Effects of Anthriscus sylvestris Hoffmann Extract on the Biosynthesis of Collagen in Human Dermal Fibroblasts (사람 섬유아세포에서 전호(Anthriscus sylvestris Hoffmann)추출물이 콜라겐 생합성에 미치는 영향)

  • In this study, we investigated the boosting effects on collagen biosynthesis of $Anthriscus$ $sylvestris$ ethanol extract (ASE) in human dermal fibroblasts. To obtain more effective fraction and subfraction for collagen biosynthesis, standard solvent partition and open column chromatography were performed. The EtOH extract, solvent fractions, and 8 EtOAc subfractions were tested for their collagen synthesis capacity by [$^3H$]Proline-incorporation and ELISA assay. ASE increased 25% of total collagen synthesis and 27% of procollagen biosynthesis. The total collagen biosynthesis was increased by EtOAc fraction and E6 subfraction to 28% and 50% respectively. Type I procollagens were also upregulated by EtOAc fraction and E6 subfraction to 30% and 47%, each. Taken together, our data suggest that potential anti-aging effect of ASE on skin is via increasing collagen biosynthesis and effective subtraction is E6 subfraction of EtOAc fraction.

Cosmetic Effects of Dietary Fiber from Mozuku, Cladosiphon novae-caledoniae Kylin (큰실말(Cladosiphon novae-caledoniae Kylin) 부산물의 화장품소재 특성)

  • To investigate the effect of dietary fiber from mozuku, Cladosiphon novae-caledoniae kylin (C. novae-caledoniae kylin) on the skin care, we measured anti-oxidant activity, anti-microbial activities, tyrosinase activity inhibition and elastic activity. B16F10 melanoma cell (MTT assay) were used to measure cell viability. MC and MI exhibited in vitro antibacterial activity against Staphyloccus aureus (S. aureus) and MRSA without antifungal activity. Mozuku extract (MS) showed excellent tyrosinase inhibition effect compared to arbutin as a positive control (to 49% for tyrosine). The wrinkle-improving effect was relatively low. However, wrinkle-improving effect was relatively low. DPPH free radical scavenging activity was 89% in a concentrations at $500{\mu}g/mL$. These results indicate that the mozuku extracts may be an effective cosmetic ingredient for skin whitening.

  • In Korea, the human skin tests to evaluate the anti-wrinkles and whitening effect have been accomplished in accordance with the KFDA guideline. Regarding the data of the visual assessment and machinery evaluation of the results for the human skin test, unpaired t-test have been used in order to compare between the test and the control groups and paired t-test for the comparison of effects for before and after. Descriptive statistics such as frequency analyses was used for the questionnaire evaluation data. In many cases of the European and American clinical test centers, the methodology and the statistical analysis were similar to ours. But, the documentation obtained by repeated application from identical individual has high relation. For this reason, it is desirable to apply RM ANCOVA and RM ANOVA to a visual assessment and machinery evaluation. We suggested that RM ANCOVA and RM ANOVA is the new approach to statistical analysis of human test data of functional cosmetics.

  • This study was carried out to develop a stable plant-derived natural sunscreen (BHC-S) that replaces the artificially synthesized organic sunscreen agents. The natural sunscreen (BHC-S), which is composed of peanut extract, Centella asiatica extract, and Ecklonia stolonifera extract, has the same level of ultraviolet shielding effect as PARSOL MCX-XR (OMC), which is a synthetic sunscreen. and has safety against skin. MultiFunctional effect such as and anti-wrinkle improvement. Thus, it can be used as raw material for natural cosmetics for ultraviolet ray blocking, and anti-aging.

