An Improved Method for Screening Rice Cultivars with Field Resistance to Bacterial Leaf Blight (벼흰빛잎마름병에 대한 포장 저항성의 새로운 검정법)

  • To improve methods of screening rice cultivars with field resistance to bacterial leaf blight, testing plant inoculation and neighbor plant inoculation were compared by using 33 rice cultivars. In the testing plant inoculation method, field resistance was evaluated by measuring the leaf areas diseased on the new leaves expanded after the inoculation. Varietal differences in field resistance were recognized more clearly by the testing plant inoculation method than by the neighbor plant inoculation method. Highly significant correlation was observed between the results of the two methods. Some rice cultivars such as, Seomjin, Hangangchal, Taebaeg, Samgang, Milyang 42, Asominori, Java 14, Chugoku 45 and 70X-46 showed remarkable field resistance to bacterial leaf blight. The testing plant inoculation method appeared desirable for screening rice cultivars for the qualitative and field resistance to bacterial leaf blight because of using less labor and less field area than neighbor plant inoculation.

  • This study was conducted to develop a easy seedling method to test large amounts of barley and wheat cultivars for resistance of powdery mildew. In addition, we also examined the resistance of genetic resources that have been used in barley and wheat breeding programs in Korea. This seedling test used paper bag with seeds can be completed within three weeks. to-day seedlings were available to inoculation and 8 days was needed for result reading. This method can test at least 180 collections at one time. we can identify the two resistant types by leave symptoms showed non infection and necrotic spots. Among the 79 Korean barley cultivars, only two cultivar, 'Sangrokbori' and 'Dajinbori' were resistant and 'Jejubori' showed moderate resistant. There was no resistant in hulless barley and wheat cultivars. It was same results in comparison of earlier resistant reports in field test. We confirm that this method could using in test of powdery mildew resistance in barley and wheat. Among the 1,401 genetic resources using in Korean breeding program, malting barley has more resistant collections comparing to 796 hulled and hulless barley and 273 wheat germplasms.

  • To find out highly resistant gene source to black rot fungus in sweetpotato varieties, a screening test was carried out with inoculated tubers and sprouts in 1976 and 1977. Fifteen varieties out of 165 and 54 varieties out of 161 were highly resistant to black rot in the tuber and sprout tests. respectively. The sprout test showed bigger variation from year to year compared to tuber test. Varieties highly resistant to black rot in both tuber and sprout tests were Suweon # 59, #81, #90, Norin #17, #23. Chilship-il cho, Sachun Jong #36, Hamkao, and Kandee.

  • 한국에서 재배되고 있는 벼 품종의 도열별에 대한 지속저항성 품종을 조기에 선발, 보급할 수 있는 방법을 보색하기 위하여 1985년부터 1994년까지 온실 유묘검정, 밭못자리 및 포장검정을 통하여 저항성 정도를 평가하고 재배면적과 재배기간을 참고로 하여 지속저항성 품종을 선발하였다. 도열병에 대한 저항성 품종으로는 다양한 레이스에 침해되면서 개체당 병반수 20개 이내, 밭못자리검정에서는 평균 발병정도 5이하, 본답에서의 잎도열병은 병반면적율 0.5%이하, 이삭도열병 발생은 이병수율 5% 이내로 병발생이 비교적 적은 섬진벼, 팔공벼 및 동진벼를 선발하였고, 지속저항성 품종으로는 재배기간이 10년 이상 유지되면서 재배면적은 전체재배 면적의 20% 이상을 차지하고 있는 품종 중 장기간 동안 저항성을 발현하는 동진벼를 선발하였다.

  • The present study was carried out to determine appropriate growth stage for evaluating resistance to septoria brown spot in field and to search resistance sources from soybean germ plasm. Disease severity expressed by log $\frac{x}{1-x}$ was different with soybean genotypes and vertical progress of the disease was related to the diseased leaf area. Correlation between diseased leaf area and the area under septoria brown spot disease progress curve (AUBC) was highest at full blooming stage, indicating a reasonable stage for measuring the disease severity to evaluate resistance in field. There was no lines highly resistant to the disease among 1,428 native soybean lines tested.

  • The objective of this research was to develop a screening system for selection of grape downy mildew (Plasmopara viticola)-resistant grape cultivars or seedlings under greenhouse condition. Inoculum concentrations ranging from $10^4$ to $10^5spores/mL$ for screening of resistance were evaluated and $5{\times}10^4spores/mL$ was optimum. Of the tested 14 grape cultivars, Campbell Early and Niagara were resistant to grape downy mildew, Sheridan and Cheongsoo were moderate resistant, while Kaiji, Red Queen and Ruby Okuyama were susceptible under both greenhouse and field conditions. Vitis vinifera grape cultivars were more susceptible to grape downy mildew than V. vinifera-labrusca hybrids and V. vinifera-labrusca-aestivalis hybrids. In V. vinifera-labrusca hybrids, tetraploid grape cultivars were more susceptible than diploid culivars. The evaluated results of grape downy mildew resistance under greenhouse condition were in accordance with those of field condition. Results of this study indicated that both greenhouse and field procedures could be used to screen grape cultivars and seedlings for downy mildew resistance.

