Preparation and Characterization of Electrospun Nanofibers Containing Natural Antimicrobials (천연 향균물질 함유 나노섬유의 제조 및 특성분석)

  • The fabrication of PHBV nanofibers containing various plant polyphenols by electrospinning has been examined. It has been found that the average diameters of fibers increased by the adding of polyphenols. The resulting fibers exhibited a uniform diameter ranging from 340 to 450 nm. As the concentration of polyphenols increased, the diameter of fibers increased due to the hydrogen bonding interaction between the ester groups of PHBV and hydroxyl groups of polyphenols. The interaction between PHBV and polyphenols, which forms a complex together in solution, was verifed by UV measurement. ATR-FTIR analysis confirmed the existence of the hydrogen bonding interaction. The semicrystalline structure of the PHBV nanofiber was observed from XRD pattern. The crystallinity of PHBV nanofibers was increased by the adding of polyphenols. PHBV nanofibers containing polyphenols showed superior antimicrobial activities.

Extraction, Component Analysis and Antibacterail Activity of Panax ginseng Absolute Essential Oil (인삼 앱솔루트 정유의 추출, 성분 분석 및 항균 활성)

  • Interests in natural products have been gradually increasing as well-being era has arrived. Panax ginseng was chosen since it represents Korean traditional medicine as its effect on many age-related symptoms. Panax ginseng absolute essential oil was prepared and analyzed for its main components. Ginseng absolute oil was extracted from Panax ginseng using organic solvent of hexane and ethanol with the yield of 0.146%. The oil was analyzed by GC-MS and main components are sesquiterpenes such as neoclovene, panasinsene, and calarene and more than 110 components were identified from it. The oil was tested for antibacterial activity against general bacteria(E. Coli, S. aureus and P. aeruginosa), yeast, acne germ(Propionibacterium acne) and dandruff germ(Pityrosporum ovale). Panox ginseng absolute essential oil showed the prominent antibacterial activity against Propionibacterium acnes ACCC 6919.

The Effect of Chrysin on the Transcriptional Activity of Vitamin D Receptor in Human Keratinocytes (각질형성세포에서 Chrysin이 Vitamin D Receptor의 전사 활성화에 미치는 영향)

  • Chrysin (5,7-dihydroxyflavone) is a natural flavonoid found in various plants and foods such as propolis and honey. It has been reported that chrysin has various biological effects including antioxidant, anti-aging, anti-inflammatory and anti-cancer. In this study, we investigated the effect of chrysin on the transcriptional activity of VDR in human epidermal keratinocytes by performing dual-luciferase assay. Chrysin significantly induced the transcriptional activity of VDR in a concentration-dependent manner. The VDR mRNA expression was investigated by quantitative real time PCR and chrysin increased the VDR mRNA expression in normal human epidermal keratinocytes. We also found that chrysin increased the expression of keratinocyte differentiation markers such as keratin 10, involucrin and filaggrin. Therefore, the results suggest that chrysin can stimulate the differentiation of human keratinocytes by increasing transcriptional activity of VDR.

Antibacterial and Antioxidant Effect of Botanical Antimicrobial Agent-Citrus product on Pollack or Ascidian Fishmeat (식물성 천연항균처리에 의한 저장 명태와 우렁쉥이 어육제품의 항균 및 항산화효과)

  • The antibiotic and antioxidant effect of botanical antimicrobial agent-citrus product(BAAC) prepared from citrus fruits were investigated by using fishmeats made from pollack and ascidian fillet. We inoculated Salmonella typhi into fishmeats that were aferwards treated with BAAC varying the concentration level (50-500ppm) including the control not-treated with BAAC. While specimens were stored at 5$^{\circ}C$ for designated period, we measured changes of total number of bacteria, peroxide level and textural properties. In addition, we performed organoleptic examination, focusing on appearance, texture and odor for fishmeats treated with BAAC and stored for designated period. For pollack and ascidian fishmeats, the initial total number of bacteria were 3.7${\times}$l0$^4$CFU/g and 7.5${\times}$l0$^4$CFU/g, respectively. After they were treated with 250 ppm of BAAC solution and stored for 5 days at 5$^{\circ}C$, the figures reduced to 2.5${\times}$l0CFU/g and 8.4${\times}$10CFU/g. No bacteria were found in fishmeats treated with above 500 ppm of BAAC, which remarkably proved antibiotic effect of BAAC solution. Furthermore, all the BAAC-treated specimens exhibited notable antioxidant effect. Fishmeats which were previously treated with 500 ppm of BAAC solution and stored for 30 days at 5$^{\circ}C$, showed the lower peroxide level of 16.5 meq/kg than 68.9 meq/kg in the control of pollack and 21.2 meq/kg than 80.7 meq/kg in the control of ascidian. In the results of texture and organoleptic evaluation we could also observe good effects of BAAC on maintaining the freshness of fisher products.

