Analysis of Genetic Diversity and Structure of Prunus salicina Lindl. Populations in Adjacent Area (자두나무(Prunus salicina Lindl.) 접경지역 집단의 유전 다양성 및 구조 분석)

  • 자두나무(Prunus salicina Lindl.)는 세계에서 5번째로 많이 생산되는 과실로, 한국에서 재배하는 자두나무의 기본종은 중국 양쯔강 유역에서 기원했다. 2016년 과수용 자두나무와는 다른 자두나무가 양구에서 발견되었다. 양구군, 인제군, 고성군 일대의 자두나무 개체군의 유전다양성 및 집단 구조 분석을 통해 본 자두나무 개체군의 유전다양성 및 개체군 구조를 확인하고자 했다. 과수용 재배종을 포함한 시료를 채취하여 GBS 분석을 진행했고 주성분 분석과 STRUCTURE 분석을 통해 개체군간 유전적 구조를 확인했다. 재배종의 유전형이 다른 개체군에서도 나타나는 것으로 보아 유의한 유전적 분화가 일어났다고 보기 힘들었다. 하지만 고성군 고진동계곡 등 DMZ에 인접한 집단이 분계도에서 재배종 개체군과 가장 유전적 거리가 있는 것으로 나타났다. 하지만 본 분석으로는 야생집단으로 발견도니 자두나무의 실체와 기원을 단정짓기에는 부족한 것으로 보이며 외군 추가 및 더 많은 시료를 확보하는 등 추가 조사와 분석이 필요하다.

Effect of Edible Coatings on Softening of the Stored Immature Plums (식용코팅이 미숙 자두의 연화에 미치는 효과)

  • Immature plums(Prunus salicina L.) were coated with edible coatings, calcium-added methyl cellulose(CaMC) and hydroxy propyl methyl cellulose-15(CaHPMC-15). Weight loss and respiration rate of immature plums coated with CaMC and CaHPMC-15 were lower only at the earlier storage period. The titratable acidity of plums was decreased and pH was increased during storage, Soluble pectin, pectinesterase, and polygalacturonase activity as the marker of softening were commonly increased with storage time, but those increases were lower in CaMC and CaHPMC-15-coated plums. Mechanical firmness became significantly soft during storage, while the coated plums were remained harder. As a result of sensory evaluation, softening of plum tissue was found during storage, but the coated plums had the harder texture. These data suggest that the application of CaMC or CaHPMC-15 coatings into immature plums can reduce the postharvest softening during storage.

Effect of Plum Wine on the Lipid Metabolism and Lipid Peroxidation of Rats (자두와인의 섭취가 흰쥐의 지질대사 및 지질과산화에 미치는 영향)

  • The effect of plum wine on lipid metabolism and lipid peroxidation in rats with chronic ethanol consumption was evaluated. Sprague-Dawley rats were randomly divided into the following 4 groups; water (NC), alcohol control (AC), low concentration plum wine (LP) and plum wine (P). The alcohol content of experimental drinking water of AC, LP and P were 6%, 6% and 12%, respectively. Animals were fed AIN-76 diet and experimental drinking water for 4 weeks. LP group showed significantly decreased liver weight per 100 g body weight, the levels of total cholesterol and atherogenic index in plasma whereas the ratio of HDL-cholesterol to total cholesterol was significantly increased in comparison to that of AC group. LP and P groups showed significantly decreased total lipid, total cholesterol in liver tissue, AST and ALT activities of plasma as compared with that of AC group. LP and P groups showed a significant decrease in the level of plasma lipid peroxidation products and LP group showed a significant decrease in the level of liver lipid peroxidation products as compared with that of AC group. These results suggest that supplementation of low concentration plum wine may exert more beneficial effects than pure alcohol beverage on lipid metabolism and lipid peroxidation products in chronically alcohol-treated animals by improving lipid profiles in plasma and liver tissues and decreasing plasma and hepatic lipid peroxidation product.

