Properties of Lipases and Palm Oil Assimilating Patterns in Palm Oil Fermentation (팜유발효에 있어서 리파제의 특성과 팜유자화와의 관계)

  • In order to elucidate the patterns of natural oils and fats assimilation by microorganisms, lipases properties of yeast and bacterium strain, Torulopsis candia Y-128 and Acinetobacter calcoaceticus KB-2, which could assimilate palm oil efficiently, were investigated. T candida Y-128 attached palm oil droplets directly, and assimilated unsaturated fatty acid more easily than saturated acids liberated by the action of its lipase. Lipase of A. calcoaceticus KB-2 was extracellular and appeared quickly from the beginning of log phase of growth, whereas lipase of f candida Y-128 appealed intracellular. The lipases of two strains seem to be only enough to utilize the lipid materials for their own growth, without accumulation of lipases in the culture broth. Lipases of the strains have 1 (3-)-positional specificities on triglycerides. The patterns of palm oil assimilation showed that two strains attached droplets of lipid materials directly and split off fatty acids at 1 (3-)-position of triglycerides first, and assimilated the reaction products via fatty acids metabolic pathway.

Antiadipogenic Effect of Korean Glasswort (Salicornia herbacea L.) Water Extract on 3T3-L1 Adipocytes (세포배양 모델을 통한 함초(Salicornia herbacea L.) 물 추출물의 항비만 효과 탐색)

  • As a natural plant ingredients, glasswort (Salicornia herbacea L.) contains various physiological activities, mainly anti-oxidative and anti-diabetic activities. However, only a few studies have been carried out on its anti-adipogenic effect. This study investigated the anti-obesity effects of Salicornia herbacea L. on 3T3-L1 adipocytes. As adipogenesis of preadipocytes to adipocytes involves proliferation and differentiation of cells, we treated three concentrations (125, 250, and $500{\mu}g/mL$) of Salicornia herbacea L. water extracts (SLW) in both pre-processing and post-processing stages. When 3T3-L1 adipocytes were differentiated and dyed with Oil Red O, adipocytes size and the value of relative Oil Red O staining were reduced by all concentrations of SLW in post-processing stage. Following adipogenic differentiation, the concentration of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in the cell supernatant significantly increased upon treatment with $125{\mu}g/mL$ of SLW and further rose at concentrations of 250 and $500{\mu}g/mL$ during post-processing stage. There was no significant difference in glycerol production upon SLW treatment. Leptin production significantly decreased at all SLW concentrations during post-processing stage, whereas peroxisome proliferator activated receptor-${\gamma}$ (PPAR-${\gamma}$) and adiponectin secretions were significantly enhanced. Overall results showed that SLW might have an anti-adipogenic effect via enhancement of TNF-${\alpha}$ production, which causes dedifferentiation and inhibits lipid accumulations in adipocyte. Furthermore, SLW might prevent diabetes and cardiovascular disease, as it reduces leptin secretion and enhances production of both PPAR-${\gamma}$ and adiponectin. However, further research is needed to elucidate the exact mechanism and bioactive compounds of glasswort.

Inhibitory Effects of Sasa borealis on Mechanisms of Adipogenesis (조릿대 에틸아세테이트 분획물의 지방세포에서 분화전사인자 조절을 통한 지방형성 저해 효능)

  • Sasa borealis is a major source of bamboo leaves used for traditional medicine in Korea. Obesity is a serious health problem in industrialized countries that has been implicated in various diseases, including type 2 diabetes, hypertension, cancer, and coronary heart disease. Recent reports have proposed mechanisms to reduce obesity by decreasing preadipocyte differentiation, and proliferation in 3T3-L1 preadipocyte. The preadipocytes play a key role by differentiation into mature adipocytes and increasing fat mass. In this study, we investigated whether ethanol-soluble extracts and ethyl acetate-soluble fractions from Sasa borealis inhibits intracellular accumulation of lipid droplets in a dose-dependent manner in 3T3-L1 cells (an important model system for studying adipogenesis). The down-regulation of PPAR${\gamma}$ and C/EBP${\alpha}$ (key adipogenic transcription factors) were confirmed by the reverse transcription polymerase chain reaction (RT-PCR). Ethyl acetate-soluble fractions from Sasa borealis attenuated the expression of PPAR${\gamma}$ and C/EBP${\alpha}$. These results suggest that Sasa borealis inhibits adipogenic differentiation by regulating adipogenic transcription factors in 3T3-L1 cells. Therefore, Sasa borealis extracts may be a good candidate for the management of obesity.

  • Human plasma low density lipoprotein (LDL) is the major factor of coronary heart disease.But recent studies suggest the normal LDL can be realdily oxidized by oxygen free radicals and not interact with LDL receptors.Lipoprotei particles consist of lipid and protein, and fatty acids are prone to oxidatioin.The fatty acid compositions of LDL from Koreans was compared with that of Westerners.From the results, the raio of unsaturated fatty acids of korean and Westerner approximately 30 and 70%, respectively.which means Westerners are more labile in the lipid oxidation of LDL than Koreams.Normal LDL was incubated with $CuSO_4$ in PBS to lead for the peroxidation of LDL, and it was tested by the detection of TBARS and free radicals.Then, ascorbate, ${\alpha}-tocopherol$ and hyaluronic acid were found to have effects of antioxidants on LDL oxidation.The amount of free radical increased as the extent of oxidation increased.The time course of free radical formation was similar to TBARS.Therefore, determination of free radical by Luminometer was much more convenient than that of TBARS.

