Research was initiated to investigate early establishment characteristics and germination pattern of fine-textured fescues (FF). Six varieties from Chewings fescue ($Festuca$$rubra$ L. ssp. $commutata$ Gaud., CF), creeping red fescue ($F.$$rubra$ L. ssp. $rubra$ Gaud., CRF), hard fescue ($F.$$ovina$ ssp. $longifolia$ Thuill., HF) and sheep fescue ($F.$$ovina$ L., SF) were evaluated in the study. An alternative environmental condition requiring a FF germination test by International Seed Testing Association (ISTA) was applied in the experiment, consisting of 8-hr light at $25^{\circ}C$ and 16-hr dark at $15^{\circ}C$ (ISTA conditions). Daily and cumulative germination patterns were measured and analyzed on a daily basis. Significant differences were observed in germination pattern, days to the first germination, days to 50% germination, days to 60% germination, and germination rate. The final germination percentage was variable with species and varieties, being 40.25 to 82.00% at the end of study. There were considerable variations in early germination characteristics and germination pattern among FF species. The first germination in all entries except HF was initiated between 5 and 6 DAS (days after seeding) under ISTA conditions, while HF between 6 and 7 DAS, being 1 day later. It was 8 to 10 DAS in days to the 50% germination, which was 2 to 4 days after the first germination date. Days to the 60% germination were 9.10 to 14.80 DAS under ISTA conditions, being 5.70 days in differences among the entries. CF 'Jamestown II' and 'Shadow II' and HF 'Aurora Gold' were the fast varieties. The slowest one was HF 'Rescue 911'. Among FF species, turf establishment speed was becoming faster in CRF, SF, HF and CF in this order. Information on differences in germination characteristics and pattern from this study would be usefully applied for golf course design and construction, when established with FF.

This study was conducted to investigate early germination characteristics and germination pattern of new cultivars of creeping bentgrass (CB, Agrostis pall/stris Huds.) under natural room temperature conditions and to analyze differences between alternative and natural conditions. Seven cultivars of CB were tested in the study. Daily and cumulative germination patterns were measured and analyzed on a daily basis. Significant differences were observed in germination pattern, days to the first germination, days to 50% germination, days to 85% germination, and germination rate among cultivars under different environments. The first germination in all entries was initiated between 4 and 5 days after seeding (DAS) under alternative conditions, while between 6 and 8 DAS under natural conditions, being 2 to 3 days later as compared with that of alternative conditions. As for the first germination percentage, it was generally 5 to 25% under alternative conditions. However, 'Penn A-1', 'Penncross', and 'Penn A-4' cultivars showed a vigorous germination, producing 50 to 70%. Under natural room temperature conditions, it was 5 to 10% for the most entries, except 'Penncross', 'L-93', and 'Penn A-4' cultivars which vigorously produced 20 to 50%. Days to the 50% germination in CB resulted in 4 to 10 DAS under alternative conditions, while 6 to 15 DAS under natural conditions, being 2 to 5 days later than under alternative conditions. Days to the 85% germination were 5.90 to 11.75 DAS under alternative conditions and 7.95 to 12.50 DAS under natural conditions. 'Penn A-1' and 'L-93' were the fastest cultivars under alternative and natural conditions, respectively. Regardless of the temperature conditions, the longest cultivar in germination was 'T-1', being over 11 DAS. In-depth comparative data on germination characteristics and germination patterns among CB cultivars would be usefully applied for a practical CB turf establishment in golf course construction etc.

