Crystallization of Escherichia coli IciA Protein An Initiation of Chroirnsomal Replication (대장균 염색체 복제 개시 저해제, IciA 단백질의 결정화)

  • Specific binding to the oric region of E, coli chromsome by IciA protein inhibits initiation of chrorrnsomal replication in vitro by blocking the opening of this region effected by the initiator DnaA protein. The IciA protein has been suggested play a critical role in a key stage of the cell cycle. In order to study the structure-function relationship of IciA protein, we are determining the three-dimensional structure of IciA Votein by X-ray crystallography, As a first step toward its structure detumination E. coli IciA protein has been crystallized using sodium formate as a precipitant.

Preparation of Ag Nano-Powder from Aqueous Silver Solution through Reductive Precipitation Method (환원침전법을 이용한 수용액으로부터 은 나노분말의 제조 연구)

  • As one of the hydrometallurgical processes available in the recycling of silver-bearing wastes, the preparation of Ag nano-powder was investigated by a reductive precipitation reaction in silver solution using sodium formaldehydesulfoxylate and ascorbic acid as a reducing agent. Silver solution was prepared by dissolving silver nitrate with distilled water, and Tamol NN8906, PVP, SDS and caprylic acid were also used respectively as the dispersant to avoid the agglomeration of particles during the reductive reaction. Ag particles obtained from the reduction reaction from silver solution were characterized using the particle size analyzer and TEM to determine the particle size distribution and morphology. It was found that about $40\%$ excess of sodium formaldehydesulfoxylate was required to reduce completely silver ions in the solution. It alto appeared that the particle size generated with sodium formaldehydesulfoxylate was much greater than that with ascorbic acid. As far as the effect of dispersant on the Ag particles was concerned, the particle size distribution showed typically bimodal distribution in case of Tamol/FVP while very broad distribution ranged from 0.01 to $100{\mu}m$ appeared in case of SDS/caprylic acid.

  • Red mud is an industrial by-product produced during the manufacturing aluminum hydroxide ($Al(OH)_3$) and aluminum oxide($Al_2O_3$) from Bauxite ores. In Korea, approximately 2 tons of red mud in a sludge form with 50% moisture content is produced when 1ton of $Al_2O_3$ is produced through the Bayer process. In the paper, dispersion characteristics of liquefied red mud that does not require heating and grinding process for recycling were investigated through viscosity and sediment index. The results showed that the sediment index of liquefied red mud increased but viscosity of that decreased with a higher W/R ratio. Also we proposed the range of initial viscosity from 2000cP to 8000cP and target sedimentation index below 20% at elapsed time 180days for stable dispersion of liquefied red mud.

A Study on Highly Dispersed Pt/$Al2O_3$ Catalyst for Preferential CO Oxidation (고분산 담지된 Pt/$Al2O_3$ 촉매의 선택적 CO 산화반응 특성에 관한 연구)

  • 선택적 CO 산화반응(PrOx)에 사용되는 촉매 중 Pt, Ru, Rh 등의 귀금속 계 촉매들은 비귀금속 계 촉매에 비해 활성이 좋은 반면 가격이 비싸다는 경제적인 제한점이 있다. 따라서 소량의 귀금속을 사용하여 높은 활성의 촉매를 제조하고자 활성금속의 고분산 담지 방법에 대한 연구가 이루어지고 있다. 본 연구에서는 담체인 ${\gamma}-Al_2O_3$ 표면에 활성금속인 Pt의 고분산 담지를 위해 증착-침전법(Deposition-precipitation)을 적용하였으며 용액의 pH 변화에 따른 Pt 금속 입자의 분산도에 대한 영향을 살펴보았다. Pt의 함량은 1wt%로 고정하였고 침전제로 NaOH를 사용하여 용액의 pH를 pH 7.5 ~ 10.5로 변화시켰다. 제조된 촉매는 세척 후 $400^{\circ}C$, 3시간 소성 하였다. 제조된 1wt% Pt/$Al_2O_3$ 촉매의 특성분석을 위해 BET, TPR, CO-chemisorption을 수행하였다. PrOx 반응 실험은 GHSV=60,000 $ml/g_{cat}{\cdot}h$, $T=100{\sim}200^{\circ}C$, ${\lambda}$=4 조건에서 수행되었으며 반응 전에 촉매는 $400^{\circ}C$, 3시간 환원 후 사용하였다. 촉매의 특성분석과 PrOx 반응 실험 결과를 통해 촉매가 담체 위에 고분산 되는 최적의 pH를 확인할 수 있었으며, 기존의 함침법으로 제조된 촉매와 성능 비교를 통해 제조방법에 따른 영향을 살펴보았다.

