaf Extract in Human Mast Cells (인간 비만세포에서 그라비올라 잎 추출물의 항알러지 효과)

  • This study is aimed at determining whether graviola (Annona muricata) beneficially influences immunoglobulin (Ig) E-mediated allergic reactions in human mast cells (LUVA cells). To examine the anti-allergic effect of graviola leaf extract (GLE), we treated antigen/IgE sensitized mast cells with extracts of various concentrations. In this study, the results of the in vitro model of IgE-mediated mast cell degranulation showed that GLE significantly inhibited the release of histamine, β-hexosaminidase, TNF-α, IL-4 and IL-6 in LUVA cells. Pretreatment with GLE suppressed the phosphorylation of antigen-induced Lyn and Syk, thus suppressing the downstream MAPKs pathways. The above results indicate GLE could suppress mast cell activation and allergic responses. Accordingly, it can be supported that GLE has the potential to be used as functional cosmetic material.