Screening of Natural Antioxidant from Plant and Their Antioxidative Effect (식물성 천연 항산화물질의 검색과 그 항산화력 비교)

  • Certain parts of 95 species of edible and medical plants were extracted with water and 75% of ethyl alcohol. After addition of those extracts to palm oil, lard and soybean oil at different level, their antioxidative activities were compared by Rancimat test. Six species among them seemed to have rather strong antioxidative activity and high extracting yields(i.e. Taraxacum platycarpum, Plantago asiatica, Rhus javanica L., Lycopus lucidus, Astragalus membranaceus, Taraxacum platycarpumH). Among them, the Rhus javanica L. ethanol extract retarded greatly the induction period of palm oil and lard. When 600 ppm of Rhus javanica L. extract were added to palm oil and lard, AI(antioxidant index was expressed as induction period of oil containing various plant extracts/induction period of control oil) of each was 1.35 and 3.03 respectively. This result indicated that the Rhus javanica L. extract was more effective on lard than the other oils.

Verification of Estrogenic Activity in Ethanol Extracts of Marine Organisms Using in vitro Test System. (In vitro 검출시스템을 이용한 해양생물 추출물로부터 에스트로겐 활성 검증)

  • In order to verify the occurrence of an estrogenic compound in natural products, the estrogenic activity was measured using an in vitro detection system. For this system, human breast cancer cell line MCF7 was transfected using an estrogen responsive CAT reporter plasmid. Estrogenic activities of photosynthetic algae spirulina and sea lettuce were evaluated using this system. Estrogenic activities of a $500\mug/ml\; and\; 50 \mug/ml$ ethanol extracts of spirulina were as much as that of $10^{-8}$M standard solution (17$\beta$-estradiol) and activity of $5\mug/ml$ ethanol extract of spirulina was as much as that of $10^{-10}$ M standard solution. However, no significant estrogenic activity was observed using sea lettuce extract. Estrogenic activities of marine animals, such as star fish and shrimp, were also evaluated using this system, however, no significant estrogenic activity was observed in these extracts. In this result, it is confirmed that spirulina extract possesses estrogenic compound.

  • 최근 환경과 건강에 대한 관심이 증대되면서, 복합기능을 갖는 다기능성 섬유에 대한 요구가 증대되고 있는 실정이다. 일반적으로 기능성은 마이크로캡슐이나 후가공제를 이용하여 일반 섬유 제품에 적용하고 있다. 마이크로캡슐이나 후가공제가 적용된 일반 섬유제품은 일상생활 속에서의 마찰 및 세탁 등으로 인해 기능 저하가 발생된다. 이러한 문제점을 개선하기 위해 특수 방사 기술을 이용하여 다기능성을 갖는 섬유 제조 기술이 개발되고 있다. 다기능성 섬유소재 및 응용제품은 그 제조 공정성이 우수하고, 염색 가공 등의 후 공정 조건에 의해 기능성 저하를 초래하지 않고, 지속시간이 오래 유지해야하는 조건을 만족해야한다. 이러한 기능성 저하 문제를 해결하고 내구성 향상 및 기능성의 지속 시간을 오래 유지하기 위하여 복합 방사 기술을 이용하여 이형 단면 원사, Sheath-Core 단면 원사 등이 개발되고 있다. 개발된 기능성 원사의 경우 일반 원사와 다른 형태의 염색성을 보일 수 있으며, 염색 온도 및 염색 시간 등 염색 조건이 달라질 수 있다. 본 연구에서는 식물 등에서 추출한 천연 항균 유기물을 마이크로캡슐화하여 이형단면의 심부에 담지된 형태인 Sheath(Nylon: 50%)-Core(PP MB: 50%) 원사의 염색성 평가 시험을 실시하였다. 시험 방법은 Sheath-Core 원사와 일반 Nylon 원사와 염착 거동 시험을 통해 비교해 나타냈고, 염색 전 후의 물성을 비교해보았다. 또한 염색성 평가 시험을 토대로 기능성 원사의 염색 온도와 시간에 따른 색차 비교 시험을 통해 기능성 원사의 염색 조건을 선정하는 시험을 실시하였다.

