Over the past 50 years, red sea bream Pagrus major has been selectively bred for more than 10 generations to improve its growth rate. However, the effect of genetic factors on growth under low- or non-fish meal diet is still unclear. Here, we analyzed 251 individuals randomly mating 21 females (dam) and 16 males (sire) to determine the difference in growth between non-fish meal (i.e., treatment group) and fish meal-based diet (i.e., control group), if any. The kinship coefficient between each group of individuals and their parents was estimated by the KING (Kinship-based Inference for GWASs) software package. The presumed parents of 74 individuals in the treatment group and 57 individuals in the control group could be effectively identified. Notably, one specific female was consistently related to high- rather than low-growth individuals in both groups. In addition, more than one parent in each group was related to either high- or low-growth individuals. Parents of both high-growth individuals in the treatment group and low-growth individuals in the control group were also identified. Although further study is required on various growth factors, this study suggests the possibility of selective breeding under non-fish meal diet.
We evaluated the effects of prebiotic (mannan oligosaccharides, Mos) and probiotic diet supplements on growth performance, innate immunity, antioxidant activity, and intestinal changes in the microbial flora of red seabream Pagrus major. A basal diet (Con) was formulated to meet the nutrient requirement of red seabream. The dietary starch in Con was replaced with 0.6% Mos, Lactobacillus plantarum, Bacillus subtilis, B. licheniformis and probiotic mixture (labeled as Mos, Pro-LP, Pro-BS, Pro-BL and Pro-Mix, respectively). We stocked 450 fish in 18 polypropylene tanks (400 L) in triplicate groups per dietary treatment. The fish were fed one of the diets twice (08:30, 18:30 h) a day for 63 days. Lysozyme activity was significantly higher in all the supplemented groups than that of the Con group. The immunoglobulin level of Pro-Mix, anti-protease activity of Pro-BL, and glutathione peroxidase and superoxide dismutase activity of Pro-BS, Pro-BL and Pro-Mix groups were significantly higher than those of the Con group. The ratio of total Vibrio/heterotrophic marine bacteria counts was significantly lower in Pro-LP, Pro-BL and Pro-Mix groups than that of the Con group. Therefore, dietary supplementation of Mos and probiotics to improves immune response and antioxidant enzyme activity and inhibits Vibrio bacteria in the intestine.
In this study, using a continuous behavior measurement technique, the short-term behavioral responses and tolerance limits of red seabream Pagrus major fingerlings to sudden exposure to low salinity in a controlled environment were observed. The activity of the fingerlings suddenly exposed to 21.4, 17.3, and 9.8 psu increased temporarily at the initial exposure to show irregular swimming behavior, but then recovered a stable activity pattern through rapid salinity adaptation. However, the organisms suddenly exposed to 7.3 and 4.3 psu could not withstand the salinity stress, and their swimming behavior was severely disturbed and all individuals died within 48 hours. The findings suggest that red seabream underwent a temporary salinity stress process at the beginning of the exposure to concentrations of 10.0 psu or higher. At these concentrations, osmotic control was possible within at least 11 hours, so stable metabolic activity was also possible. However, organisms suddenly exposed to concentrations below 5.0 psu exceeded the tolerance to low salinity and the sublethal limit. In red seabream exposed to this concentration range, severe behavioral and metabolic disturbances were observed, and death was observed due to osmotic control failure. In conclusion, a salinity range of 5.0 to 10.0 psu can be predicted to correspond to a concentration range in which the osmotic control ability of the red seabream fingerlings is lost, and sub-lethal reactions occur.
In this study, the correlation between clinical changes and RSIV genome copy number was investigated to determine the quantitative criteria for the characteristics of RSIV infection. The rock bream (Oplegnathus fasciatus) was intraperitoneally injected with three different doses (1.0×101, 1.0×103 and 1.0×105 viral genome copies/fish) as low, medium, and high doses, respectively. The clinical signs (spleen enlargement, death) observation and real-time PCR were conducted at 5, 10 and 14 days post-injection. During the experiment, spleen index as a quantitative indicator for spleen enlargement was continuously increased in the medium- (up to 2.26) and high-dose (up to 4.99) challenge groups, respectively. Notably, when the spleen index was over 1.5, 2.0, 2.5 and 3.0, a positive correlation was revealed with average viral genome copy numbers of 2.51, 3.37, 4.97 and 5.43×107 viral genome copies/mg, respectively. Moreover, the threshold of spleen index over 1.5 was 1.0×106 viral genome copies/mg, while the thresholds of spleen index over 2.0 and dead was 2.51×107 viral genome copies/mg and the thresholds of spleen index over 2.5 and 3 was 3.98×107 viral genome copies/mg. These findings suggest the possibility of quantitatively analyzing the characteristics and development process of RSIV infection.