Effect of Oyster Shell Addition on the Dissolved Air Flotation and Sedimentation of Bulking Sludge (팽화슬러지의 용존공기부상과 침전에 미치는 굴패각 첨가의 영향)

  • The objective of this study is to examine the effect of the waste oyster shell powder as the addition agent in bulking sludge thickening of paper manufacturing plant using DAF(Dissolved Air Flotation) and gravitational sedimentation. The effect of parameters such as dosage and size distribution of oyster shell were examined. The results showed that the optimum dosage of mixed oyster shell(size range : $\sim250{\mu}m$) was 0.8 g/L. The oyster shell addition of 5.0 g/L in sedimentation process was increased thickening concentration of 3.25 times. When 5.0 g/L of oyster shell was added in DAF process, water content of sludge was decreased from 95.5% to 82.7% in dewatering process using Buchner funnel test device. When size of oyster shell was divided four ranges($\sim53{\mu}m$, $53\sim106{\mu}m$, $106\sim150{\mu}m$, $150\sim250{\mu}m$), optimum size range for the flotation and dewatering was $53\sim106{\mu}m$.

  • The concentration and determination of trace phosphate ion was studied by $La(OH)_3$ coprecipitaiton. Phosphate ions in 1.0L samples were coprecipitated with lanthanium hydroxide at pH 9.5 adjusted with ammonia solution. The precipitates were floated with the aid of mixed surfactant(1:8 sodium oleate/sodium dodecyl sulfate) and nitrogen gas bubbles. The floated precipitate was collected in suction flask from the solution. The precipitate were washed with dil. ammonia solution and dissolved in sulfuric acid. The phosphate ion in the concentrated solution was finally determinated by UV/VIS spectrophotometry using the molybdenium blue method. The proposed method could be applied to the determination of phosphate ion in tap water and river water.

  • As a way of utilizing cheese whey to fortify sulfur-bearing amino acids to soybean protein, whey-soybean coagulum was made from whey-soy milk mixture and optimum conditions for coprecipitation of the two proteins were determined. Mixture of whey and soymilk in 1 : 1 volume ratio was coagulated at $0.005{\sim}0.5M$ of $CaCl_2$ concentration, pH $1.5{\sim}8.0$, and at $60{\sim}100^{\circ}C$, and absorbance at 500nm of filtrate from the coagulum was measured. Optimum conditions for the coprecipitation were 0.0125M of $CaCl_2$ concentration, pH $6.5\sim7.5$, and $70{\sim}80^{\circ}C$ which resulted in the minimum absorbance and also good physical properties of the curd.

  • Zirconia nanoparticles were synthesized by hydrothermal process, and experimental parameters such as reaction temperature, reaction time, kind and concentration of precipitator, kind of precursor were varied. Particle sizes and crystalline phases of each synthesized nanoparticles were analyzed with X-ray diffraction and FE-scanning electron microscope (SEM). The particle size and crystallization of zirconia increased with increasing concentration of precipitator. The growth rate of particle sizes when NaOH as a precipitator was used also increased more than that of KOH. Therefore, the use of KOH rather than NaOH was more effective in the control of particle sizes. An amorphous zirconia nanoparticle was found in 4 h of hydrothermal reaction, but the monoclinic zirconia nanoparticle was found in 8 h and over of hydrothermal reaction, and the width of nanoparticles was slightly slimmed and the length of nanoparticles was slightly extended with increasing reaction time. The smallest particle size was produced at the same synthesis condition when zirconium chloride among the precursors such as zirconium (IV) acetate, zirconium nitrate and zirconium chloride was used.

Preparation of Ultra Fine Ammonium Perchlorate Crystals (초미세 크기의 과염소산암모늄 결정체 제조)

  • The precipitation of ultra fine ammonium perchlorate(UFAP) crystals from the N-methyl pyrrolidone(NMP) solutions of ammonium perchlorate(AP) was studied. The characteristics of the precipitated crystals were investigated by means of scanning electron micrograph(SEM), X-ray diffraction(XRD), and thermogravimetric analysis(TGA). When chloroform, methylene chloride and toluene were the precipitants, the product crystals had a mean particle size less than 2$\mu\textrm{m}$. The crystallographical property and thermal decomposition behavior of the prepared UFAP were almost the same as those of the commercial AP.

The Preparation of PZT Fine Powder Using Ammonia Gas as a Precipitator (Ammonia gas를 침전제로 이용한 PZT 미분말의 합성)

  • In this study, the synthesis of PZT powder by bubble column reactor was investigated at various reaction conditions. As a result, the volume % of $NH_3$ gas used as a precipitator had no effect on the synthesis, but the more research is needed to control particle size.As a carrier gas, Ar, $O_2$ and air only increased the stirring effect but had no effect chemically on the synthesis. The calcination temperature of prepared PZT powder was about $500-600^{\circ}C$ and the meanparticle size of synthesized PZT powder was about $0.17{\mu}m$. The grain size of sintered body is about $0.5~3{\mu}m$ and this is similar with the value of commercial products.
