홍화의 부위별 화학성분과 DPPH radical 소거 활성

  • 홍화의 식품재로로서 이용성을 높이고자 홍화의 부위별 성분분석 및 DPPH radical 소거 활성을 조사하였다. 단백질은 어린싹 부위에 28.39%, 지방은 씨 부위에 20.47%로 많이 함유되어 있었다. Glucose는 어린싹에 1,253.6 mg%, fructose는 이린싹에 970.7 mg%, sucrose는 꽃봉오리 부위에 912.0 mg%로 많이 함유되어 있었다, Succinic acid는 꽃잎 부위에 2,795.3 mg%, malic acid는 잎과 어린싹 부위에 각각 2,054.8 mg%와 934.2 mg%로 많이 함유하고 있었다. 무기질로는 K이 잎과 어린싹 부위에 각각 2,826.8 mg%와 l1999.8 mg%, Ca 또한 잎과 어린싹 부위에 각각 2,613.6 mg%와 1160.9 mg%로 많이 함유하고 있었다. 불포화지방산인 linoleic acid는 어린싹과 씨 부위에 각각 80.01%와 78.21%로 가장 많이 함유하고 있었다. 총페놀함량은 꽃잎, 어린싹, 잎 부위에 각각 5.8%, 4.4% 및 2.5%, 총플라보노이드 함량은 꽃잎, 어린싹, 잎 부위에 각각 4.7%, 6.5% 및 2.0%로 많이 함유하고 있었다. Serotonin(5-bydrnxyoyptamine, 5-HT)화합물인 serotonin-I은 홍화씨 부위에 147.7 mg%, serotonin-II 또한 홍화씨 부위에 155.4 mg%를 함유하고 있었고, flavonoid화합물인 acacetin도 홍화씨 부위에 116.5 mg%을 함유하고 있었다. 또한, luteolin은 어린싹 부위에 388.3 mg%, luteolin 7-glucoside은 잎 부위에 692.3 mg%로 많이 함유하고 있었다. DPPH radical 소거 활성은 꽃잎과 잎의 80% 에탄을 추출물이 114.2%와 113.6%의 높은 활성을 나타내어 기존의 합성항산화제인 BHA 100 ppm 농도의 88.05%의 활성 보다 높은 항산화 활성을 가지고 있음을 확인하였다.

Characterization of Plants Induced by in vitro Culture of Leaf Blade-segments in a Variegated Tobacco (Nicotiana tabacum L. cv. BY-4) (Variegated 담배 (Nicotiana tabacum L. cv. BY-4)의 잎 절편 배양에 따른 재생 식물체의 특성)

  • Plantlets derived from leaf blade-segment culture of a variegated tobacco (Nicotiana tabacum L. cv. BY-4) that was induced by a heavy-ion ($^{14}N$) beam irradiation to proembryos, were characterized. When explants from both white and green sections of leaves of the variegated plant were cultured on MS medium containing 0.1 mg/L NAA and 1.0 mg/L BAP, the white sections yielded only white shoots, whereas the green sections generated approximately 47.2% green, 37.4% white and 15.4% variegated shoots. In the F1 generation of a green tobacco derived from the leaf blade-segment culture, the segregation ratio of green to white was 1,651:54. Furthermore, reciprocal crosses showed that all of the progenies was green, indicating that the variegation is not maternally inherited. When the signal intensity of photosynthesis genes was determined by DNA gel blot analysis using the variegated leaves derived from green sections of variegated leaves, there were more of the rbcL, psbA, 16S rDNA and 23S rDNA chloroplast genes in the white sections than the chloroplast genes in wild type and green sections of the variegated plants.