Antioxidant Activity of Momordica charantia L. Extracts (여주 추출물의 항산화 활성)

  • This study was carried out to determine the antioxidant activity and vitamin C contents in plant extracts of the Momordica charantia L. The vitamin C was detected as the highest content in immature fruit (92.2 mg/100 g), while the content in stem (2.5 mg/100 g) was lower 40 times than that of immature fruit. Antioxidant activity for the dried sample was investigated by TBA method. The lowest TBARS values were obtained from extracts of dried leaf and followed by ascorbic acid and BHT, showing that the extracts from dried leaf possess the strongest antioxidant activity. Compared with fresh tissues, SOD activity, ATX activity and CAT activity were high level in the dried tissue. These results suggest that the Momordica charantia L. would be a promising antioxidant source as an alternative antioxidant, based on natural plant resources.

Evaluation of Genotoxicity and 14-day Repeat Dose Toxicity of Water Extract of the Mixture of Natural Plants, Anemarrhena and Phellodendron (천연식물자원 지모와 황백피 혼합 수추출물의 안전성평가)

  • The safety of a new natural plant composition (ADP) was assessed on the genotoxicity study and 14-day repeat dose toxicity study. ADP contains a mixed water extract obtained from the mixture of Phellodendron cortex (Phellodendron amurense) and Anemarrhena rhizoma (Anemarrhena asphodeloides), and poses the contractile properties mediated by alpha-adrenoceptor of the prostate and urethra as well as antioxidant and anti-inflammatory properties. In order to evaluate genetic safety, in vivo micronucleus test was performed in ICR mice orally administered with three dose levels of 1250, 2500, 5000 mg/kg body weight, and vehicle and positive control. In the 14 days study, Sprague-Dawley rats were treated with ADP at the dose levels of 500, 1000, 2000 mg/kg once a day, and clinical signs, body weights, hematology, serum biochemistry, necropsy findings and organ weights were monitored and examined. In experimental results, ADP treatment, compared with vehicle control, did not induce the micronucleated erythrocytes from mouse bone marrow. In the 14 days study, any significant and toxicological differences in all measurements of parameters were not observed in ADP treatment groups of animals, compared with vehicle treatment. The No-Observed-Adverse-Effect-Level (NOAEL) of ADP in the 14 days study was determined to be greater than 2000 mg/kg/day in both sexes.

Anti-inflammatory Activity of Herbal Extracts through Inhibition of TNF-α, IL-6 and IL-8 (TNF-α, IL-6, IL-8 저해를 통한 천연약용식물 추출물의 항염증 특성)

  • This study evaluated the cytotoxicity and anti-inflammatory activities of natural herbal extracts including Houttuynia cordata and Eucommia ulmoides against human mast cell (HMC-1). Houttuynia cordata (HC) and Eucommia ulmoides(EU) were extracted with distilled water (at $75^{\circ}C$) and then freeze-dried for 5 days. Finally, the mixture of HC and EU were sterilized by ${\gamma}$-rays irradiation. Cytotoxicity of the mixture against HMC-1 cell was measured using cell counting kit-8 (CCK-8) assay. In addition, inflammatory mediator cytokines such as TNF-${\alpha}$, IL-6 and IL-8 were evaluated by ELISA kit on the HMC-1 cells with calcimycin A23187 and phorbol 12-myristate 13-acetate (PMA). The results showed that mixture of HC and EU had no cytoxicity and reduced TNF-${\alpha}$, IL-6, IL-8 response on HMC-1 cells.
