Molecular Breeding of Tobacco Plants Resistant to TMV and PVY (분자생물학적 TMV 및 PVY 저항성 연초 육종)

  • Plant viruses of tobacco including tobacco mosaic virus (TMV) and potato virus Y (PVY) cause severe economic losses in leaf-tobacco production. Cultural practices do not provide sufficient control against the viruses. Use of valuable resistant cultivars is most recommendable for the control of the viruses. However, conventional breeding programs are not always proper for the development of virus-resistant plants mostly owing to the frequent lack of genetic sources and introduction of their unwanted properties. Therefore, we tried to develop virus-resistant tobacco plants by transforming commercial tobacco cultivars, NC 82 and Burley 21, with coat protein (CP) or replicase (Nlb) genes of TMV and PVY necrosis strain (PVY-VN) with or without untranslated region (UTR) and with or without mutation. Each cDNA was cloned and inserted in plant expression vectors with 1 or 2 CaMV 35S promotors, and introduced into tobacco leaf tissues by Agrobacterium tumefaciens LBA 4404. Plants were regenerated in kanamycin-containing MS media. Regenerated plants were tested for resistance to TMV and PVY In these studies, we could obtain a TMV-resistant transgenic line transformed with TMV CP and 6 genetic lines with PVY-VN cDNAs out of 8 CP and replicase genes. In this presentation, resistance rates, verification of gene introduction in resistant plants, stability of resistance through generations, characteristics of viral multiplication and translocation in resistant plants, and resistance responses relative to inoculum potential and to various PVY strains will be shown. Yield and quality of leaf tobacco of a promising resistant tobacco line will be presented.

Selection of Representative Magnaporthe oryzae Isolates and Rice Resistant Gene Types for Screening of Blast-resistant Rice Cultivars (우리나라 벼 도열병균의 대표 균주 및 벼의 저항성 유전자형 선발)

  • Rice blast is one of the most serious disease threatening stable production of rice. Breeding of resistant cultivars has been used as the most effective and useful method to controll rice blast caused by Magnaporthe oryzae. To collect rice blast isolates in fields and test their pathogenicity on new cultivars are important for establishment of new resistant cultivars breeding program of rice. Pathotypes of Korean rice blast isolates have been categorized to Korean differential race system developed in 1985. However, it is little known about genetic background of Korean differential cultivars, so that it is hard to understand for relationship between each pathogen and each host plant at genetic level. In this study, we suggested necessity of a new differential system by analyzing pathogenic responses between 24 monogenic rice lines and 200 Korean rice blast isolates. In addition, we determined the nine representative resistant genes based on the resistance responses of the monogenic lines to rice blast isolates, indexed resistant responses of the monogenic lines to ten representative rice blast isolates and selected 30 Korean representative rice blast isolates proper to Korean system. We think the newly developed differential race system can be broadly used to select resistant cultivars to rice blast in Korea.

Development of Cleaved Amplified Polymorphic Sequence (CAPS) Marker for Selecting Powdery Mildew-Resistance Line in Strawberry (Fragaria×ananassa Duchesne) (딸기 흰가루병 저항성 계통 선발을 위한 분자마커 개발)

  • Powdery mildew (PM) caused by Podosphaera aphanis is a major disease that can result in significant yield losses in strawberry (Fragaria ${\times}$ ananassa Duchesne). For preventing PM, pesticides are usually applied in strawberry. In this study, molecular markers were developed to increase breeding efficiency of PM-resistance cultivars by marker-assisted selection (MAS). An $F_2$ population derived from a cross between PM-resistance 'Seolhyang' and PM-susceptibility 'Akihime' was evaluated for disease resistance to PM and RAPD (random amplification of polymorphic DNA)-BSA (bulked segregant analysis). Among 200 RAPD primers tested, OPE10 primer amplified a 311bp-band present in with 331bp. Sequence alignment performed for searching polymorphisms and six single nucleotide polymorphism (SNP) were found in amplified regions. To develop polymorphic marker for distinguishing between resistant and susceptible, RAPD was converted to cleaved amplified polymorphic sequence (CAPS) marker. Among restriction enzymes associated with six SNPs, Eae I (Y/GGCCR) was successfully digested to 231bp in susceptible. The results suggest that the selected CAPS marker could be used for increasing efficiency of selecting powdery mildew resistant strawberry in breeding system.

Development of Molecular Markers Conferring Bacterial Leaf Pustule Resistance Gene, rxp, using Resistant and Susceptible Cultivars in Soybean (콩 불마름병 저항성 및 감수성 품종을 이용한 rxp 유전자 근접 분자표지 개발)

  • Bacterial pustule (BP) is a leaf disease of soybean that is most common in Korea. Inoculation of 8ra, pathogen strain, to resistant and susceptible cultivars for finding the BP resistance gene (rxp) was much tried but the sequence of the exact gene is not found. This research performed in order to confirm the rxp gene near molecular marker by using the resistant and susceptible cultivars. Soybean BP resistance gene which related to region of near molecular marker could select the resistant cultivar. For the near molecular marker of rxp, reference genomics data available at sequenced Phytozome was used for designing molecular markers. The rxp was mapped between Satt372 and Satt486 on chromosome 17. According to previous study, rxp released in find mapping 7.2 Mbp to 7.3 Mbp on chromosome 17. In this study, we developed 3 random markers near from 6.6 Mbp to 7.3 Mbp on chromosome 17 identified to increase the genetic resolution of the rxp gene region using resistant and susceptible cultivars. Particularly, Rxp17-700 marker was mostly coincided resistance and susceptible genotype to rxp. This result suggests that Rxp17-700 marker will be more tightly linked to rxp gene.