Vitamin content in Rosa davurica Pall. (생열귀나무의 채취부위 및 시기별 비타민 함량)

  • Vitamin contents in different plant parts and harvesting time of Rosa davurica Pall. were determined to examine their applicability for a new medicinal supply. Among plant parts analysed, leaf contained the highest ascorbic acid concent of 1973.9 mg/100g. Fruit contained ascorbic acid (714.6mg/100g) 2. 7 times less than leaf. Vitamin contents in leaf parts also differed depending on harvesting time. Vitamin content in leaves harvested in November was much higher than that in September. Spectrophotometric analysis of total ${\beta}-carotene$ in leaf showed higher contents than that in fruit.

Physicochemical Properties and Antioxidant Activities of Different Parts of Kkujippong (Cudrania tricuspidata Bureau) from Miryang (밀양산 꾸지뽕 나무(Cudrania tricuspidata Bureau)의 부위별 이화학적 특성 및 항산화 활성)

  • Various parts of Kkujippong (Cudrania tricuspidata Bureau) tree (leaves, stems, roots, and fruit) were evaluated to determine their properties. The general components of Kkujippong tree were fat 1.3-5.9 g /100 g, protein 6.3-18.7 g /100 g, and ash 2.8-9.0 g /100 g. The fiber content of the stem was the highest amount at 69.2 g/100 g, followed by protein in the roots (49.9), leaves (45.5), and fruit (15.6). In addition, Kkujippong tree was found to contain high concentrations of several minerals (Ca, Fe, Mg, Na, P, Zn) in the leaves. The total polyphenol content was significantly higher in the leaves than in other areas at 571.3 mg% (p<0.05). Electron donating ability was also highest in the leaves at 83.55% (p<0.05).

Optimization of polyphenol extraction from non-edible parts of Aronia melanocarpa for the development of functional bioconvergence materials (아로니아 비가식 부위로부터 기능성 바이오융복합 소재 개발을 위한 폴리페놀의 추출 공정 최적화)

  • Polyphenols extraction conditions including extraction solvent, temperature, pH and time were optimized for the development of functional bio-convergence materials using non-edible parts of Aronia melanocarpa including its berry. Water, ethanol, and methanol were used for the extraction of polyphenol from aronia leaves, stem and twigs. Water was selected as an extraction solvent because water gave the highest extraction yields. Among the non-edible parts, aronia leaves had the highest total polyphenol content. The polyphenol extraction conditions from aronia leaves were statistically optimized using a experimental design method: reaction time of 4.5 h, extraction temperature of 79.3℃, and pH 7.2. These optimized extraction conditions could be used for the production of functional bio-material.

Composition Analysis and Antioxidative Activity from Different Organs of Cirsium setidens Nakai (부위별 고려엉겅퀴(Cirsium setidens Nakai)의 이화학적 성상 및 항산화 활성 효과)

  • In the present study, we investigated the compositions including protein, lipid, ash, carbohydrate, and minerals as well as antioxidant activity of Cirsium setidens Nakai in order to detect the biological activities and develop novel functional resources. Different organs commonly had the highest concentration of potassium among 7 minerals evaluated in this study. The leaf had K at the concentration of 5371.97 mg/100 g, while the flower, and stem, the root and the boiled leaf at the concentration of 1770.62 mg/100 g, 1983 mg/100 g, 6096.74 mg/100 g and 1604.2 mg/100 g, respectively. The monosaccharides were composed of the xylose, galactose and glucose. The xylose was only detected in the flower and stem and the galactose was only detected in the stem. DPPH scavenging activity was measured at the $88.22\;{\mu}g/mL$ and $111.19\;{\mu}g/mL$ in root and leaf at $IC_{50}$ value in ethanol extracts, while $53.27\;{\mu}g/mL$, $75.84\;{\mu}g/mL$ and $257.48\;{\mu}g/mL$ in flower, boiled leaf and stem at $IC_{50}$ value, respectively, in water extracts. These results suggest that extracts from Cirsium setidens Nakai can be potentially used as novel resources for antioxidant and biological active substances.