Days to the 60% germination were 12.1 to 19.0 DAS, being 3.0 to 4.2 days slower than that under ISTA conditions. CF 'Jamestown II' was the fastest cultivar, while the slowest CRF 'Audubon'. Turf establishment speed was faster with CRF < SF < HF and < CF in order. Information on differences in germination characteristics and pattern between alternative and natural conditions would be practically useful for a golf course construction etc., when established with FF.
Calli were induced from leaf base region of germinated rice(Oryza sativa L. cv. Nakdong) with high frequency of up to 65% on LS medium supplemented with $2.5mg/{\ell}2$, 4-D in the dark at $27^{\circ}C$. Embryogenic calli of pale yellow, globular type were selected and used for the initiation of cell suspension cultures in AA2 liquid medium with $2mg/\ell$ 2,4-D, 0.2mg/$\ell$ kinetin arid $0.1mg/\ell$ GA3. Protoplasts were isolated from the embryogenic cell suspensions after 4 months of culture and then were electroporated with 400V/cm for 1 msec. Electroporated protoplasts divided with plating efficiency of 1.1% on PCM liquid medium supplemented with $2.5mg/\ell$ 2, 4-D, $0.1mg/\ell$ kinetin and 10mM proline. The protoplasts-derived microcalli were cultured on $0.2{\mu}m$ membrane fitter placed onto LS2.5 solid medium containing fine suspension cells as a feeder cells, for 2 weeks in the dark at $27^{\circ}C$. After an additional 2 weeks of culture under fluorescent light of $30{\pm}/3{\mu}E$.m^{-2}S^{-1}, yellow calli of 2mm diameter were transferred to regeneration medium. Shoots were produced from the green spot of protoplasts-derived calli and plants were regenerated form protoplast-derived green calli with frequencies of 11∼33%.
Gentiana scabra Bunge var buergeri Max, can be widely used such as cut-flower, ornamental and medicinal plant among the native plants. Microscopic and Scanning electron microscopic observation plant were carried out to estimates the characteristics correlated with germinability and germination process, and the results were as follows. The seeds were germinated just after harvest, and it means that there is no or little dormancy of the seeds. Matured seeds showed higher germinability than those of immatured. As the storage period was longer, immatured seeds showed shorter longevity than those of matured. The average longevity was about 190 days. According to the observation results of morphological changes during the germination, seed coat was expanded with water absorption immediately after seeding, and radicals were observed 2 days after seeding. Root hairs were formed behind the growing tip of the root on 4 days after seeding and cotyledon were unfolded on 6 days after seeding. In the microscopic observation of leaf tissues, it could be easily distinguished between midrib, and leaf blade, and also observed upper epidermis, palisade parenchyma and spongy parenchyma consisted as a sing1e layer. Palisade parenchyma was consisted of a single layer of cell. Stomata were restricted lower surface, arranged as anamocytic type, and surrounded by guard cells. In the microscopic anatomy of stem were consisted of cortical layer, forming a single layer of epidermis and parenchyma, and vascular bundle which was consisted of continuous cylinder of vascular tissues.

Experiment was conducted under a room temperature condition of $5^{\circ}C$ to $25^{\circ}C$(natural conditions). Daily and cumulative germination patterns were measured and analyzed on a daily basis. Significant differences were observed in germination patterns, days to the first germination, days to 50% germination, days to 75% germination, and germination percentage with different environments and varieties. Germination percentage was variable with varieties at the end of study. It was 85.25 to 97.00% under natural conditions. There were considerable variations in early germination characteristics and patterns among the entries according to different conditions. The first germination was initiated between 7 and 9 DAS(days after seeding) under natural conditions, being 2 to 3 days later as compared with ISTA conditions. It was 11 to 12 DAS in days to the 50% germination, which was 3 to 4 days after the first germination. Days to the 75% germination were 12.38 to 14.29 DAS, being 1.55 to 4.77 days slower as compared with ISTA conditions. Olympic Gold was regarded as the fastest variety and Tar Heel II was slowest. Data from this study demonstrated that information on differences in germination characteristics and patterns among varieties would be usefully applied for TF establishment such as golf course construction.