  • In this study, alcohol fermentation condition for production of plum wine was investigated and further, sensory evaluation and nutraceutical properties of the plum wine and liquors were also determined. Ethanol was produced maximally when 5% Saccharomyces cerevisiae was added in red plum juices and fermented at $25^{\circ}C$ for 5 days. Among the red plum wine and various kinds of plum liquors, red plum wine showed the best acceptability and its ACE inhibitory activity, fibrinolytic activity and antioxidant activity were also excellent.

  • In 1998, bacterial shot hole of peach (Prunus persica) and Japanese plum(Prunus salicina) was found in Naju and Milyang. Five isolates of bacteria isolates from the diseased leaves and fruits of peach and Japanese plum were classified into genus Erwinia and Xanthomonas on diagnostic characteristics. Of five isolates, two were identified as X. campestris pv. pruni, three as E. nigrifluens. E.nigrifluens is the first description of bacteria which causes the disease on peach and Japanese plum in Korea. the symptoms caused by E. nigrifluens were hardly distinguished from those caused by X. campestris pv. pruni. In addition, it was observed that two pathogenic bacteria were isolated from most of naturally infected plants at the same time. from the reason mentioned above, we proposed to use a single common name \"bacterial shot hole of peach and Japanese plum\" for the both bacterial diseases, hereafter.

Molecular Diagnosis of Grapholita molesta and Grapholita dimorpha and Their Different Occurrence in Peach and Plum (복숭아순나방과 복숭아순나방붙이의 분자동정법 개발 및 복숭아와 자두에서의 발생차이)

  • The plume fruit moth, Grapholita dimorpha Komai, a fruit tree pest occurring in the northeast Asia, was firstly reported to infest apple in Korea in 2009, but its direct damage to other fruit trees has been poorly studied. In this study, we investigated shoots and fruits of both peach and plum trees and compared their damage rates by G. dimorpha to those by G. molesta, a congeneric species. In order to discriminate the two moth species, we developed a molecular diagnosis method using species-specific primer sets on different PCR conditions and distinguished the two species collected from the damaged shoots or fruits. The shoots and fruits of peach were infested mostly by G. molesta. However, in plums, the shoots were damaged by G. molesta and the fruits mostly by G. dimorpha. In addition, these two species showed a clear difference in host preference in fruit damage, where 92.5% of the Grapholita moths collected in peach fruits were identified as G. molesta, but 97.0% of the moths in plum fruits were G. dimorpha. The difference of the damage between the two fruit trees may give important information for monitoring of the two moth species in these orchards.

Effect of Calcium-Added Cellulose Coatings on the Firmness of Plums (칼슘을 첨가한 셀롤로우스 코팅이 자두의 경도에 미치는 효과)

  • Plums (Prunus salicina L.) were coated with calcium-treated methylcellulose (CaMC) and hydroxypropylmethylcellulose-15 (CaHPMC-15). Respiration rate, weight loss, pH and titratable acidity, texture, and sensory characteristics were compared during 8-day storage period at $30^{\circ}C$. Weight loss and respiration rate of plums coated with CaMC and CaHPMC-15 were lower than those of control plums during storage while pH increased and titratable acidity slowly deceased during storage. Mechanical firmness was significantly deceased during storage, in which CaMC and CaHPMC-15-coated plums were harder than the control. Amount of soluble pectin, and pectin esterase and polygalacturonase activities as the markers of softening commonly increased with storage, but both CaMC and CaHPMC-15-coated plums were not found those increases. Sensory evaluation showed that the flesh softening of plums decreased during storage, and that in the coated plums exhibited the harder flesh. In conclusion, introduction of CaMC and CaHPMC-15 coatings on plums decreased respiration rate and contributed to delay of flesh softening, and the significant difference in postharvest storage quality of the both plums were not found between CaMC and CaHPMC-15 coatings.

