In the synthesis of water-based polyurethane using self-emulsification process, after being neutralized, polyurethane pre-polymers containing ionic pendant groups are dispersed by simple convective mixing. Preparation of dispersion is followed by chain extension reaction, which is considered as important step for growth of the molecular weight of polyurethane. In this work, pH variations in the aqueous phase were monitored during the chain extension process in the presence of water-soluble diamines. The optimum degree of chain extension and the average particle size in the polyurethane dispersions were examined with varying ionic pendent group contents, type of chain extenders, and feed rate of chain extenders, The initial pH value in the aqueous phase linearly increased and the optimum chain extension point could be obtained from the intersection of two linear lines having different slopes, All average particle sizes before chain extension reaction were almost same, however, the final average particle size increased as feed rates of chain extenders increased, In addition, as the ionic pendant group contents increased, the particle size decreased since the hydrophilicity and hydrodynamic volume increased. As carbon numbers of the chain extenders increased, the final particle size increased significantly. From the results, it was concluded that the chain extension reaction took place among the particles not only in a particle.

As a fundamental study to apply the new flocculation method using ballast in water treatment process, the optimal conditions for general and ballast coagulant dosage, and pH, which are known to have a significant influence, were derived by response surface methodology. Poly aluminum chloride (PAC) and magnetite ballast were used as a general coagulant and ballast, respectively. Coagulation experiments were performed by jar-tester using the kaolin based synthetic water. The effects of three independent variables (pH, PAC, and ballast) on response variables (turbidity removal rate and average settling velocity of flocs) and the optimum condition of independent variables to induce the optimum flocculation were obtained by 17 experimental conditions designed by Box-Behnken procedure. After performing experiments, the quadratic regression model was derived for each of response variables, and the response surface analysis was conducted to explore the correlation between independent variables and response variables. The $R^2$ values for the turbidity removal rate and the average settling velocity were 0.9909 and 0.8295, respectively. The optimal conditions of independent variables were 7.4 of pH, 38 mg/L of PAC and 1,000 mg/L of ballast. Under these conditions, the turbidity removal rate was more than 97% and the average settling velocity exceeded 35 m/h.
The microbial properties and acid tolerance of the three kinds of Acetobacter sp. isolated from persimmon vinegar were investigated. Acid tolerance was also evaluated. Acetobacter sp. were gram negative, short rod, nonspore forming and motile. They reacted positively catalase, methyl red, oxidation fermentation, Voges-Proskauer and nitrate reduction tests and negative to hydrogen sulfide test and ONPG. Acetobacter sp. showed normal growth curve in Carr broth and there was no significant difference between isolates and (standard on) typical strains such as Acetobacter aceti (KCTC1010), Acetobacter liquefaciens (KCTC2804), Acetobacter diazotrophicus (KCTC 2859). Optimum temperature and initial ethanol concentration in incubation were $30^{\circ}C$ and 6%, respectively. Growth and acid production of Acetobacter sp. were suppressed by the concentration of above 4% acetic acid. The amount of $Mg^{++}$ release from Acetobacter sp. cells in medium was increased by acetic acid, and almost in the concentration of 6% acetic acid. Glycolysis by Acetobacter sp. had optimal pH about 6.0 to 7.0 and more stable in acidic condition than in alkalic. The $H^+-ATPase$ of Acetobacter sp. S-1 and S-3 showed a maximal activity between pH values of approximately 5.5 to 7.5 and 6.0 to 7.5, respectively.
With the increase of the use of antibiotics and invasive procedures, infections caused by multidrug-resistant Acinetobacter baumannii(MRAB) are increasing. We screened the antibiotic producing strain B-51 for antibacterial activity against MRAB from the soils and studied the effects of culture medium on the antibiotic production of B-51. The medium conditions for maximum antibiotic productivity of B-51 was 2% glycerol, 0.5% soybean meal, 0.01% $CaCl_2$, 0.01% $MgSO_4{\cdot}7H_2O$ and 0.01% $KH_2PO_4$ at an initial pH of 6.0, at $30^{\circ}C$ for 76 h.
For selective elimination of salt in the boiled extract of baby neck clam (Tapes variegata, BEBNC), electrodialyzer was used and the desalination conditions of BEBNC were determined at various concentrations, pH's and volumes of BEBNC. The ion-exchange membrane with a molecular weight cut off 100 Da was used for desalting of BEBNC. The desalination times on BEBNC concentrations of 0.5% and 4% were 100 min and 170 min and the final desalting ratio was above 90%. 1'he desalination of BEBNC at pH 4.0 was 12% higher than that at pH 9.0 for 60 min. The electrodialysis process could remove above 90% of the initial salt content in 2% BEBNC of 1 L for 80 min. The results obtained prove that electrodialysis is a practical solution to the problem of selective elimination of salt from BEBNC.

