Comparative Analysis of Nutritional Components of Protaetia brevitarsis Larvae Fed Soybean Curd Cake (비지박 함유 먹이원 급이 흰점박이꽃무지 유충의 영양성분 비교분석)

  • To verify the nutritional superiority and safety of Protaetia brevitarsis larvae fed a food source containing soybean curd cake, the nutrients and harmful substances of larvae reared only with fermented oak sawdust and a food source containing soybean curd cake were comparatively analyzed. The crude protein content was 50.0% in the larvae fed with a food source containing soybean curd cake (SCC), which was 1.1 times higher than that of the larvae fed with fermented oak sawdust (FOS) at 44.7%. Potassium, which showed the highest content among macrominerals, was 2.5 times higher in SCC (3,169.6 mg/100 g) than in FOS (1,245.9 mg/100 g). Among trace minerals, zinc showed 12.6 times higher content than FOS (8.2 mg/100 g) in SCC (103.0 mg/100 g). As a result of the analysis of harmful substances, all heavy metals met the edible insect control standards, and pathogenic microorganisms, such as E. coli and Salmonella spp, were not detected. According to the above study, the larvae of P. brevitarsis fed soybean curd cake contain various nutrients, and safety has also been confirmed, so it is judged to be suitable for use as food.

Artificial Cultivation and Antioxidant Activity of Korean Native Mushroom Pycnoporus coccineus (한국산 간버섯의 인공재배 및 항산화 활성 평가)

  • The mycelial growth of P. coccineus strain was good in PDA and YMA, but mycelial growth was low in MEA. Light irradiation during the incubation period affected the pigment formation and density of mycelia. Mushroom of P. coccineus strain was able to produce fruiting bodies in both bottle and bag cultivation, and oak sawdust was found to be the most suitable substrate for spawn culture and cultivation. In artificial cultivation using sawdust medium, fruiting body was grown to the extent that visual observation was possible from the 15th day, and it formed about 5 days fast in the treatment group with low relative humidity. From 40 to 45 days of mushroom development, mature fruiting bodies could be harvested, and the lower relative humidity of the growing room favored mushroom development and growth. Antioxidant activity of fruiting bodies harvested from artificial cultivation showed that ABTS radical scavenging activity of bottle-cultivated and wild fruit bodies were shown at 505㎍/㎖ and 515㎍/㎖, respectively. However, fruiting bodies harvested in bag cultivation were high at 910㎍/㎖. As a result of DPPH radical scavenging activity, all extracts were found to be inactive, exhibiting IC50 value of more than 2,000㎍/㎖ concentration. The ethyl acetate extract of mushrooms obtained from bottle cultivation showed the highest activity with 1,550㎍/㎖ IC50 value. Methanol extract of wild fruit bodies had the highest ABTS radical scavenging activity at the same concentration (10mg/㎖).

Characteristics of Mycelial Growth for Artificial Cultivation in Termitomyces albuminosus (흑피흰개미버섯(Termitomyces albuminosus) 인공재배를 위한 균사배양 특성)

  • The investigation of mycelial growth for artificial cultivation in T. albuminosus determined that within PDA, YM, MEA medium, both KNCAF 9001 and 9002 strains showed the highest mycelial growth in MEA media with 89, 81 mm, respectively, followed by YM, PDA in respected order. Furthermore, optimal mycelial growth of 89 and 77 mm in 9001 and 9002, respectively, at 25 ℃. Mycelial growth rapidly decreased at 30 ℃, but it was confirmed that it survived at 40 ℃. At pH 6, mycelial growth was fast with growth of 89 and 88 mm for both 9001 and 9002, respectively. In the case of 9001, the addition of with cotton seed hull(80%) and rice bran (20%) each other resulted in maximum productivity of 99 mm/10 days. The result differed in order of corn cob(97 mm), oak sawdust(96 mm) and poplar sawdust(93 mm). Ultimately, the result confirmed that addition of rice bran in comparison to bran resulted in higher yield. Even with identical medium component, 9002 showed the same tendency as 9001 but its growth was relatively slower than 9001. The effect of mycelial growth on the moisture content of the medium was best at 101. 94 mm/10 days when the moisture content of 9001 and 9002 was 65%. Mycelial growth rate tended to be slower when the moisture content was higher or lower than 65%.

Selection of Highly Bitter taste Strains on Ganoderma sp. (영지버섯 고미성 균주 선발에 관한 연구)

  • A study was conducted on the characteristics of 11 strains of Ganoderma sp. to select the strains with more bitterness. Among the flat type strains, ASI 7071 and 7091 showed higher bitterness, while among the branched type strains ASI 7074 and 7094 were found to more bitter than other strains. The growth of ASI 7091 was best on Ganoderma complete media(GCM), while ASI 7010, 7048 and 7075 performed best in oak saw dust media. Among the branched type strains the esterase isozyme band patterns were similar. On the other hand among the flat type strains, the esterase isozyme band patterns differed from each other. Out of 11 strains, ASI 7004 was found to have the heaviest fruiting body, ASI 7071 the thickest cap and ASI 7094 the biggest cap.