In this study, we analyzed the phylogenetic classification, epitope prediction, and pathogenicity of red sea bream iridovirus (RSIV) isolated from rock bream between 2019 and 2023. Phylogenetics based on genes encoding MCP and ATPase indicated that all five RSIV isolates belonged to RSIV subtype II. The deduced amino acid sequence of the MCP for the amplicons (1362 bp) obtained from RSIV isolates had a length of 453 amino acids. Among these, the amino acid sequences of the RSIV-19, 21, 22, and 23 isolates showed 100% identity, while the RSIV-20 isolate showed 99.78% identity with one residue difference at position 306. As a result of antigenicity analysis based on amino acid sequence, the antigenicity score of the RSIV-20 isolate was 0.6386 and the other RSIV isolates were 0.6365. Additionally, the prediction of their antigenic determinants resulted in a total of 17 identical antigenic plots. When each RSIV was inoculated into rock bream, no significant differences were observed with 100% cumulative mortality in all groups. This study provides data on the potential for genetic variation of RSIV isolated in the same marine area over the past five years, and the antigenicity and pathogenicity results of each isolate are expected to be useful information for selecting future vaccine strains.
This study evaluated the effects of defatted and non-defatted black soldier fly meal (BSFM) as a fish meal replacement in growing red seabream. Four isonitrogenous and isolipidic diets were formulated: 0% BSFM (D1), 5% defatted BSFM (D2), 5% non-defatted BSFM (D3), and 5% defatted + non-defatted BSFM (1:1, D4). A total of 360 growing red seabreams (mean ± SD body weight, 98.9±0.29 g) were equally distributed into 12 circular polyethylene tanks (1,000 L; 30 fish per tank; N=3 tanks per treatment). The red seabream were fed until satiation twice daily for 12 weeks. After 12 weeks, growth, feed utilization, whole-body proximate composition, blood parameters, and immune related parameters were measured. No significant differences were observed in weight gain, specific growth rate, feed conversion ratio, morphological parameters, plasma metabolites, plasma lysozyme, glutathione peroxidase, and superoxide dismutase among the experimental groups. However, immunoglobulin M (IgM) in fish fed D2 and D3 were significantly higher than those in fish fed D1. Additionally, the fish in D2 group showed higher IgM levels than those in the other treatment groups. These results indicate that defatted and non-defatted BSFM could be utilized as a potential feed ingredient for fishmeal replacement for red seabream.
Red sea bream iridovius (RSIV)는 우리나라와 일본의 참돔 (Pagrus major), 돌돔(Oplegnathus fasciatus) 등 주로 돔류에 질병을 유발하는 어류 병원바이러스로 알려져 왔으나, 돔류 이외에도 감수성을 나타내는 어종이 매우 다양하며 아시아를 중심으로 고수온기에 대량 폐사를 유발하는 중요한 원인체로 보고되고 있다. 본 연구는 RSIV에 대한 자연산어류에서 검출동향의 모니터링을 통해 금후 red sea bream iridoviral disease (RSIVD)에 대한 예방학적 접근의 일환으로서 자연산 어류로부터의 RSIV 검출 및 지리적 분포, 보균 어종에 대한 유전학적인 조사를 실시하였다. 조사는 2003년과, 2005년도에 동중국해역 3지점, 서해안 5지점 그리고 남해안 5지점에서 어류를 채집하여 PCR을 이용한 검출결과, 자연산 해산어류 160마리 중 39마리에서 RSIV가 검출되어 24.3%의 높은 검출율을 보였으며 특히 조사된 어류 중 상어목에서 가장 높은 검출율을 보였다. 자연산 어류에서 분리한 RSIV 분리주는 양식산어류에서 분리된 RSIV-K strain과 염기서열 분석에서 97.2%~100%, 아미노산 분석에서 94.7~100% 유사성을 나타내었으며 megalocytivirus Subgroup Ⅲ에 속함을 확인하였다.
