Cholinesterase Inhibitors Isolated from the Fruits Extract of Evodia officinalis (오수유의 Cholinesterase 저해활성 성분)

  • The MeOH extract of Evodiae Fructus exhibited a significant inhibition on the acetylcholinesterase (AChE) and butyrylcholinesterase (BChE), in a dose dependent manner, respectively. The extensive bioactivity-guided fractionation process with the MeOH extract finally isolated four compounds, as rutaecarpine (1), evodiamine (2), limonin (3) and dehydroevodiamine (4). Among them, compound 2 exhibited specific inhibitory activity on BChE with the $IC_{50}$ values 1.7 ${\mu}g/ml$, whereas compound 4 showed the potent inhibition upon both AChE and BChE.

  • Extensive phytochemical investigation of the methanol extract from the whole plant of Youngia japonica (Asteraceae) led us to the isolation of a new guaiane-type sesquiterpene (1), together with three related guaianolides, youngiajaponicoside A (2), crepiside H (3) and crepeside E (4). The chemical structure of 1 was elucidated by the aid of spectroscopic analyses including 2D-NMR experiments (COSY, HMBC, HMQC and ROESY). The isolated components (1-4) were evaluated for the inhibitory effect on the proliferation of four cultured human tumor cell lines such as A549, SK-OV-3, SK-MEL-2 and HCT-15, in vitro.

  • The gene analysis of resistance in rice cultivars, Daeanbyeo, Hwasunchalbyeo, Daejinbyeo, Naepungbyeo, Hwajinbyeo and Surabyeo to strains of Xanthomonas oryzae pv. oryzae was studied. F$_1$ plants and F$_2$ populations from the crosses between six cultivars and near isogenic lines carrying the single bacterial blight(BB) resistance gene were analyzed using Korean and Japanese BB races. Daeanbyeo, Hwasunchalbyeo, Daejinbyeo, Naepungbyeo, Hwajinbyeo and Surabyeo are alleic with IRBB101 but are non-alleic with IRBB104 and IRBB105. The allelic tests indicated that Daeanbyeo, Hwasunchalbyeo, Daejinbyeo, Naepungbyeo, Hwajinbyeo and Surabyeo have the Xal gene for resistance.

Relationship between the Production of Fermentational Off-flavor and Presence of Microbial Endophytes in Bloody Watermelon (수박의 이상 발효(피수박)와 내생세균의 존재와의 연관성)

  • The bloody watermelon exhibiting dark red and fermentation off-flavor results in a great economic loss. As an effort to clarify the cause of the bloody watermelon, relationship between the fermentational off-flavor and the presence of endophytic bacteria was studied. The number of endophytes was 2.2-37.0 ${\times}10^3$ cfu/g fw (fresh weight) in normal watermelons, compared to 1.26-1.75 ${\times}10^6$ cfu/g fw in bloody ones. Seventeen bacteria among 56 isolates from bloody watermelons could induce bloody watermelons. The bacteria responsible for bloody watermelons were mainly Gram negative: aerobic Pseudomonas spp and some anaerobic bacteria. The results in this study strongly suggested that the bloody watermelons were produced by abnormal fermentation and growth of endophytic Gram negative bacteria.

Anti-proliferative Effect of the Rhizome Extract of Alpinia officinarum on Cultured Human Tumor Cell Lines (고량강 추출물의 암세포증식 저해 효과)

  • The methanol (MeOH) extract of the rhizome of Alpinia officinarum Hance (Zingiberaceae) demonstrated a potent inhibition on the proliferation of cultured human tumor cell lines such as MES-SA (human uterine carcinoma cell line), MESSA/DX5 (multidrug resistant subline of MES-SA), HCT-15 (human colorectal adenocarcinoma cell line), HCT15/CL02 (multidrug resistant subline of HCT15). The MeOH extract was fractionated into four portions by serial solvent partition, ie., methylene chloride (CH2Cl2) soluble part, ethylacetate (EtOAc) soluble part, n-butanol (BuOH) soluble part and remaining water layer. Among them, the $CH_2Cl_2$ soluble part of the extract exhibited a most potent inhibition on the proliferation of tested tumor cell lines. Bioassay-guided fractionation of the $CH_2Cl_2$ soluble part led to the isolation of five diarylheptanoid and two flavonoid constituents, i. e., galangin (1), 7-(4"-hydroxy-3"-methoxyphenyl)-1-phenylhept-4-en-3-one (2), 1,7-diphenyl-5-hydroxy-3-heptanone (3), trans,trans-1-(3'-methoxy-4'-hydroxyphenyl)-7-phenyl-5-ol-4,6-dien-3-heptanone (4), 5-methoxy-7-(4"-hydroxy-3"-methoxyphenyl)-1-phenyl-3-heptanone (5), kaempferide (6), 5-hydroxy-7-(4"-hydroxy-3"-methoxyphenyl)-1-phenyl-3-heptanone (7). Structures of the isolated active components (1 - 7) were established by chemical and spectroscopic means.