Cultural Characteristics of Pycnoporus coccineus and P. cinnabarinus (간버섯과 주걱간버섯의 배양특성)

  • Basic studies on the cultural characteristics of Pycnoporus coccineus and P. cinnabarinus were performed. They exhibited $30{\sim}40{\circ}C$ optimal temperature ranges and optimal pH ranges of $5{\sim}6$. Among 6 media, they were good at mycelial growths on PDA, LBA and YMA.P. coccineus grew more than P.cinnabarinus on the same medium. Among 10 sawdust media, they were good at mycelial growth on three oak trees and Alnus hirsuta. However, the sawdust of Castanea crenata was bad at mycelial growth. Among 3 coniferous trees, Larix leptolepis showed better growth than the other trees such as Pinus densiflora and P. koraiensis. The fruit body production P. coccineus was about twice better than P. cinnabarinus on Quercus spp. sawdust cultivation.

  • This study was carried out to select the suitable substrates for bag cultivation of Lentinula edodes. We investigated the optimal additive materials and its mixing ratio in bag cultivation of L. edodes, Sanjo 701 ho. The suitable substrates for L. edodes bag cultivation were oak sawdust as new material plus deffatted corn flour, and corn husk as an additive at the ratio 8:1:1(v/v), as the result of shorter mycelial growth, higher biological efficient, and a higher yield than any other substrates.

  • Cellulose-digesting bacteria were isolated from hindgut of Allomyrina dichotoma (Coleoptera: Dynastidae). The bacterial isolates were identified as Yersinia sp. and Bacillus sp. The addition of the identified bacteria to diet increased growth rate of the cetoniid beetle, Protaetia brevitarsis senlensis (Coleoptera: Cetoniidae), probably by digesting cellulose nutrient contained in the oak tree sawdust diet. An additive of wheat flour at more than 10% to the sawdust diet significantly enhanced growth of P. brevitarsis senlensis. Trimmed branches of apple trees have been disposed in the apple farms and could be used for a diet component of the cetoniid beetle when the cellulose-digesting bacteria were mixed with the derived-sawdust. Resulting manure from mass rearing of P. brevitarsis senlensis contained high organic matters and trace amounts of toxic metals. When the manure were splayed on soil, it was effective as a natural compost and significantly stimulated lettuce growth. This research suggests a model technology to use cellulose-digesting bacteria to use for culturing grub, which results in natural recycles of trimmed branches in apple farms as grub diet, and to use grub manure as a natural compost.

  • Researches were carried out to find the optimal conditions of carbon sources, nitrogen sources and pH for the maximum production of sporophore of Lentinus edodes. Dextrin, aspartic acid and pH 4.0 were the best conditions for yield of sporophore by using replacement culture technique. The production of sporophore was stimulated by addition of 0.8% triacylglycerol in NS medium. Coffee waste was chosen for the best substrate among the poplar, oak, white aspen saw dust and coffee waste. Increased growth of mycelim and yield of sporophore was obsewed by adding tannin up to 0.1% as substrate.

Antioxidant and antitumor activities of ethanol extracts from the fruiting body of Phellinus gilvus grown in oak and artificial sawdust (참나무 원목 및 톱밥에서 재배된 마른진흙버섯 자실체의 에탄올 추출물에 대한 항산화 및 항암활성)

  • The purpose of this study is to examine the elemental compositions, antioxidant and antitumor activity of water, 20%, 40%, 60%, and 80% ethanol extracts obtained from the fruiting body of Phellinus gilvus. In electron donating ability test, the strong activities more than 70% were observed in $80{\mu}g/ml$ of 20%, 40%, 60% and 80% ethanol extracts from the fruiting body of P. gilvus grown in oak and sawdust. The antitumor activity was evaluated by sulforhodamine B (SRB) in terms of cell survival level. The tumor cells (sarcoma 180) were treated with various ethanol extracts (water, 20, 40, 60 and 80%). The results showed that all extracts inhibited proliferation showing a dose-dependent manner against tumor cells.

Cultivation and characterization of commercial strain "Hambak" derived by di-mono crossing in Grifola frondosa (Di-mono 교잡에 의한 잎새버섯 품종 "함박"의 특성 및 재배)

  • A new commercial strain "Hambak" of Grifola frondosa was developed by di-mono crossing between dikaryon of ASI 9031 and monokaryotic strain derived from ASI 9021. It can be cultivated in plastic bottle and bag filled with oak and poplar sawdust substrate which supplemented with 15% rice bran or 10% corn bran. Improvement of cultivation methods was required for shortage of mycelium incubation period and management of humidity in growing room. The optimum temperature of mycelial growth was $25^{\circ}C$ and that of fruiting body development was $15{\sim}18^{\circ}C$. The color of pileus surface was brown and fine pores, whitish to cream-colored, were underside. Primordia formation of Hambak was two days faster than that of Yipsae 1. The yield was $97g/850m{\ell}$ bottle, a bunch of fruiting body was bigger and the quality was better than Yipsae 1.