This study was conducted to establish the system of effective in vitro propagation by various explant sources culture of Bippeastrum hybridum Hort, 'Dazzler'. We tested the effects of optimal explant source, plant growth regulators on bulblet formation and plant regeneration. Callus was readily produced on the different tissues excised from floral buds whereas, bulbs and shoots were formed only on pedicel explants as compared with anthers, styles and ovaries. Pedicel is the best optimal explant for in vitro propagation. Two distinct pathways, organogenesis through callus and direct bulblet formation, could be recognized in pedicel culture. Up to the $80-100\%$ of bulblet formation and shoot organogenesis from the pedicel in fifteen days before anthesis were effectively induced by MS medium supplemented with 0.5 mg/L NAA and 1.0 mg/L BA. Plantlet regeneration was successfully achieved from pedicel-derived callus, via shoot bud induction or direct bulblet formation. The bulblets with blooming flower were produced within 2 years.
Useful Dianthus species were collected and selected from two native and seven foreign species distributed in Gangwon province. For in vitro breeding,. callus was induced from the explants of apical meristem, leaf, stem and the in vitro adventitious shoots on MS basal medium with 2.0 mg/L 2,4-D and 0.5 mg/L BA at 27$^{\circ}C$ under continuous light. After 3 weeks of culture, calli initiated the most highly from the leaf explants of D. chinensis Organogenic calli were able to be selected from the adventitious shoot-derived calli. For shoot regeneration, these organogenic calli were cultured on MS medium with the combination of 0.1 mg/L NAA+1.0 mg/L BA under continuous light. Multiple shoots were proliferated with low frequency (about 30%) from those adventitious shootderived calli. Also, shoots initiated directly from the adventitious shoot explants without callus formation at high frequency of 52% when cultured on N6 medium containing 0.1 mg/L NAA and 1.0 mg/L BA in D. gratianopol. Multiple shoots and plantlets grew well and rooted on MS medium supplemented with 0.1 mg/L NAA. Regenerants with well-developed roots were transferred to 8-cm pots containing vermiculite at 85% relative humidity and 27$^{\circ}C$ These plantlets were acclimatized in artificial soil mixture and transferred to the greenhouse for flowering with normal phenotypes. M28 Mutant line was selected with white flowers from 0.03M EMS-treated organogenic calli derived from in vitro adventitious shoot explants of D. chinensis and set seeds.
This study was conducted to establish a practical masspropagation system of Phalaenopsis clones from basal shoot segments. The frequency of PLB (protocorm like body) induction was compared with various explants. Basal shoot segments showed the most successful result of 45%, while root tips, stalk node segments, stalk leaves and mature leaves represented low frequency (below 5%). The PLB induction ratio in the culture of basal shoot segments was examined with 11 different Phalaenopsis varieties, and the majority of varieties, including pink flower lines, showed an about 30% rate of PLB formation. Especially, when whole basal shoot parts without cutting were inoculated onto PLB induction medium, giant PLB was induced from explant. This giant PLB was green color and big in size compared with normal PLB. When dissected giant PLB segments inoculated onto PLB multiplication medium, only normal size of PLBs were induced from them. PLBs induced by basal shoot culture were transferred onto proliferation medium and then shooting medium, from which normal plants were formed. Therefore, this culture method is considered as effective and practical protocol for Phalaenopsis mericlone production. In addition, it is suggested that clones of an infinite number can be produced consecutively by this culture system through repeated cycles of PLB induction and proliferation using the basal shoot segment of flask plant.

This study was conducted to find out optimum cultivars, and growth temperature and stage in peanut sprout for resveratrol production. Morphological characteristic, days to 50% of the final germination percentage($T_{50}$) and early growth vigor were measured in 8 different peanut varieties. In result 'Jopyeong' appeared to be the most appropriate cultivar for peanut production because of its lower contamination rate, lesser lateral root formation, and thicker hypocotyl length. Optimum temperature for growing peanut sprouts was determined 27. Content of resveratrol was examined by high performance liquid chromatography(HPLC) to investigate appropriate growth stage of peanut sprouts for resveratrol production. Resveratrol was higher than $17.0{\mu}g/g$ in peanuts sprouts 9 days after plating. Considering peanut sprout's shapes and content of resveratrol into account, it was most appropriate to harvest in 9-day after germination.