Bioefficacy Evaluation of Non-edible Parts of Aronia melanocarpa for the Use of Functional Biomaterials (기능성 바이오소재 활용을 위한 아로니아 비가식 부위 추출물의 생리활성 평가)

  • Total polyphenol content and biological activities of water and methanol extract from Aronia melanocarpa berry, leaf, stem and twig were analyzed to investigate the potential for a bio-functional material. The polyphenol content of leaf and twig extracts were higher than those of berry and stem extracts. DPPH radical and ABTS radical scavenging activities of both extracts, were measured in order of twig, leaf, berry and stem. Tyrosinase inhibitory activities of water extract were measured in order of berry, leaf, twig and stem. Those of methanol extract were measured in order of twig, leaf, berry and stem. The nitrite-scavenging ability of water and methanol extract were measured in order of berry, leaf, twig and stem. Consequently it was shown that the non-edible parts of aronia, leaf and twig, had antioxidant activities and nitric oxide scavenging activities. It is expected that these materials could be used as functional bio-materials in bio-health care products including cosmetic products.

Physiological Activities of Ethanol Extracts from Different Parts of Allium hookeri (삼채부위별 에탄올 추출물의 생리활성탐색)

  • Biological compounds such as crude saponin and total polyphenol were determined in Allium hookeri roots and leaves. In addition, activities of DPPH radical scavenging activity, hydroxyl radical scavenging activity, ${\alpha}$-amylase inhibition activity and nitrite scavenging were detected in concentrates of 70% ethanol extracts of Allium hookeri roots and leaves. The crude saponin content of the roots and leaves were 4.28 mg/g, and 4.17 mg/g, respectively. Total polyphenol and total flavonoid content was higher in leaves than roots. Furthermore the DPPH radical scavenging activity and hydroxyl radical scavenging activities of leaves was higher than roots. For ${\alpha}$-glucosidase inhibitory activity, 70% ethanol extracts of Allium hookeri roots and leaves showed 60% activity inhibition at a 10 mg/mL concentration. Nitrite scavenging abilities under acidic conditions were most effective for Allium hookeri leaves extracts. These results suggested that Allium hookeri leaves might be used as the components of health functional food.

Anatomical Observation of Leaf of Gerbera hybrida Hort. Injured by Liriomyza trifolii (아메리카잎굴파리에 의한 거베라 피해잎의 조직학적 관찰)

  • This study was conducted to obtain a basic information on the structural and histological characteristics of Gerbera hybrida Hort. leaf injured by american serpentine leafminer (ASL), Liriomyza trifolii, by using light and scanning electron microscope (SEM). Based on the anatomical observation of leaf blade injured by L. trifolii, the injury process could be divided into three stages. In the initial stage, the punctured tiny holes where ASL layed eggs after suction in the upperside of leaf were observed in the palisade parenchyma. In the middle stage, the hatched larvae made mines in the palisade parenchyma only. In the final stage, the mature larvae grew up making the mines bigger, and just before going out from the epidermis, it injured the inside of leaf containing one layer palisade parenchyma and two layers of spongy parenchyma.

Quality Characteristics of Kimchi Prepared with Different Part of Chinese Cabbage and Its Quality Change by Freeze-drying (배추의 부위에 따른 김치의 품질특성과 동결건조에 의한 품질변화)

  • Acid production and growth of lactic acid bacteria, sensory properties, volatile odor components, and effects of freeze-drying on quality of kimchi prepared with whole, midlib, and leaves of Chinese cabbages ripened at $20^{\circ}C$ for 3 days were investigated. Salinities of whole, midlib, and leaf were 2.31, 2.03, and 2.68%, respectively, In kimchi pH of midlib was the lowest and that of leaf was the highest, while acidities of whole and midlib were higher than that of leaf. Numbers of lactic acid bacteria in whole and midlib were slightly higher than that of leaf. Overall acceptability, taste, and odor of kimchi and freeze-dlied/rehydrated kimchi prepared with whole or midlib were better than those of leaf. Volatile odor components such as ethanol and sulfur-containing components of kimchi prepared with whole or midlib were generally higher than those of kimchi prepared with leaf.
