Electrochemical determination of hydrogen peroxide using carbon paste biosensor bound with butadiene rubber (부타디엔 고무로 결합된 탄소반죽 바이오센서를 이용한 과산화수소의 전기화학적 정량)

  • When polybutadiene dissolved in toluene was a binder of carbon powder, the volatility of solvent just after electrode fabrication assured the mechanical solidity of the carbon paste electrode. This characteristic met the qualifications for practical use of carbon paste electrodes. A new enzyme electrode bound with butadiene rubber was constructed. In order to confirm whether it shows quantitative electrochemical behaviors or not, its electrochemical kinetic parameters, e.g. the symmetry factor, the exchange current density, the capacitance of double layer, the time constant, the maximum current, the Michaelis constant and other factors were investigated. These experimental facts showed that butadiene rubber is a recommendable binder for practical use of a carbon paste electrode.

  • Even though the carbon paste electrode bound with mineral oil is useful for research about the characteristics of enzymes, it remains far from practical uses because the lack of mechanical hardness limits its practical use. When the rubber liquefied in toluene was used as a binder of carbon powder in lab, it is confirmed that the mechanical robustness of the electrode is guaranteed. In order to confirm whether it shows quantitative electrochemical behaviors or not, its kinetic parameters, e.g. the symmetry factor (${\alpha}=0.28$), the exchange current density ($i_0=4.06{\mu}A/cm^2$), the capacity of the double layer ($C_d=2.11{\times}10^{-3}F$), the Michaelis constant ($K_M=2.45{\times}10^{-3}M$), and the time constant (${\tau}_B=0.077sec$) were investigated. Our experimental observations prove that the chloroprene rubber is a promising binder for the practical use of a carbon paste electrode.

  • A strip sensor based on $PbO_2$/carbon paste electrode was prepared by a screen-printing method, and employed to electrochemically determine the concentration of fruit sweeteners(i.e. glucose, sucrose, and fructose). The $PbO_2$/carbon paste electrode could monitor electrocatalytic oxidation of organic compounds such as carbohydrates, and measure the levels of natural sweeteners without enzyme. Severe interference from ascorbic acid was effectively reduced by modifying the electrode surface with a Nafion membrane. The response level of the Nafion/$PbO_2$/carbon paste electrode increased in the order of fructose, sucrose, and glucose, which corresponds to the order of sweetness perceived by humans.

  • An HRP immobilized carbon paste electrode, which was bound by EPDM, was newly fabricated and its electrochemical properties were investigated for the purpose of validating the new possibility for the practical use of biosensor. In the experimental range of substrate concentration, Lineweaver-Burk plot of the signal showed a good linearity. This means that HRP was embedded effectively to preserve its identity in the bulk of composite electrode materials and EPDM is a recommendatory binder. When the electrode was run at low operating potential($0.0\sim-1.0$ V vs. Ag/AgCl), it showed a high sensitivity and a good reproducibility. Especially the mechanical stability of the dried rubber was a remarkable breakthrough to get over a difficulty in putting the carbon-paste electrode bound with silicon oil to real use.

  • All array of carbon paste-based electrodes was prepared by screen printing the carbon paste modified with metal oxides $(TiO_2,\;RuO_2,\;PbO_2,\;Ni(OH)_2)$ and Prussian blue (PB). An electronic tongue system was assembled with the carbon-paste electrode array, and applied to discriminate the tastes of various commercial beverages and foods by measuring the chronoamperometric responses to the samples diluted in 0.1M carbonate buffer (pH 9.6). The results were analyzed with principal component analysis(PCA) method and plotted on two dimensional PCA coordinate; it was apparent that the amperometric electronic tongue system could discriminate the types of various foods and beverages.

  • A biosensor including the homogenized tissue of mugwort embedded in carbon paste, which senses hydrogen peroxide, was constructed and its electrochemical properties were validated using voltammetry. The good linearity of Hanes-Woolf plot implied that the reduction reaction of substrate was catalyzed by mugwort peroxidase at the electrode surface. Also the small value of symmetry factor, 0.28, indicated that electrochemical kinetics of the sensor is very sensitive to the change of electrode potential. Many experimental results collected above proved that the dissociation of hydrogen peroxide is dependent on the catalytic power of mugwort peroxidase qualitatively and quantitatively at the surface of the mugwort electrode. It is our firm belief that the marketed HRP can be replaced with mugwort tissue.

  • An activated carbon paste electrode was modified with the N-Hydroxysuccinimide(NHS) layer and applied to determine the dopamine in the presence of an excess ascorbic acid using square-wave voltammetry. The electrochemical properties of the modified electrode were examined in the solution containing dopamine/ascorbic acid using cyclic voltammetry(CV): the separation between the oxidation peaks of dopamine and ascorbic acid was largely dependent on the pH of the sample solution and became maximum at pH 4.0. Hence, the square-wave voltammetric determination of dopamine was carried out in a pH 4.0, 100mM phosphate buffer saline(PBS) containing 140mM NaCl. The detection limit and response slop were improved from $1.0{\mu}M\;to\;5.0\times10^{-2}{\mu}M\;and\;from\;0.93{\mu}A/{\mu}M\;to\;6.1{\mu}A/{\mu}M$, respectively, upon modification of the electrode surface by NHS.

Spinach Root-Tissue Based Amperometric Biosensor for the Determination of Hydrogen Peroxide (시금치 뿌리 조직 바이오센서를 이용한 과산화수소의 정량)

  • The response characteristics of the bioelectrode developed by the co-immobilization of spinach root tissue and ferrocene in a carbon paste matrix for the amperometric determination of hydrogen peroxide were evaluated. In the range of electrode potential examined (-0.3~0.0V vs. Ag/AgCl). the response time was relatively short ($t_{95%}=11.8$ sec) and it responded in the wide range of pH. Also, its detection limit was $2.25{\times}10^{-6}M$ (S/N=3) and a relative standard deviation of the measurements which were repeated 15 times using $1.0{\times}10^{-3}M$ hydrogen peroxide was 1.87%. The bioelectrode sensitivity decreased to 40% of the original value in 19 days of continuous use.