자두(Dried Plums)를 첨가한 White sausage의 물성 및 저장성에 관한 연구

  • 본 연구는 파슬리 등의 야채를 첨가하여 제조되는 화이트소시지의 물성 및 저장성에 미치는 자두 분말의 효과를 조사하기 위해 실시되었다. 물성의 변화로서 pH는 자두의 첨가율이 증가함에 따라 상대적으로 감소 하였는데 3% 첨 가시 전 저장 기간 중 가장 낮게 ($pH\;5.11\;{\sim}\;4.44$) 나타났다. 육색(meat color)은 자두의 첨가량에 비례하여 적색도(redness)를 나타내는 a-value에서 높게 나타났으며 명도 (light ness)를 나타내는 L-value에서는 상대적으로 낮은 수치를 보여주었다. 전단력(Shear stre ngth) 측정에서는 저장 기간 중 자두 분말의 첨가량에 비례 하여 근소한 차이를 나타냈는데 저장 7일째에서 가장 높았다. 자두 분말 첨가구 에서의 다소 높은 전단력의 결과는 보수력 (WHC) 측정 에서도 같은 경향을 나타내었는데 자두 분말의 증가량에 따라 3% 첨가구가 대조구 (0%)보다 낮게 나타나 약한 보수성을 보여 주었다. 저장성에 미치는 결과로서 총세균수의 측정결과 저장 3일이 경과한 구간에서 Sorbic acid (0.07%)의 첨가구와 자두 분말(3%) 첨가구 에서 비슷한 항균 효과를 나타내었다. 이 같은 동일한 수준의 항균 효과는 저장 9일째에도 같은 경향으로 나타났다. VBN 측정에는 저장 기간에 따라 증가된 수치를 나타냈는데 저장 7일 및 8일, 9일째 측정 결과는 3% 자두분말 첨가구와 0.07% Sorbic acid 첨가구 와 비슷한 수준을 나타내어 동일한 저장효과를 보여 주었다. 관능검사 실시 결과 총체적인 풍미에 있어서는 자두 분말 첨가구 1.5%가 가장 우수 했으며 다음으로는 3%, Sorbic acid 첨가구 및 공시료 순으로 나타났다.간에 유의차는 나타나지 않았다.pm을 spiking한 후 SPE상에서 SCX(Strong cation exchange column)을 통한 clean-up과정을 거친 후의 STP의 limit of quantification(LOQ)는 약 0.44ppm이었으며, 이에 대한 회수율은 89.7${\pm}$2.3%(n=6)를 나타냈다. 실제 CODEX에서 권장한 우유의 MRL이 0.6ppm인 점을 감안하면 CODEX권고치에 도달할 수 있는 것으로 판단되었다. 따라서 본 연구에서 개발 된 시험법은 지금까지 국내적으로 STP에 대한 시험법이 확립되어 있지 않은 것으로 이와 아울러 간편한 parallux와 병용해 STP에 대한 정량 및 정성 분석을 유도체화 장치 및 형광검출기를 이용해 잔류항생물질 STP에 대한 분석시험법을 개발하였다.화와 네트워크 조직망 구축을 위한 지역중심의 복합 생활문화 공간이 필요할 것이며, 이를 촉진키 위한 mentor academy시스템 접근을 통해 점점 고령화되어 가고 있는 재래시장에 대해 차별적 특성이 반영된 종합적이고 체계적 접근 방법연구가 필요하다./TEX> 자체기술개발, 선진기술 도입, 산업간 및 산업내 기술확산, 국제기술협력 ${\rightarrow}$ 기술혁신의 촉진 ${\rightarrow}$ 총요소생산성과 기업경쟁력(자원 및 역량, 프로세스 경쟁력, 품질경쟁력, 시장경쟁력, 고객성과, 시장성과, 재무성과)의 제고 ${\rightarrow}$ 가격경쟁력(임금, 금리, 물류비용, 환율 등)과 비(非)가격경쟁력(디자인, 에프터서비스, 품질, 운송 등)의 제고 ${\rightarrow}$ 국가경쟁력의 제고 ${\rig

  • A larval attack leading to severe loss to fruit production of Japanese plum (Prunus salicina Lindl.) was reported in the orchards in Gwangyang, Jeonnam province, South Korea during the spring seasons of 2018 and 2019. As the result of investigation, the damage was identified to be caused by Monocellicampa pruni Wei of the family Tenthredinidae. In this study, we present the first report of this pest in Korea and describe the damage caused by the larvae as well as taxonomic information by illustrating the characteristics of male genitalia.
