A Study on the Alkaline Protease Produced from Bacillus subtilis (Bacillus subtilis가 생산하는 Alkaline Protease에 관한 연구)

  • The alkaline protease producing bacteria isolated from soil and identified as Bacillus subtilis. The optimum medium for alkaline protease production from the microorganism was as follows; soluble starch, 1.5% ; proteose peptone, 0.5% ; $K_2HPO_4$, 0.1% ; $MgSO_4{\cdot}7H_2O$, 0.02% and sodium carbonate, 1.0%. The optimum temperature for alkaline protease production was $35^{\circ}C$, and the initial pH of medium was pH 10.5. The alkaline protease activity was about 2,300 U per ml of culture broth by Casein-Folin Method. A 9.2 fold purification of alkaline protease was obtained from culture broth. The recovery was 14% and purified enzyme was identified as single band, and its molecular weight was about 19,000. The optimum temperature for enzyme reaction was $70^{\circ}C$, and optimum pH was 12. The activity of purified enzyme was inhibited by metal ion ($Fe^{++}$), and Phenylmethylsulfonyl Fluoride, a serine protease inhibitor.

Effects of Medium, Temperature and pH on Mycelial Growth and Cellulase Activity of Ectomycorrhizal Fungi from Korean Forests (우리나라 산림에서 분리한 외생균근균의 균사생장에 있어 배지, 온도, pH의 영향과 셀룰라아제 활성)

  • Mycelial growth of ectomycorrhizal fungi (27 strains of 8 species) collected from Korean forests was observed on various culture conditions (media, temperature, pHs). After 60 days of incubation, all strains grown on potato dextrose agar (PDA) and modified Melin-Norkran's agar (MMNA), whereas no mycelial growth was observed on malt extract agar (MEA) or sabouraud dextrose agar (SDA) in some strains including Tricholoma matsutake. Mycelial growth on PDA was poor at high temperature ($30^{\circ}C$) than the low temperature ($10^{\circ}C$). The optimal temperature on PDA and pH in potato dextrose broth (PDB) for mycelial growth in most strains were $20-25^{\circ}C$ and pH 4-5, respectively. All strains tested showed the carboxymethyl cellulase (CM-cellulase) activity and the maximal cellullase activity was expressed by the mycelium of T. matsutake (KFRI 1266) on the CMC agar plate with pH 5.0.

Characterization of Optimum Conditions Affecting on Multi-Color Dyeing of Cut Rose (Rosa hybrida 'Akito') (절화장미(Rosa hybrida 'Akito') 복색염색에 미치는 몇 가지 요인의 최적조건 구명)

  • This experiment was conducted to examine the ideal condition for multiple colors dyeing of cut rose, 'Akito'. Four different colors of dye were applied to make multiple flower colors in cut rose by absorption method. Acidic condition around pH 3.5 showed the fastest absorption speed in all four colors. There is no difference at each pH levels, at part of leaves dyeing, it is more affected colors rather than pH levels in purple and blue colors. From vary concentration of each dyes, $10g{\cdot}L^{-1}$ concentration of dyeing solution gave the best color definition. But it's not significant and result was different by each color in a same concentration. 10 minutes of dyeing performed pastel color and 30 minutes performed the best color definition, but the 30 minutes treatment accompanied with significant dyeing on the leaves also. As a result of this experiment, optimum condition for dyeing cut rose, 'Akito' requires pH 3.5, $5-10g{\cdot}L^{-1}$ of dye concentration and 10-30 minutes treatment.

  • Aspergillus nidulans가 생산하는 naringinase를 acrylamide gel을 이용한 entrapping moth길에 의하여 고정화하여 그 고정화 효소의 물리, 화학적인 성질에 대하여 검토한 결과 다음과 같았다. 1) Acrylamide의 농도는 10% 일때 enzyme activity가 가장 좋았다. 2) Immobilized enzyme의 최적온도는 $50^{\circ}C로서$ 유리효소의 경우보다 $10^{\circ}C정도$ 증가 하였고 최적 pH는 유리효소의 경우와 마찬가지로 pH 40이었다.(중략)

  • The present investigation was conducted to determine the chemical oxygen demand (COD) removal efficiency by Pseudomonas aeruginosa EMS1. The COD removal efficiency in the medium containing 1% metal working fluid (MWF) was 12% after cultivation of 4 days. The composition of optimum medium for the COD removal efficiency of 1% MWF by P. aeruginosa EMS1 were NH$_4$Cl 0.3%,$ K_2HPO_4\; 0.05%,\; KH_2PO_4\; 0.04%,\; MgSO_4.7H_2O\; 0.05%,\; CaCl_2.2H_2O 0.03%$ and $FeSO_4.7H_2O$ 0.04% at initial pH 7.0 and $30^{\circ}C$. Under this condition, the highest the COD removal efficiency was observed after 4 days.