본 연구에서는 담수사육 감성돔을 대상으로 수온을 상승시켰을 때, 세포적 스트레스 측면에서 HSP90 및 HSP70 mRNA의 발현 정도를, 신경-내분비적 스트레스 측면에서 혈장 cortisol 및 glucose 농도를 조사하였다. RT-PCR법을 이용하여 생식소로부터 HSP90 (891 bp) 및 HSP70 (465 bp) cDNA 단편을 클로닝 하여, 타 종과 그 상동성을 비교해 본 결과, 감성돔 HSP90은 참돔 HSP90과 99%, 무지개송어 HSP90과 95%, 대서양 연어HSP90과 94%, zebrafish HSP90과 94%로 나타났으며, 감성돔 HSP70은 무지개송어 HSP70과 96%, silver seabream HSP70과 95%, zebrafish HSP70과 95%의 상동성을 나타내었다. 감성돔의 사육수온을 $30\;^{\circ}C$로 상승시켰을 때, HSP90 mRNA는 모든 조직에서 그 발현 정도가 $20\;^{\circ}C$ 실험구에 비하여 $7{\sim}9$배 정도 높았으나, HSP70 mRNA는 생식소에서만 발현하는 것으로 나타났다. 혈장 cortisol 및 glucose 농도는 $20\;^{\circ}C$ 실험구에 비하여 $30\;^{\circ}C$ 실험구에서 유의하게 증가한 것으로 나타났다.
Growth performances of the selected and the non-selected red sea bream (Pagrus major) lines were evaluated up to marketable size. The offspring were mass-produced either from the selected line (the fourth generation of broods from mass selection) or the non-selected line (the second generation of wild broods). Early (59 to 103 days old, trial I) and late growth performances (4 to 24 months old, trial II) were evaluated based on communal rearing in land-based tanks and sea cages, respectively. In trial I, the selected line exhibited significantly higher values in both weight gain (WG) and specific growth rate (SGR) when compared to those observed in the non-selected line (P<0.05). In trial II comprising of three growth phases (4-9, 9-16 and 16-24 months old), selected line showed only slightly improved in WG and SGR at the beginning two growth phases (4-9 and 9-16 months old) (P>0.05), however, displayed significantly higher values at the last growth phase (16-24 months old) (P<0.05). Overall WG and SGR throughout the experiment (4-24 months) were also significantly higher in the selected line than the non-selected line (P<0.05). Consequently, mean body weight of the selected line at 24 months old was 1.4-fold (40.8{\%}\;increased) as compared to that of the non-selected line. This present result indicates that mass selection of this species against the growth traits might be quite effective, and the present advance has important Implication in the productivity enhancement of red sea bream aquaculture.
본 연구는 동물용 생화학 분석장치를 이용하여 분석한 어류 혈액검사 항목 가운데 적용할 수 있는 분석항목의 활용도를 평가함과 동시에 어류 혈액학적 건강진단에 필요한 분석항목의 참고자료를 얻고자 하였다. 경남 지역의 해상가두리 어류 양식장에서 사육중인 육안적으로 보아 건강한 조피볼락 (Sebastes schlegeli), 참돔 (Pagrus major), 돌돔 (Oplegnathus fasciatu) 및 감성돔 (Acanthopagrus schlegeli) 총 522마리를 대상으로 hematocrit 와 간편하게 혈액성분을 측정할 수 있는 FUJI DRI-CHEM 3000 기종을 이용하여 혈장화학적 성분 분석을 실시하였다. Hematocrit (Ht)는 microhematocrit법에 의해 그리고 hemoglobin (Hb), 총단백 (TP), 알부민 (ALB), 알칼리성포스파타제 (ALP), 요소질소 (BUN), 젖산탈수소효소 (LDH), 중성지방 (TG), 총콜레스테롤 (TCHO), 크레아티닌 (CRE), aspartate aminotransferase (AST), alanine aminotransferase (ALT), 포도당(GLU)은 정상범위의 표준자료가 이미 프로그램화되어 장치된 동물용 생화학 분석장치인 FUJI DRI-CHEM 3000 기종을 이용하여 분석하였다. 그 결과, 측정된 Hb와 혈장화학성분의 실제 값들은 본 기종의 하한 및 상한 한계치를 상당히 벗어났다. 본 장비로부터 분석값을 검출하지 못하였던 비율을 살펴보면, ALB 및 ALT는 68~66%, LDH, TP, ALP 및 GLU는 42~21%, TCHO, AST, TG, Hb 및 CRE는 18~3%의 범위를 나타내었으나, BUN의 분석값은 모두 검출한계를 벗어나지 않았다.
