Discrimination of Geographical Origin far Ligusticum Root (Ligusticum wallichii) by Capillary Electrophoresis (Capillary electrophoresis(CE)를 이용한 천궁의 원산지 판별)

  • Optimal extraction, separation and capillary rinsing conditions for capillary electrophoresis (CE) were established to discriminate the geographical origin of ligusticum root (Ligusticum wallichii) using 113 samples (domestic sample n = 62, foreign sample n = 51). Ligusticum root was extracted with 30% ethanol and separated on a uncoated fused-silica $(50\;{\mu}m{\times}27\;cm)$ capillary. Conditions for optimal analysis include: temperature, $40^{\circ}C$; voltage, 10 kV; and pressure injection time, domestic and foreign samples were 5 sec and 2 sec, respectively. The optimal separation buffer was 0.1 M phosphate buffer (pH 2.5) containing 15 mM iminodiacetic acid with 40% methanol. Under the optimal conditions established for CE, the ratio of specific peak area (peak LW-1) to other peak area (peak LW-5) was effective in discrimination geographical origin of ligusticum root. The mean accuracy for correct discrimination of geographical origin of domestic and foreign ligusticum roots were 65% and 63%, respectively.

Anti-inflammatory and antimicrobial effects of Jeju rosemary essential oil against skin flora (제주산 로즈마리 에센셜 오일의 항염 및 피부 상재균에 대한 항균 활성)

  • The purpose of this study was to investigate the anti-inflammatory effect and the antimicrobial activity to skin flora of essential oil from rosemary that naturally grown in Jeju. rosemary essential oil was extracted by water distillation essential oil extraction method. In order to confirm the anti-inflammatory activity of rosemary essential oil, it was confirmed that the production of NO and $PGE_2$ induced by LPS in RAW 264.7 cells was inhibited in a concentration-dependent manner. Western blot analysis showed that the expression of iNOS and COX-2, which are biosynthetic enzymes, decreased in a concentration-dependent manner. In addition, production of $TNF-{\alpha}$ and IL-6 the pro-inflammatory cytokines were inhibited. Antimicrobial activities of three S. epidermidis and three P. acnes strains including two antibiotic resistant strains were observed in paper disc method and MIC and MBC tests showed inhibition of bacterial growth and death. From the results of the experiment, we confirmed that rosemary essential oil has the anti-inflammatory and antimicrobial efficacy and it could be used as a cosmetic and skin care material in the future.

Studies on Distribution, Characterization and Detoxification of Shellfish Toxin in Korea 3. Detoxification of Paralytic Shellfish Poison of Sea Mussel, Mytilus edulis (한국산 주요패류에 대한 독의 분포, 특성 및 제독에 관한 연구 3. 마비성패류독의 제독에 관하여)

  • We have veen already reported the distribution of PSP of bivalve mollusca in southern coast of Korea and also analyzed their characteristics. The purpose of this study was to develop detoxification method for PSP infested sea mussel, Mytilus edulis, by rearing methods or processing treatments. There was no significant detoxification effect when the PSP infested sea mussel was reared in a tank with water recirculation system, but the toxicity of sea mussel rapidly decreased during the rearing time in a water flow system with filtered water. The detoxification rate of PSP during the rearing for 5 days in a water flow system tank with $15-17^{\circ}C$ of sea water was $94\%$ in case of high toxic sample with more than $2,600{\mu}g/100g$ and about $40\%$ in case of low toxic sample with less than $100{\mu}g/100g$. The toxicity of PSP extracted from the sample with 0.1N/ HCl solution was about 2-5 times higher than that extracted with distilled water. When sea mussel contained $100-150{\mu}g-PSP$ per 100g of edible meat was boiled for 30 min with tap water, the toxicity was destroyed as the level of PSP undetected by mouse assay. We can suggest that boiling of sea mussel with tap water was one of the most significant detoxification methods, but it was not enough to be safe in case of extremely high intoxicated sea mussel with PSP. For example, the digestive gland of sea mussel contained more than $9593{\mu}g/100g$ was heated in a can with tap water at $116^{\circ}C$ for 65 min. the residual PSP was more than $170{\mu}g$.

  • Volatile flavor components in powdered Katsuobushi were simultaneous trapped by steam distillation-extraction method, and these were fractionated into the neutral, the phenolic, the acidic and the basic fraction. Volatile flavor components in these fraction were analyzed by the high-resolution GC and GC-MS equipped with a fused silica capillary column. The whole steam volatile concentrate consisted of $48\%$ neutral fraction(NF), $35\%$ phenolic fraction(PF), $12\%$ acidic fraction(AF) and $5\%$ basic fraction(BF). Thirty components such as 8 hydrocarbons, 8 aldehydes, 6 furans, 5 alcohols and 3 ketones were identified from NF. And sixteen components such as phenol, guaiacol, dimethoxy phenol, eugenol in PF, twelve components such as propionic, butanoic, isopentanoic, n-hexanoic, heptanoic, octanoic acid in AF, ten components such as 2,6-dimethylpyrazine, 2-nethylpyridine, 2,4-dimethylthiaBole in BF were identified. NF and PF gave a much higher yield than others and were assumed to be indispensable for the reproduction of aroma of powdered Katsuobushi. It was also identified eight components of volatile carbonyl compounds such as ethanal, propanal, butanal, pentanal by 2,4-DNPH method.