  • The purpose of this study is to examine and analysis the problems in managing 'Business Transaction(Danwi-gwaje)' of regional local government, and propose implications. The study points out the problems emerged in the operation process of Business Transaction including creating of Business Transaction and Business Transaction Profile, setting retention period and filing records, and gives concrete example of them. It concludes with presenting improvements and considerations in managing Business Transaction for systematic and efficient records management.

Preventive Effect of Poricoic Acid against Nonalcoholic Steatohepatitis (Poricoic acid의 비알코올성 지방간염 억제 효능)

  • Nonalcoholic steatohepatitis (NASH) is the progressive stage of nonalcoholic fatty liver disease (NAFLD) that highly increases the risk of cirrhosis and liver cancer, and there are few therapeutic options available in the clinic. Poricoic acid (PoA), a component of Poria cocos Wolf, has a wide range of pharmacological activities; however, little is known about its effects on NASH. The preventive effects of PoA on NASH were examined in vivo and in vitro by analyzing triglyceride synthesis, inflammation and fibrosis. In the high fat and methionine-choline deficient diet (HFMCD)-induced NASH mice, PoA reduced the liver weight and the levels of alanine aminotransferase and aspartate aminotransferase compared with non-treated HFMCD group. The staining with Oil Red O and hematoxylin and eosin revealed that PoA administration reduced red staining and the size of lipid droplet. qPCR analysis showed that PoA also reduced the expression of genes related to triglyceride synthesis. Further, immunostaining with CD68 and qPCR analysis revealed that PoA reduced the staining with CD68 and the expression of inflammatory genes induced by HFMCD. Moreover, PoA reduced the staining with sirius red and antibody of α-smooth muscle actin and also reduced the expression of genes related to fibrosis. The treatment of PoA to AML12 cells reduced the increase in triglyceride amount and expression of genes associated with triglyceride synthesis, inflammation and fibrosis. Taken together, our study indicate that PoA has therapeutic effect on NASH through preventing triglyceride synthesis, inflammation and fibrosis.

  • Vegetable oils (triacylglycerols) produced mainly in seeds of plants are used for valuable foods that supply essential fatty acids for humans as well as industrial raw materials and biofuel production. As the demanding for vegetable oils has increased, plant metabolic engineering to produce triacylglycerols in biomass such as leaves has been considered and explored for alternative source of vegetable oils. Leaves are genetically programmed to supply the fixed carbon by photosynthesis to other organs for plant development and growth. Therefore, in order to produce and accumulate triacylglycerols in leaves, one should take account of multiple metabolic pathways such as carbon flux, competition of carbohydrate and fatty acid biosynthesis, and triacylglycerols turnover in leaves. The recent metabolic engineering strategy has showed potential in which the co-expression of three genes WRINKLED1, DGAT1, and OLEOSIN involved in the critical step for increasing the fatty acid synthesis, accumulating triacylglycerols, and protecting triacylglycerols, respectively produced higher amount of vegetable oils in leaves. Developing of genetically engineered plants producing vegetable oil in biomass at non-agricultural lands will be promising to the future success of the field.

Inhibitory Effects of Chrysanthemum boreale Makino on 3T3-L1 Preadipocyte Differentiation and Down-regulation of Adipogenesis and Lipogenesis (산국(Chrysanthemum boreale Makino) 꽃 유래 에센셜오일(Essential oil)이 지방세포 분화 및 지방생성에 미치는 영향)

  • Obesity is associated with an increased risk of many diseases including type 2 diabetes mellitus, hypertension, and hyperlipidemia. The flowers of Chrysanthemum boreale have been used as traditional medicines for the treatment of diseases such as obesity and type 2 diabetes mellitus. This study aimed to evaluate the effect of C. boreale Makino flower essential oil (CFEO) on adipocyte differentiation using preadipocyte cell line 3T3-L1. CFEO at concentrations between 0.1 and $5{\mu}g/ml$ did not affect 3T3-L1 cell viability. A CFEO concentration of between 0.1 and $1{\mu}g/ml$ significantly inhibited lipid accumulation during MDI-induced differentiation in 3T3-L1 cells in a dose-dependent manner, reaching a maximal level at $1{\mu}g/ml$ ($28.94{\pm}2.01%$; approximately 30% of control treated with MDI alone). Western blot analysis revealed that CFEO concentrations between 0.1 and $1{\mu}g/ml$ suppressed the activations of three adipogenic transcription factors in the MDI-stimulated 3T3-L1 cells: peroxisome proliferator-activated receptor ${\gamma}$; CCATT/enhancer binding protein ${\alpha}$; and sterol regulatory element binding protein-1. Moreover, the expressions of lipogenic enzymes, acetyl-CoA carboxylase, and fatty acid synthase were also inhibited by treatment with CFEO between 0.1 and $1{\mu}g/ml$. CFEO may therefore be a promising functional material for obesity prevention.
