Surveys of elecirophoretic variations in isozymes and proteins encoded by 25 persumtive loci were conducted to estimate the degree of genic variation and to assess species recognition for three species of the Family Gasterosteidae (Pisces) in Korea. The average degree of genic variation of Gasterosteus aculeatus was HD = 0.086, HG = 0.084, and P =36% and it was similar to Pungitius sinensis (HD = 0.080, HG = 0.091, P = 29%). But P.kaibarae was less variable than these species (HD = 0.034; HG = 0.034, P = 13%). The average genetic similarities among the populations in each species of Gasterosteus aculeatus pungitius sinensis and P. kaibarae were S = 0.963, S = 0.953 and S = 0.800 respectively.Yongchon and Kyongiu populations of P. kaibarae were genetically remote from the rest of the conspecific populations (S = 0.690). Chae (1988) also observed morphological differences between them. The value of genetic similariiy coefficients of S = 0.606 between P. sinensis and P. kaibarae is low enough to consider as specific level of genetic difference, but moderate rate of hybridization (44%) between them at a synpatric area of Kangnung indicates that isolating mechanism is incomplete. Therefore it is concluded that P. sinensis and P. kaffarae are semispecitic status. Intergeneric genetic similarities between the genus Gasrerosteus and Pungitius were S = 0.052 and this value is the lowest estimate up to date in freshwater fishes.
An area comprising Juwangsan National Park and its adjacent mountains (southeastern Korean Peninsula) is a good model system for testing the effects of physical barriers to gene flows in plant populations. We predicted that plant species consisting of isolated populations are genetically more differentiated than those that are rather continuously distributed. Most populations of Sedum polytrichoides occur in four isolated valleys, and we assessed the genetic variability and structures using twelve allozyme loci in ten populations. We also compared the present results to earlier findings pertaining to the two co-occurring herbs Hylotelephium ussuriense (${\equiv}$ Sedum ussuriense) (growing only in the four isolated valleys) and S. kamtschaticum (rather continuously distributed). We found moderate levels of within-population genetic variation in S. polytrichoides ($H_{e}=0.112$). Estimates of among-population divergence in S. polytrichoides were also moderate ($F_{ST}=0.250$) and, as expected, very similar to that of H. ussuriense (0.261) but considerably higher than the variation in S. kamtschaticum (0.165). An analysis of molecular variance (AMOVA) revealed that S. polytrichoides and H. ussuriense had higher percentages of among-valley variation (19% each) than S. kamtschaticum (4%). Most of this variation, as also indicated by the STRUCTURE program, was due to differences in genetic profiles between the two central valleys. We concluded that the genetic differences observed between species (S. kamtschaticum vs. S. polytrichoides and H. ussuriense) are mainly due to differences in their distribution within the study area.
The aim of this study was to evaluate the genetic diversity and relationships of Ogye populations in Korea. A total of 243 genomic DNA samples from 6 Ogye population (Yeonsan Ogye; YSO, Animal Genetic Resources Research Center Ogye; ARO, Chungbuk Ogye; CBO, Chungnam Ogye; CNO, Gyeongbuk Ogye; GBO, Seoul National University Ogye; SUO) and 3 introduced chicken breeds (Rhode Island Red; RIR, White Leghorn; LG, Cornish; CN) were used. Sizes of 25 microsatellite markers were decided using GeneMapper Software(v 5.0) after analyzing ABI 3130XL. A total of 153 alleles were observed and the range was 2 to 10 per each locus. The mean of expected and observed heterozygosity and PIC (Polymorphism Information Content) value was 0.53, 0.50, 0.46 respectively. The lowest genetic distance (0.073) was observed between YSO and SUO, and the highest distance (0.937) between the RIR and CBO. The results of clustering analysis suggested 3 clusters (${\Delta}K=7.96$). Excluding GBO population, 5 Ogye populations (YSO, ARO, CBO, CNO, SUO) were grouped in same cluster with high genetic uniformity (0.990, 0.979, 0.989, 0.994, 0.985 respectively). But GBO population was grouped in cluster 1 with low genetic uniformity (0.340). The results of this study can be use to basic data for the genetic evaluation and management of Ogye populations in Korea.
This study was conducted to analyze the genetic characteristics of Korean Native Pigs(KNP), and to establish an individual identification system comprising many microsatellite markers located on different pig autosomes. Genotype data from 13 microsatellites typed in 446 animals was used to determine the validation of a method of individual identification in 4 KNP. A total of 112 alleles of the 13 microsatellites were detected and average heterozygosities(polymorphic information content) ranged from 0.286(0.423) to 0.686(0.796) in this study. Comparing the pattern of allele frequency among the KNP, Yorkshire, Landrace and Duroc breeds, there was specific differentiation between populations at multi-allelic loci. The cumulative power of discrimination(CPD) was 99.999% by including 10 microsatellite loci for the individual identification system. The probability that two different individuals incidentally have same genotype was estimated to be $0.36{\times}10^{-9}$. The system employing these 10 markers therefore proved to be applicable to the individual identification of KNP.

The Grey-capped greenfinch (Chloris sinica) is a polytypic species that exhibits complicated geographical variation in morphology. This study provides an intraspecific phylogeographic variability of C. sinica populations in Korea with their morphometric data. The observed morphometric variations were that Ulleung island population was morphologically distinct in bill length and depths as compared to the mainland populations. Phylogenetic relationships among mitochondrial COX1 regions provided evidence for genetic differentiation between Ulleung and mainland populations. However, their genetic distances and nucleotide diversities were very low, highlighting their recent divergence. The needs for additional research is heightened to substantiate if the genetic clines in different localities may arise in C. sinica subspecies, each of which could have different breeding and wintering habitats, distribution patterns, and migration pathways.