Establishment of the Text Method for Evaluating the Fideld Resistance of Rice Varieties to Rice leaf Blight (벼 흰잎마름병 포장저항성 검정방법 체계 확립+)

  • To establish the simple and practical test method of estimating resistance of rice varieties against rice leafblight, trials were made to correlate among the four test methods such as true resistance test,secondaryinfection test, secondary epidemic test and field test. The results obtained through the experiment on 25 ricevarieties and 3 pathogenic isolates designated to HB9O11, HB9022 and HB9033 can be summarized asfollows. 1 .Based on the results of true resistance test, 25 rice varieties can be classified into four groups:1 )Samgang variety group ;resistant to HB9011, 13 varieties, 2)Pungsan variety group ; resistant to HB9011 and HB9022, 5 varieties, 3)Samgang variety group ; resistant to all three isolates, 2 varieties, 4)Unbongvariety group: sensitive to all three isolates, 12 varieties. 2. The responses of rice varieties to isolates showed some discrepancies among on the test methods. These examples were found in 3 varieties including Yeongdeog varieties to HB9011, 3 varieties including Taebaeg variety to HB9022 and Taebaek vaieties to HB9033. 3. Correlation coefficiences between the secondary infection test and the secondary epidemic test for HB9011, HB9022 and HB9033 were 0.972, 0.894 and 0.919, respectively. It suggests that the two methods are not significantly different so that one of the two methods are not significantly different so that one of the two methods can be omitted from resistant test without affecting the result. 4. Between the true resistance test and the field test at the disease common area, there were no significant correlations. Unbong, Chucheong and Yeongdeog varieties are appeared as resistant varieties in the true resistant test, but their responses in the field test were different and appeared as sensitive varieties. 5. The disease index was used to express theresults from four test methods. The disease index was calculated as the sum of each numerical values of theresults from the four test methods by giving the same weights(0 to 25)to each test method. If the disease index for certain variety is less than 15, then the variety is considered to be resistant. 4 varieties such as Seohae, Hwajin, Yeongdeog and Pungsan varieties-disease indices were less than 15 were selected as field resistance varieties.

  • Fusarium head blight (FHB) is a severe disease problem that affects the quality and yield of barley grain. The evaluation of FHB resistance is difficult because environmental conditions greatly influence FHB infection and development. The objectives of this study were to: 1) establish an efficient screening method for selecting resistant barley to FHB, 2) compare FHB severity between the cut-spike method and pot-plant method for development of mass screening, and 3) estimate FHB resistance for barley germplasms. Barley cultivars and lines were evaluated for reaction to FHB in controlled-greenhouse condition. Spikes were spray-inoculated with a suspension $(5.0\times10^5\;macroconidia\;mL^{-1})$ of Fusarium graminearum SCK-O4 strain, and then kept in a greenhouse at $18-25^{\circ}C$ with $80-100\%$ relative humidity. Inoculation were employed at 3 different heading growth stages (heading date, three days after heading, and five days after heading). The inoculation was performed in 2 consecutive days in order to avoid escapes. The inoculated plants were maintained in the greenhouse at 4 different free moisture periods (1, 3, 5, and 7 days). The percentage of FHB severity was scored from 0 to 9 according to the rate of infected kernels per spike, and three spikes were evaluated per replication with 3 replicates. There were significant differences of FHB severity depending on the different free moisture periods, but not by the inoculation at different heading stages. The optimum evaluation point of FHB severity in the greenhouse condition was on the 7th day under free moisture condition after inoculation at the heading date. Infection level in cut-spike method highly correlated with that in pot-plant method. This suggested that cut-spike method is useful in evaluating of FHB resistance in barley. Six cultivars, such as Jinkwang, Buheung, Atahualpha 92, Chevron-b, Gobernadora-d, and MNBrite-c, were selected as resistant varieties to FHB. Correlation coefficient for the FHB severity evaluated by the pot-plant method between two seasons was 0.794, indicating the stability and accuracy of the screening method.

Development of a Simple and Effective Bioassay Method to Evaluate Resistance of Watermelon Plants to Fusarium oxysporum f. sp. niveum (수박 덩굴쪼김병에 대한 간편 저항성 검정법 확립)

  • Root-dipping inoculation method has been widely used to determine the resistance of watermelon to Fusarium oxysporum f. sp. niveum causing Fusarium wilt. Although this method leads to the precise results of plant disease responses, more rapid and efficient assay methods have been still required because the root-dipping inoculation method is labor-intensive and time-consuming. In this study, we established a simple and effective bioassay method based on the comparison of various inoculation methods and growth conditions. To develop the system, the occurrence of Fusarium wilt on four resistant and susceptible cultivars was investigated by four different inoculation methods, root-dipping, scalpel, tip and soil-drenching methods. Of these inoculation methods, scalpel method resulted in clear plant disease resistance responses with the simplicity. With the use of scalpel method, we also explored the disease development of the cultivars depending on inoculum concentration, growth stage of seedlings, and incubation temperature after inoculation. Furthermore, we found that the resistance degrees of 23 cultivars derived by scalpel inoculation method were similar to the results by root-dipping method established previously.

  • Sixteen recommended soybean cultivars in Korea were tested for resistance to Heterodera glycines. The nubmer of females which developed on roots of cultivars were fewer on 'Hwanggeum kong' and greater on 'Geumgangdaelib'. But there was no resistant cultivar to Heterodera glycines. Four field populations of Heterodera glycines tested for ability to reproduce on five host differentials. Geographical differentiations of H. glycines were newly founded in Korea. These were identlified to race 1 at Hwasun, race 5 at Yangsan and Suweon and Unknown race(or race C) at Seonsan.