  • The culture condition and medium composition for the enhanced mycelial growth of Inonotus mikadoi IMSNU 32058 were investigated. The optimal temperature and pH for the mycelial growth were $27^{\circ}C$ and pH 4.5, respectively. Among the complex media tested, the malt extract medium and Phellinus igniarius medium were very good for mycelial growth of I. mikadoi. When Czapek-Dox medium was used as a minimal medium for cultivation of mycelia, xylose, raffinose and carboxymethyl cellulose were very excellent as a carbon and energy source. With respect to carbohydrate, sucrose and glucose were very good carbon sources. In general, organic complex nitrogen sources were better than other inorganic ones. As the organic complex nitrogen sources tested, yeast extract, soytone, proteose peptone and bacto peptone were the best as a source of organic nitrogen. When ammonium sulfate as an inorganic source of nitrogen was used, the enhanced mycelial growth was shown. p-Aminobenzoic acid was proved to be most appropriate source of vitamin. After the mycelia of I. mikadoi was cultivated at $27^{\circ}C$ for 5 days in MEM broth (pH 4.5), the activities of both exomycelial and endo-mycelial enzymes were determined. Among endomycelial enzymes assayed, the specific activity of laccase was much higher than those of other enzymes. When the fungus was grown in MEM broth, exomycelial specific enzyme activity of laccase was comparatively high. However, little or no enzyme activities of protease, chitinase and lipase were found.

Studies on the Celluloytic Enzymes Produced by Stropharia rugosoannulata in Synthetic Medium (합성배지에서 Stropharia rugosoannulata가 생산하는 섬유소분해효소에 관한 연구)

  • For the purpose of utilizing cellulose resources by cellulolytic enzymes of Stropharia rugosoannulata, it's cultural conditions for the prodution of cellulolytic enzymes in synthetic media were investigated. The optimum pH for the production of Avicelase and ${\beta}-glucosidase$ was pH 5.0, while that of CMCase was pH 4.0. The optimum temperature for the production of Avicelase, CMCase and ${\beta}-glucosidase$ was $40^{\circ}C$. Among the carbon sources, xylose was good for the production of CMCase and ${\beta}-glucosidase$, but maltose was good for the production of Avicelase. The optimum concentration of the carbon sources for the production of CMCase, Avicelase and ${\beta}-glucosidase$ was 1.0, 0.8 and 1.1%, respectively. As inorganic nitrogen sources, $NH_4Cl$ was good for the production of all the three cellulolytic enzymes. The optimum concentration of $NH_4Cl$ for the production of CMCase was 0.3% while that of Avicelase and ${\beta}-glucosidase$ was 0.4%. As organic nitrogen sources, malt extract was good for the production of all the three cellulolytic enzymes. The optimum concentration of organic nitrogen for the production of ${\beta}-glucosidase$ was 1.3% while that of CMCase and Avicelase was 1.0%. As the mineral sources, $CoCl_2$ good for the was good for the production of all the three cellulolytic enzymes. The optimum concentration of $CoCl_2$ for the production of all the three enzymes was 0.35%.

Optimum pH Condition of Defatted Rice Protein Extraction by Alkaline Method (알칼리 추출법에 의한 탈지 미강 단백질 추출의 최적 pH 조건)

  • For efficient extraction of protein from defatted rice bran, the 5 ranges of extraction pH (8, 9, 10, 11 and 12) and the 3 ranges of isoelectric precipitation pH (2, 4 and 6) were used. The protein content, browning reaction, the electrophoresis pattern and the recovery yield of soluble protein at each pH range were compared each other. The recovery yield of soluble protein increased in proportion to extraction pH, but at the same time, browning reaction became more conspicuous. The most amount of protein was recovered at the precipitation pH of 4. The SDS-PAGE patterns of the extracted proteins showed no significant correlations between pH and the protein content, but the highly alkaline condition was more advantageous to extract protein less than 35 kDa. In each pH range, the recovery yield of soluble protein averagely reached 32.5% on the basis of extraction. In result, it was found that combination of extraction pH 10 and precipitation pH 4, which resulted in 37.65% of recovery yield and low level of browning reaction, was the optimum condition for the extraction of protein from defatted rice bran.

Monitoring of pH and dissolved oxygen in microorganism fermentation processes using a 24-well microplate (24-웰 마이크로 플레이트를 이용한 미생물 발효공정에서 pH 및 용존산소의 모니터링)

  • In this study, the sensing membranes for detection of pH and dissolved oxygen(DO) were prepared by immobilizing 6-aminofluorescein or ruthenium complex onto the sol-gel matrixes of GPTMS, MTMS, and TEOS and then recoated with the mixture of hydrophobic sol-gel and graphite for light insulation. The pH and DO sensing membranes recoated with the light insulation layer showed a higher sensitivity than those without light insulation layer. The sensing membranes were immobilized on the wells of 24-well microplate and used to monitor the fluorescence intensity for pH and DO in E.coli JM109 and P.pastoris X-33 fermentation processes. The change of the fluorescence intensity in the DO sensing membrane agreed with the growth patterns of microorganisms, that the membranes are valuable to monitor the DO in fermentation processes. In the case of pH monitoring, the fluorescence intensity has showed good correlation to the off-line pH data, that the pH membranes are valuable to monitor pH values in fermentations.