The genome size is one of the basic characters of an organism, and it is widely applied in various fields of biology, such as systematics, breeding biology, population biology, and evolutionary biology. This factor was recently highlighted in genome studies because choosing a representative of a plant group having the smallest genome size is important for the efficiency of a genome project. For the estimation of the genome size, flow cytometry has recently been highlighted because it is a convenient, fast, and reliable method. In this study, we report the genome sizes of 15 taxa of Lamiaceae from nine genera distributed in Korea using flow cytometry. Data pertaining to the genome size for all of our species have not been reported thus far, and the data from Agastache, Clinopodium, Elsholtzia, and Isodon are the first reported for each genus. The genome sizes of 15 genera and 39 species were reported to the Plant DNA C-values Database (http://data.kew.org/cvalues/). Scutellaria indica L. has a genome size of 0.37 pg (1C). This is the fourth smallest value among the 98 Lamiaceae taxa in the Angiosperm DNA C-value Database, indicating that this taxon can be used as a reference species in the genome studies in Lamiaceae as a native Korean species. The largest genome size observed in this study is in Phlomis umbrosa Turcz. (1C=2.60 pg), representing the possible polyploidy origin of this species in the family.
The 886-bp sequence of the mitochondrial region encoding the cytb gene was used to identify the origin of the Goryeong (GR) population of Pungitius kaibarae and to characterize genetic diversity and structure among wild populations. The GR population showed the lowest haplotype diversity (Hd=0.000), while the highest haplotype diversity was confirmed at 0.755 among the Goseoung (GS) population. Nucleotide diversity ranged was the highest diversity at 0.00291 in the GS population and the lowest diversity at 0.00000 in the GR population. The GR population was genetically closest to the Pohang (PH) population. The haplotype network confirmed that the GR population was most similar to the PH population. The GR population also clustered with the PH population with high bootstrap support (98%) in a phylogenetic tree. We thus conclude that the GR population is derived from a population similar to the PH population.
This study examined the consumers' willingness to purchase Genetically Modified Organism(GMO) and the factors to distinguish subjects with differential levels of their willingness to buy GMO. Data were collected from 506 male and female adults in Seoul, Daegu and Susan by the self-administered questionnaire. Frequencies, X²tests, F tests and discriminant analysis were conducted to analyze the data by SPSS Windows. The results of this study were as follows: (1) the concerns and the information-seeking behavior about GMO were generally high, but recognition of GMO was low, (2) the willingness to purchase GMO was generally high in that 2/3 of the subjects were willing to purchase it, and (3) the concerns about GMO, the information-seeking behavior, age and sex were the factors to distinguish acceptors from non-acceptors.
Morphological and genetic variation of Cottus populations in Korea were compared to each other in the watershed. Morphological variation was analyzed by meristic and morphometric characters, and by fertilized egg sizes. Genetic variation was assessed by amplified fragment length polymorphism (AFLP) fingerprinting. Cottus koreanus populations of rivers and streams running towards the eastern coasts of the Korean Peninsula were significantly different in morphometric characters from populations in rivers and streams running towards the western and southern coasts of the Korean Peninsula, but did not differ in meristric characters and fertilized egg sizes. Cottus populations in the upper streams-Yangyangnamadae Stream, Jusu Stream and Samcheogosip Stream-were the same in meristic and genetic characters. However, the Cottus sp. population from Baebong Stream is related to C. hangiongensis in meristic characteristics and to C. koreanus in the morphometric characteristics of length of the ventral fin ray and in the fertilized egg size. Pairwise genetic distances assessed by the AFLP method among C. koreanus populations were in the range of 0.110 to 0.221. Genetic distances between C. hangiongensis and C. koreanus populations varied from 0.542 to 0.621, and those between the Cottus sp. population of Baebong Stream and C. koreanus populations from 0.222 to 0.304. The result of the UPGMA dendrogram shows the Cottus sp. population of Baebong Stream was clearly separated of other C. koreanus populations.
This study was conducted to determine the inheritance of the ability of callus formation and plant regeneration in seed cultures of rice. The culturabilities of three Japonica rices,'Chucheongbyeo', 'Nagdongbyeo', and 'Daeribbyeo 1', were higher than those of Tongil type cultivars, 'Milyang 23' and 'Samgangbyeo 'The frequency for callus growth in F₂ populations of the three crosses, 'Milyang 23/Chucheongbyeo', 'Milyang 23/Daeribbyeo 1', and' Samgangbyeo/Nagdongbyeo', revealed a nearly normal distribution. The broad-sense heritability estimated from the ability of callus formation in the crosses were ranged from 83.8% to 90.1%. The frequency distribution of plant regenerability in F₂ population of 'Milyang 23/Daeribbyeo 1' showed a continuous variation. But the segregation mode of plant regenerability from seed-derived callus in the F₂ progenies of 'Milyang 23/Chucheongbyeo' and 'Samgangbyeo/Nagdongbyeo' appeared to fit the expected 3 : 1 ratio for the high and low regenerability. These results suggest that the high plant regenerability of 'Chucheongbyeo' and 'Nagdongbyeo' was regulated by a single dominant gene.