The effect of on the cell growth, the elaboration of pullulan, the morphology and were the effect of on the molecular weight of pullulan were investigated. A. pullulans showed maximum pullulan production when initial pH 6.5 was 11.98 g/l in shake-flask culture. In batch culture, the maximum pullulan production of 15.16 g/l was obtained at an aeration rate of 0.5 vvm. The mixture of yeast-like form and mycelial form of cells was found at the constant pH 4.5, at which condition, the elaboration of pullulan was high, about 13.31 g/l. However, pullulan with its higher molecular weight (>1,000,000) was produced at the constant pH 6.5.

The strain producing biosurfactant was isolated from soil smples. The isolated strain was identified as the genus Nocardia through its morphological, cultural and physiolgical characteristics. A high concentration of the biosurfactant by Nocardia sp. L-417 was obtained after 4 days of cultivation in the culture medium containing 3% n-hexadecane, 0.1% $NaNO_3$, 0.02% $K_2HOP_4$, 0.01% $H_2PO_4$, 0.01% $MgSO_4$.$7H_2O$, 0.01% $CaCl_2$, 0.02% yeast extract, and 0.02% tryptone. The optimum pH and temperature for biosurfactant production were pH 6.0 and $30^{\circ}C$, respectively. Furthermore, most biosurfactans were produced during the exponential growth phase, and this fact indicated that the biosurfactans production was growth-associated. The biosurfactant showed the good emulsification activities on various emulsifying substrates such as bunker A, paraffin, corn oil which are used widely in industries.
This study was carried out to analyze the differences in enzyme activity and stability between the native Fe superoxide dismutase (FeSOD) and the 6xHis-tagged superoxide dismutase (6xHis-FeSOD) of Streptomyces subrutilus P5. The optimum pHs for both native FeSOD and 6xHis-FeSOD were 7, while the pH range of the activity was narrower for the 6xHis-FeSOD. The native FeSOD was stable at pH 4-9, but the 6xHis-FeSOD lost its stability at pH > 9. The temperatures of the optimum activities were same for both types of enzymes. However, the heat stability of the 6xHis-FeSOD was clearly decreased; even at $20^{\circ}C$ the enzyme lost the activity after 360 min. In contrast, the native FeSOD was stable after 720 min at below $40^{\circ}C$. $H_2O_2$ inhibition was occurred already at 0.5 mM for the 6xHis-tagged enzyme. Therefore, from the results that the 6xHis-FeSOD retained the enzyme activity at pH 6-7 and $20-40^{\circ}C$, it can be assumed that the protein structure became destabilized under different storage conditions and sensitive to the enzyme inhibitor.
This study was conducted to investigate the effect of operational parameters such as dosage of magnesium and phosphate, pH, reaction time and existence crystal core for the removal of nitrogen and phosphorus in anaerobic fermentation supernatant by struvite crystallization. Optimal mole ratio of $Mg^{2+}:NH_4{^+}:PO_4{^{3-}}$ was 1.2:1.0:1.2. Under the optimal molar ratio, removal ratio and reaction rate constant of $NH_4{^+}-N$ and $PO_4{^{3-}}-P$ were 79.2, 96.8%, 0.157 and $0.344min^{-1}$, respectively. Optimal pH and reaction time were 11 and 10 minutes, respectively, in the optimal molar ratio. Residual concentration of $NH_4{^+}-N$ and $PO_4{^{3-}}-P$ showed lowest value with 1 g/L of crystal core addition. SEM analysis of struvite crystallization with crystal core showed higher crystal core growth than that of without crystal core. Struvite precipitate proved to be orthorhomic crystal structure by XRD analysis.
The putative endochitinase gene, yheB of Escherichia coli K-12 is not expressed under lab culture conditions. The endochitinase gene was amplified by PCR and subcloned into pET28c vector and pQE9 vector, respectively. The endochitinase produced in E. coli harboring pET28c containing yheB or pQE9 vector containing yheE was partly released into the growth medium. The overproduced endochitinase was partially purified by His affinity column chromatography and DE-52 column chromatography. The apparent molecular weight of the endochitinase determined by SDS-polyacrylamide gel electrophoresis was about 97,000. The purified E. coli endochitinase showed maximal chitinolytic activity at pH 6 and $40^{\circ}C$.