Effect of Hydrosol Extracted from Chrysanthemum boreale Makino Flower on Proliferation and Migration in Human Skin Keratinocyte (산국화(Chrysanthemum boreale Makino) 유래 Hydrosol의 피부 각질형성세포 증식 및 이주 유도 활성에 미치는 효과)

  • In the present study, we extracted the hydrosol from flower of Chrysanthemum boreale Makino (CBMF hydrosol) by steam distillation and tested the effect of the CBMF hydrosol on skin regeneration using normal human keratinocytes (HaCaTs). CBMF hydrosol induced proliferation as well as migration in HaCaTs in a dose-dependent manner. Treatment with $1{\mu}g/mL$ CBMF hydrosol increased proliferation to $143.71{\pm}3.37%$ and migration to $139.98{\pm}5.72%$ compared with a control group. CBMF hydrosol also significantly enhanced the phosphorylations of extracellular signal-regulated kinase (Erk) 1/2 and serine/threonine-specific protein kinase (Akt) in HaCaTs. Moreover, CBMF hydrosol dose-dependently induced sprout outgrowth in HaCaTs. These results demonstrate that CBMF hydrosol has skin regeneration and wound healing activity in HaCaTs. Therefore, CBMF hydrosol could be used as a potential cosmetic material.

Changes in Properties of Silk Monofilament Caused by Drawing and Hydrolysis (견 Monofilament의 연신과 가수분해에 의한 특성변화)

  • The middle silk gland, that is a liquid silk thread gland consisting of silk protein, was taken out and a silk monofilament was made by drawing rapidly to approximately 3 times. In order to deteriorate the inter molecular hydrogen bonding force and to stretch in, the drawn silk filament was swoolen in boiling water. The results obtained are as follows ; The silk gland sample that just dried silk gland was occupied in crystalline region of silk-I type and random amorphous region. According to the examination of X-ray diffraction and thermal analysis, silk-II type crystal begins to appear partially in monofilament sample and spread to almost complet silk-II type crystal in 65.2% drawn sample. And, orientation of silk fibroin mlecule increased suddenly in early stage with a rise of drawing ratiofrom birefringence and density, and it was found that orientation of fibroin molecule was completed. As drawing ratio increases relation with time of hydrolysis, birefringence appeared almost fixed a tendency. Crystallization collapse by hydrolysis was not found in X-ray diffraction and thermal analysis. But, amorphous region began to flow by treated hydrolysis, that orientation of crystallization part was disturbed was supposed.

  • The toxicitiy of organic sludge such as municipal sewage sludge (MSS), industrial sewage sludge (ISS), alcohol fermentation processing sludge (AFPS) and leather processing sludge (LPS) were evaluated with three environmental biomarkers as acetylcholinesterase, cytochrome P450, and heat shock protein 70 extracted from earthworm (Eisenia fetida). Their toxicities were compared with those of pig manure compost (PMC). MSS, ISS, LPS, and AFPS did not significantly affect the acetylcolinesterase activity, whereas only the elutriate of PMC slightly was increased the activity. MSS, AFPS, and PMC tended to slightly inhibit the cytochrome $P_{450}$ activity, but ISS and LPS showed significantly the inhibitory effect on cytochrome $P_{450}$. The hsp70 expression began to increase after treatments and showed high induction at 6 hour, followed by zero level at around 12 hour. The quantity of the hsp70 expressed by elutriate treatments of PMC, AFPS, MSS, ISS, and LPS was 1.9, 3.0, 3.3, 4.4, and 4.7 fold higher than that of distilled water. These results indicate that in toxicity tests of five organic waste materials, four kinds of sludge materials appeared more toxic than PMC. Results of AChE, P450, and hsp70 of earthworm might be useful for expecting or assessing an effect by exposure of organic wastes to earthworms in soil.

Antioxidant Effects of Fermented Anchovy (멸치액젓의 항산화효과)

  • The antioxidative activity of Fermented Anchovy on linoleic acid autooxidation was investigated in an aqueous system at pH 7.0. All solvent fractions from Fermented Anchovy were exhibited the strong antioxidative activity. Especially, BuOH and aqueous fractions were gained large amounts with strong antioxidative activity. Ultrafiltration, dialysis, heat treatment of aqueous fraction indicated that water-soluble antioxidants of Fermented Anchovy were heat-resistant, amino acid related compounds with smaller molecular weights than 1,000. Unbound fractions from DE-52 anion exchange chromatography were exhibited antioxidative activity with or without $15\;{\mu}M\;Fe^{+++}\;ion$. We were able to purify one methionine derivative from lots of antioxidative substances in Fermented Anchovy aqueous fraction by gel filtration, anion-exchange chromatography, TLC and HPLC, successfully. These data suggest that Fermented Anchovy aqueous fraction is a mixture of fermented small molecules with strong antioxidative activities.

Physicochemical Characteristics of Steamed Prunus mume Powder Granules in a Fluid-Bed Granulator (유동층조립기를 이용한 금매분말과립의 물리·화학적 특성)

  • $Prunus$ $mume$ was steamed for easier removal of the pulp. The steamed fruit pulp was vacuum dried and powdered. The steamed $Prunus$ $mume$ powder (SPP) was passed through a 250 ${\mu}m$ sieve, fluidized in a fluid-bed granulator, and then granulated by top-spraying with water (SPPGW) or the extract obtained from steam (SPPGE). Then the physicochemical and sensory properties of SPP, SPPGW, and SPPGE were evaluated. The flowability of powder (angle of repose $^{\circ}$) of SPP, SPPGW and SPPGE was $23.59^{\circ}$, $11.07^{\circ}$, and $13.94^{\circ}$, respectively. The water dispersibility of SPP, SPPGW, and SPPGE was 18.69, 10.04 and 6.00 sec, respectively. Also, the overall acceptance of SPP, SPPGW and SPPGE was 3.00, 3.44 and 6.56, respectively. In conclusion, SPPGE can be used as granular steamed whole fruit pulp with good powder flowability and dispersibility, and therefore consumer acceptance.