A Study on the Skin Anti-wrinkle Effect of Novel Palmitoyl Tripeptide (신규한 Palmitoyl Tripeptide의 피부 주름개선 효과에 관한 연구)

  • Collagen is the most abundant animal protein in mammals, accounting for about 30 % of all proteins. It is present in connective tissue and contributes to the structural framework of most organs. The tripeptide (glycineproline-hydroxyproline) with the INCI name Tripeptide-29 is main component of collagen type I. The palmitoyl tripeptide (palmitoyl-glycine-proline-hydroxyproline) with the INCI name Paimitoyi Tripeptide-29 is a synthetic material that was designed as a topical agent to stimulate collagen production. We synthesized the palmitoyl tripeptide as a potential anti-wrinkle compound. This compound has been characterized using HPLC. This compound proved, through in vitro tests, to stimulate collagen production and fibroblast proliferation. These results were very promising, so human studies were subsequently performed. We investigated the skin improvement effect of the palrnitoyl tripeptide on human skin by using non-invasive instruments. We measured physiological effects such as skin wrinkles and elasticity after volunteers applied the cosmetic products for 8 weeks. We observed significant improvement in skin wrinkles and elasticity after use of the cosmetic products for 8 weeks. We concluded that the palmitoyl tripeptide had an anti-aging effect on human facial skin.

Evaluation of Antiaging Activity on Lycium chinense Miller. Extracts (구기자 추출물의 항노화 활성 평가)

  • The purpose of this study was to investigate the functionality of Lycium chinense Miller. as a functional cosmetic material. The total polyphenol content in the extract was 17.3 mg/mL, and as a result of confirming cytotoxicity through MTT assay before the enzyme experiment, it was confirmed that it did not affect cell growth at all concentrations below 1000 ㎍/mL. As a result of conducting DPPH radical scavenging tests as an antioxidant efficacy test, it showed 84.97% scavenging ability at a concentration of 1000 ㎍/mL, and showed very excellent antioxidant efficacy in SOD-like activity test with a result of 80.54% at 1000 ㎍/mL. The measured elastase inhibitory activity, the inhibition rate of 50.93% was observed at 1000 ㎍/mL, indicating that the extract of Lycium chinense Miller. was effective in inhibiting elastase. All these findings suggested that 70% ethanol extract of Lycium chinense Miller. was confirmed as a functional cosmetic material by confirming its antioxidant and anti-wrinkle efficacy.

Functional Cosmetic Effect of Sargassum Extracts (모자반추출물의 기능성 화장품소재 특성)

  • There is growing interest in natural cosmetic ingredients as natural cosmetics become popular. As a part of effort to look for natureal cosmetic agent, seaweed Sargassums were tested for functional cosmetic agents. Effective materials were extracted from Sargassum coreanum. Sargassum hemiphyllum, and Sargassum patens by simple hot water extraction. Antioxidation, whitening, anti-wrinkle, UV absorption and anti-inflammation effects were studied for functional cosmetic agents. Sargassum extracts indicated excellent cell viability, strong anti-oxidation effect by DPPH radical scavenging activity and showed significant whitening effect from tyrosinase inhibition. However, effects of antiwrinkle, UV absorption and anti-inflammation were negligible. In conclusion, Sargassum coreanum extracts showed good possibility for anti-oxidation and whitening cosmetic agent.

Beauty food activities of wild-cultivated Ginseng (Panax ginseng C.A. Meyer) ground part (산양삼(Panax ginseng C.A. Meyer) 지상부위의 미용 식품 활성)