Breeding of a Mid Maturing Watermelon Cultivar, 'Hangyeol' with Resistance to Anthracnose Race 3 (수박 탄저병 Race 3 저항성 중생종 수박 '한결' 육성)

  • The incidence of Anthracnose causing severe damage to the foliage and fruit in watermelon has increased in some major watermelon producing areas in Korea. To develop anthracnose resistant line, 'AU-Producer' having resistant gene to anthracnose was selected from germplasm and crossed with high quality line '920533'. Following the initial cross, backrossing and disease screening were performed to select resistant lines that produced high yields with excellent quality fruit. As a results of these procedure, a mid maturing watermelon line, 'Hangyeol' with resistance to anthracnose (Colletotrichum orbiculare) race 1 and 3 was developed at the National Institute of Horticultural and Herbal Science, Rural Development Administration (RDA). It has red flesh and commonly produces fruit with clear stripes on skin between 5.3 to 10.1 kg. Average soluble solid contents are ranged from 9.8 to 11.8$^{\circ}$Bx. The yield and quality of 'Hangyeol' is comparable to or better than those harvested from the popular commercial cultivars. The achievement of this experiment could contribute to provide the resistant parents in an anthracnose resistance breeding program in watermelon.

Analysis of QTLs Related to Resistance to Brown Planthopper in Rice (DH 집단을 이용한 벼멸구 저항성 연관 QTLs 분석)

  • This study was conducted to develop a japonica-type rice cultivar with brown planthopper (BPH) resistance using DNA markers. A doubled haploid (DH) population consisting of 120 pure-lines was established by anther culture of $F_1$ hybrids between 'Samgang', a Tongil type BPH resistance cultivar, and 'Nagdong', a japonica cultivar. To determine the map position of genes responsible for BPH resistance in rice, a genetic map was constructed based on 120 DH lines. A total of 162 molecular markers were classified into 12 linkage groups, covering 1,884 Kosami centimorgan (cM) with an average of 11.6 cM. Five QTLs (qBPR3, qBPR6, qBPR7, qBPR8, and qBPR12) associated with BPH resistance were identified and mapped on chromosomes 3, 6, 7, 8, and 12, respectively, using the genetic map constructed in this study. To analyze the relationship between BPH resistance and agronomic traits, a total of eight QTLs related to the agronomic traits were detected on 12 rice chromosomes. In an analysis of relationships, three QTLs (qBPR3, qBPR7, and qBPR8) showed a linkage with tested agronomic traits. A QTL (qBPR3) located on chromosome 3 (RM282-3023) was closely linked to culm length (qCL3). The QTL (qBPR8) for BPH resistance on the short arm of chromosome 8 also overlapped the region detected in culm length (qCL8).

Evaluation and Selection of Breeding Lines of Pepper Developed by Incorporation of Resistance to Phytophthora capsid into Local Cultivars of Gyeungbuk Province (고추 역병 저항성 도입 경북지역 재래종 육성계통의 평가와 선발)

  • Lines bred for resistance to Phytophthora capsici by incorporation of resistance to P. capsici in PI201234 or CM334 into 'Subi' and 'Chilseong', land races in Youngyang, and 'Punggak', a land race in Cheongdo in Gyeungbuk province, and lines bred for fortification of one of them above with resistance to viral complex, and tolerant selections from another landrace collection from Punggak (KC268) were evaluated for resistance to P. capsici by inoculation at seedling stage. Almost all the breeding lines showed high level of resistance to P. capsid and selections from KC268 showed tolerance or moderate resistance to P. capsid. The selected plants were grown in a net cage in an outdoor nursery for seed production. Utilization of the lines in breeding was discussed.

Resistance Degree of Cucurbits Cultivars to Colletotrichum orbiculare (탄저병(Colletotrichum orbiculare)에 대한 박과작물의 저항성)

  • Anthracnose disease caused by Colletotrichum orbiculare, induces severe damage to cucurbits worldwide. Resistance of 112 commercial cultivars of cucurbits to C. orbiculare was evaluated. Seedlings of each cultivar at 2- to 3-leaf stage were inoculated with C. orbiculare KACC 40809 by spraying spore suspension of the fungus at a concentration of $4.0{\times}10^5$ spores/mL. Among the 36 cultivars of cucumber, 'Asiastrike', 'Tongilbaedadagi', 'Daeseon', 'Cheongrokmatjjang', 'Nokyacheongcheong', and 'Asianogak' were moderately resistant and the others were susceptible. All the tested cultivars of melon (33) and watermelon (4) showed highly susceptible response to C. orbiculare. On the other hand, the squash cultivars (17) represented less susceptibility to the fungus than the other cucurbits. Of the squash cultivars, 'Gammirak' and 'Teotbat' were resistant and 12 cultivars were moderately resistant. Among the rootstocks for cucurbits, ten cultivars including 'JjeuyakaEX', 'Nunbusyeo', 'Union', 'RS111', 'Ganggeuntoza', 'Hwangjaetoza', 'NO.8', 'Shintoza', 'Bulpaetoza', and 'Newtype' showed high resistance to the anthracnose pathogen. From the results, the resistant cultivars could be used as sources of resistance to cucurbits anthracnose (C. orbiculare) in the future breeding programs.

  • Natural infection of purple stain seemed to be higher in early maturing lines comparing to late maturing group. It was positively correlated with length of the period from flowering to maturity and significantly associated with weather conditions during the maturing period.