  • This study aimed to investigate the beauty food activities of wild-cultivated ginseng (Panax ginseng C.A. Meyer). wild-cultivated ginseng extracts were analyzed for antioxidant, skin whitening, anti-wrinkle effect was measured in water and 70% ethanol extract. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging and 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical decolorization activities of water and 70% ethanol extracts were 16.69 and 2.18% as well as 4.04 and 3.25% at a solid content of $200{\mu}g/mL$, respectively. The antioxidant protection factors (PF) of water and 70% ethanol extracts at a solid content of $200{\mu}g/mL$ were 1.06 PF and 1.09 PF, respectively. Thiobarbituric acid reactive substance (TBARs) were both 96% at a solid content of $200{\mu}g/mL$. As PF and TBARs showed higher activity than DPPH and ABTS, we could know that antioxidant activity in the lipophilic component of wood-cultivated ginseng were superior to water-soluble component of wood-cultivated ginseng. Tyrosinase inhibitory activity was 10.97 and 52.39% in water and 70% ethanol extracts at a solid content of $200{\mu}g/mL$. The collagenase and elastase inhibitory activities as anti-wrinkle effect were 15.71 and 20.43% in water extracts as well as 32.26 and 86.74% in 70% ethanol extract at a solid content of $200{\mu}g/mL$. The results show that anti-wrinkle effect was the best among the other experiments. This extracts from wood-cultivated ginseng, therefore, seems to be a potent beauty food resource against wrinkles.

Pupal Drone Extracts for Anti-wrinkle and Skin-lightening Materials (수벌번데기 추출물의 주름개선 및 미백효과 구명)

  • In this study, we created pupal stage extracts of Apis mellifera L. drones for use in cosmetic materials. The effect of the drone pupae extract (DPE) on HDF cells was assessed for analysis of anti-wrinkle activity by collagen or collagenase gene expression, and the skin-lightening effect was studied by in vitro tyrosinase inhibition and B16F10 melanoma assay; the two cells were found to be non-cellular when the concentration of DPE was 100 ㎍/ml. Albutin concentration (positive control) in the whitening test was set at a capacity of 100 ug/ml and m-melanocyte stimulating hormone (α-MSH). A melanin-producing induction material was set at a concentration of 100 nM, and the expression of collagen type I and MMP1 collagenase was measured using HDF cells. MMP1 expression was seen to reduce in a concentration-dependent manner in treatment with DPE. Inhibiting melanin generation with B16F12 cells indicated a tendency to decrease in the DPE treatment group. Both L-Tyrosine and L-DOPA as DPE were used in an in vitro tyrosinase induction test to demonstrate the effects of tyrosinase suppression on concentrations. The higher the concentration of DPE, the greater the wrinkle reduction and whitening effect. In conclusion, it was found that DPE is an effective smoothing and whitening material by increasing collagen generation and inhibiting collagenase expression and reducing melanin production.

The anti-oxidant, whitening and anti-wrinkle effects of Castanea crenata inner shell extracts processed by enzyme treatment and pressurized extraction (효소처리 및 가압추출 공정을 이용한 율피 추출물의 항산화, 피부 미백 및 주름개선 효과)

  • In this study, the anti-oxidant, whitening, and anti-wrinkle effects of Castanea crenata inner shell extracts processed by enzyme treatment and pressurized extraction were investigated. The Castanea crenata inner shell was first hydrolyzed using celluclast, viscozyme, or hemicellulase. Then, it was subjected to pressure extraction for different durations (30, 60, and 120 min). The yields of the Castanea crenata inner shell extracts processed by different enzyme treatments followed by pressurized extraction for different times are in the range of 12.42-29.80%. The total polyphenol, flavonoid, and tannin contents of the C30m (celluclast enzyme and autoclave extracts at 30 min) extract were 15.48, 10.82, and 15.82 g/100 g, respectively. The total sugar content of the H120m(hemicellulase enzyme and autoclave extracts at 120 min) extract is 61.07 g/100 g. The 1,1-diphenyl-2-pycrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities of the C30m extract at $1,000{\mu}g/mL$ are 89.20, and 81.96%, respectively. The superoxide radical scavenging and ferric-reducing antioxidant power of the C30m extract at $1,000{\mu}g/mL$ are 67.63% and $1,324.79{\mu}M$, respectively. Further, the tyrosinase and elastase inhibition activity of the C30m extract at $1,000{\mu}g/mL$ are 61.32, and 61.06%, respectively. Our results indicate that the Castanea crenata inner shell extracts processed by enzyme treatment followed by pressurized extraction could have beneficial effects on facial skin and they should be considered for use in new functional cosmetics.
